Protective Effect of Gingerol Oxime on Glu-Induced Neurotoxicity in PC12 Cells

Abstract:[Objective] To investigate the effect of glutamate (Glu) on cultured PC12 cells and the protective effect of gingerol oxime. [Methods] The toxicity model was established by treating PC12 cells with Glu in vitro. Cell viability was assayed with MTT method. The cell apoptotic rate was measured by flow cytometry (FCM) and Hoechst 33258 DNA staining method. [Results] After treated with Glu (5 mmol/L) for 24 h, the viability of PC12 cells depressed and the viable cells was only 58.3%when compared with cell control group tested by MTT assay. In a certain range of concentration, gingerol oxime can protect PC12 cells from the injury of Glu, althogh it showed some toxicity to cell at high concentration. Pretreated with gingerol oxime solution at concentrations of 3.125, 6.25, 12.5, and 25 μg/mL, respectively, the cell viabilities were all increased significantly. The protective effect of gingerol oxime on cells increased with its concentration decreased. At the concentration of 6.25 μg/mL, it showed the highest protective effect and the survival rate of PC12 cells reached to 75.2% (P 0.01), following with 70.2% (P 0.01) when preincubated with 3.125 μg/mL of gingerol oxime. Glu (5 mmol/L) induced apoptosis in PC12 cells, and the apoptotic rate was 20.1% after 24 h of treatment. Pretreated with different dose of gingerol oxime at 3.125, 6.25, 12.5 and 25 μg/mL, the cell apoptotic rates reduced and reached at 3.5%(P 0.01), 2.8%(P 0.01), 9.6%(P 0.01), and 17.7%(P 0.05), respectively. [Conclusion] Glu can induce apoptosis in PC12 cells, while gingerol oxime can protect PC12 cells from the jnjury induced by Glu, especially at lower concentration, and 6.25 μg/mL of gingerol oxime exerted the best effect.
