Abstract:Endothelial progenitor cells (EPCs) improve neovascularization and endothelium regeneration. Transplantation with EPCs is a therapeutic strategy for the treatment of ischemic diseases. However, the transplanted EPCs are susceptible to adverse environments such as hypoxia, inflammation and oxidative stress. Oxidative stress-induced apoptosis of transplanted EPCs greatly reduces their therapeutic efficacy. Lipopolysaccharide (LPS) is a highly immunogenic antigen. Recent findings suggest that low dose of LPS pretreatment has protective effect against apoptosis. In this study, the role of LPS in apoptosis of EPCs was investigated. Pretreatment with 1µg/ml LPS prevented oxidative stress-induced EPCs apoptosis and ROS generation, which effects were abolished by TAK-242, a specific TLR4 antagonist. Further investigation of the mechanisms demonstrated that the activation was mediated by TLR4, and that PI3K/Akt/ NF-κB p65 signaling pathway may play a critical role in the process.
Hypoxia Confers Protection Against Apoptosis Via the PI3K/Akt Pathway in Endothelial Progenitor Cells.

Abstract:AIM:The recruitment of endothelial progenitor cells (EPC) to ischemia has recently been suggested as an important mechanism of tissue repair. Although tissue ischemia can facilitate EPC mobilization, recruitment, and retention at the hypoxic site, the effects of hypoxia on EPC survival are not well known. In the present study, we examined whether hypoxia (2% O2) would suppress apoptosis induced by serum withdrawal and whether survival signals, such as the phosphatidylinositol 3-kinase (PI3K)/Akt and extracellular signal-regulated protein kinase (ERK) pathways, were involved in this process.METHODS:After being serum-starved for 24 h, EPC were cultured under normoxic or hypoxic conditions (2% O2) for 24 h. Cell survival was assessed by 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, annexin V-propidium iodide dual-color flow cytometry, and terminal deoxynucleotidyl transferase-mediated digoxigenin-dUTP nick-end labeling assay. The expressions of signaling proteins were evaluated by Western blot analysis.RESULTS:Under hypoxic conditions, EPC were resistant to apoptosis induced by serum starvation. The inhibition of the PI3K/Akt pathway using the LY294002 inhibitor prevented hypoxia-inhibited apoptosis in EPC and altered the phosphorylation state of glycogen synthase kinase-3beta, an effector protein involved in regulation of EPC apoptosis. However, ERK inhibitor PD98059 had no significant effect on cell survival.CONCLUSION:Our data demonstrated that hypoxia inhibited serum withdrawal-induced apoptosis in EPC, which might be associated with the activation of the PI3K/Akt pathway.
Tongxinluo Inhibits Palmitic Acid-Induced Apoptosis of Human Endothelial Progenitor Cells by Down——regulating Expression and Activity of Caspase-3 Protein

Abstract:AIM:To explore the effect of tongxinluo(TXL)on apoptosis,expression and activity of caspase-3 in palmitic acid(PA)-induced peripheral blood-derived human endothelial progenitor cells in vitro.METHODS:Total mononuclear cells were isolated from peripheral blood in vitro by Ficoll density gradient centrifugation and plated on fibronectin-coated culture dishes.After 7 days,the attached cells were divided into seven groups:one control group(normal cells),three PA groups [attached cells were incubated with different concentrations of PA(200,400 and 800 mol/L)for 48 h] and three TXL groups [attached cells were incubated with different concentrations of TXL(100,200 and 400 mol/L),respectively,for 2 h before incubation with 400 μmol/L PA for 48 h].Flow cytometry was used to detect apoptosis.Expression and activity of caspase-3 protein were detected by Western blot and spectrophotometry,respectively.RESULTS:After exposure to PA,the apoptosis rate in treatment groups was dose-dependently higher than in the control group(P0.05).TXL was added at different concentrations before PA incubation and cells in the TXL groups(200,400 mol/L)showed lower apoptosis rate than in PA groups(P0.05).Expression of caspse-3 protein and activity of EPCs in TXL groups was lower than in PA groups(P0.05).CONCLUSION:PA promotes cell apoptosis by up-regulating expression and activity of caspase-3.TXL partly inhibits the effects of PA.
Insulin-like Growth Factor-1 Receptor Activation Prevents Hydrogen Peroxide-Induced Oxidative Stress, Mitochondrial Dysfunction and Apoptosis

Abstract:Vascular disease is the leading cause of morbidity and mortality. Oxidative stress can cause endothelial cell apoptosis. Low insulin like growth factor-1 (IGF-1) has been linked to adverse risk profile and increased vascular disease incidence. Since IGF-1 acts as an important survival factor for multiple cell types, we undertook this study to investigate whether IGF-1 favorably affects oxidative-stress mediated apoptosis of vascular endothelial cells. Exposure to hydrogen peroxide induced apoptotic changes (e.g. DNA fragmentation, altered mitochondrial membrane potential and caspase-3 activity) in human umbilical vein endothelial cells (HUVECs) in a time dependent manner. Addition of IGF-1 blocked the oxidative-stress effect parallel to IGF-1 receptor (IGF-1R) expression, and silencing the IGF-1R with small interference RNA attenuated the IGF-1 influence. Our findings show that enhanced IGF-1 signaling inhibits oxidative-stress induced apoptosis in HUVECs by reducing mitochondrial dysfunction. Specifically the protective mechanism of IGF-1 involves preserving the mitochondrial membrane potential, maintaining the mitochondrial retention of cytochrome-c, and reducing caspase-3 activity. These results may have therapeutic implications in preventing/reducing vascular disease associated endothelial dysfunction.
A redox-sensitive pathway mediates oxidized LDL-induced downregulation of insulin-like growth factor-1 receptor

Abstract:Oxidized low density lipoprotein (OxLDL) has multiple proatherogenic effects, including induction of apoptosis. We have recently shown that OxLDL markedly downregulates insulin-like growth factor-1 receptor (IGF-1R) in human aortic smooth muscle cells, and that IGF-1R overexpression blocks OxLDL-induced apoptosis. We hypothesized that specific OxLDL-triggered signaling events led to IGF-1R downregulation and apoptosis. We examined OxLDL signaling pathways and found that neither IGF-1R downregulation nor the proapoptotic effect was blocked by inhibition of OxLDL-triggered extracellular signal-regulated kinase, p38 mitogen-activated protein kinase (MAPK), or peroxisome proliferator-activated receptor gamma ( PPAR gamma) signaling pathways, as assessed using specific inhibitors. However, antioxidants, polyethylene glycol catalase, superoxide dismutase, and Trolox completely blocked OxLDL downregulation of IGF-1R and OxLDL-induced apoptosis. Nordihydroguaiaretic acid, AA-861, and baicalein, which are lipoxygenase inhibitors and also have antioxidant activity, blocked IGF-1R downregulation and apoptosis as well as reactive oxygen species (ROS) production. These results suggest that OxLDL enhances ROS production possibly through lipoxygenase activity, leading to IGF-1R downregulation and apoptosis. Furthermore, anti-CD36 scavenger receptor antibody markedly inhibited OxLDL-induced IGF-1R downregulation and apoptosis as well as ROS production. In conclusion, our data demonstrate that OxLDL downregulates IGF-1R via redox-sensitive pathways that are distinct from OxLDL signaling through MAPK- and PPAR gamma-involved pathways but may involve a CD36-dependent mechanism.
SIRT1 Protects Against Oxidative Stress-Induced Endothelial Progenitor Cells Apoptosis by Inhibiting FOXO3a Via FOXO3a Ubiquitination and Degradation.

Abstract:Cell loss due to apoptosis induced by oxidative stress is a major hurdle for endothelial progenitor cells (EPCs)‐based therapy. Sirtuin 1 (SIRT1) plays important roles in many pathophysiological processes by deacetylating various substrates, including forkhead transcription factor (FOXO). However, after deacetylation, the fate of FOXO protein remains to be explored. In the present study, we investigated whether SIRT1 exerted a protective effect on hydrogen peroxide (H2O2)‐induced EPCs apoptosis and, if so, what the underlying mechanism might be. EPCs were isolated and obtained from human umbilical cord blood by density gradient centrifugation and identified by morphology, tube formation ability, cell surface markers, and the ability to take up acetylated low‐density lipoprotein (Dil‐Ac‐LDL) and bind ulex europaeus agglutinin 1 (FITC‐UEA‐1). Immunofluorescence showed that SIRT1 is localized in the nucleus of EPCs in the presence or absence of H2O2. SIRT1 protein level in EPCs was increased by the treatment with H2O2 for 24 h. Incubation of EPCs with H2O2 dose dependently induced EPCs apoptosis. SIRT1 overexpression reduced the rate of EPCs apoptosis induced by H2O2, whereas SIRT1 downregulation and EX527, a specific SIRT1 inhibitor, exerted the opposite effect. SIRT1 overexpression decreased the total FOXO3a protein expression, whereas SIRT1 downregulation and EX527 increased the amount of FOXO3a protein. SIRT1 reduced FOXO3a transcriptional activity according to Bim expression. Co‐immunoprecipitation assay showed that SIRT1 could bind to FOXO3a, reduce its acetylation level and increase its ubiquitination level. To sum up, our work demonstrated that SIRT1 had a pivotally protective role in the regulation of EPCs apoptosis induced by H2O2 and that SIRT1 protected against apoptosis by inhibiting FOXO3a via FOXO3a ubiquitination and subsequent degradation. IGF-1 Alleviates Ox-Ldl-induced Inflammation Via Reducing HMGB1 Release in HAECs.

Abstract:Atherosclerosis, a multifactorial chronic inflammatory response, is closely associated with oxidatively modified low-density lipoprotein (ox-LDL). High-mobility group box 1 (HMGB1) is a DNA-binding protein, which upon release from cells exhibits potent inflammatory action. Insulin-like growth factor 1 (IGF-1) can elicit a repertoire of cellular responses including proliferation and anti-apoptosis. However, the role of IGF-1 in inflammation is still unclear. In the present study, we aimed to investigate the role of IGF-1 in inflammation and the underlying mechanism. Human aortic endothelial cells were stimulated by ox-LDL (50 μg/ml) to induce inflammation. The expression of intercellular adhesion molecule 1 (ICAM-1) was assessed by western blot analysis and immunofluorescence. The release of HMGB1 was determined by enzyme-linked immunosorbent assay. IGF-1 receptor (IGF-1R) expression was assessed by reverse transcription-polymerase chain reaction and western blot analysis. IGF-1R phosphorylation was determined by western blot analysis. Ox-LDL stimulation reduced IGF-1R mRNA and protein expression but increased HMGB1 release. IGF-1 treatment decreased ox-LDL-induced ICAM-1 expression potentially through reducing HMGB1 release, while picropodophyllin, an IGF-1R specific inhibitor, increased the inflammatory response. In conclusion, IGF-1 can alleviate ox-LDL-induced inflammation by reducing HMGB1 release, suggesting an unexpected beneficial role of IGF-1 in inflammatory disease.
Role of Caspases in Ox-LDL–Induced Apoptotic Cascade in Human Coronary Artery Endothelial Cells

Abstract:Oxidized low-density lipoprotein (ox-LDL) induces apoptosis in endothelial cells. However, steps leading to ox-LDL–induced apoptosis remain unclear. We examined the role of ox-LDL and its newly described receptor LOX-1 in the expression of intracellular pro- and antiapoptotic proteins and caspase pathways in human coronary artery endothelial cells (HCAECs). Cells were cultured and treated with different concentrations (10 to 80 μg/mL) of ox-LDL for different times (2 to 24 hours). Ox-LDL induced apoptosis in HCAECs in a concentration- and time-dependent manner. Ox-LDL also activated caspase-9 and caspase-3, but not caspase-8. After ox-LDL treatment, there was a significant release of activators of caspase-9, including cytochrome c and Smac from mitochondria to cytoplasmic compartment, and their release was not affected by treatment of cells with inhibitors of either caspase-8 or caspase-9. Ox-LDL also decreased expression of antiapoptotic proteins Bcl-2 and c-IAP (inhibitory apoptotic protein)-1, which are involved in the release of cytochrome c and Smac and activation of caspase-9, in a concentration- and time-dependent manner. On the other hand, ox-LDL did not change the expression of Fas-associated death domain-like interleukin-1β–converting enzyme-inhibitory protein (FLIP) and proapoptotic protein Fas, which are required for the activation of caspase-8. Further, ox-LDL did not cause the truncation of Bid, which implies the activation of caspase-8. In other experiments, pretreatment of HCAECs with the caspase-9 inhibitor z-LEHD-fmk, but not the caspase-8 inhibitor z-IETD-fmk, blocked ox-LDL–induced activation of caspase-3 and apoptosis. As expected, pretreatment with the caspase-3 inhibitor DEVD-CHO inhibited ox-LDL–induced activation of caspase-3 and resultant apoptosis. The proapoptotic effects of ox-LDL were mediated by its receptor LOX-1, because pretreatment of HCAECs with antisense-LOX-1, but not sense-LOX-1, blocked these effects of ox-LDL. These findings suggest that ox-LDL through its receptor LOX-1 decreases the expression of antiapoptotic proteins Bcl-2 and c-IAP-1. This is followed by activation of apoptotic signaling pathway, involving release of cytochrome c and Smac and activation of caspase-9 and then caspase-3.

Abstract:Endothelial regeneration play an important role in restoring vascular tone in respone to vascular injury. And the increased level of angiotensin II (Ang II) is an important risk factor in endothelial injury, which has been considered to play a key role in the initiation and development of atherosclerosis. Endothelial progenitor cells (EPCs) have been shown to enhance the formation of a new endothelim and reendotheliazation. But in previous study, Ang II affects the apoptosis of the EPCs. However, the precise mechanism is still unclear. Therefore, we focused in this study to understand whether Ang II can induce the eNOS (and caspase-3 activity in the EPCs. EPCs were isolated from peripheral blood and characterized. The EPCs were cultured then were treated with Ang II for 48 h. The apoptosis of the endothelial cells was checked by the flow cytometry and, while caspase-3 activity and eNOS was measured simultaneously. After the EPC were treated, the mean of apoptotic rates and caspase-3 enzyme activity and eNOS significantly decreased. Ang II marked inducd a cell cycle at G0/G1 phase and cell apoptosis. and the Angiotension II type1 receptor mRNA is upregulation. therefore Ang II stimulated caspase-3 enzyme leading to apoptosis of the EPCs.
Hydrogen Peroxide Induced Impairment of Endothelial Progenitor Cell Viability is Mediated Through a FoxO3a Dependant Mechanism

Abstract:Objectives: Increased oxidative stress has been suggested to contribute to the functional impairment of endothelial progenitor cells (EPCs). The Forkhead box O transcription factors (FoxOs) are critical regulators involved in various cellular processes including cell apoptosis. Here, we investigated whether FoxOs are required in oxidative stress induced EPC apoptosis.Methods and results: EPCs were cultured from cord blood derived mononuclear cells and treated with hydrogen peroxide (H2O2) for induction of oxidative stress. Incubation with H2O2 dose dependently reduced viability and increased apoptosis in EPCs. Western blotting showed that EPCs predominantly expressed FoxO3a and the expression was markedly increased upon H2O2 treatment. Transduction with adenoviral vectors expressing either a wide-type or a non-phosphorylatable, constitutively active mutant of FoxO3a led to further increased apoptosis of EPCs after H2O2 treatment. Conversely, FoxO3a silencing rescued EPCs from these H2O2 induced deleterious effects. Overexpression of FoxO3a also increased the level of the pro-apoptotic protein Bim, whereas FoxO3a silencing downregulated H2O2 induced Bim expression. Furthermore, Matrigel assay demonstrated that FoxO3a overexpression significantly impaired the tube forming ability of EPCs, whereas its silencing completely protected EPCs from H2O2 induced decrease of capillary formation.Conclusions: These data suggest that oxidative stress induced impairment of EPC survival is mediated through a FoxO3a dependant mechanism, possibly by transcriptional regulation of Elm. Role of PERK/eIF2α/CHOP Endoplasmic Reticulum Stress Pathway in Oxidized Low-density Lipoprotein Mediated Induction of Endothelial Apoptosis.

Abstract:OBJECTIVE:PERK/eIF2α/CHOP is a major signaling pathway mediating endoplasmic reticulum (ER) stress related with atherosclerosis. Oxidized LDL (ox-LDL) also induces endothelial apoptosis and plays a vital role in the initiation and progression of atherosclerosis. The present study was conducted to explore the regulatory effect of ox-LDL on PERK/eIF2α/CHOP signaling pathway in vascular endothelial cells.METHODS:The effects of ox-LDL on PERK and p-eIF2α protein expression of primary human umbilical vein endothelial cells (HUVECs) were investigated by Western blot analysis. PERK gene silencing and selective eIF2α phosphatase inhibitor, salubrinal were used to inhibit the process of ox-LDL induced endothelial cell apoptosis, caspase-3 activity, and CHOP mRNA level.RESULTS:Ox-LDL treatment significantly increased the expression of PERK, PERK-mediated inactivation of eIF2α phosphorylation, and the expression of CHOP, as well as the caspase-3 activity and apoptosis. The effects of ox-LDL were markedly decreased by knocking down PERK with stable transduction of lentiviral shRNA or by selective eIF2α phosphatase inhibitor, salubrinal.CONCLUSION:This study provides the first evidence that ox-LDL induces apoptosis in vascular endothelial cells mediated largely via the PERK/eIF2α/CHOP ER-stress pathway. It adds new insights into the molecular mechanisms underlying the pathogenesis and progression of atherosclerosis.
ox-LDL-Induced Endothelial Progenitor Cell Oxidative Stress via p38/Keap1/Nrf2 Pathway

Abstract:Background. Nrf2 which was recently reported to regulate the antioxidant genes and cellular redox regulators was highly expressed in EPCs. However, its role in ox-LDL-induced EPC oxidative stress and apoptosis has not been fully illustrated. Methods. EPCs isolated from human peripheral blood mononuclear cells were treated with different concentrations of ox-LDL, Keap1 siRNA, and a specific p38 MAPK inhibitor SB203580 and then used to assay the cytoplasmic Nrf2, nuclear Nrf2, NAD(P) H:quinone oxidoreductase 1 (NQO1) and Bax/Bcl-2 levels with Western blot, NQO1 mRNA levels with RT-PCR, ROS levels with H2DCF-DA, loss/disruption of mitochondrial membrane potential with JC-1, apoptosis with Annexin V and PI, migration with transwell chambers, and tube formation with Matrigel. Results. ox-LDL decreased the nuclear Nrf2/Histone H3 to cytoplasmic Nrf2/GAPDH ratio, NQO1 mRNA, and protein levels. ox-LDL enhanced ROS production, induced the loss of membrane potential, and increased the cell shrinkage, pyknotic nuclei, and apoptosis of EPCs. Keap1 siRNA increased Nrf2 nuclear translocation, NQO1 mRNA transcription, and protein expression and prevented ROS generation and formation of JC-1 monomers. ox-LDL increased the activation of p38. SB203580 significantly eliminated ox-LDL induced inhibition of Nrf2 nuclear translocation, depression of NQO1 mRNA transcription, generation of ROS, and formation of JC-1 monomers in EPCs. Keap1 siRNA decreased the Bax/Bcl-2 ratio which was increased by ox-LDL in EPCs. ox-LDL decreased EPC migration and tube formation. Keap1 siRNA preserved the migration and tube formation of EPCs. Conclusion. ox-LDL activated EPCs p38/Keap1/Nrf2 pathway and induced oxidative stress, dysfunction, and apoptosis of EPCs.

Abstract:Background: Insulin-like growth factor-1 (IGF-1), a member of the insulin family, has a high degree of homology with insulin and exhibits anti-inflammatory and anti-oxidative stress properties. However, the potential protective effect of IGF-1 on hyperoxia-induced lung injury remains unknown. In this study, we aimed to explore the effects and mechanism of action of IGF-1 in hyperoxia-induced lung injury in neonatal rats. Materials and Methods: Hematoxylin-eosin staining was used to observe pathological changes in lung tissue; transmission electron microscopy was used to examine the ultrastructure, and ELISA was used to detect the level of pro-inflammatory cytokines in bronchoalveolar lavage fluid. Further, malondialdehyde, glutathione, and superoxide dismutase activities in lung tissue were evaluated. TUNEL staining was used to detect cell apoptosis, and western blot analysis was used to detect the expression of Bax, Bcl-2, Caspase-3, p-PERK, p-eIF2α, ATF4, and CHOP in the lung tissue. Moreover, the wet/dry weight ratio of lung tissue was determined. Results: Intraperitoneal injection of IGF-1 effectively reduced lung tissue damage induced by hyperoxia; production of inflammatory cells and release of pro-inflammatory cytokines, oxidative stress, and cell apoptosis. Further, IGF-1 down-regulated the expression of ATF4, CHOP, and Bax/Bcl-2, and inhibited the phosphorylation of PERK and eIF2α. Conclusion: The results suggest that IGF-1 reduces hyperoxia-induced lung inflammation and oxidative stress in neonatal rats through the PERK/eIF2α/ATF4/CHOP signaling pathway and inhibits cell apoptosis.

Abstract:Oxidative stress contributes to tissue injury and cell death during the development of various diseases. The present study aims at investigating whether oxidative stress triggered by the exposure to hydrogen peroxide (H2O2) can induce apoptosis of induced pluripotent stem cells (iPS cells) in a mechanism mediated by insulin-like growth factor (IGF-1) and microRNA-1 (miR-1). iPS cells treated with H2O2 showed increases in miR-1 expression, mitochondria dysfunction, cytochrome-c release and apoptosis, Addition of IGF-1 into the iPS cell cultures reduced the H2O2 cytotoxicity. Prediction algorithms showed that 3′-untranslated regions of IGF-1 gene as a target of miR-1. Moreover, miR-1 mimic, but not miR-1 mimic negative control, diminished the protective effect of IGF-1 on H2O2-induced mitochondrial dysfunction, cytochrome-c release and apoptosis in iPS cells. In conclusion, IGF-1 inhibits H2O2-induced mitochondrial dysfunction, cytochrome-c release and apoptosis. IGF-1′s effect is, at least partially, regulated by miR-1 in iPS cells.
Ox-LDL Induces Endothelial Cell Apoptosis Via the LOX-1-dependent Endoplasmic Reticulum Stress Pathway.

Abstract:Objective: To investigate the effect of lectin-like ox-LDL receptor-1 (LOX-1) on oxidized low-density lipoprotein (ox-LDL)-induced apoptosis and the involvement of the endoplasmic reticulum (ER) stress response pathway.Methods and results: Human umbilical vein endothelial cells were treated with 50, 100, or 200 mu g/ml ox-LDL and cultured for 12, 24, or 48 h for concentration- and time-dependent studies. Cells were transfected with LOX-1 or Nox-4 shRNAs, and target proteins were inhibited with the corresponding antibodies for mechanistic studies. Active proteins and mRNAs were analyzed by Western blotting and RTPCR, respectively. Cell apoptosis was analyzed by Annexin and Hoechst staining assays. Ox-LDL induced both apoptosis and protein expression of LOX-1 and Nox-4 through activation of ER stress sensors IRE1 and PERK, and nuclear translocation of ATF6 and their subsequent pathways were indicated by JNK, eukaryotic initiation factor 2 phosphorylation, XBP-1, and chaperone GRP78 expression; up-regulation of proapoptotic proteins CHOP and Bcl-2; and caspase-12 activity. LOX-1 gene silencing and treatment with an anti-LOX-1 antibody attenuated the effects of ox-LDL. Pretreatment with irestatin 9389, salubrinal, or AEBSF also blocked ox-LDL-induced expression of CHOP and Bcl-2 and activation of caspase-12 activity, leading to an attenuation of endothelial cell apoptosis. Furthermore, Nox-4 siRNA attenuated the up-regulated expression of GRP78, PERK, IRE1, and XBP-1 to reduce ox-LDL-induced endothelial cell apoptosis.Conclusions: LOX-1 plays a critical role in ox-LDL-induced endothelial cell apoptosis via the ER stress pathway. Abstract:Studies have shown that oxidized low density lipoprotein (ox-LDL) elicits both necrotic and apoptotic cell death and several mechanisms have been proposed. Ox-LDL induces reactive oxygen species (ROS), a second messenger that might be involved in apoptosis, formation in different types of cells including endothelial cells (ECs) and smooth muscle cells (SMCs). As lectin-like ox-LDL receptor-1 (LOX-1) was the main receptor for ox-LDL, this study was designed to determine whether the apoptosis induced by ox-LDL was mediated by LOX-1 in cultured human umbilical vein endothelial cells (HUVECs) and whether there is an association between LOX-1 mediated apoptosis and the production of ROS. After exposure to ox-LDL (50, 100, and 150 mu g/ml for 18 h), HUVECs exhibit typical apoptotic characteristics as determined by transmission electron microscopy and flow cytometry analysis in a dose-dependent pattern. Ox-LDL increases intracellular ROS formation including superoxide anion (O-2(-) ) and hydrogen peroxide (H2O2) in a dose-dependent and time-dependent manner. Pretreatment with anti-LOX-1 mAb, Vitamin C, apocynin or catalase significantly reduced ROS production and prevented ox-LDL-induced apoptosis, while indomethacin or allopurinol had no effect. These results suggest that LOX-1 mediates ox-LDL-induced apoptosis in endothelial cells and that ROS production and NADPH oxidase might play an important role in ox-LDL-induced apoptosis. Abstract:Icariin belongs to the family of flavonoids that is extracted from Epimedium brevicornum Maxim, and exhibits antioxidative, antitumorigenic, antiosteoporotic, immunoregulatory and antiatherosclerotic properties. To understand the mechanisms underlying the antiatherosclerotic properties of icariin, the present study investigated the effects of icariin on human vascular endothelial cells (HUVECs) following treatment with oxidized low‑density lipoprotein (ox‑LDL). Thus, following pretreatment with icariin at four various concentrations (0, 10, 20 and 40 µM), HUVECs were stimulated with ox‑LDL (100 µg/ml). The viability of cells was evaluated via an MTT assay and flow cytometry was performed to assess apoptosis. Additionally, the protein and mRNA expression levels of apoptosis regulator Bcl‑2 (Bcl‑2) and caspase‑3 were determined by western blotting and reverse transcription‑quantitative polymerase chain reaction. The findings of the present study indicated that icariin prevented injury and apoptosis in HUVECs following ox‑LDL treatment, in particular via the regulation of protein and mRNA expression levels of Bcl-2 and caspase-3.
Hxeroderma pigmentosum mediates the effect of ox-LDL on promoting the apoptosis of HUVEC

Abstract:Aim To explore whether the hxeroderma pigmentosum D(XPD) could mediate the apoptosis of human umbilical vein endothelial cells (HUVEC) promoted by oxidized low density lipoprotein (ox-LDL).Methods The model of apoptosis of HUVEC was established by ox-LDL.XPD-siRNA was transfected into HUVEC with liposome,followed by treatment with ox-LDL.This experiment was divided into six groups:blank control group;negative control siRNA group;XPD-siRNA group;ox-LDL group;ox-LDL+negative control siRNA group;ox-LDL+XPD-siRNA group.Cell vitality was detected by MTT;Cell apoptosis rate and cell cycle were assessed with flow cytometry;The expressions of the XPD,Bax and Bcl-2 were detected by RT-PCR and Western blot.Results The optimal concentration of ox-LDL to establish the model of apoptosis of HUVEC was 100 mg/L.Compared with negative control siRNA group,the cell apoptosis rate of XPD-siRNA group was significantly decreased (P＜0.05),the survival rate was significantly increased (P＜ 0.01),the cell number in G0/G1 phase decreased (P＜0.05),while increased in S phase (P＜0.05),the expressions of XPD and Bax were declined (all P＜0.05),while the expression of Bcl-2 was elevated (P＜0.05);Compared with blank control group,the cell apoptosis rate of ox-LDL group increased significantly (P＜0.01),the survival rate was decreased (P＜0.05),the cell number in G0/G1 phase increased (P＜0.05),while decreased significantly in S phase (P＜0.01),the expressions of XPD and Bax were elevated (all P＜0.05),while the expression of Bcl-2 was declined (P＜0.05).Compared with ox-LDL+negative control siRNA group,the cell apoptosis rate of ox-LDL+XPD-siRNA group was reduced (P＜0.05),the survival rate was increased (P＜0.01),the cell number in G0/G1 phase decreased (P＜0.05),while increased in S phase (P＜0.05),the expressions of XPD and Bax were declined (all P＜0.05),while the expression of Bcl-2 was elevated (P＜0.05).Conclusion XPD can mediate the effect of ox-LDL on promoting the apoptosis of HUVEC.
Irisin Attenuates Oxidized Low‐density Lipoprotein Impaired Angiogenesis Through AKT/mTOR/S6K1/Nrf2 Pathway

Abstract:It is known that irisin increases total body energy expenditure, decreases body weight, and enhances insulin sensitivity. Although previous studies have demonstrated that irisin induces vascular endothelial cell (EC) angiogenesis, the molecular mechanisms underlying irisin-induced angiogenesis under conditions reflecting atherosclerosis are not known. The aim of the present study is to investigate whether irisin could inhibit oxidized low-density lipoprotein (oxLDL) impaired angiogenesis. We investigated the effect of irisin on angiogenesis in vitro by evaluating cell viability, cell migration, and the capacity to form capillary-like tubes using human umbilical vein endothelial cells and human microvascular endothelial cells (HUVECs and HMEC-1) that were treated with oxLDL. We also evaluated the effects of irisin on angiogenesis in vivo by Matrigel plug angiogenesis assay and in a chicken embryo membrane (CAM) model. Our results demonstrated that irisin increased oxLDL-treated EC viability as well as migration and tube formation. Moreover, oxLDL inhibited angiogenic response in vivo, both in the Matrigel plug angiogenesis assay and in the CAM model, and was attenuated by irisin. Furthermore, irisin decreased apoptosis, inflammatory cytokines, and intracellular reactive oxygen species (ROS) levels in oxLDL-treated EC. In addition, we found that irisin upregulated pAkt/mTOR/Nrf2 in oxLDL-treated EC. Both mTOR/Nrf2 shRNA and LY294002 could inhibit the protective effect of irisin. Taken together these results, they suggested that irisin attenuates oxLDL-induced vascular injury by activating the Akt/mTOR/Nrf2 pathway. Our findings suggest that irisin attenuates oxLDL-induced blood vessel injury.
IGFBP7 mediates oxLDL-induced human vascular endothelial cell injury by suppressing the expression of SIRT1

Abstract:Endothelial cell injury plays an important role in initiating atherosclerotic lesion formation. Insulin-like growth factor binding protein 7 (IGFBP7) is known to modulate the behaviors of tumor-associated endothelial cells. This study was conducted to test whether IGFBP7 is involved in endothelial cell injury during atherosclerosis. Oxidized low-density lipoprotein (oxLDL) treatment was used to mimic atherosclerosis-related endothelial cell apoptosis and inflammation response. Small interfering RNA (siRNA) technology was employed to deplete IGFBP7 expression in human aortic endothelial cells (HAECs). HAECs were exposed to recombinant human IGFBP7 protein to evaluate the function of IGFBP7. Notably, IGFBP7 expression in HAECs was induced by oxLDL treatment. Knockdown of IGFBP7 or treatment with anti-IGFBP7 abolished oxLDL-induced apoptosis and inflammation in HAECs. Moreover, recombinant IGFBP7 (40 ng/mL but not 25 ng/mL) promoted apoptosis and inflammation in HAECs. IGFBP7 co-localized with CD93 on the surface of HAECs. A mechanistic investigation uncovered that IGFBP7 induced endothelial cell injury through interaction with CD93 and reduction of SIRT1 expression via an autocrine manner. Overexpression of SIRT1 rescued IGFBP7-induced phenotype in HAECs. Taken together, IGFBP7 is induced by oxLDL and mediates oxLDL-induced endothelial cell apoptosis and inflammation, likely through downregulation of SIRT1. These observations support a rationale to prevent atherosclerosis by targeting IGFBP7 activity.
