Abstract:Objective:To investigate the expression and clinical significance of hypoxia-inducible factor 1α(HIF-1α) in hypoxic human lung cancer cell lines and human lung carcinoma tissues. Methods:Transwell assay was used to test the invasive capacity of hypoxic lung cancer cells. The expression of HIF-1α mRNA and protein in hypoxic lung cancer cells in vitro was determined by reverse transcription-polymerase chain reaction(RT-PCR) and Western blot. The expression of HIF-1α was detected using immunohistochemistry and reverse transcription-polymerase chain reaction analyses(RT-PCR) in 60 specimens of lung carcinoma and 20 specimens of corresponding adjacent tissues. Results:The effect of hypoxia on the invasion of lung cancer cells increased. After hypoxia exposure,the HIF-1α mRNA and protein expression in lung cancer cells increased time-dependently. Compared with those in the control group,the mRNA and protein levels of HIF-1α in the hypoxic groups had more significant difference(P 0.05 or P 0.01). In lung carcinoma tissues,the positive expression rates of HIF-1α mRNA and proteins were 90.0% and 75.0% ,while 30.0% and 20.0% in normal lung tissues respectively,showing the expression of HIF-1α in cancer tissues were significantly different from that in normal tissues(P 0.01). Comparing with the mRNA and protein levels of HIF-1α in lung cancer tissues of ⅠandⅡstage,there was marked increase in Ⅲ and Ⅳstage. The expression of HIF-1α in group of lymph node metastasis was higher than that in group without lymph node metastasis(P 0.05). Conclusion:CoCl2 can up-regulate the expression of HIF-1α in lung cancer cell lines by inducing hypoxia. These hypoxia-mediated effects have the potential impact on lung cancer cells to promote its hypoxic tolerance. HIF-1 overexpression plays important roles in invasion and metastasis of lung carcinoma. HIF-1 may be a useful marker for evaluating the biological behaviors of lung carcinoma.
The Expressions of EphB4 and Ephrinb2 in Lung Adenocarcinomas: a High Level of the EphB4 Protein is Associated with Lymph Node Metastasis.

Abstract:Eph tyrosine kinase receptors and their ephrin ligands are involved in normal development and tumorigenesis. Eph receptors are frequently over expressed in a wide variety of human malignant tumors, being associated with tumor growth, invasion, angiogenesis, and metastasis. This study aimed to evaluate the clinical significance of EphB4 and its ligand of ephrinB2 protein expressions in human lung adenocarcinomas. EphB4 and ephrinB2 protein expressions were assessed immunohistochemically on paraffin embedded tissues obtained from 93 patients with lung adenocarcinoma. Fifty-one out of 93 (54.8%) specimens were negative for EphB4 expression, and 42 out of 93 (45.2%) were positive for EphB4 expression. EphrinB2 expression was consistently negative in all tissues. EphB4 expression was significantly associated with tumor differentiation (P = 0.001), lymph node metastasis (P = 0.021), and Ki67 (P = 0.012). No significant relationship was found between EphB4 expression and gender, age, or ALK mutation. Our data show that EphB4 is differentially expressed in lung adenocarcinoma tissues. A high level of EphB4 protein expression was associated with lymph node metastasis in lung adenocarcinoma.
High Expression of EphA4 is Associated with Invasion and Lymph Node Metastasis in Colorectal Carcinomas.

Abstract:Eph receptors and ephrin ligands have been implicated in development of embryo, especially in vascular and nervous system. Increasing evidence suggests that Eph/ephrin signaling plays important roles in carcinogenesis. EphA4 is a member of Eph receptors and its role has been found in development of nerve and in tumorigenesis. Till now, EphA4 expression and its clinical significance in colorectal carcinoma have not been well investigated. In the present study, a set of 102 colorectal carcinoma tissue specimens were subjected to immunohistochemistry by using a specific polycolonal antibody for EphA4. Expression of EphA4 protein was more often detected in colorectal carcinoma than in normal mucosa (P<0.001). High expression of EphA4 protein in colorectal carcinomas was positively associated with tumor size (P=0.008, rs =0.260), depth of invasion (P=0.004, rs =0.286), lymph node metastasis (P=0.013, rs =0.244), and TNM stage (P=0.005, rs =0.279), and was negatively associated with age of patients (P=0.027, rs =-0.219). No significant relationship between expression of EphA4 and sex and tumor location was found. The survival analyses showed that patients whose carcinoma exhibited high expression of EphA4 had a poor outcome (P=0.006). Our results suggest that EphA4 play a role in the invasion and lymph node metastasis of colorectal carcinoma. The EphA4 protein expression level may be used as a potential prognostic marker in colorectal carcinoma. Furthermore, the EphA4 gene may provide a novel target for therapy of colorectal carcinoma.
Ephrin-A3 Promotes Hepatocellular Carcinoma Cell Proliferation and Metastasis by Interacting with EphA2

Abstract:Background: Emerging evidence suggests that Eph/ephrin-system involves different facets of tumorigenesis and cancer progression. Whether ephrin-A3, a cognate ligand to several Eph receptors, plays a role in HCC cell proliferation and metastasis remains unclear. Methods: The expression of ephrin-A3 was detected by immunohistochemistry and qRT-PCR in human HCC tissues. The clinical significance of ephrin-A3 was analyzed using multiple databases. The biological functions of ephrin-A3 were investigated in vitro and in vivo. Immunofluorescence and co-immunoprecipitation were used to detect the interaction between ephrin-A3 and EphA2. Findings: Elevated expression of ephrin-A3 was positively correlated with the histological grade, metastasis and poor prognosis in human HCC. Knockdown of ephrin-A3 led to significantly decreased proliferative activity and metastatic potential, and re-introduction of ephrin-A3 rescued the decreased proliferation, migration, and invasion induced by ephrin-A3 knockdown in HCC cells. Ephrin-A3 interacted with EphA2 and positively regulated EphA2 and AKT phosphorylation. Knockdown of EphA2 attenuated HCC cell proliferation, migration, and invasion induced by ephrin-A3 overexpression. Furthermore, ephrin-A3-induced EphA2 phosphorylation is both a substrate and a positive regulator of AKT. In HCC tissues, ephrin-A3 expression was positively correlated with EphA2, and patients with high co-expression of ephrin-A3 and EphA2 exhibited poorer prognosis. Interpretation: Ephrin-A3 promotes HCC metastasis by interacting with EphA2 and activating EphA2 and AKT. Thus, our study implicates ephrin-A3 and EphA2 as potential prognostic biomarkers, and provides valuable information for HCC prognosis and treatment. Abstract:Ezrin is known to regulate cellular survival, adhesion, migration, and invasion and has been identified as 1 of the key components of tumor progression and metastasis. The purpose of this study was to evaluate the clinicopathologic and prognostic significance of ezrin expression in non-small-cell lung cancer (NSCLC).We investigated the expression pattern of ezrin immunohistochemically in 89 paraffin samples of NSCLC between January 1998 and December 2006 and conducted survival analyses. In addition, 73 frozen specimens (including tumorous and precancerous tissues) of NSCLC and 28 frozen specimens of benign pneumonic diseases collected between January 2009 and December 2009 were analyzed by Western blot and reverse transcriptase-polymerase chain reaction (RT-PCR).In 89 paraffin samples, ezrin was expressed in 40 cases, either in the cytoplasm or on the membrane. Ezrin-positive expression was significantly associated with increased tumor stage and lymph node (LN) metastasis. The positive rate of cytoplasm expression was significantly associated with LN metastasis. Importantly, ezrin-positive expression independently predicted inferior overall survival (OS) and disease-free survival (DFS). In 73 frozen specimens of NSCLC and 28 frozen specimens of benign pneumonic diseases, the ezrin mRNA, protein, and phospho-ezrin protein expressions in tumorous tissues were higher than they were in precancerous tissues and benign pneumonic tissues.These results suggested that high-level ezrin expression contributed to NSCLC progression and that phosphorylation and subcellular translocation of ezrin might be the important mechanisms. Ezrin might be a potential prognostic marker of progression in NSCLC.

Abstract:Objective To evaluate the expression of HIF-1α in breast cancer and the influence to prognosis.Methods The expression of HIF-1α was assayed by immunohistochemistry on tissue specimens from 112 patients with breast cancer,then analysis the relationship of HIF-1α with VEGF,clinical and pathological features,and also the influence of the prognosis.Results HIF-1α positive expression was observed in 62.5% patients,and the 5-year survival rate in patients with HIF-1α positive expression was 54.3%,significant lower than HIF-1α negative breast cancer patients(85.7%,P = 0.001).HIF-1α expression was associated with tumor size,TNM stage,histological grade,axillary lymph node metastasis and VEGF positive expression.Conclusions HIF-1α promote the expression of VEGF,HIF-1α positive patients had worse prognosis and shorter survival time,HIF-1α may become the indicator to assess the prognosis of breast cancer.

Abstract:Vascular endothelial growth factor-C(VEGF-C) plays a critical role in tumor-induced lymphangiogenesis and contributes to lymph node metastasis.Human antigen R(HuR) is one of the firstly identified RNA-binding proteins.It can increase the stability of a variety of growth factors and cytokines and upregulate protein expression.The aim of this study is to investigate the expression of HuR and VEGF-C protein in non-small cell lung cancer(NSCLC),and explore the relationship between the expression of HuR and VEGF-C and clinicopathological factors.HuR and VEGF-C protein levels were detected in 81 NSCLC tissues and 15 control benign pulmonary lesion tissues by immunohistochemistry method(SP method).In NSCLC tissues,positive rate of cytoplasmic HuR,nuclear HuR and VEGF-C was 45.7%(37/81),82.7%(67/81) and 70.4%(57/81),respectively.There was a significant difference in positive expression of HuR and VEGF-C between NSCLC and benign pulmonary lesion tissues(P < 0.05).The expression of cytoplasmic HuR was closely related to pTNM stages,differentiation degree and lymph node metastasis(P < 0.05),but not correlated with sex,age and histological classification(P > 0.05).Furthermore,cytoplasmic immunoreactivity for HuR protein(P < 0.05) but not nuclear HuR expression(P > 0.05) was associated with high VEGF-C expression.Cytoplasmic HuR and VEGF-C are overexpressed in NSCLC,and are related to tumor development.HuR may mediate the modulation of VEGF-C gene expression in NSCLC.
Expression of Glucose Transporter in Non-Small Cell Lung Carcinoma and Its Clinical Significance

Abstract:OBJECTIVE:To investigate the expression of glucose transporter (Glut)1, Glut3, and hypoxia inducible factor (HIF)-1 alpha in human non-small cell lung carcinoma (NSCLC), and its clinical significance. METHODS:Specimens of cancer tissues and paracancerous lung tissues from 34 cases of NSCLC and 17 specimens of benign lung lesions were collected. The expressions of Glut1, Glut3, and HIF-1 alpha were detected with immunohistochemical staining, RT-PCR, and Western blot. RESULT:The relative mRNA expressions of Glut1 and HIF-1 alpha were 0.689 +/-0.245, 0.693 +/-0.248 in cancer tissues; and 0.338 +/-0.157, 0.351 +/-0.184 in paracancerous lung tissues (P <0.001); while those of Glut3 were 0.506 +/-0.246 in cancer tissues and 0.482 +/-0.238 in paracancerous tissues (P >0.05). The relative protein expressions of Glut1 and HIF-1 alpha were 0.582 +/-0.247, 0.525 +/-0.246 in cancer tissues and 0.288 +/-0.151, 0.261 +/-0.135 in paracancerous lung tissues (P<0.001), but the protein expressions of Glut3 were 0.551 +/-0.251 and 0.436 +/-0.224 respectively (P>0.05). Glut1 and HIF-1 alpha expressions were higher in poor differentiation group and in stage III group, than those in medium and well differentiation group and stage I and II group. Moreover, there was a significant correlation between the expression of Glut1 and HIF-1 alpha (r=0.854, P<0.01). CONCLUSION:Glut1 and HIF-1 alpha are highly expressed in NSCLC, and their expressions are associated with tumor differentiation and clinical stage.

Abstract:Objective To investigate the relationship between the expression of vascular endothelial growth factor (VEGF), its receptors FLT-1, FLK-1/KDR and tumor angiogenesis, clinical staging, lymph node metastasis in non-small cell lung cancer (NSCLC). Methods Immunohistochemical staining and enzyme-linked immunosorbent assay (ELISA) were used to detect the levels of VEGF and its receptors FLT-1,FLK-1/KDR in the tissue and serum of 112 patients with NSCLC, and analysed their clinical significances. Results The expression of VEGF and its receptors FLT-1, FLK/KDR in lung tissue of NSCLC were significantly higher than that in normal alveolar tissue and paracancerous tissue (P<0.05) and they increased significantly in proportion with clinical stages Ⅰ, Ⅱ, Ⅲ-Ⅳ (P<0.05). The positive rates of VEGF and its receptors FLT-1, FLK-1/KDR in lymph node metastasis group were significantly higher than those without lymph node metastasis group (P<0.05). Se- rum VEGF in NSCLC was significantly higher than that in the control group and also increased according to clinical staging (Ⅰ, Ⅱ, Ⅲ-Ⅳ; P<0.05). Serum VEGF was significantly higher in the lymph node metastasis group than that of the absence group (P<0.05) . Conclusions The expression of VEGF and its receptors FLT-1, FLK/KDR in NSCLC increased significantly, and positively correlated with tumor development, progression and lymph node metastasis. Serum VEGF is an important marker of hyperplasia and metastasis in NSCLC.
Expression of SRF, E-cadherin and β-catenin in hepatocellular carcinoma and its clinical significances

Abstract:Objective: To investigate the expression of serum response factor(SRF),E-cadherin and βcatenin in hepatocellular carcinoma and its relation to clinicopathological characterisitics of the tumor and prognosis.Methods: Tissue microarray and immunohistochemical study(EnVision method) for SRF,E-cadherin and βcatenin was performed on tissue microarray which consisted of 182 cases of hepatocellular carcinoma,92 cases of adjacent non-neoplastic liver tissues.Results: In the hepatocellular carcinoma group 75.8% tumor samples expressed SRF,and the levels is significantly higher than that in surrounding liver tissue(21.7%,P<0.05).The expression of SRF was positively correlated with the TNM stage,tumor metastasis and recurrence(P<0.05).In the hepatocellular carcinoma group,aberrant expression of E-cadherin and β-catenin was 59.3% and 58.8% respectively,and these levels were higher than those in the adjacent non-neoplastic liver tissue group(30.4% and 26.1% P<0.05).Aberrant expression of E-cadherin andβ-catenin was related to vascular cancer embolus,the TNM stage,tumor metastasis and recurrence(P<0.05).The expression of SRF correlated with aberrant expression of E-cadherin and β-catenin in hepatocellular carcinoma.Conclusion: In hepatocellular carcinoma,the overexpression of SRF and the downregulated expression of E-cadherin and β-catenin may be important mechanisms for promoting the invasion and metastasis of hepatocellular carcinoma and may also play a key role in the occurrence and development of hepatocellular carcinoma.
Comprehensive analysis of Epha10 as a predictor of clinical prognosis and immune checkpoint therapy efficacy in non-small cell lung cancer

Abstract:The EphA family belongs to a large group of membrane receptor tyrosine kinases. Emerging evidence indicates that the EphA family participates in tumour occurrence and progression. Nonetheless, the expression patterns and prognostic values of the nine EphAs in non-small cell lung cancer (NSCLC) have rarely been studied before. In the current study, we comprehensively analysed the expression and prognostic role of EphA family members by different means. The Cancer Genome Atlas and Gene Expression Profiling Interactive Analysis databases were used to investigate the expression of EphAs in NSCLC. The cBioPortal database was applied to analyse the prognostic values and genetic mutations of EphAs.We discovered that the expression of EphA10 was significantly higher in NSCLC tissues than in adjacent noncancerous tissues, and survival analyses showed that a higher level of EphA10 predicted poor prognosis. Further exploration into the role of EphA10 by ESTIMATE, CIBERSORT, and ssGSEA analysis found that it was also related to immune infiltration and higher expression of targets of ICI targets. In conclusion, this study revealed that among the EphA family members, EphA10 played an oncogenic role and was a promising biomarker for poor prognosis and better immunotherapy response in NSCLC.
The expression of HIF-1α in primary hepatocellular carcinoma and its correlation with radiotherapy response and clinical outcome

Abstract:The purpose of this study was to evaluate the relationship between hypoxia-inducible factor-1α (HIF-1α) protein expression in hepatocellular carcinoma (HCC), and responses of abdominal metastatic lymph nodes (LNs) from HCC patients treated with external beam radiotherapy (EBRT). HIF-1α immunohistochemical staining was performed on tissue microarrays (TMAs) of primary HCC specimens from 69 HCC patients with abdominal LN metastases. All patients received abdominal metastatic LN EBRT at the Department of Radiation Oncology at Zhongshan Hospital. A receiver-operating characteristic (ROC)-based approach and logistical regression analysis were used to determine the predictive value of HIF-1α expression in primary tumors with HCC metastatic LN EBRT response. Kaplan–Meier curves and log-rank tests were used to analyze patient survival. Cox proportional hazards regression model was used to analyze independent prognostic factors. HIF-1α expression was correlated with blood hemoglobin (Hb: r = −0.280, P = 0.020), response of abdominal metastatic LNs to EBRT ( r = 0.286, P = 0.017), locoregional recurrence ( r = 0.278, P = 0.021), and cancer-specific deaths ( r = 0.298, P = 0.013). HIF-1α expression was predictive of EBRT response of metastatic LNs [area under the curve (AUC): 0.646; 95% confidence interval (CI): 0.499–0.793; P = 0.047], locoregional recurrence (AUC: 0.657; 95% CI: 0.509–0.805; P = 0.049) and cancer-specific deaths (AUC: 0.671; 95% CI: 0.531–0.812; P = 0.035). Patients with tumors exhibiting high HIF-1α expression had significantly poorer overall survival (OS) than those with low tumor expression of HIF-1α ( P = 0.016). Multivariate analysis showed that Hb ( P = 0.035), vascular invasion ( P = 0.026), Child-Pugh score ( P < 0.001), intrahepatic tumor control ( P < 0.001), and HIF-1α ( P = 0.020) were independent prognosis factors for OS of HCC patients after receiving abdominal metastatic LN EBRT. HIF-1α expression in primary HCCs was associated with EBRT response of abdominal metastatic LNs and poor prognosis.
EphB 4 Gene Polymorphism and Protein Expression in Non-Small-cell Lung Cancer

Abstract:The objective of this study was to identify new diagnostic, prognostic or therapeutic molecules for non-small-cell lung cancer (NSCLC). We investigated the expression of EphB4, a tyrosine kinase receptor which has been shown to act as a tumor promoter in other cancers. Using immunohistochem-istry, we visualized EphB4 expression in 28 samples of NSCLC and 12 samples of adjacent normal tissues. Additionally, we assessed a single-nucleotide polymorphism in EphB4 to determine its effect on protein expression. The correlation of both genotype and protein expression with disease severity was determined. EphB4 was expressed in 53.6% of patients with lung cancer, a significant increase compared to control lung samples (0.0%, P<0.05). Furthermore, EphB4 expression was correlated with differentiation, lymph node metastasis and TNM stage of tumors (P<0.05). Additionally, the polymorphism in EphB4 at rs314310 appeared to correspond to protein expression and disease susceptibility. While the frequencies of CC, CA and AA genotypes were not different between lung cancer patients and healthy controls, the frequencies of C and A alleles were significantly different between these groups (P<0.05). Further analysis showed that the positive rate of EphB4 expression in patients with the AA genotype was significantly higher compared to that in patients with other genotypes (P<0.05). Overexpression of EphB4 plays a role in the occurrence and development of NSCLC, and the polymorphism at rs314310 may predispose individuals to this disease.

Abstract:Objective: Explore and discuss the expression of hypoxia-inducible factor-1α( HIF-1α) and vascular endothelial growth factor( VEGF) in hepatocellular carcinoma as well as their correlations with tumor angiogenesis. The anti-human mouse CD31 monoclonal antibody was adopted to mark the tumor vascular endothelial cells and assess the micro-vessel density( MVD). Results: The positive expression rates of HIF-1α and VEGF were 82. 9%( 34 /41),37. 5%( 6 /16),80. 5%( 33 /41) and 25. 0%( 4 /16) and the MVD values were 41. 0 ± 20. 2 and 20. 1 ± 9. 1 in HCC tissues and adjacent noncancerous tissues respectively; the differences between the two groups were statistically significant for all three variables( P 0. 05). The expression levels of HIF-1α,VEGF and the MVD value are correlated with each other in HCC tissues. Moreover,HIF-1α and VEGF expression,as well as the MVD value,were correlated with the pathological degree of HCC. Conclusion: HIF-1α and VEGF are frequently over expressed in HCC,and play an important role in tumor angiogenesis.

Abstract:Background: Erythropoietin-producing human hepatocellular (Eph) receptors stand out as the most expansive group of receptor tyrosine kinases (RTKs). Accumulating evidence suggests that within this expansive family, the EphA subset is implicated in driving cancer cell progression, proliferation, invasion, and metastasis, making it a promising target for anticancer treatment. Nonetheless, the extent of EphA family involvement across diverse cancers, along with its intricate interplay with immunity and the tumor microenvironment (TME), remains to be fully illuminated. Methods: The relationships between EphA gene expression and patient survival, immunological subtypes, and TME characteristics were investigated based on The Cancer Genome Atlas (TCGA) database. The analyses employed various R packages. Results: A significant difference in expression was identified for most EphA genes when comparing cancer tissues and non-cancer tissues. These genes independently functioned as prognostic factors spanning multiple cancer types. Moreover, a significant correlation surfaced between EphA gene expression and immune subtypes, except for EphA5, EphA6, and EphA8. EphA3 independently influenced the prognosis of papillary renal cell carcinoma (KIRP). This particular gene exhibited links with immune infiltration subtypes and clinicopathologic parameters, holding promise as a valuable biomarker for predicting prognosis and responsiveness to immunotherapy in patients with KIRP. Conclusion: By meticulously scrutinizing the panorama of EphA genes in a spectrum of cancers, this study supplemented a complete map of the effect of EphA family in Pan-cancer and suggested that EphA family may be a potential target for cancer therapy.

Abstract:Background: Eukaryotic initiation factor 4E (eIF4E), an important regulator of translation, plays important roles in tumor transformation, progression and metastasis. However, the clinical significance of eIF4E expression in lung adenocarcinoma (AdC) remains unclear. The aim of this study was to explore the expression of eIF4E gene in lung adenocarcinoma cell lines and tissues, and to investigate its relationship with clinical characteristics and prognosis of patients with lung adenocarcinoma in combination with p53 status.Methods: Semi-quantitative RT-PCR and Western blotting assays were performed to detect the expression of eIF4E mRNA and protein in normal human lung epithelial cell line, immortalized lung epithelial cell line and lung adenocarcinoma cell lines. Additionally, the expression of eIF4E gene was also detected in 32 cases of lung adenocarcinoma tissues, tumor adjacent tissues and tumor surrounding normal tissues by the same methods. Moreover, expression of eIF4E and the status of p53 in specimens from 76 patients with lung adenocarcinoma were examined by immunohistochemical staining. Correlations between eIF4E expression and clinicopathological features, and the effect of eIF4E on prognosis of patients with lung adenocarcinoma were evaluated by statistical analysis.Results: The levels of eIF4E mRNA and protein expression were higher in lung adenocarcinoma cell lines and in telomerase-immortalized lung epithelial cell line than in the normal lung epithelial cell line. The expression of eIF4E gene showed statistical difference between tumor tissues, tumor adjacent tissues and tumor surrounding normal tissues (P < 0.05). Moreover, the higher levels of eIF4E expression were correlated with poorer differentiation (P=0.012), higher pathological stage (P<0.0001) and clinical stage (P=0.002), a higher incidence of hematogenous metastasis (P=0.007) and cancer-related death (P=0.036). The 5-year survival rate of patients with higher eIF4E expression was significantly lower than that of patients with lower eIF4E expression (P=0.0045). Furthermore, in a multivariate analysis by Cox regression model, high eIF4E expression was confirmed to be an independent prognostic factor (HR: 2.258; unfavorable, P=0.0056), while lymph node (HR: 2.033; unfavorable, P=0.0440) and hematogenous metastasis (HR: 3.489; unfavorable, P<0.0001) were also significant prognostic factors.Conclusion: High eIF4E expression was correlated with poorer overall survival in lung adenocarcinoma patients. eIF4E might be a better clinical marker predicting the prognosis for lung adenocarcinoma patients in combination with p53 status. Abstract:Objective To investigate the expression of EphA1 in colorectal cancers and its clinical significance.Methods Quantitative real-time RT-PCR and immunohistostaining were employed to detect the expression level of EphA1 in colon cancer cell lines and CRC specimens.Methylation specific PCR (MSP) was also performed.Results The expression of EphA1 transcript,methylated DNA of EphA1 was detected in cell lines.EphA1 mRNA expression in the corresponding normal mucosae is not significantly different from that in tumor tissues (P=0.496).In adenomas,the immunostaining for EphA1 was positive diffusely and strongly.However,in CRCs,the expression of EphA1 protein was predominately down-regulated.Reduced or lost expression of the EphA1 protein was significantly associated with poor differentiation (P=0.027),greater depth of wall invasion (P=0.002) and advanced TNM stage (P=0.002),and preferentially occurred in patients with lymph node metastasis (P= 0.019 ).Patients with reduced EphA1 expression had a tendency of poorer prognosis than those with EphA1 upregulation.Concolusion EphA1 may have tumor suppressor roles in CRCs and may be an important biomarker by which malignancy,biological behavior and prognosis are evaluated.

Abstract:Lung adenocarcinoma (LUAD) remains a challenge within the realm of non-small cell lung cancer (NSCLC), demanding innovative diagnostic and therapeutic solutions. In this study, we systematically detected the correlation between the expression of hypoxia-induced factor 1A (HIF1A) and the clinical characteristics of LUAD, alongside lung squamous cell carcinoma (LUSC). Our bioinformatic analysis reveals that HIF1A mRNA expression is significantly upregulated in both LUAD and LUSC samples compared to non-tumorous lung tissues. The overexpression is positively correlated with increased copy number variation and negatively associated with promoter methylation. However, meta-analysis and survival analyses revealed a pronounced association between elevated HIF1A expression and poor clinical outcome specifically within the LUAD subset, with no such correlation evident in LUSC. Additionally, we explored the interplay between HIF1A expression, leukocyte infiltration, and the presence of immunosuppressive markers, revealing HIF1A's suppressive role in cytotoxicity against cancer cells. Furthermore, we performed in silico prediction to explore the correlations between HIF1A and its interacting proteins, associated pathways, glycolysis, and m6A modification, and the feasibility of targeting HIF1A with specific drugs. In summary, our study revealed the prognostic significance and therapeutic potential of HIF1A in LUAD.
EZH2 regulates PD-L1 expression via HIF-1α in non-small cell lung cancer cells

Abstract:Lung cancer is the most commonly diagnosed cancer and accounts for most cancer-related mortalities worldwide. The high expression of programmed death ligand 1 (PD-L1) is an important factor that promotes immune escape of lung cancer, thus aggravates chemotherapy resistance and poor prognosis. Therefore, understanding the regulatory mechanism of PD-L1 in lung cancer is critical for tumor immunotherapy. Enhancer of Zeste homolog2 (EZH2), an epigenetic regulatory molecule with histone methyltransferase activity, promotes the formation of an immunosuppressive microenvironment. This study aimed to investigate the role of EZH2 in PD-L1 expression and in the progression of lung tumors. We found that EZH2 was upregulated in lung cancer tissues and positively correlated with PD-L1 levels and poor prognosis. Further, shRNA-expressing lentivirus mediated EZH2 knockdown suppressed both the mRNA and protein expression level of PD-L1, thus delaying lung cancer progression in vivo by enhancing anti-tumor immune responses. Moreover, the regulatory effect of EZH2 on PD-L1 depended on HIF-1α. The present results indicate that EZH2 regulates the immunosuppressive molecule PD-L1 expression via HIF-1α in non-small cell lung cancer cells.
