Abstract:Virus-induced gene silencing offers a powerful reverse-genetic tool for the study of gene function in plants. We have previously reported effective gene silencing of plant genes using a viral satellite DNA associated with Tomato yellow leaf curl China virus (TYLCCNV). In this study, we further modified the viral satellite DNA-based vector. The modified vector can induce sulfu (Su) gene silencing as effective as the original vector in Nicotiana benthamiana plants, but the new system simplifies procedures for construction of vector derivative. Furthermore, a fragment of petunia Su or chalcone synthase (CHS) endogenous gene was inserted into the modified vector. When petunia plants were agroinoculated with the modified vector carrying a Su or CHS gene, the Su silenced plants started to appear yellowing in veins of systemically infected upper leaves two weeks after agroinoculation, while the CHS silenced plants started to show flower color change one month after agroinoculation and later single-color flowers became mosaic.

Abstract:The research progress about anthocyanin biosynthetic genes cloning,application and exogenous gene transformation of Petunia hybrida were reviewed in this paper.At least 12 enzymes were involved in the petunia anthocyanin biosynthesis.Except AAT and AMT,the most of synthetic enzyme genes,regulatory genes An2,An4 and enzyme activity regulation gene difF,have been cloned.A variety of exogenous color genes such as DFR,F3′5′H,CHS,CHI,CHR and Lc were introduced into petunia,the changes of flower color were observed.Some researchers also reported that the transformants with CHS were male sterility.The application prospects about petunia anthocyanin biosynthetic genes were also discussed.

Abstract:Chalcone synthase A is a key enzyme in the anthocyanin biosynthesis pathway. Expression ofchsA gene in transgenicPetunia hybrida resulted in flower color alterations and co-suppression of transgenes and endogenous genes. We fused the β-glucuronidase (uidA) gene to the C-terminal ofchsA gene, and transferred the fusion gene intoPetunia hybrida viaAgrobacterium tumefaciens. GUS histochemical staining analysis showed that co-suppression occurred specifically during the development of flowers and co-suppression required the mutual interaction of endogenous genes and transgenes. RNAin situ hybridization analysis suggested that co-suppression occurred in the entire plant, and RNA degradation occurred in the cytoplasm.

Abstract:We attempted to overexpress chalcone synthase (CHS) in pigmented petunia petals by introducing a chimeric petunia CHS gene. Unexpectedly, the introduced gene created a block in anthocyanin biosynthesis. Forty-two percent of plants with the introduced CHS gene produced totally white flowers and/or patterned flowers with white or pale nonclonal sectors on a wild-type pigmented background; none of hundreds of transgenic control plants exhibited such phenotypes. Progeny testing of one plant demonstrated that the novel color phenotype co-segregated with the introduced CHS gene; progeny without this gene were phenotypically wild type. The somatic and germinal stability of the novel color patterns was variable. RNase protection analysis of petal RNAs isolated from white flowers showed that, although the developmental timing of mRNA expression of the endogenous CHS gene was not altered, the level of the mRNA produced by this gene was reduced 50-fold from wild-type levels. Somatic reversion of plants with white flowers to phenotypically parental violet flowers was associated with a coordinate rise in the steady-state levels of the mRNAs produced by both the endogenous and the introduced CHS genes. Thus, in the altered white flowers, the expression of both genes was coordinately suppressed, indicating that expression of the introduced CHS gene was not alone sufficient for suppression of endogenous CHS transcript levels. The mechanism responsible for the reversible co-suppression of homologous genes in trans is unclear, but the erratic and reversible nature of this phenomenon suggests the possible involvement of methylation.
Effects of GhMADS3 Gene Constitutive Expression on Flower Shape and Color of Petunia

Abstract:An AGAMOUS homologue(GhMADS3)was introduced into petunia via Agrobacterium tumefaciens mediated method. Forteen independent kanamycin-resistant petunia plants were alive. GUS staining revealed that ectopic gene was expressed in transgenic plants. PCR detection showed that the GhMADS3 was integrated into their genomes. The expression of the GhMADS3 was detected by RT-PCR. Constitutive expression of GhMADS3 resulted in visible phenotypic alterations in floral organ. In transgenic petunia plants, GhMADS3 resulted in the sepals swelling, corolla smaller and corolla deep spliting. It was noteworthy that the color of transgenic floral organ became weaker or entirely pale. The measuration of anthocyanin and chlorophyll showed that the content in transgenic plants were greatly lower than wild-type. It also indicated that GhMADS3 in plants has some relationship with the formation of flower color.
Overexpression of a Freesia hybrida flavonoid 3-O-glycosyltransferase gene, Fh3GT1, enhances transcription of key anthocyanin genes and accumulation of anthocyanin and flavonol in transgenic petunia (Petunia hybrida)

Abstract:UDP glucose flavonoid 3-O-glycosyltransferase(3GT) is a key enzyme in biosynthesis of stable anthocyanins, which are generally required for flower coloration. Fh3GT1, a cDNA encoding the 3GT from Freesia hybrida, was overexpressed in Petunia hybrida to investigate the influence on flavonoid accumulation. Flowers of transgenic petunia showed color alteration from white to pink and were subjected to further analysis by HPLC-ESI-MS. The results indicated that overexpression of Fh3GT1 resulted in evident increase of cyanidin, peonidin, and kaempferol content in transgenic flowers, whereas the content of quercetin increased slightly. Expressions of endogenous flavonoid biosynthetic pathway genes in transgenic flowers were analyzed by real-time qPCR. The results showed that transcript levels of PhCHS, PhCHI, PhF3H, and Ph3GT were significantly upregulated. Meanwhile, overexpression of Fh3GT1 also led to enhanced expression of AN1 (bHLH) and MYBx (R3MYB) genes, as the mRNA levels of other transcription factors remained constant in control and overexpression lines. Thus, Fh3GT1 overexpression increased the accumulation of anthocyanins and flavonols by modulating the expressions of various flavonoid biosynthesis genes in flowers of transgenic petunia. These results also suggest that Fh3GT1 is a key gene involved in anthocyanin and flavonol biosynthesis and should serve as a useful candidate to modify the components of flavonoid in other plants.

Abstract:Dihydroflavonol 4-reductase (DFR) genes from Rosa chinensis (Asn type) and Calibrachoa hybrida (Asp type), driven by a CaMV 35S promoter, were integrated into the petunia (Petunia hybrida) cultivar 9702. Exogenous DFR gene expression characteristics were similar to flower-color changes, and effects on anthocyanin concentration were observed in both types of DFR gene transformants. Expression analysis showed that exogenous DFR genes were expressed in all of the tissues, but the expression levels were significantly different. However, both of them exhibited a high expression level in petals that were starting to open. The introgression of DFR genes may significantly change DFR enzyme activity. Anthocyanin ultra-performance liquid chromatography results showed that anthocyanin concentrations changed according to DFR enzyme activity. Therefore, the change in flower color was probably the result of a DFR enzyme change. Pelargonidin 3-O-glucoside was found in two different transgenic petunias, indicating that both CaDFR and RoDFR could catalyze dihydrokaempferol. Our results also suggest that transgenic petunias with DFR gene of Asp type could biosynthesize pelargonidin 3-O-glucoside.

Abstract:Flower breeders are continually refining their methods for producing high-quality flowers. Phalaenopsis species are considered the most important commercially grown orchids. Advances in genetic engineering technology have provided researchers with new tools that can be used along with traditional breeding methods to enhance floral traits and quality. However, the application of molecular techniques for the breeding of new Phalaenopsis species has been relatively rare. In this study, we constructed recombinant plasmids carrying flower color-related genes, Phalaenopsis Chalcone synthase (PhCHS5) and/or Flavonoid 3′,5′-hydroxylase (PhF3′5′H). These genes were transformed into both Petunia and Phalaenopsis plants using a gene gun or an Agrobacterium tumefaciens-based method. Compared with WT, 35S::PhCHS5 and 35S::PhF3′5′H both had deeper color and higher anthocyanin content in Petunia plants. Additionally, a phenotypic comparison with wild-type controls indicated the PhCHS5 or PhF3′5′H-transgenic Phalaenopsis produced more branches, petals, and labial petals. Moreover, PhCHS5 or PhF3′5′H-transgenic Phalaenopsis both showed deepened lip color, compared with the control. However, the intensity of the coloration of the Phalaenopsis lips decreased when protocorms were co-transformed with both PhCHS5 and PhF3′5′H. The results of this study confirm that PhCHS5 and PhF3′5′H affect flower color in Phalaenopsis and may be relevant for the breeding of new orchid varieties with desirable flowering traits.

Abstract:Recently, CRISPR/Cas9 technology has emerged as a powerful approach for targeted genome modification in eukaryotic organisms from yeast to human cell lines. Its successful application in several plant species promises enormous potential for basic and applied plant research. However, extensive studies are still needed to assess this system in other important plant species, to broaden its fields of application and to improve methods. Here we showed that the CRISPR/Cas9 system is efficient in petunia (Petunia hybrid), an important ornamental plant and a model for comparative research. When PDS was used as target gene, transgenic shoot lines with albino phenotype accounted for 55.6%–87.5% of the total regenerated T0 Basta-resistant lines. A homozygous deletion close to 1 kb in length can be readily generated and identified in the first generation. A sequential transformation strategy—introducing Cas9 and sgRNA expression cassettes sequentially into petunia—can be used to make targeted mutations with short indels or chromosomal fragment deletions. Our results present a new plant species amenable to CRIPR/Cas9 technology and provide an alternative procedure for its exploitation.
Silencing of Chalcone Synthase Genes by Artificial Microrna in Petunia

Abstract:Transgene-mediated gene silencing is a useful technique for elucidating biological function of genes and modifying crops by genetic engineering. In plants, amiRNAs make an effective tool for specific gene silencing. We designed the amiRchsl for silencing the petunia chalcone synthase genes according to the amiRNA design principles refined from the structure characters of plant microRNA. We used the precursor for Arabidopsis miR319a as backbone for the amiRchs 1 expression under the control of 35S promoter, exchanging the natural miR319a sequence with the sequence of amiRchsl by overlapping PCR. The transgenic petunia plants expressing the amiRchsl precursor produced flowers with varying amounts of diffused white dots or irregular white patches, and nearly white flowers. Both the contents of anthocyanin and CHS mRNA in corollas of the transgenic plants were reduced significantly. These results showed that the precursor for Arabidopsis miR319a can be used to express amiRNA in petunia, and structure genes can be knocked down effectively by the amiRNA technology.
Transgene‐mediated suppression of chalcone synthase expression in Petunia hybrida results from an increase in RNA turnover

Abstract:Summary Co-suppression of the pigmentation gene chalcone synthase (chs) in Petunia hybrida by chs transgenes leads to white or variegated flowers and is characterized by a reduction in steady-state mRNA levels. To determine the level at which suppression occurs different petunia transformants were analysed containing CaMV-35S RNA promoter-driven hybrid genes consisting of the β-glucuronidase gene (uidA) linked to the full-length chsA cDNA, the 5′ half or to the 3′ half. With these transgenes one out of 12–15 primary transformants showed suppression of the transgenes and of the resident chs genes throughout the flower or in sectors. The reduction in steady-state chs mRNA was not the result of a transcriptional inactivation event. As determined by nuclear run-on experiments, transcription of suppressed chs genes was similar to that of non-suppressed genes. This indicates that co-suppression occurs post-transcriptionally. Among individual transformants the transgenes were transcribed at different levels but neither a high nor a low level correlated with a particular degree or pattern of suppression. Surprisingly, even a promoterless chs transgene construct was found to suppress the endogenous chs genes in three out of 15 transformants. It remains, however, unknown whether or not transcription of the transgene locus is required to induce co-suppression. The data suggest that properties of the chs transgene locus other than the expression level are important for inducing co-suppression. The possible role of antisense RNA, which was detected in all transformants, ectopic pairing and the structure of the integrated T-DNAs in the mechanism of the selective increase in chs RNA turnover are discussed.

Transient Gene Expression in Phalaenopsis aphrodite Petals via Agrobacterium tume faciens Infiltration

Abstract:The study of gene function in Phalaenopsis aphrodite is hampered by the low efficiency of trans-formation systems and the long time span needed for the generation of transgenic plants.Therefore ,the es-tablishment of transient expression system of Phalaenopsis aphrodite petals will be an effective method for gene function analysis.An experiment was conducted to investigate the transient expression mediated by Agrobacterium tumefaciens using the sepals and petals of Phalaenopsis aphrodite (P.'Big chili'). Effects of A.tume faciens concentration ,infection time ,acetosyringone concentration ,and co-culture length on the transient expression of β-glucuronidase(GUS)gene were analyzed to determine the optimal transfor-mation condition.The chalcone synthetase gene RNAi interference vector was transiently transformed into the petals of Phalaenopsis aphrodite.After 3 days of co-cultivation ,changes in the phenotype and pig-mentation of the flower color were observed ,and RT-PCR was used to detect the expression level of CHS gene transcription.(1)A higher transient expression level of GUS gene could be obtained as OD600 value of A.tume faciens 0.6 ,infiltrating for 60 seconds ,adding 150 μmol/L acetosyringone to co-culture medium , and co-culture for 3 days in darkness.It reaches 85.01%.(2)The color of the petals of transgenic Phalae-nopsis aphrodite was significantly lighter and pigment content decreased.(3 ) Semi-quantitative PCR showed that the transcriptional activity of CHS gene was significantly lower than that of the control group.The experiment successfully established a rapid gene function verification method ,providing techni-cal support for the study of gene function and breeding work of the Phalaenopsis aphrodite.
Advances in Application of Transgenic Breeding Technology in the Traits Improvement of Chrysanthemum

Abstract:Following the discovery of chrysanthemum sensitivity to Agrobacterium in 1989,the technique of Agrobacterium-mediated genetic transformation has been successfully used in improvement of traits such as flower color,plant style,flowering time,disease-insect resistance and abiotic stress tolerance.In this review,the advances of Agrobacterium-mediated genetic transformation in chrysanthemum were summarized,including Agrobacterium strains,explants for infection,the addition of chemical compounds and selection markers.Also,the improvement of traits,exploration of important genes was reviewed and the perspective of enhancing transformation efficiency and developing environment-friendly transgenic technology were discussed.The main goal is to provide reference for molecular breeding of chrysanthemum.
Flower color modification through expression of Aquilegia buergeriana F3′5′H in Petunia hybrida

Abstract:Aquilegia buergeriana is a native plant in Korea with blue flowers. Flavonoid 3′,5′ hydroxylase ( F3′5′H ) is a key gene involved in the synthesis of delphinidin pigment responsible for the flower's blue color. We isolated the F3′5′H from the petals of A. buergeriana ( AbF3′5′H ) and introduced the AbF3′5′H gene into Petunia hybrida using Agrobacterium-mediated transformation. Forty-five plants were acquired from a kanamycin-supplemented medium. Fifteen of these were identified as transgenic plants using polymerase chain reaction (PCR). Quantitative real-time PCR (qRT-PCR) analysis revealed that the AbF3′5′H was expressed in the petal, corolla tube, and stigma of P. hybrida . AbF3′5′H -transgenic plant (T 0 ) flower color was darker than that of non-transgenic plants (NTs). Particularly, the stigma color was dramatically changed, from light yellow green (145C) to purple (N77C or N79D). The segregation ratio of the three transgenic (T 1 ) lines was identified as 3:1 by PCR analysis of AbF3 ′ 5′H and neomycin phosphotransferase-II. The flower color change of the transgenic lines (T 1 ) was similar to that of T 0 . qRT-PCR analysis showed that AbF3′5′H -transgenic T 1 lines had a higher AbF3′5′H expression than NT in all floral organs. Moreover, delphinidin was confirmed to be accumulated in both corolla tube and stigma and was enhanced in the petals of AbF3′5′H -transgenic T 1 lines through UPLC analysis. Our findings indicate the role of AbF3′5′H in flower color change. These results also indicate the functionality of AbF3′5′H in bluish flower modifications.

Abstract:Anthocyanins are important secondary metabolites in plants. After the formation of anthocyanidins, Flavonoid 3‐O‐glucosyltransferase (3GT) mediated glycosylation first occurs at the C‐3 site, forming a stable anthocyanin 3‐O‐glucoside. Several studies have investigated the function of 3GT using biochemical methods. However, it is necessary to provide further genetic evidence for the role of Ph3GT in petunia (Petunia hybrida). In addition, there is no information regarding the subcellular localization of Ph3GT and the regulation of transcription factors on Ph3GT. In this study, the full‐length Ph3GT gene from petunia (Petunia hybrida) was isolated. We found that Ph3GT is localized in the cytoplasm. Ph3GT exhibited high expression levels in the corollas during the coloring period of petunia flowers. VIGS‐mediated Ph3GT silencing resulted in a lighter corolla color and a significant decrease in the anthocyanin content in six petunia cultivars. The silencing of Ph3GT affected the expression levels of eight key genes in the anthocyanin synthesis pathway. Additionally, dual luciferase and yeast one‐hybrid assays showed that R2R3‐MYB transcription factor PhAN2 directly regulates the transcript of Ph3GT.

Improvement of the Hydroponic Growth and Waterlogging Tolerance of Petunias by the Introduction of Vhb Gene

Abstract:The coding sequence of Vitreoscilla hemoglobin (vhb) was cloned with PCR technique from Vitreoscilla stercoraria Pringsheim. The plant expression vector with vhb gene under the control of CaMV 35S promoter was constructed and used in the transformation of Petunia hybrida Vilm by the Agrobacterium mediated procedure. The results of PCR amplification and Southern hybridization indicated that the vhb gene had been integrated into the petunia genome and the vhb gene expression had been detected by RT-PCR amplification. In hydroponic culture the transgenic petunias grew much better than non-transgenic controls. For further analysis of hypoxia tolerance of transgenic petunia, the petunia plants with vhb gene were submerged into liquid MS medium. The transgenic plants survived in hypoxic condition and grew out of the liquid surface in a few weeks, while non-transgenic plants were still submerged and suffocated in culture solution without ability to grow out of liquid medium in submersed culture for four to five weeks. The vhb gene transformed petunia plants had been planted and tested in a simulated flooding condition, and showed obvious tolerance to waterlogging. It seems that hemoglobin gene from Vitreoscilla might have the potential use in molecular breeding for the improvement of plant resistance to hypoxia and flooding.

Abstract:The research was conducted using petals of ground-cover chrysanthemum(Rudbeckia hirta Linn.)cultivar 'Zaoxiaochun' as explants to establish a highly efficient regeneration system and its stable Agrobacterium-mediated transformation system.The result showed that the rate of callus formation was 95%,on the improved callus induction medium containing MS+2.0 mg· L-1 ZT+0.5 mg·L-1 2,4-D.The differentiation rate of adventi-tious buds on differentiation medium was 87%.The genetic transformation was performed.The results showed that dilutcd Agrobacterium with MS was the best medium,the suitable infection duration was around 8 min,and the appropriate co-culture duration was 3 d;the optimum concentrations of kanamycin and cephalosorine for transformed leaves of 'Zaoxiaochun' were 40 and 500 mg·L-1 respectively.
Genetic Transformation Efficiency Enhanced by Ipt Gene and Heat Shock Promoter Driven Gene Excision in Petunia

Abstract:In this research,we studied the influence of petunia shoot regeneration when transformed with ipt gene,and gene deletion effect of the heat shock inducible ＂gene deletion＂ construction in which flp gene was driven by the promoter hsp18.2.We transformed constructions of pBin-hsp18.2：flp-35S：ipt and pBin-hsp18.2：flp into petunia axilaris,to gain transgenic plant.We analyzed shoot regeneration between pBin-hsp18.2：flp-35S：ipt and pBin-hsp18.2：flp transformed plant,studied gene deletion efficiency with heat shock.The results showed that ipt gene can enhance adventitious shoots regeneration of petunia.The shoots inductivity of pBin-hsp18.2：flp-35S：ipt lines is 21.5% evidently higher than pBin-hsp18.2：flp lines which are 8.7%.Furthermore,the phenotype of transgenic plant became recovery by heat shock,which treatment conditions was heat shock under temperature of 44℃ for 6 h,and heat shock for 6 times.It proved the exogenous gene has been deleted in transgenic plants by GUS and PCR analysis.The highest gene deletion efficiency is 43.8% in this reasearch.This study provide evidence for application of ipt gene in transformation with plants that difficult to transform.
