Isolation and Identification of a Novel cDNA that Encodes human Phosphopantethein Binding Protein

Abstract:In a suppression subtractive hybridization experiment,attempted to reveal the character differences of adhesion and apoptosis between AD293 and HEK293,a cDNA fragment was isolated from an AD293 high expression library,which encodes a putative human phosphopantetheine binding protein(hPPB) inferred by electronic annotation.The 2745bp full-length cDNA was isolated from human fetal brain library,which encodes a protein of 518 amino acids.The protein contains an apoptosis inhibitory protein 5(API5) domain,an involucrin repeat,and a copper binding octapeptide repeat.Blast alignment against human genomic database revealed that the gene contains eight exons,and is assigned to human chromosome 2p22.3.A multiple tissue cDNA based RT-PCR showed that hPPB is ubiquitously expressed in human normal tissues and human tumor tissues.The transcript of the PPB gene is highly conserved during evolution.
