Effects of proadrenomedullin N-terminal 20 peptide on the proliferation and collagen synthesis in vascular smooth muscle cells induced by angiotensinⅡ

Abstract:AIM: To observe the effect of proadrenomedullin N-terminal 20 peptide on the proliferation and collagen synthesis in vascular smooth muscle cells (VSMCs) induced by angiotensinⅡ. METHODS: This experiment was carried out at Central Hospital, Zhujiang Hospital, Southern Medical University from September 2003 to September 2004. ① VSMCs of rats were cultured with tissue adhere method. ② Grouping: blank control group; angiotensinⅡ groups of 1×10-9,1×10-8,1×10-7,1×10-6 mol/L; proadrenomedullin N-terminal 20 peptide groups of 1×10-9,1× 10-8,1×10-7,1×10-6 mol/L; groups of 10-7 mol/L angiotensinⅡ + 1×10-9,1× 10-8,1×10-7,1×10-6 mol/L proadrenomedullin N-terminal 20 peptide . ③ The collagen synthesis of VSMCs was measured with the incorporation of 3H-proline , cell proliferation was measured with methylthiazolyl tetrazolium (MTT) assay . Effects of different concentrations of proadrenomedullin N-terminal 20 peptide and angiotensinⅡ on the proliferation of VSMCs and collagen synthesis were observed respectively. RESULTS: ① Effect of proadrenomedullin N-terminal 20 peptide and angiotensinⅡ on collagen synthesis: There was no significant difference of incorporation rate of 3H-proline between proadrenomedullin N-terminal 20 peptide group and control group (P > 0.05). With the increase of concentration of angiotensinⅡ, the incorporation rate of 3H-proline of VSMCs presented decreasing tendency. In the angiotensinⅡ + proadrenomedullin N-terminal 20 peptide group, when the concentration of angiotensinⅡ kept the same, the incorporation rate of 3H-proline of VSMCs presented decreasing tendency with the concentration increase of proadrenomedullin N-terminal 20 peptide. There was significant difference among groups (P < 0.01). ②Effects of proadrenomedullin N-terminal 20 peptide and angiotensinⅡ on the proliferation of VSMCs: With the increase of concentration of proadrenomedullin N-terminal 20 peptide, A490 values of proadrenomedullin N-terminal 20 peptide groups were not changed significantly (all P > 0.05). AngiotensinⅡ increased A490 values in VSMCs by concentration-dependent manner (P < 0.01). There was significant difference among groups (all P < 0.01). In the group of 10-7 mol/L angiotensinⅡ + 1×10-9,1× 10-8,1×10-7,1×10-6 mol/L proadrenomedullin N-terminal 20 peptide, when the concentration of angiotensinⅡ kept the same, A490 values were significantly decreased with the concentration of proadrenomedullin N-terminal 20 peptide increasing (P < 0.01). CONCLUSION: Proadrenomedullin N-terminal 20 peptide obviously inhibits the proliferation and collagen synthesis in VSMCs induced by angiotensinⅡ.
