Abstract:Intracellular Ca2+ homeostasis is a prerequisite for a healthy cell life. While cells from some mammals may suffer dysregulation of intracellular Ca2+ levels under certain deleterious and stressful conditions, including hypothermia and ischemia, cells from mammalian hibernators exhibit a remarkable ability to maintain a homeostatic intracellular Ca2+ environment. Compared with cells from non-hibernators, hibernator cells are characterized by downregulation of the activity of Ca2+ channels in the cell membrane, which helps to prevent excessive Ca2+ entry. Concomitantly, sequestration of Ca2+ by intracellular Ca2+ stores, especially the sarcoplasmic/endoplasmic reticulum, is enhanced to keep the resting levels of intracellular Ca2+ stable. An increase in stored Ca2+ in heart cells during hibernation ensures that the levels of Ca2+ messenger are sufficient for forceful cell contraction under conditions of hypothermia. Maintenance of Na+ gradients, via Na+-Ca2+ exchangers, is also important in the Ca2+ homeostasis of hibernator cells. Understanding the adaptive mechanisms of Ca2+ regulation in hibernating mammals may suggest new strategies to protect nonhibernator cells, including those of humans, from Ca2+-induced dysfunction.
Transcriptional Regulation of Intermolecular Ca2+ Signaling in Hibernating Ground Squirrel Cardiomyocytes: the Myocardin–junctophilin Axis

Abstract:The contraction of heart cells is controlled by the intermolecular signaling between L-type Ca2+ channels (LCCs) and ryanodine receptors (RyRs), and the nanodistance between them depends on the interaction between junctophilin-2 (JPH2) in the sarcoplasmic reticulum (SR) and caveolin-3 (CAV3) in the transversal tubule (TT). In heart failure, decreased expression of JPH2 compromises LCC-RyR communication leading to deficient blood-pumping power. In the present study, we found that JPH2 and CAV3 transcription was concurrently regulated by serum response factor (SRF) and myocardin. In cardiomyocytes from torpid ground squirrels, compared with those from euthermic counterparts, myocardin expression was up-regulated, which boosted both JPH2 and CAV3 expression. Transmission electron microscopic imaging showed that the physical coupling between TTs and SRs was tightened during hibernation and after myocardin overexpression. Confocal Ca2+ imaging under the whole-cell patch clamp condition revealed that these changes enhanced the efficiency of LCC-RyR intermolecular signaling and fully compensated the adaptive down-regulation of LCCs, maintaining the power of heart contraction while avoiding the risk of calcium overload during hibernation. Our finding not only revealed an essential molecular mechanism underlying the survival of hibernating mammals, but also demonstrated a "reverse model of heart failure" at the molecular level, suggesting a strategy for treating heart diseases.

Abstract:Hibernation is an adaptive behavior for some small animals to survive cold winter. Hibernating mammals usually down-regulate their body temperature from ~37°C to only a few degrees. During the evolution, mammalian hibernators have inherited unique strategies to survive extreme conditions that may lead to disease or death in humans and other non-hibernators. Hibernating mammals can not only tolerant deep hypothermia, hypoxia and anoxia, but also protect them against osteoporosis, muscle atrophy, heart arrhythmia and ischemia-reperfusion injury. Finding the molecular and regulatory mechanisms underlying these adaptations will provide novel ideas for treating related human diseases.

Abstract:Significance The efficiency of excitation–contraction coupling in heart cells depends partially on the interaction between junctophilin-2 in the sarcoplasmic reticulum and caveolin-3 on T-tubules. Decreased junctophilin-2 has been linked to heart failure. Here, we found that the transcription of junctophilin-2 and caveolin-3 is controlled concurrently by the serum response factor and myocardin. During the entree into hibernation in ground squirrels, the up-regulation of myocardin boosts junctophilin-2/caveolin-3 expression and enhances the efficiency of excitation–contraction coupling. This finding not only reveals an adaptive mechanism underlying the blood-pumping power in hibernation, but also offers a strategy to correct deficient Ca 2+ signaling in heart diseases.

Recording of Calcium Transient and Analysis of Calcium Removal Mechanisms in Cardiac Myocytes from Rats and Ground Squirrels

Abstract:With confocal microscopy, we recorded calcium transients and analyzed calcium removal rate at different temperatures in cardiac myocytes from the rat, a non-hibernator, and the ground squirrel, a hibernator. The results showed a remarkable increase of the diastolic level of calcium transients in the rat but no detectable change in the ground squirrel. Calcium transient of the ground squirrel, compared with that of the rat at the same temperature, had a shorter duration and showed a faster calcium removal. As indicated by the pharmacological effect of cyclopiazonic acid, calcium uptake by sarcoplasmic reticulum (SR) was the major mechanism of calcium removal, and was faster in the ground squirrel than in the rat. Our results confirmed the essential role of SR in hypothermia-tolerant adaptation, and negated the importance of Na-Ca exchange. We postulated the possibility to improve hypothermia-tolerance of the cardiac tissue of non-hibernating mammals.

Abstract:Heart tissues from hibernating mammals, such as ground squirrels, are able to endure hypothermia, hypoxia and other extreme insulting factors that are fatal for human and nonhibernating mammals. This study was designed to understand adaptive mechanisms involved in intracellular Ca(2+) homeostasis in cardiomyocytes from the mammalian hibernator, ground squirrel, compared to rat. Electrophysiological and confocal imaging experiments showed that the voltage-dependence of L-type Ca(2+) current (I(Ca)) was shifted to higher potentials in ventricular myocytes from ground squirrels vs. rats. The elevated threshold of I(Ca) did not compromise the Ca(2+)-induced Ca(2+) release, because a higher depolarization rate and a longer duration of action potential compensated the voltage shift of I(Ca). Both the caffeine-sensitive and caffeine-resistant components of cytosolic Ca(2+) removal were more rapid in ground squirrels. Ca(2+) sparks in ground squirrels exhibited larger amplitude/size and much lower frequency than in rats. Due to the high I(Ca) threshold, low SR Ca(2+) leak and rapid cytosolic Ca(2+) clearance, heart cells from ground squirrels exhibited better capability in maintaining intracellular Ca(2+) homeostasis than those from rats and other nonhibernating mammals. These findings not only reveal adaptive mechanisms of hibernation, but also provide novel strategies against Ca(2+) overload-related heart diseases.
[Experimental Analysis of the Calcium Source for Cardiac Excitation-Contraction Coupling in Hibernator Citellus Dauricus].

Abstract:The force-interval and force-frequency relationships and the effects of Cd2+ and ryanodine on the myocardial action potential and contraction were compared between hibernating (HGS) and active (AGS) ground squirrels Citellus dauricus. (1) Raising the driving frequency caused a negative inotropic effect in the AGS group, but a biphasic change with an increase of the peak force followed by a decrease in the HGS group. The contraction in HGS group exhibited a more pronounced post-interval potentiation, and was more significantly modulated by the driving frequency. (2) HGS animals displaged action potentials of shorter early-stage duration and stronger contractions of shorter time course in comparison with the AGS animals. Both the action potential and the contraction of HGS group were less affected by Cd2+, but the contraction was more significantly inhibited by ryanodine than that of the AGS group. Our results suggested that dependence of cardiac excitation-contraction coupling on calcium influx was weakened, while the function of sarcoplasmic reticulum as a source of activator Ca2+ was enhanced during hibernation, which might take an important part in the cold-tolerant adaptation of hibernating hearts.
Up-regulation of a non-kinase activity isoform of Ca2+/calmodulin-dependent protein kinase kinase beta1 (CaMKKβ1) in hibernating bat brain

Abstract:Hibernation is an adaptive strategy that is utilized by some animals to survive the harsh environments of low temperature and food scarce. Hibernators, however, can survive in frequent and dramatic fluctuation of body temperature and blood flow causing by periodic arousals during hibernation without brain insult, and this indicates that it must have some unique adaptive aspects of hibernation physiology. To find out the up-regulated genes of bat brain during hibernation and explore the brain function adaptive mechanism of bat, the suppression subtractive hybridization (SSH) library was constructed from the brain tissue of greater horseshoe bats. Dot blot screening was carried out and the up-regulated genes in hibernating state were obtained. Then RT-PCR and RQ-PCR were performed to test the expression patterns of selected cDNAs. Here we first show that the functional and non-functional isoforms of bat CaMKKβ1 display distinct expression patterns between hibernating and active states. The up-regulation of non-functional form of CaMKKβ1 may represent a new neuroprotective strategy adopted by bats or even other hibernators to avoid the CNS damage during hibernation. Our results showed that bat CaMKKβ1 gene has four transcript isoforms and these transcript variants differ primarily in exons b and d, which are 129 bp and 43 bp respectively. Statistical analyses indicated that these isoforms display distinct expression patterns at different states, in which only isoform 3, the non-functional form, increased 300% at hibernating state. These results suggest that distinct expression patterns of transcript isoforms of a gene, which have different activity, may represent a new potential adaptive mechanism in hibernation, except for the simple up-regulation of selected genes/proteins and the reversible protein phosphorylation.

Abstract:1. The hibernating mammals inherited a stable cardiovascular function as a result of adaptation to extreme external and internal environments during hibernation.2. The cardiovascular function of a hibernator is characterized by three distinct features: (i) tolerance of hypothermia; (ii) resistance to dysrhythmia; and (iii) endurance of hypoxia. All these have clinical relevance in cardiovascular disorders in humans.3. We suggest that the hibernating mammal can serve as a useful animal model for research into the cardiovascular system.
A Unique Energy-Saving Strategy during Hibernation Revealed by Multi-Omics Analysis in the Chinese Alligator

Abstract:Many ectotherms hibernate in face of the harsh winter conditions to improve their survival rate. However, the molecular mechanism underlying this process remains unclear. Here, we explored the hibernation mechanism of Chinese alligator using integrative multi-omics analysis. We revealed that (1) the thyroid hormone biosynthesis, nutrition absorption and metabolism, muscle contraction, urinary excretion and immunity function pathways are overall downregulated during hibernation; (2) the fat catabolism is completely suppressed, contrasting with the upregulation of hepatic fatty-acid-transporter CPT1A, suggesting a unique energy-saving strategy that differs from that in hibernating mammals; (3) the hibernation-related genes are not only directly regulated by DNA methylation but also controlled by methylation-dependent transcription networks. In addition, we identified and compared tissue-specific, species-specific, and conserved season-biased miRNAs, demonstrating complex post-transcriptional regulation during hibernation. Our study revealed the genetic and epigenetic mechanisms underlying hibernation in the Chinese alligator and provided molecular insights into the evolution of hibernation regulation.
Molecular mechanism of Chinese alligator (Alligator sinensis) adapting to hibernation

Abstract:Hibernation is a physiological state for Chinese alligators to cope with cold weather. In mammals, gene expression changes during hibernation and their regulatory mechanisms have been extensively studied, however, these studies in reptiles are still rare. Here, integrated analysis of messenger RNA (mRNA), microRNA (miRNA), and long noncoding RNA (lncRNA) reveals the molecular mechanisms of the hypothalamus, liver, and skeletal muscle in hibernating and active individuals. During hibernation, the number of genes increased in the hypothalamus, liver, and skeletal muscle was 585, 282, and 297, while the number of genes decreased was 215, 561, and 627, respectively, as compared with active individuals. Through Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis, the differential expressed genes were mainly enriched in DNA damage repair, biological rhythm, energy metabolism, myoprotein degradation, and other related items and pathways. Besides, 4740 miRNAs were identified in three tissues. Through the comprehensive analysis of miRNA and mRNA abundance profiles, 12,291, 6997, and 8232 miRNA-mRNA pairs all showed a negative correlation in the hypothalamus, liver, and skeletal muscle, respectively. Some miRNA target genes were related tobiological rhythm and energy metabolism, suggesting that miRNA may play an important role in the physiological metabolism of the hibernating adaptability of Chinese alligators. Moreover, 402, 230, and 130 differentially expressed lncRNAs were identified in the hypothalamus, liver, and skeletal muscle, respectively. The targeting relationship of four lncRNA-mRNA pairs were predicted, with the main function of target genes involved in the amino acid transportation. These results are helpful to further understand the molecular regulatory basis of the hibernation adaptation in Chinese alligators.
A Temporal Examination of Cytoplasmic Ca2 + Levels, Sarcoplasmic Reticulum Ca2 + Levels, and Ca2 + -Handling-Related Proteins in Different Skeletal Muscles of Hibernating Daurian Ground Squirrels

Abstract:To explore the possible mechanism of the sarcoplasmic reticulum (SR) in the maintenance of cytoplasmic calcium (Ca2+) homeostasis, we studied changes in cytoplasmic Ca2+, SR Ca2+, and Ca2+-handling proteins of slow-twitch muscle (soleus, SOL), fast-twitch muscle (extensor digitorum longus, EDL), and mixed muscle (gastrocnemius, GAS) in different stages in hibernating Daurian ground squirrels (Spermophilus dauricus). Results showed that the level of cytoplasmic Ca2+ increased and SR Ca2+ decreased in skeletal muscle fiber during late torpor (LT) and inter-bout arousal (IBA), but both returned to summer active levels when the animals aroused from and re-entered into torpor (early torpor, ET), suggesting that intracellular Ca2+ is dynamic during hibernation. The protein expression of ryanodine receptor1 (RyR1) increased in the LT, IBA, and ET groups, whereas the co-localization of calsequestrin1 (CSQ1) and RyR1 in GAS muscle decreased in the LT and ET groups, which may increase the possibility of RyR1 channel-mediated Ca2+ release. Furthermore, calcium pump (SR Ca2+-ATPase 1, SERCA1) protein expression increased in the LT, IBA, and ET groups, and the signaling pathway-related factors of SERCA activity [i.e., β-adrenergic receptor2 protein expression (in GAS), phosphorylation levels of phospholamban (in GAS), and calmodulin kinase2 (in SOL)] all increased, suggesting that these factors may be involved in the up-regulation of SERCA1 activity in different groups. The increased protein expression of Ca2+-binding proteins CSQ1 and calmodulin (CaM) indicated that intracellular free Ca2+-binding ability also increased in the LT, IBA, ET, and POST groups. In brief, changes in cytoplasmic and SR Ca2+ concentrations, SR RyR1 and SERCA1 protein expression levels, and major RyR1 and SERCA1 signaling pathway-related factors were unexpectedly active in the torpor stage when metabolic functions were highly inhibited.

Convergent accelerated evolution of mammal-specific conserved non-coding elements in hibernators

Abstract:Mammals maintain their body temperature, yet hibernators can temporarily lower their metabolic rate as an energy-saving strategy. It has been proposed that hibernators evolved independently from homeotherms, and it is possible that the convergent evolution of hibernation involved common genomic changes among hibernator-lineages. Since hibernation is a seasonal trait, the evolution of gene regulatory regions in response to changes in season may have been important for the acquisition of hibernation traits. High-frequency accumulation of mutations in conserved non-coding elements (CNEs) could, in principle, alter the expression of neighboring genes and thereby contribute to the acquisition of new traits. To address this possibility, we performed a comparative genomic analysis of mammals to identify accelerated CNEs commonly associated with hibernation. We found that accelerated CNEs are common to hibernator-lineages and could be involved with hibernation. We also found that common factors of genes that located near accelerated CNEs and are differentially expressed between normal and hibernation periods related to gene regulation and cell-fate determination. It suggests that the molecular mechanisms controlling hibernation have undergone convergent evolution. These results help broaden our understanding of the genetic adaptations that facilitated hibernation in mammals and may offer insights pertaining to stress responses and energy conservation.

Anti-apoptotic signaling as a cytoprotective mechanism in mammalian hibernation

Abstract:In the context of normal cell turnover, apoptosis is a natural phenomenon involved in making essential life and death decisions. Apoptotic pathways balance signals which promote cell death (pro-apoptotic pathways) or counteract these signals (anti-apoptotic pathways). We proposed that changes in anti-apoptotic proteins would occur during mammalian hibernation to aid cell preservation during prolonged torpor under cellular conditions that are highly injurious to most mammals (e.g. low body temperatures, ischemia). Immunoblotting was used to analyze the expression of proteins associated with pro-survival in six tissues of thirteen-lined ground squirrels, Ictidomys tridecemlineatus. The brain showed a concerted response to torpor with significant increases in the levels of all anti-apoptotic targets analyzed (Bcl-2, Bcl-xL, BI-1, Mcl-1, cIAP1/2, xIAP) as well as enhanced phosphorylation of Bcl-2 at S70 and T56. Heart responded similarly with most anti-apoptotic proteins elevated significantly during torpor except for Bcl-xL and xIAP that decreased and Mcl-1 that was unaltered. In liver, BI-1 increased whereas cIAP1/2 decreased. In kidney, there was an increase in BI-1, cIAP and xIAP but decreases in Bcl-xL and p-Bcl-2(T56) content. In brown adipose tissue, protein levels of BI-1, cIAP1/2, and xIAP decreased significantly during torpor (compared with euthermia) whereas Bcl-2, Bcl-xL, Mcl-1 were unaltered; however, Bcl-2 showed enhanced phosphorylation at Thr56 but not at Ser70. In skeletal muscle, only xIAP levels changed significantly during torpor (an increase). The data show that anti-apoptotic pathways have organ-specific responses in hibernators with a prominent potential role in heart and brain where coordinated enhancement of anti-apoptotic proteins occurred in response to torpor.

Temperature Dependence of Intracellular Free Calcium in Cardiac Myocytes from Rat and Ground Squirrel Measured by Confocal Microscopy

Abstract:The temperature-dependence of intracellular free calcium ([Ca 2+ ] i ) was investigated in indo-1 loaded ventricular myocytes from the rat, a non-hibernator, and from the ground squirrel, a hibernator. The dissociation constant of indo-1 at different temperatures was calibrated both at pH-stat and at α-stat, and the result demonstrated that the α-stat calibration should be preferred. Analysis of the fluorescent image showed a striking increase of [Ca 2+ ] i as well as spontaneous calcium waves in rat cells, indicating an overloaded calcium. In contrast, cardiac myocytes of the ground squirrel were found to keep a constant [Ca 2+ ] i without calcium overload regardless of temperature variation. It is believed that understanding of the mechanisms underlying the intercellular calcium homeostasis of hibemators may lead to solutions of some medical questions.
Life in the cold: links between mammalian hibernation and longevity

Abstract:The biological process of aging is the primary determinant of lifespan, but the factors that influence the rate of aging are not yet clearly understood and remain a challenging question. Mammals are characterized by >100-fold differences in maximal lifespan, influenced by relative variances in body mass and metabolic rate. Recent discoveries have identified long-lived mammalian species that deviate from the expected longevity quotient. A commonality among many long-lived species is the capacity to undergo metabolic rate depression, effectively re-programming normal metabolism in response to extreme environmental stress and enter states of torpor or hibernation. This stress tolerant phenotype often involves a reduction in overall metabolic rate to just 1-5% of the normal basal rate as well as activation of cytoprotective responses. At the cellular level, major energy savings are achieved via coordinated suppression of many ATP-expensive cell functions; e.g. global rates of protein synthesis are strongly reduced via inhibition of the insulin signaling axis. At the same time, various studies have shown activation of stress survival signaling during hibernation including up-regulation of protein chaperones, increased antioxidant defenses, and transcriptional activation of pro-survival signaling such as the FOXO and p53 pathways. Many similarities and parallels exist between hibernation phenotypes and different long-lived models, e.g. signal transduction pathways found to be commonly regulated during hibernation are also known to induce lifespan extension in animals such as Drosophila melanogaster and Caenorhabditis elegans. In this review, we highlight some of the molecular mechanisms that promote longevity in classic aging models C. elegans, Drosophila, and mice, while providing a comparative analysis to how they are regulated during mammalian hibernation.

Abstract:Cardiac excitation-contraction (E-C) coupling is controlled by the signaling between L-type Ca2+ channels (LCCs) in the cell membrane/T-tubules (TTs) and ryanodine receptor (RyR) Ca2+ release channels in the sarcoplasmic reticulum (SR). In heart failure, decreased expression of junctophilin-2 (JPH2) decreased the efficiency of LCC-RyR signaling and compromised the gain of E-C coupling. In contrast, we found that the LCC-RyR signaling efficiency and the E-C coupling gain were increased significantly during hibernation in ground squirrels. The structural consolidation of LCC-RyR signaling apparatus was in parallel with increased expression of JPH2 and caveolin-3 (CAV3), the interaction between which anchors the SR to TTs. Interestingly, the promoters of both JPH2 and CAV3 exhibited binding sites for the serum response factor (SRF), which initiated JPH2 and CAV3 transcription by binding the transcription co-factor myocardin. During hibernation, the expression of myocardin was increased while that of microRNA-24, which mediates post-transcriptional suppression of JPH2 expression, was decreased. Adenovirus-directed overexpression of myocardin, but not that of SRF, increased JPH2 and CAV3 expression, improved LCC-RyR signaling efficiency and enhanced E-C coupling gain in ground squirrel cardiomyocytes. The myocardin-mediated conjunct regulation of JPH2/CAV3 transcription explained the cardiac specificity of JPH2 expression, and revealed a stoichiometry-optimized mechanism for E-C coupling regulation. This finding not only elucidated the mechanism for maintained cardiac contractility in hibernation, but also provides new ideas for studying the “loss-of-function” remodeling in heart diseases, such as heart failure.

Abstract:Hibernation, an evolved survival trait among animals, enables them to endure frigid temperatures and food scarcity during the winter months, and it is a widespread phenomenon observed in mammals. The gut microbiota, a crucial component of animal nutrition and health, exhibits particularly dynamic interactions in hibernating mammals. This manuscript comprehensively evaluates the impacts of fasting, hypothermia, and hypometabolism on the gut microbiota of hibernating mammals. It suggests that alterations in the gut microbiota may contribute significantly to the maintenance of energy metabolism and intestinal immune function during hibernation, mediated by their metabolites. By delving into these intricacies, we can gain a deeper understanding of how hibernating mammals adapt to their environments and the consequences of dietary modifications on the symbiotic relationship between the gut microbiota and the host. Additionally, this knowledge can inform our comprehension of the protective mechanisms underlying long-term fasting in non-hibernating species, including humans, providing valuable insights into nutritional strategies and health maintenance.

Abstract:During hibernation, mammalian cells are exposed to severe environmental stressors such as low temperature, lowered O2 supply, and glucose deficiency. The cellular metabolic rate is markedly reduced for adapting to these conditions. AMP-activated protein kinase (AMPK) senses the cellular energy status and regulates metabolism. Therefore, we examined AMPK signaling in several brain regions and peripheral tissues in hibernating chipmunk. Eukaryotic elongation factor 2 (eEF2) is a downstream target of AMPK. Phosphorylation of eEF2, indicating its inactivation, is enhanced in the cerebral cortex of hibernating chipmunks. The study indicated that the sequential regulation of AMPK-mammalian target of rapamycin complex 1-eEF2 signaling was altered and protein synthesis ability was reduced in the cerebral cortex of hibernating chipmunks.
