Changes of blood flow and formation of blood vessels during wound healing after deep Ⅱ degree scald in rats

Abstract:Objective To explore the relationship between blood flow and formation of blood vessels during wound healing process after deep Ⅱ degree scald injury in rats. Methods 48 Wistar rats were randomly divided into pathological observing group (group A, 42 rats) and blood flow detecting group (group B, 6 rats). Group A was further divided into 6 subgroups according to the postburn period, which were immediately after scald, 1, 3, 7, 14 and 21 days after scald and normal control. Deep Ⅱ degree scald in rats was made on either side of back by using a metal scald machine. Rats in group A were sacrificed at different time points and specimens were collected for pathological and immunohistochemistry observation. Rats in group B were kept alive for detecting blood flow at the same position of the wound at different time points. Results Single endothelial cell was observed and increased to form the cavity from 3 to 7 days after injury, the endothelial cell numbers increased in accordance with the formation of granulation tissues. 14 days after injury, blood capillary with cavums in granulation tissues increased and even mature small vessels with intact structures were found. The shape and numbers of blood vessels were near to normal after 21 days. Blood flow decreased dramatically immediately after thermal injury, which was about 51% of the normal value. However, it reached to 68% after 3 days and 88% after 14 days of normal value. After 14 days, the value was almost back to normal. Conclusions During the wound healing process after deep Ⅱ degree scald injury in rats, blood flow increased gradually in accordance with the changes of endothelial cells in granulation tissues. The numbers and structure of endothelial cells affecting the changes of blood flow.
