Abstract:Two new spirostanoides, filiasparosides E (1) and F (2), one new furostanoside, filiasparoside G (3), and one new ecdysterone, stachysterone A-20, 22-acetonide (4), together with six known steroidal saponins, asparagusin A (5), filiasparoside A (6), filiasparoside B (7), aspafilioside A (8), aspafilioside B (9), and filiasparoside C (10) were isolated from the roots of Asparagus filicinus Buch.-Ham. Their structures were elucidated on the basis of spectroscopic and chemical evidence. Compounds 1–10 were investigated for their cytotoxicities against human breast adenocarcinoma MDA-MB-231 cell line and compounds 8–10 exhibited cytotoxic activities with IC50 values ranging from 3.4 to 6.6μM.

Abstract:Four new steroidal saponins, filiasparosides A-D (1-4), together with known aspafiliosides A (5) and B (6) were isolated from the roots of Asparagus filicinus. The structures of these new compounds were elucidated by detailed spectroscopic study and chemical analysis. Compounds 1-6 were cytotoxic against human lung carcinoma (A549) and breast adenocarcinoma (MCF-7) tumor cell lines with EC(50) values of 2.3-16.8 microg/mL. Compound 3 showed the most potent cytotoxicity, with EC(50) values of 2.3 and 3.0 microg/mL toward A549 and MCF-7 cell lines, respectively.

Abstract:Objective To study the steroidal saponins from Allii Macrostemonis Bulbus.Methods The silica gel and ODS column chromatographies were used for the isolation and purification of compounds from Allii Macrostemonis Bulbus.Their structures were identified through comparing with references and spectral analyses.Results Three steroidal saponins were isolated from the 70% ethanol extract from Allii Macrostemonis Bulbus and identified as 5β-spirostane-25(27)-en-3β,12β-diol-3-O-β-D-glucopyranosyl(1→2)-β-D-galactopyranoside(1),(25R)-5β-spirostane-3β,12β-diol-3-O-β-D-glucopyranosyl-(1→2)-β-D-galactopyranoside(2),and 5β-spirostane-25(27)-en-2β,3β-diol-3-O-β-D-glucopyranosyl-(1→2)-β-D-galactopyranoside(3).Conclusion Compound 1 is a new compound named macrostemonoside S.Compound 2 is isolated from Allii Macrostemonis Bulbus for the first time.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:Two novel aspochalasins, tricochalasin A (1) and aspochalasin A2 (2), along with three known compounds (3-5) have been isolated from the different culture broth of Aspergillus sp., which was found in the gut of a marine isopod Ligia oceanica. Compound 1 contains a rare 5/6/6 tricyclic ring fused with the aspochalasin skeleton. The structures were determined on the basis of electrospray ionisation mass spectroscopy (ESIMS), nuclear magnetic resonance (NMR) spectral data, and the absolute configurations were further confirmed by modified Mosher's method. Cytotoxicity against the prostate cancer PC3 cell line were assayed by the MTT method. Compound 3 showed strong activity while the remaining compounds showed weak activity.

Abstract:Abstract Phytochemical investigation on the roots of Asparagus cochinchinensis led to the isolation of one new furostanol saponin, named 26- O - β - d -glucopyranosyl-22 α -hydroxyl-(25 R )-Δ 5(6) -furost-3 β ,26-diol-3- O - α - l -rhamnopyranosyl-(1 → 2)-[ β - d -glucopyranosyl-(1 → 4)- α - l -rhamnopyranosyl-(1 → 4)]- β - d -glucopyranoside ( 1 ), along with three known congeners ( 2 ‒ 4 ). The structure of new saponin was elucidated via comprehensive inspection of its HRMS and NMR spectral data as well as chemical technology, whereas those of known ones were identified by comparison of their NMR and MS spectral data with those reported in literatures. All isolated saponins were evaluated for their cytotoxic effects on two human liver (MHCC97H) and lung adenocarcinoma (H1299) cancer cells in vitro. Among them, both 1 and 2 showed significant cytotoxicity against above mentioned cell lines. Further studies revealed that these two saponins could significantly inhibit their proliferation of MHCC97H and H1299 cells. Graphic Abstract

Abstract:A new sesquiterpenoid derivative, named aspergiketone (1), along with seven known compounds (2-8) were isolated from the coastal saline soil fungus Aspergillus fumigatus. Their structures were elucidated by spectroscopic analysis, and by comparison of experimental and reported data. The absolute configuration of compound 1 was defined by X-ray diffraction analysis. Compound 1 was cytotoxic towards HL-60 and A549 cell lines with IC50 values of 12.4 and 22.1 mu M, respectively.

Abstract:OBJECTIVE:To isolate and determine the chemical constituents from Astilbe chinensis. METHOD:Four compounds were isolated and purified by extraction and column chromatography on silica gel. The chemical structures were determined on the basis of physical-chemical evidence and extensive spectral analysis(HR-EIMS, EIMS, 13C-NMR, 1H-NMR, DEPT, 1H-1H-COSY, HMQC and HMBC). RESULT:Their structures were elucidated as beta-sitosterol palmitate I, daucosterol II beta-sitosterol III and Bergenin IV, respectively. CONCLUSION:Compound I and II were isolated from A. chinensis for the first time.

Abstract:A preliminary chemical investigation on 70% MeOH extract of the roots of Asparagus cochinchinensis resulted in the isolation of nine steroids. These isolates comprised of four new C21 (1-4) and one new pregnane (5) glycosides, and four known C27 (6-9) spirostanol steroids. Their structures were identified via analysis of the spectroscopic data and the results of hydrolytic cleavage. The cytotoxic activities of the compounds were tested toward the human tumor cell line Hela (cervical cancer), and compounds 7 and 8 displayed moderate activity with IC50 values of 35.5 and 39.6 μM, respectively.

Abstract:A new steroidal saponin, 26-O-β-d-glucopyranosyl-(25R)-furost-5-ene-3β,22α,26-triol 3-O-(1−4)-β-d-glucopyranosyl-α-l-rhamnopyranosyl-(1−2)-[α-l-rhamnopyranosyl-(1−4)]-β-d-glucopyranoside [asparacochioside A (1)] was isolated from a hot water extract of the roots of Asparagus cochinchinensis, together with the known steroidal saponins protodioscin (2), methyl protodioscin (3), aspacochioside A (4), aspacochioside C (5), 15−hydroxypseudoprotodioscin (6), and chamaedroside E (7). The structure of the new compound 1 was determined by interpretation of its spectroscopic data (1D- and 2D-NMR and HR−Q−TOF−MS) and sugar analysis. The isolated compounds 1−7 were tested for their in vitro cytotoxicity against human ovarian cancer cell lines (A2780 and SKOV3). Asparacochioside A (1) exhibited a significant cytotoxicity against both A2780 and SKOV3 cells with IC50 values of 5.25 ± 2.2 and 46.82 ± 9.43 μM, respectively. Furthermore, asparacochioside A (1) significantly increased the percentage of Annexin V-positive cells (apoptotic cells), suggesting that asparacochioside A induces ovarian cancer cell death via apoptosis.

Abstract:In the ongoing research on potent antitumor agents from the rhizomes of Asparagus cochinchinensis, seven undescribed steroidal saponins asparagusoside A–G (1–7), along with twenty known ones (8–27), were isolated and elucidated via analyzing their 1D, 2D NMR, mass spectroscopic data and chemical methods. All isolated compounds were evaluated for their cytotoxic effects against human large cell lung carcinoma cells (NCI-H460) in vitro. Among them, compounds 7, 9 and 27 showed more significant antitumor activities than the positive control cisplatin (11.56 μM) with IC50 values of 1.39, 3.04, and 2.25 μM, respectively. Further research about asparagusoside G (7) showed G0/G1 arrest in NCI-H460 cell line cycle and induced cell death by apoptosis in a dose‑dependent way.

Abstract:Two new sulfated steroidal compounds (1 and 2), along with three known steroidal saponins (3, 4, and 5) were isolated from the starfish Asterias amurensis Lutken. The structures of new compounds were elucidated as 3beta-O-sulfated-cholest-5-ene-7alpha-ol (1) and (E) 25-O-beta-d-xylopyranosyl-26, 27-dinor-24(S)-methyl-22-ene-15alpha-O-sulfated-5alpha-cholest-3beta,6alpha-ol (2) by extensive NMR experiments and chemical evidence. Their effects on UMR106 cell proliferation were screened by MTT method. The results indicated that compounds 2 and 2a (0.01-100 microM) significantly promoted the osteoblastic proliferation. The initial structure-activity relationship analysis suggests that the sugar moiety is the necessary group for the activity.

Abstract:Two new steroidal saponins, chinenoside VI (1) and neomacrostemonoside D (8b), together with ten known saponins were isolated from the bulbs of Allium chinense. On the basis of chemical evidence and spectral analysis (IR, H-1-NMR C-13-NMR H-1-H-1 COSY, HMQC, HMBC and SI-MS), chinenoside VI was established as (25R)-24-O-beta-D-glucopyranosyl-3 beta,24 beta-dihydroxy-5 alpha-spirost 3-O-alpha-arabinopyranosyl-(1-->6)-beta-D-glucopyranoside, and neomacrostemonoside D was elucidated as neotigogenin 3-O-beta-D-glucopyranosyl-(1-->2)[beta-D-glucopyranosyl-(1-->3)]-(6-acetyl-beta-D-glucopyranosyl)-(1-->4)-beta-D-galactopyranoside, respectively.

Abstract:OBJECTIVE:To study the chemical constituents of the roots and rhizomes of Asarum longerhizomatosum.METHOD:Chromatography and spectral analysis were used to isolate the constituents and elucidate their structures.RESULT:Five compounds were isolated from the ethanol extracts of the roots and rhizomes, and identified as asarone(I), beta-sitosterol(II), 2,4,5-trimethoxybenzaldehyde(III), 4-(2,4,5-trimethoxyphenyl)-3-en-butylone(IV) and 3 beta-hydroxystigmast-5-en-7-one(V).CONCLUSION:All the compounds were isolated from the plant for the first time, and IV is a new natural product.
