Wnt/ Beta -Catenin Signaling in the Pathogenesis of Osteoarthritis

Abstract:Objective: To investigate the Wnt/β-catenin signaling pathway related proteins and gene expression in the articular cartilage of osteoarthritis (OA) samples from patients undergoing total knee arthroplasty. Methods: The samples of articular cartilage were divided into three groups by using the modified Mankin score including control group, mild OA and moderate OA group. The levels of β-catenin and MMP-13 were detected by immunohistochemistry staining. Total RNA was extracted from articular cartilage tissues. Real Time reverse transcriptase polymerase chain reaction (Real Time RT-PCR) was performed to test the expression of bmp-2, col-10 and mmp-13, and also the expression of β2m was tested for reference. Results: There were 8 cases in control group, 15 cases in mild OA and 27 cases in moderate OA. The result of β-catenin immunohistochemistry showed negative in control group, 9 positive in mild OA group and 7 positive in moderate OA group. The result of MMP-13 immunohistochemistry showed one positive in control group, 14 positive in mild OA and 21 positive in moderate OA group. The results of Real Time RT-PCR showed that the expression levels of bmp-2, col-10 and mmp-13 were significantly increased in mild OA group (3.01-, 2.89-and 4.04-fold) than that of moderate OA group (P 0.01). Conclusion: Overexpression of β-catenin in articular chondrocytes leads to the articular cartilage damage and the development of OA.
