Abstract:Angiotensin-converting enzyme (ACE) is best known for the catalytic conversion of angiotensin I to angiotensin II. However, the use of gene-targeting techniques has led to mouse models highlighting many other biochemical properties and actions of this enzyme. This review discusses recent studies examining the functional significance of ACE tissue-specific expression and the presence in ACE of two independent catalytic sites with distinct substrates and biological effects. It is these features which explain why ACE makes important contributions to many different physiological processes including renal development, blood pressure control, inflammation, and immunity.

Abstract:Angiotensin-converting enzyme (ACE), a zinc metalloprotein, is a central component of the renin–angiotensin system (RAS). It degrades bradykinin and other vasoactive peptides. Angiotensin-converting-enzyme inhibitors (ACE inhibitors, ACEIs) decrease the formation of angiotensin II and increase the level of bradykinin, thus relaxing blood vessels as well as reducing blood volume, lowering blood pressure and reducing oxygen consumption by the heart, which can be used to prevent and treat cardiovascular diseases and kidney diseases. Nevertheless, ACEIs are associated with a range of adverse effects such as renal insufficiency, which limits their use. In recent years, researchers have attempted to reduce the adverse effects of ACEIs by improving the selectivity of ACEIs for structural domains based on conformational relationships, and have developed a series of novel ACEIs. In this review, we have summarized the research advances of ACE inhibitors, focusing on the development sources, design strategies and analysis of structure-activity relationships and the biological activities of ACE inhibitors.

Abstract:Angiotensinogen (AGT) is the unique precursor of all angiotensin peptides. Many of the basic understandings of AGT in cardiovascular diseases have come from research efforts to define its effects on blood pressure regulation. The development of novel techniques targeting AGT manipulation such as genetic animal models, adeno-associated viral approaches, and antisense oligonucleotides made it possible to deeply investigate the relationship between AGT and cardiovascular diseases. In this brief review, we provide contemporary insights into the emerging role of AGT in cardiovascular diseases. In light of the recent progress, we emphasize some newly recognized features and mechanisms of AGT in heart failure, hypertension, atherosclerosis, and cardiovascular risk factors.

Abstract:OBJECTIVE:To investigate the expression of angiotensin converting enzyme 2 (ACE2) and the changes treated with angiotensin converting enzyme inhibitor (ACEI), and its signal transduction pathway.METHODS:Atrial tissues were obtained from 47 patients with RHD undergoing cardiac surgery. The mRNA of ACE2 and ACE were semi-qualified by RT-PCR and normalized to the gene beta-actin. Western blot analysis was employed to examine the expressions of ACE2, ACE, ERK1/2 and phosphorylated ERK (pERK1/2). The atrial tissue angiotensin II (Ang II) content was determined by radioimmunoassay detection.RESULTS:The expression of ACE2 was significantly decreased (P < 0.05), the expression of ACE and pERK1/2 were significantly increased (P < 0.05), and the level of atrial tissue Ang II was significantly increased in patients with chronic atrial fibrillation group (CAF) compared with sinus rhythm group (SR) (P < 0.05). Compared with CAF patients treated without ACEI, the expression of ACE2 significantly increased (P < 0.01), and the relative activity of ERK1/2 significantly decreased (P < 0.05), whereas the expression of ACE and the level of atrial tissue Ang II remained unchanged in CAF patients treated with ACEI.CONCLUSIONS:The study suggested that the dysequilibrium of ACE/ACE2 might play an important role in the process of atrial fibrillation, which may be related to the activation of ERK1/2 pathway. The clinical effect of long-term treatment of ACEI maybe associated with elevated ACE2 expression but not ACE expression.

Abstract:OBJECTIVE To introduce the effects of epoxide hydrolase and its inhibitions for the antihypertensive therapy.METHODS Consulting relative references,something about the role of epoxide hydrolase in hypertension control and its mechanisms was summarized. RESULTS Soluble epoxide hydrolase(sEH) is widely distributed in mammalian tissues.It is the primary enzyme that is responsible for the conversion of epoxyeicosatrienoic acids(EETs) which are endogenous antihypertensive eicosanoids.Disruption of the sEH gene or inhibition of sEH decreases blood pressure.CONCLUSION Potent selective sEH inhibitors have potentiality to become a novel class of antihypertensive agent.

Abstract:Angiotensin-converting enzyme inhibitors (ACEIs) can improve the fibrotic processes in many internal organs. Recent studies have shown a relationship between ACEI with cutaneous scar formation, although it has not been confirmed, and the underlying mechanism is unclear. In this study, we cultured mouse NIH 3T3 fibroblasts with different concentrations of ACEI. We measured cell proliferation with a Cell Counting Kit-8 and collagen expression with a Sirius Red Collagen Detection Kit. Flow cytometry and western blotting were used to detect transforming growth factor β1 (TGF-β1) signaling. We also confirmed the potential antifibrotic activity of ACEI in a rat scar model. ACEI reduced fibroblast proliferation, suppressed collagen and TGF-β1 expression, and downregulated the phosphorylation of SMAD2/3 and TAK1, both in vitro and in vivo. A microscopic examination showed that rat scars treated with ramipril or losartan were not only narrower than in the controls, but also displayed enhanced re-epithelialization and neovascularization, and the formation of organized granulation tissue. These data indicate that ACEI inhibits scar formation by suppressing both TGF-β1/SMAD2/3 and TGF-β1/TAK1 pathways, and may have clinical utility in the future.

Abstract:OBJECTIVE: To analyze the adverse reactions induced by angiotensin converting enzyme inhibitor (ACEI) so as to deepen our understanding on the ACEI-induced ADR and maximize its therapeutic effect. METHODS: ACEIinduced ADR cases reported in domestic medical or pharmaceutical journals (not including the Review) were collected and analyzed. RESULTS: ACEI-induced ADR manifested itself as irritable cough,parageusia,gastrointestinal tract discomfort, skin rash,itch of skin,headache,dizziness,hematuria and angioneurotic edema etc. Of all the ACIE,Captopril took the lead in terms of ADR incidence. CONCLUSION: In most cases,ACEI-induced ADR disappeared after discontinuation or symptomatic treatment. Some patients presented with the same symptoms when taking ACEI again,suggesting the importance of patients’history of allergies when taking ACEIs to prevent misdiagnosis.

Abstract:The angiotensin-converting enzyme (ACE) was discovered in 1956 and has been actively studied to date. It has a unique structure of two homologous domains, each containing a catalytic zinc ion. Domains have different substrate specificity. In terms of function, ACE is a zinc metallopeptidase widely present on the surface of endothelial and epithelial cells. The gene encoding ACE is located on the long arm of chromosome 17 (17q23) and is 21 kb long, including 26 exons and 25 introns. The structure of ACE may be the result of an ancient gene duplication that occurred approximately 700 million years ago. The main function of ACE is the conversion of AngI to the vasoconstrictor AngII, which is the main active product. In addition, ACE metabolizes bradykinin, which is a potent vasodilator. ACE is involved in the metabolism of other angiotensins, in particular Ang(1–7), forming, together with ACE 2 and other components of the renin-angiotensin-aldosterone system (RAAS), a complex balanced system for maintaining blood pressure, water and electrolyte balance, and many other components of systemic, tissue and cellular homeostasis that have not yet been fully studied. More data are accumulating confirming the role of ACE for the renal development, early hematopoiesis, normal male fertility, erythropoiesis, myelopoiesis. ACE plays important roles in the immune response, intracellular signaling.

Abstract:The renin-angiotensin system (RAS) plays a central role in the maintainance of homeostasis of blood pressure and electrolyte balance. Recently,the RAS has been implicated in the pathogenesis on the pathogenesis of respiratory disease,especially in pulmonary fibrosis,pulmonary artery hypertension.Angiotensin-converting enzyme 2 was cloned as a first homolog of human ACE and functioned as an negative regulator of RAS.Recent studies has confirmed the protective effect of ACE2 against acute lung injury.In this review we shall discuss the roles for ACE2 in disease pathogenesis and the possibility of ACE2 as a novel approach for the treatment of lung diseases.

Abstract:Angiotensin receptor neprilysin inhibitor (ARNI) decreases renin-angiotensin-aldosterone system (RAAS) and sympathetic nervous systems (SNS) activity promoting vasodilation, decreasing myocardial hypertrophy and fibrosis. Beyond the SNS, RAAS and natriuretic peptide systems, ARNI results in increased circulatory and myocardial nitric oxide levels activating cGMP and protein kinase G, which reduces oxidative stress, myocyte hypertrophy, cell death and has anti-thrombotic effects. ARNIs have a class I indication by heart failure (HF) guidelines in HFrEF patients with NYHA class II to III symptoms. Beyond HFrEF, the use of ARNIs has also been expanded to other clinical settings including HF with preserved ejection fraction (EF, HFpEF), acute HF, advanced HF, hypertension, arrhythmias and chronic kidney disease. This paper reviews the clinical benefits of ARNIs in both HF and the aforementioned cardiovascular conditions. We also discuss the combined use of ARNI with SGLT2i and their potential synergistic benefits on cardiovascular outcomes.

Abstract:Angiotensin-converting enzyme 1 (ACE1) is a peptide involved in fluid and blood pressure management. It regulates blood pressure by converting angiotensin I to angiotensin II, which has vasoconstrictive effects. Previous studies have shown that certain compounds of natural origin can inhibit the activity of angiotensin-converting enzymes and exert blood pressure-regulating effects. Surface Plasmon Resonance (SPR) biosensor technology is the industry standard method for observing biomolecule interactions. In our study, we used molecular simulation methods to investigate the docking energies of various herbal metabolites with ACE1 proteins, tested the real-time binding affinities between various herbal metabolites and sACE1 by SPR, and analyzed the relationship between real-time binding affinity and docking energy. In addition, to further explore the connection between inhibitor activity and real-time binding affinity, several herbal metabolites' in vitro inhibitory activities were tested using an ACE1 activity test kit. The molecular docking simulation technique's results and the real-time affinity tested by the SPR technique were found to be negatively correlated, and the virtual docking technique still has some drawbacks as a tool for forecasting proteins' affinities to the metabolites of Chinese herbal metabolites. There may be a positive correlation between the enzyme inhibitory activity and the real-time affinity detected by the SPR technique, and the results from the SPR technique may provide convincing evidence to prove the interaction between herbal metabolites and ACE1 target proteins.

Abstract:Angiotensin (Ang)-(1-7) is recognized as a new bioactive peptide in renin-angiotensin system (RAS). Ang-(1-7) is a counter-regulatory mediator of Ang-II which appears to be protective against cardiovascular disease. Recent studies have found that Ang-(1-7) played an important role in reducing smooth muscle cell proliferation and migration, improving endothelial function and regulating lipid metabolism, leading to inhibition of atherosclerotic lesions and increase of plaque stability. Although clinical application of Ang-(1-7) is restricted due to its pharmacokinetic properties, identification of stabilized compounds, including more stable analogues and specific delivery compounds, has enabled clinical application of Ang-(1-7). In this review, we discussed recent findings concerning the biological role of Ang-(1-7) and related mechanism during atherosclerosis development. In addition, we highlighted the perspective to develop therapeutic strategies using Ang-(1-7) to treat atherosclerosis.

Abstract:The angiotensin-converting enzyme inhibitors (ACEi) is a widely prescribed anti-hypertensive drugs targeting the renin-angiotensin system (RAS). These drugs still have limitations due to their side effects. This study aims to identify the potential of bioactive compounds from Andaliman fruit (Zanthoxylum acanthopodium DC.) as natural ACEi for the treatment of hypertension with an in-silico approach. Phytoconstituents from Z. acanthopodium fruits were screened using the SwissADME web server for their bioavailability. The selected compounds were then paired to human ACE (PDB ID: 1O86) by site-specific docking to the ACE binding pocket. The stability of the interaction between ligand and protein was evaluated using molecular dynamics simulations in YASARA software. From a total of 39 compounds, we predict that 5 compounds namely kaempferol, quercetin, abscisic acid, benzophenone, and cis-3,5,3,4-tetrahydroxystilbene can bind to ACE with good affinity and form interactions with key residues. The presence of these compounds may reduce the binding affinity of angiotensin I to ACE and alter the interaction site. Molecular dynamics simulations show the compounds have good interaction stability with ACE and remain in the active site until the end of the simulation. This study showed that five bioactive compounds from Andaliman fruit act potentially as natural ACE-inhibiting agents.
