Abstract:In order to isolate the strain with highly thermostable exo-inulinase,and to investigate its culture conditions optimization with the maximal enzyme production and analyse enzyme characterization,experiments were carried out. The results showed that the newly isolated strain with highly thermostable extracellular exo-inulinase(EC 3.2.1.80)enzyme has been investigated to improve industrial processed of inulin. Production of Exo-beta-D-fructosidase by Penicillium notatum isolated from dahlia rhizosphere was mutated and designated Penicillium notatum MH1. Half fractional factorial design (HFFD)and Central composite design(CCD)were used to optimize the crucial factors of medium composition. The highest production average was 32.46 U/(mL·min)after 48 h with media composition g/L-1〔Jerusalem artichoke root powder without prior treatments(JA)24,Beef extract(BE)5,Yeast extract(YE)1,Soy bean cake powder(SB)1,KH2PO4 3,MgSO4·7H2O 0.2〕at 34℃,initial pH 3.5(phosphate buffer)and shaking speed 200 r/min. Properties of the enzyme partially studied,pH range was 4 to 7 and optimum pH and temperature were 4.2 and 55℃ respectively. The crude enzyme was purified by fractional precipitation with ammonium sulphate and the highest activity was achieved at 20%～58% saturation,followed by two chromatographic(DEAE-cellulose and Sephadex G-100 column chromatographies). The purified enzyme had MW(108±1)KDa as judged by SDS-PAGE.
