Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.

Abstract:7β-Amino-3-[(1-Methyl-1H-tetrazo5-yl)thiomethyl]-3-cephem-4-carboxylic acid(7-AMCA) was synthesized by using 7-Aminocephalosporanic acid(7-ACA) and 5-mercapto-1-methyl-1,2,3,4-tetrazole(5-MMT) as the materials.In this paper,influencing factors of nucleophilic substitution reaction such as the molar ratio,the amount of solvent,the amount of catalysis,the reaction time and temperature were studied.The better experimental conditions were the molar ratio(1∶1.20) of 7-ACA to 5-MMT,the reaction time 100 min and the temperature about 65 ℃ when the acetic acid used as the solvent,the methylsulfonic acid used as the catalyst.The yield of 7-AMCA was 89.87 % based on 7-ACA,the purity of HPLC was 99.21 %, and its molecular structure!was exactly identified by 1H-NMR,FT-IR and elementary analysis.

Abstract:An efficient and improved process for the preparation of diphenylmethyl 7 beta-phenylacetamido-3-hydroxymethyl-3-cephem-4-carboxylate was developed. With the commercially available 7-aminocephalosporanic acid (7-ACA) as starting material, up to 73.5% overall isolated yield of the tided compound was synthesized in two steps via direct phenylacetylation with phenylacetyl chloride, followed by basic hydrolysis and esterification with diphenyldiazomethane. The newly developed process obviated the use of protecting groups, reduced the environmental footprint, and could be easily controlled and conveniently scaled up for this pivotal intermediate in cephalosporin chemistry.

Abstract:7-amino-3-(1-methyl-1-pyrrolidino)methylceph-3-em-4-carboxylate hydrochloride is synthsized from 7-ACA by five steps:protected by silane,replaced by iodine,displacement,romove the protected group,form salt.The characteristics of the title compound is investigated with HNMR.The effect of some factors such as silane,solvent,and the ratio of raw material on the synthesis the title compound are studied.The result shows that the suitable volume ratio of methylene chloride and cyclopentane is 1∶1;the optimum ratio is 7-ACA∶chlorotrimethylsilane∶imidazole∶N-methyl pyrrole is 1∶2.8∶2.4∶1.5;the silane reactiong temperature is 45 ℃,the reaction time is 2.5 h;the reaction temperature of displacement is-10 ℃,the reaction time is 2 h;the overall yield can acheives 96%.

Abstract:S)-(+)-2,2-dimethylcyclopropane carboxylic acid is the key intermediate of Cilastatin (an excellent inhibitor of dehydropeptidase-I). Using 2-methylbutenoic acid as raw material, 2,2-dimethylcyclopropane carboxylic acid was achieved via esterification, cyclopropanantion and hydrolysis, the yield is 44.1%. Specially, using zinc powder/copper (I) chloride-acetyl chloride as catalysts, the cyclopropanation proceedes easily with dibromomethane. Then 2,2-dimethylcyclopropane carboxylic acid is converted to acid chloride and reacted with L-carnitine oxalate (the chiral reagent for resolution). After fractional crystallization and hydrolysis, (S)-(+)-2,2-dimethylcyclopropane carboxylic acid was prepared with 16.7% yield. Thus a new synthesis route of 2,2-dimethylcyclopropane carboxylic acid was designed and its chiral resolution procedure was improved, this new route and improved procedure are clean, convenient to operate and apt for industrialization with lower cost.
[Determination of Residual Solvents in 7-Amino-3-chloro Cephalosporanic Acid by Gas Chromatography].

Abstract:OBJECTIVE:To develop a gas chromatography method for determination of residual solvents in 7-amino-3-chloro cephalosporanic acid (7-ACCA).METHODS:The residual levels of acetone, methanol, dichloromethane, ethyl acetate, isobutanol, pyridine and toluene in 7-ACCA were measured by gas chromatography using Agilent INNOWAX capillary column (30 m × 0.32 mm,0.5 μm). The initial column temperature was 70° maintained for 6 min and then raised (10°C/min) to 160°C for 1 min. Nitrogen gas was used as carrier and FID as detector. The flow of carrier was 1.0 ml/min, the temperature of injection port and detector was 200°C and 250°C, respectively.RESULT:The limits of detection for acetone, methanol, dichloromethane, ethyl acetate, isobutanol, pyridine, toluene in 7-ACCA were 2.5 μg/ml, 1.5 μg/ml, 15 μg/ml, 2.5 μg/ml, 2.5 μg/ml, 2.5 μg/ml and 11 μg/ml, respectively. Only acetone was detected in the sample, and was less than the limits of Ch.P.CONCLUSION:The method can effectively detect the residual solvents in 7-ACCA.

Abstract:Here we describe an efficient and cost-effective chemoenzymatic synthesis of the beta-lactamase inhibitor avibactam starting from commercially available ethyl 5-hydroxypicolinate hydrochloride. Avibactam was synthesized in 10 steps with an overall yield of 23.9%. The synthetic route features a novel lipase-catalyzed resolution step during the preparation of (2S,5S)ethyl 5-hydroxypiperidine-2-carboxylate, a valuable precursor of the key intermediate ethyl (2S,SR)-5-((benzyloxy)amino)piperidine-2-carboxylate. Our synthetic route was used to produce 400 g of avibactam sodium salt.
Synthesis and characterization of cephalosporin nucleus 7-ACCA

Abstract:7-amino-3-chloro-cephems-3-en-4-acid (7-ACCA) is an important intermediate in preparation of cephalosporin. The process for preparing 7-ACCA using 3-chloro cephalosporin as a raw material via two reactions of hydrolyzation and enzyme cleavage was described. The total yield was up to 85.0%. The product was assayed by HPLC, and the content was up to 99%, greater than that of the standard 7-ACCA. The product was also characterized by IR, element analysis, and NMR, and the results of the sample 7-ACCA was consistent with that of the standard 7-ACCA. It is found that the optimum temperature of hydrolyzation reaction is 60~65°C, the optimum temperature of enzyme cleavage reaction is 30~32°C and the optimum pH is 9.0. Under the best reactive conditions, a batch experiment was carried out for 400~500 times, and the yield did not reduce evidently. The preparation procedure is efficient, the yield is high, the reagents used are cheap, and convenient to get with low pollution, and it is very easy to be industrialized.

Abstract:3-Decarbamoyl-cefuroxime（DCC）, the key intermediate for the synthesis of cefuroxime, is obtained by condensing the 3-deacetyl-7-aminocephalosporanic acid（D-7-ACA） solution with the acid chloride. The dissolution of D-7-ACA in water and methanol using high concentration NaOH solution is the first step. However,the β-lactam ring in D-7-ACA is prone to ring-opening reaction under strong alkaline condition, resulting in low yield. The D-7-ACA dissolution has low concentration and low efficiency in a traditional tank, due to the imprecise pH control and poor micro-mixing. Therefore, a new membrane dispersion microreactor was designed and used to dissolute D-7-ACA. Firstly, the degradation kinetics of D-7-ACA at different concentration, temperature and pH value were investigated. Then the effects of the pH, temperature, and circulation flow rate on the dissolution concentration were investigated carefully. The results showed that the concentration of the D-7-ACA solution is about 76.50 mg/ml, and the target product DCC yield is about 91.90% under the optimal conditions using membrane dispersion microreactor, which were increased by 5.91% and 5.61%, respectively, compared to using traditional reactor. The microreactor provides a good choice for industrially dissolving pH-sensitive drugs.

Abstract:Cefcapene pivoxil monohydrochloride monohydrate, a broad spectrum third-generation cephalosporin for oral administration, was prepared by reacting three centers of 7 beta-amino-3-(hydroxymethyl)cephalosporinic acid (7-HACA), derived from 7 beta-aminocephalosporanic acid. The coupling of 7-HACA and (Z)-2-[2-(Boc-amino)thiazol-4-yl]pent-2-enoic acid, followed by carbamylation with chlorosulfonyl isocyanate, and precipitation with diiso-propylamine gave the key intermediate, in which thiazole side chain and carbamoyl group had been introduced into the 7-amino and 3-hydroxymethyl groups of 7-HACA, respectively, in a continuous procedure. Afterwards, the intermediate was esterified with pivaloyloxymethyl iodide to complete the modification of the C4 carboxy group affording Boc-cefcapene pivoxil, from which the desired product was finally obtained by an economical Boc removal and successive formation of the hydrochloride in 36% overall yield on a decagram scale. This synthesis promises an easy entry to cefcapene pivoxil monohydrochloride monohydrate with convenient manipulation, simple isolation, and good yields; it is of potential value for production on an industrial scale.

Abstract:Cefpodoxime proxetil, a third-generation cephalosporin for oral administration, was synthesized by a method based on the following sequence of reactions: acylation of 7-aminocephalosporanic acid (7-ACA) with S-benzothiazol-2-yl(2-amino-4-thiazolyl)(methoxyimino)thioacetate (MAEM), chloroacetylation of the cefotaxime formed with chloroacetyl chloride, esterification of the acid function with 1-iodoethyl isopropyl carbonate and final cleavage of chloroacetamide protective group by treatment with thiourea in N,N-dimethylacetamide. The developed procedure allows us to obtain better yields of cefpodoxime proxetil and to eliminate the final purification step by column chromatography, necessary during the synthesis of this antibiotic by the previously reported methods.

Possibilities to acylate 7-aminocephalosporanic acid to obtain some cephalosporins

Abstract:Aim: Acylation of 7-aminocephalosporanic acid with an adequate acyl chloride in order to obtain cefotaxime sodium salt. Material and methods: Cefotaxime sodium salt was synthesized by acylating 7-amino cephalosporanic acid with 2-[2'-chloracetamidothiazole-4-yl]-2-(syn)-methoxy-imino acetic chloride in four steps. The melting point was determined, and IR spectral analysis and elemental analysis were performed to confirm cefotaxime structure. The quantitative determination was performed. Results: The reaction conditions were established. The yield of the synthesis phases (73-80%) and actual yield (45-47%) were very good. The structure of the obtained cefotaxime sodium salt was confirmed by the IR spectral analysis and by elemental analysis (C, H, N). The melting point was 163 degrees C. The purity of the synthesized cefotaxime sodium salt was 98.9%. Conclusions: Cefotaxime sodium salt was synthesized by acylation of 7-aminocephalosporanic acid with 2-[2'-chloracetamidothiazole-4-yl]-2-(syn)-methoxy-imino acetic chloride, in aqueous solution, then transformed into sodium salt with sodium 2-ethylhexanoate. The method proved to be very good, yields were good, it is reproducible and simple, and does not involve high risks, so it is also safe.

Abstract:The invention discloses a synthesis method of a cefpodoxime proxetil intermediate, namely (6R,7R)-7-[2-(2-amino-4-thiazolyl)-(Z)-2-(methoxyimino)acetamido]-3-methoxymethyl-8-oxo-5-thio-1-azabicyclo[4.2.0]oct-2-ene-2-methanoic acid. The synthesis method includes: enabling chlorosulfonic acid and methanol to react to prepare methoxy sulfonic acid; under the action of the methoxy sulfonic acid and dimethylformamide, etherifying 7-ACA (7-aminocephalosporanic acid) and trimethyl borate prior to aftertreatment, adding into a water and methanol solution reversely to guarantee that the obtained intermediate isn't sticky and is loose, drying and grafting with AE active ester so as to obtain a target product, namely the cefpodoxime proxetil intermediate. The synthesis method of the cefpodoxime proxetil intermediate has the advantages that synthesis steps of 3-position and 7-position protection and desorption of 7-ACA can be omitted, so that low step cost, high yield and high purity are achieved, all materials are cheap and available, and industrial production and little pollution are benefited.

Abstract:OBJECTIVE:To study the production process of ceftizoxime sodium.METHODS:Ceftizoxime acid was prepared with 7-Amino-3-norcephem-4-carboxylic acid(7-ANCA)and AE-ester ceftizoxime acid as raw material;then ceftizoxime sodium was obtained through the reaction between ceftizoxime acid and sodium carbonate.The effects of parameters including reaction time,and amount and dripping velocity of ethanol,the amount of sodium carbonate,and pH of solution on quality of product were studied.RESULTS:The reaction process could be realized successfully.The optimal parameters for the synthesis of ceftizoxime sodium were as follows:reaction time=1.5h,the velocity of dripping ethanol=400mL· h-1,reaction temperature 30℃,the ratio of ethanol:ceftizoxime acid=30mL∶ 1g,the ratio of sodium carbonate:ceftizoxime acid=0.15∶ 1.00,and pH=6.9± 0.1.CONCLUSION:This process is practicable,and which has provided a sound basis for the industrialized production of ceftizoxime sodium.

Abstract:Cephalosporin enzymatic hydrolysis technology is a green technology for recovering 7-amino-3-deacetoxycephalosporanic acid (7-ADCA) from cephalosporin mother liquor. Solubility is critical for the production and purification of 7-ADCA. In this paper, the solubility of 7-ADCA and phenylglycine was measured. Solubility-temperature correlation model and solubility-pH correlation model were investigated, and Akaike information criterion (AIC) analysis was performed. The kinetic parameters of the enzymatic hydrolysis reaction of cephalexin, cefradine, and cefadroxil were determined, and the reaction rates under different substrate concentrations were measured, and the Lineweaver–Burk double-reciprocal equation was used to draw a graph. The Michaelis constants Km/(mg/mL) were 73.98, 583.84, 38.66, Vmax/(mg/mL·min) 4.20, 16.00, 1.96, respectively. The experimental results show that amphoteric compounds and buffers can prompt the reaction, low concentration of methanol promotes the reaction, while high concentration of methanol inhibits the reaction, and ethanol, isopropanol, tetrahydrofuran (THF), dimethyl sulfoxide (DMSO), ethylene glycol (EG), 1,4-dioxane all have different degrees of inhibition on the reaction speed. Finally, based on thermodynamic and kinetic studies, a process technology for the preparation of 7-ADCA by hydrolysis catalyzed of cephalexin was developed. It was confirmed that the proposed process route of preferential removal of phenylglycine by elution and/or cooling crystallization was reasonable and effective. The 7-ADCA crystal products obtained by crystallization were characterized by PXRD, thermal analysis, infrared, electron microscope, and high-performance liquid chromatography (HPLC).
