The Role of CD4+CD25high Regulatory T Cells in the Pathogenesis of Ankylosing Spondylitis

Abstract:Objective To get a better understanding of the level of circulating CD4~+CD25~(high)regulatory T cells associated with ankylosing spondylitis(AS), in order to investigate the role of CD4~+CD25~(high)regulatory T cells in the pathogenesis of AS. Methods Peripheral blood CD4~+CD25~+ regulatory T cells and CD4~+CD25~(high) regulatory T cells in 50 AS patients and 20 healthy volunteers were analyzed by 2-color flow cytometry. Levels of IgA, IgG, IgM, ESR and CRP were also measured. Results In comparison to the control group, the blood CD4~+CD25~+ regulatory T cells in patients with AS did not show a significant increase(P0.05), but the level of CD4~+CD25~(high)regulatory T cells in AS patients was significantly lower than that of the control group(P0.05), CD4~+CD25~+ regulatory T cells and CD4~+CD25~(high)regulatory T cells from AS showed no correlation with IgG, IgM, ESR, CRP, course of disease and the grade of sacroiliac joint damage(P0.05), But the CD4~+ CD25~(high)regulatory T cells was associated negatively with IgA level(P0.05). Conclusion These findings demonstrate that, comparing to healthy control subjects, blood CD4~+CD25~(high)regulatory T cells tends to de- crease in AS patients and is also associated with IgA. As a result, CD4~+CD25~(high)regulatory T cells may play an important role in the pathogenesis of AS.
