Abstract:Liquid crystal-based sensors offer the advantage of high sensitivity at a low cost. However, they often lack selectivity altogether or require costly and unstable biomaterials to impart this selectivity. To incur this selectivity, we herein integrated a molecularly imprinted polymer (MIP) film recognition unit with a liquid crystal (LC) in an optical cell transducer. We tested the resulting chemosensor for protein determination. We examined two different LCs, each with a different optical birefringence. That way, we revealed the influence of that parameter on the sensitivity of the (human serum albumin)-templated (MIP-HSA) LC chemosensor. The response of this chemosensor with the (MIP-HSA)-recognizing film was linear from 2.2 to 15.2 μM HSA, with a limit of detection of 2.2 μM. These values are sufficient to use the devised chemosensor for HSA determination in biological samples. Importantly, the imprinting factor (IF) of this chemosensor was appreciable, reaching IF = 3.7. This IF value indicated the predominant binding of the HSA through specific rather than nonspecific interactions with the MIP.

Abstract:Molecularly imprinted polymer(MIP) films for hemoglobin detection were prepared onto the Au/Cr coated surface plasmon resonance(SPR) sensor chips by the in situ electropolymerization of 3-aminophenylboronic acid(3-APBA). The formation of the films and rebinding processes of hemoglobin were monitored by in situ electrochemical-SPR(EC-SPR) spectroscopy, with allowed real-time observation of the simultaneous changes in electrochemical and optical properties of the films. Scanning electron microscopy(SEM) and atomic force microscopy(AFM) were used to characterize the surface morphologies of the MIP films. The effects of pH, ion strength, different metal ions on rebinding Hb, the specific binding and the selective recognition were investigated. The results obtained with the molecular imprinted SPR chips indicate a good adsorption of Hb in a range of 0.0005–5 mg/mL in 0.05 mol/L sodium phosphate buffer at pH=7.0. A linear calibration curve(R 2=0.94) of the SPR sensor for Hb detection was obtained in a range of 0.05–5 mg/mL. The detection limit for hemoglobin by this method was 0.000435 mg/mL(S/N=3). Interference studies indicate that the MIP films have a good selectivity compared with the referenced proteins. The stability of the sensor was also established. Results indicate that the SPR sensor chip keeps 87.6% of its original response after 14 d of storage under dry and ambient conditions.

Abstract:In this paper, a novel strategy of protein molecularly-imprinted electrochemical sensor was proposed to determine Bovine Hemoglobin selectively. Gold microdendrites and prussian blue particles were electrodeposited on the surface of glassy carbon electrode respectively to improve the sensitivity of MIECS. The molecularly imprinted polymers film is synthesized by electrochemically induced redox polymerization of acrylamide in the presence of template protein. The surface feature of the modified electrode was characterized by cyclic voltammetry. The reduced electrochemical intensity which was produced by prussian blue is related to the concentration of Bovine Hemoglobin. The times of washing and incubation of modified electrode were investigated and optimized. The MIECS with improved conductivity and electrochemical performances showed good response in the range of low concentrations from 0.1to 1.0×103μg/mL and the detection limit (LOD, 3σ, RSD≤4.31%) is 0.05μg/mL. The result demonstrates significant specific recognition toward the template protein via selective test and a reasonable regeneration in five recyclings. The proposed sensor exhibited high sensitivity and selectivity, acceptable reproducibility, and could be extended to recognize other protein.

Abstract:A bio-mimic bulk acoustic wave (BAW) sensor was fabricated by coating it with the epinephrine template molecular imprinting polymer (MIP). This sensor exhibited high selectivity and sensitive response to epinephrine. A linear relationship between 5.0×10−8 and 2.0×10−5M was revealed. The detection limit was 2.0×10−8M. Recoveries were 93.2–108.7%. Influencing factors were investigated in detail and optimized. Analytical applications of this bio-mimic sensor were performed.

Abstract:Mono‐dispersed molecularly imprinted hollow spheres (MIHSs) for hemoglobin (Hb) were prepared by employing silica nanospheres as the sacrificial templates. The obtained hollow spheres with uniform particle size of 360 nm in diameter were characterized by transmission electron microscopy. The outstanding affinities of these MIHSs to the target protein were confirmed by adsorption experiment in aqueous solution. Adsorption equilibrium was achieved within 10 min while the binding capacity (Qmax) of Hb was 8.84 µmol g–1 at pH7.0. Furthermore, the MIHSs were successfully assembled into a closely‐packed 3D colloidal array. The molecularly imprinted hollow sphere array (MIHSA) can selectively recognize Hb. As the concentration of Hb increased, the structure color of the MIHSA changed from blue to green, and turn to white finally with maximum red shift for 43 nm. The MIHSA showed promising potential for the naked‐eye detection of target Hb. Reflection spectra of the MIHSA in response to different Hb concentrations (0.075–15 µmol L–1)

Abstract:A simple and efficient molecularly imprinted polymers (MIPs) based electrochemical sensor were proposed and prepared by electropolymerization of pyrrole in the presence of bovine hemoglobin (BHb) in an aqueous solution. The fabrication process of the sensor was characterized by differential pulse voltammetry and electrochemical impedance spectroscopy, in which [Fe(CN)6]3−/[Fe(CN)6]4− was used as an electrochemical active probe. The influence factors including electropolymerization cycles, scan rate, the concentration of pyrrole, and the extraction conditions were investigated in detailed. Under the optimized conditions, the experimental results showed that the MIPs based sensor possessed a fast rebinding dynamics and an excellent recognition capacity to BHb, compared to the other non-template proteins. Moreover, the prepared sensor also exhibited a dependent relationship between the concentration of template protein and peak current of [Fe(CN)6]3−/[Fe(CN)6]4−.

Abstract:Using acrylamide and methylacrylic acid as functional monomers, human hemoglobin molecular-imprinting-polymers (HHb-MIPs) were successfully prepared. The studies showed that the specific molecular recognition properties of HHb-MIPs were related to both the interactions between the functional groups of polymers and imprinted protein, and geometric complementation of the cavities in the polymers. Further studies showed that the content of methyacrylic acid in the polymers could affect the specific binding properties of MIPs. The supplementation of methylacrylic acid can increase the specific adsorption capacity of the HHb-MIPs on the one hand, but decrease the elution capacity on the other hand.

Abstract:A simply hemoglobin (Hb) molecularly imprinted polymer (MIP) was prepared using Hb as the imprinted molecule, acrylamide as the functional monomer and cross-linked chitosan beads as the supporting matrix. The MIP was achieved by entrapment of the selective soft polyacrylamide gel in the pores of the cross-linked chitosan beads by letting acrylamide monomer and the protein diffuse into the pores of chitosan beads before starting the polymerization. The chitosan beads were freed from the surrounding polyacrylamide gel by washing. The Langmuir and Freundlich adsorption models were applied to describe the equilibrium isotherms. Langmuir analysis showed that an equal class of adsorption was formed in the MIP and the adsorption equilibrium constant and the maximum adsorption capacity were evaluated. The MIP has much higher adsorption capacity for Hb than the non-imprinted polymer with the same chemical composition, and the MIP also has a higher selectivity for the imprinted molecule. The MIP can be reused in an easy way and the reproduction coefficient was approximately 100% at low concentration.

Abstract:An electrochemical sensor based on loading molecularly imprinted polymers (MIP) on the material surface can improve the specificity towards the object. In this work, a T-shaped PbTiO3 with a high active-exposed (110) facet was prepared by a hydrothermal process. Then, Ag nanoparticles (Ag NPs) modified T-shaped PbTiO3 was obtained by in-situ photocatalytic reduced method under UV irradiation, where a hetero-junction was formed with a well lattice matching between the (111) facet of Ag-0 and the (110) facet of PbTiO3. A MIPs modified by Ag nanoparticles (NPs)/PbTiO3 (MAP) electrodes was prepared via electro polymerization process by o-Phenylenediamine (o-PD) in the presence of the template molecule, bovine hemoglobin (BHb), i.e., the detected molecule. The response peak current and concentration of BHb is demonstrated with a good linear relationship in the range of 0.00294-0.41 nM (R-2 =0.98), and the detection limit at 0.23 pM (S/N = 3). A heterojunction between Ag NPs and high- active facet of PbTiO3 is beneficial to oxidizing electroactive material ([Fe (CN)(6)](3-/4-)), generating more BHb-imprinting cavities on the modified electrode and improving the sensitivity of sensor. The electrochemical sensor is with a simple, stable structure and high sensitivity to BHb detection. Furthermore, the sensor was successfully applied to detect BHb in the bovine serum samples.

Abstract:Considering the importance of determining the levels of hemoglobin (Hb) as a vital protein in red blood cells, in this work a highly sensitive electrochemical sensor was developed based on a gold electrode (AuE) modified with Ag metal–organic framework mesoporous carbon (Ag-MOF@MC) and molecularly imprinted polymers (MIPs). To that end, the MIP layer was formed on the Ag-MOF@MC by implanting Hb as the pattern molecule during the polymerization. The modified electrode was designed using electrochemical approaches including differential pulse voltammetry (DPV), electrochemical impedance spectroscopy (EIS), and cyclic voltammetry (CV). Using a response level experimental design method, the most important parameters affecting the reaction of the sensing system including pH, incubation time, and scanning rate were optimized. Following the same route, the Hb concentration, pH, temperature, and elution times were optimized to prepare the imprinted polymer layer on the Ag-MOF@MC surface. By exploiting DPV techniques based on the optimal parameters, the electrochemical response of the AuE/Ag-MOF@MC-MIPs for Hb determination was recorded in a wide linear dynamic range (LDR) of 0.2 pM to 1000 nM, with a limit of detection (LOD) of 0.09 pM. Moreover, the Ag-MOF@MC-MIP sensing system showed good stability, high selectivity, and acceptable reproducibility for Hb determination. The sensing system was successfully applied for Hb determination in real blood samples, and the results were compared with those of the standard methods for Hb determination. Acceptable recovery (99.0%) and RDS% (4.6%) confirmed the applicability and reliability of the designed Hb sensing system.

Abstract:A novel molecular imprinting method was used to prepare twice-coated silica particles with specific recognition sites for hemoglobin. Chitosan was used as an intermedium to be coated on silica particles via phase inversion process, and the abundance of exposed amine groups (NH2) were active sites for introducing aldehyde groups. After hemoglobin was covalently immobilized by forming imine bonds with the aldehyde groups, acrylamide was then polymerized onto chitosan-coated silica particles to form the recognition sites. The obtained hemoglobin imprinted [molecularly imprinted polymer (MIP)] beads were characterized by FTIR spectroscopy, thermogravimetric analysis (TG), and scanning electron microscopy (SEM). Self-polymerization silica nanoparticles based molecularly imprinted polymers for selective recognition of protein

Abstract:Molecularly imprinted polymers (MIPs) are promising for precise protein separation and purification. However, challenges persist due to their large size, variable configuration, and instability during preparation. Here, a simple silicon self-assembly program was designed to synthesize MIPs without any organic reagents and acid-base catalysis, avoiding the structural damage of protein under severe conditions. In this method, employing hemoglobin (Hb) as a model protein, with tween-20 in emulsification, and tetraethyl orthosilicate (TEOS) as the cross-linking agent, along with co-functional monomers 3-aminopropyltriethoxysilane (APTES) and benzyl(triethoxy)silane (BnTES), enhanced binding efficacy was achieved. Successful imprinting was evidenced through surface morphology observation and physical/chemical property evaluations of the synthesized MIPs. A series of adsorption experiments were performed to investigate the recognition performance of Hb-MIPs. The Hb-MIPs not only exhibited large adsorption capacity (400 μg/mg) and good imprinting factor (6.09) toward template protein, but also showed satisfactory selectivity for reference proteins. Five cycles of adsorption proved that the Hb-MIPs had good reusability. In addition, the successful isolation of HB from bovine blood indicated that Hb-MIPs were an excellent separation and purification material. The mild preparation conditions and good adsorption capacity demonstrated the potential value of this method in separation and purification research.
Interconnectivity of Macroporous Molecularly Imprinted Polymers Fabricated by Hydroxyapatite-Stabilized Pickering High Internal Phase Emulsions-Hydrogels for the Selective Recognition of Protein

Abstract:Hydroxyapatite hybridized molecularly imprinted polydopamine polymers with selective recognition of bovine hemoglobin (BHb) were successfully prepared via Pickering oil-in-water high internal phase emulsions-hydrogels and molecularly imprinting technique. The emulsions were stabilized by hydroxyapatite of which the wettability was modified by 3-methacryloxypropyltrimethoxysilane. The materials were characterized by SEM, IR and TGA. The results showed that the BHb imprinted polymers based on Pickering hydrogels (Hydro-MIPs) possess macropores ranging from 20μm to 50μm, and their large numbers of amino groups and hydroxyl groups result in a favorable adsorption capacity for BHb. The maximum adsorption capacity of Hydro-MIPs for BHb was 438mg/g, 3.27 times more than that of the non-imprinted polymers (Hydro-NIPs). The results indicated that Hydro-MIPs possessing well-defined hierarchical porous structures exhibited outstanding recognition behavior towards the target protein molecules. This work provided a promising alternative method for the fabrication of polymer materials with tunable and interconnected pores structures for the separation and purification of protein in vitro.

Abstract:Through years of extensive research and development, molecularly imprinted polymers (MIPs) are still inferior to their biological rivals such as antibodies, enzymes etc. In this study we report a protein-imprinted cryogel, showing antibody-like affinity and selectivity against the protein template (bovine serum albumin, BSA). The MIP was synthesized from the co-polymerization of acrylamide, N,N-methylenebisacrylamide, acrylic acid and diallylamine. Due to the participation of the ionizable monomers (acrylic acid and diallylamine), imprinted cavities with inner surface-clung charged groups were created to recognize BSA. Therefore each cavity appears like a molecular capacitor charged by carboxyl and amino groups. As the cavities are all of a molecule-size volume, a membrane made of the MIP contains a huge array of the molecular capacitors. This will produce a synergistic effect and greatly amplify the impedance signal deviations when template sorption/desorption takes place on the sensor. When the MIP was used as an artificial antibody to make an electrochemical sensor, high sensitivity and selectivity were achieved at the same time. Results indicate that BSA could be determined in a linear range of 1.5 x 10(-16)-10(-12) mol-L-1. Meanwhile a low limit of detection was achieved at 7.2 x 10(-8) mol L-1. Conclusively protein-imprinted amphoteric polyacrylamide cryogels are materials of a great potential to sense and determine charged objects like molecules, cells, microorganisms or other particles.
Preparation of Molecularly Imprinted Polymers on Hemin-Graphene Surface for Recognition of High Molecular Weight Protein.

Abstract:Surface molecular imprinting for proteins, especially for the high molecular weight protein, are facing a great challenge today. We report a novel design to prepare molecular imprinting polymers (MIPs) on the surface of hemin-graphene nanosheets (H-GNs) for the recognition of thyroglobulin (Tg, 660 KD). H-GNs with an intrinsic peroxidase-like activity could effectively immobilize the protein template on its surface to improve the number of imprinted sites per unit surface area and then directly initiate free-radical polymerization to synthesize MIPs. The results indicated MIPs film could be prepared on the surface of H-GNs/filter paper, implying the advantages of surface imprinting, and exhibiting high adsorption capacity (400 mg g(-1)). The MIP composites clearly distinguished the proteins on the basis of the synergistic effect of shape complementarity and multiple interactions, not even anti-Tg antibodies and sample matrix. The gray intensity of the colorimetric paper-based sensor proved to be proportional to the concentration of Tg in the range of 5-100 ng mL(-1) with a detection limit of 1 ng mL(-1) (15 fM). The design and direct synthesis of MIPs on the H-GNs/filter paper provide a new perspective for the surface imprinting materials with potential in the recognition and detection of proteins.

Abstract:In this work, molecularly imprinted electrospun chromogenic membrane(MIP-ECM) was synthesized using P-Hydroxybenzene propanoic acid as imitate template molecule and Poly (vinyl alcohol)(PVA) as substrate. Ninhydrin is used as chromogenic agent for visualizing detection of target molecule l-tyrosine(Tyr). The results showed that molecularly imprinted membrane exhibited selectivity and specific adsorption capacity for l-tyrosine. The influences of environmental factors, such as reaction temperature, the amount of ninhydrin and elution time, on chromogenic properties were examined by SEM and optical microscope. Gray value analysis is proceeded by software Image J. Due to the facilitate preparation and intuitive visualization, the proposed method was a promising candidate for selective recognition of bio-generated acid and amino molecules.

Direct detection of fluorescence intensity of absorbed lysozyme(Lyz) on the MIMs was achieved for the fast detection of lysozyme. The surface morphology of polymer was characterized by SEM. Adsorption time and rebinding solution pH were optimized to improve recognition ability of MIMs for lysozyme. The results showed that the MIMs had a high selection and recognition ability for the lysozyme protein.
Chemically Modified Chitosan Beads As Matrices for Adsorptive Separation of Proteins by Molecularly Imprinted Polymer

Abstract:A simple molecularly imprinted polymer (MIP) was prepared using hemoglobin as the imprinted molecule and acrylamide as the functional monomer. The MIP was achieved by grafting of the selective soft polyacrylamide gel to the maleic anhydride modified chitosan beads by letting the monomers and the protein diffuse into the pores of the chemically modified chitosan matrix before starting the polymerization. The chitosan beads were freed from the surrounding polyacrylamide gel by washing. Langmuir analysis showed that an equal class of adsorption was formed in the MIP, the adsorption equilibrium constant and the maximum adsorption capacity were evaluated to be 1.47 and 35.7 mg/g wet MIP beads, respectively. The MIP has much higher adsorption capacity for hemoglobin than the non-imprinted polymer with the same chemical composition, and also has a higher selectivity for the imprinted molecule. The MIP can be reused and the reproducibility was approximately 100% at low concentration.
