Study of Growth and Invasiveness Inhibition by Rhodiola in Human Breast Cancer Cells

Abstract:OBJECTIVE: To study the effect of Rhodiola on proliferation and invasiveness of human breast cancer cells in vivo and vitro. METHODS: The growth inhibition of Rhodiola extract on breast cancer cells MDA-MB-435 was analyzed by WST (water-soluble tetrazolium salt) methods. Flow cytometry was used to examine the inhibition of drug on cell cycle stages. In human breast cancer MDA-MB-435 xenografts, the effects of Rhodiola extarct on the expression of proteolytic enzymes MMP-2 and cathepsin D(Cath D)were evaluated by the immunohistochemistry staining.RESULTS: Rhodiola extract treatment induced S-stage arrest in MDA-MB-435 cells with concentration dependent. The drug clearly inhibited the cell proliferation with both dose and time dependent. Comparred with the control group, the expression of Cath D was decreased in xenografts of Rhodiola-treated group. The average H-score of the immunohistochemistry staining of the treated group was 72±50.70, and the control group was 168±64.19, P=0.03; the expression of MMP-2 was also decreased in xenografts of Rhodiola-treated group with the average staining H-score 90±29.15 vs 176±65.80 in the control group, P=0.04. CONCLUSION: Rhodiola can inhibit human breast cancer cells through multi-path by cytostatic and down-regulating expressions of proteolytic enzymes.
