Cloning and Functional Characterization of An AcERF1 Gene from Atriplex canescens

Abstract:The ethylene responsive factors (ERF) belong to a subfamily of the AP2/ERF superfamily in plants. The ERF subfamily were shown to specifically bind the GCC-box presented in many promoters of pathogenesis related protein (PR) genes, and implicated in various responses to environmental stimuli. In this study, a new member (AcERF1) of ERF subfamily was isolated from an Atriplex canescens cDNA library. The AcERF1 contains an open reading frame of 1161 bp which encodes a protein of 387 amino acid residues with a conserved N-terminal MCGGAI (I/L) -like motif. To investigate its function associated with abiotic and biotic stress, AcERF1 was transformed into Saccharomyces cerevisiae INVSc1 and treated with saline, alkali, drought, high temperature, freezing, as well as paraquat, heterologous expression of AcERF1 in transgenetic yeast showed a relatively higher stress resistance, particularly in high temperature, alkali and paraquat conditions when compared with control. These results indicated that AcERF1 may involve in response to environmental stress and exhibit multiple, complex and flexible expression patterns in A. canescens.
