Abstract:Metal ions have a significant impact on the environment and the growth and development of organism,therefore,the recognition and detection of metal ions in the environment and organism is gaining widespread attention. Among different analytical methods,fluorescence analysis has the advantages of high sensitivity,good selectivity,real-time and in situ detection,etc.,which is a good tool to achieve the recognition and detection of metal ions in the environment and organism. This paper is aimed to review the coumarin-based fluorescent sensors for the recognition and detection of heavy metal ions (Hg2+、Pb2+、Cd2+、Ni2+、Ag+),biologically important transition metal ions (Cu2+、Zn2+、Fe3+) and their applications in recent five years,focusing on the design and synthesis of sensors, recognition mechanism,sensing characteristics and applications. With the higher requirement for metal ions detection , the design of coumarin-based fluorescent sensors will be developed for better performance with higher sensitivity,selectivity and stronger anti-interference in the future. In addition, applications of coumarin-based fluorescent sensors in biological detection are expected to be further developed.

Abstract:A new double Schiff base, carbazole, and diaminomaleonitrile-based chemosensor for Fe3+ with high sensitivity and selectivity was designed and synthesized. Addition of only Fe3+ ions induced the fluorescence quenching of the chemosensor in DMF solution. The chemosensor formed a 1:1 complex with Fe3+ with the high binding constant of 7.98·10–6 M–1. The chemosensor displayed low detection limit (3.75·10–8 M) and fast response time (15 s) to Fe3+.

Abstract:A novel coumarin-based fluorogenic probe bearing the tris unit (DAT-1) with high selectivity and suitable affinity toward Fe3+ over other cations tested was developed in biological systems. The sensing mechanism was studied by quantum calculations. The probe can be applied to the monitoring of Fe3+ in aqueous solution with a pH span of 3–8. In addition, biological imaging, membrane permeability and nontoxicity demonstrate that DAT-1 could act as a turn-off fluorescent chemosensor for Fe3+ in living cells.

Abstract:A novel coumarin-based fluorescent probe L ((4E)-4-((7-hydroxy-4-methyl-2-oxo-2H-chromen-8-yl) methyleneamino)-1,2-dihyydro-2,3-dimethyl-1-phenylpyrazol-5-one) has been developed as a simple and efficient chemosensor which exhibits a significant fluorescence reduction in the presence of metal cations. This sensor exhibits high selectivity and sensitivity toward Cu2+ over other common cations. The mechanism for detecting copper was evaluated by time-dependent density functional theory (TD-DFT) calculations and the coordination mode was also confirmed by density functional theory (DFT) calculations. Furthermore, results of cell imaging in this study indicate that this new probe may be useful for detection and monitoring of Cu2+ in biological applications.

Abstract:The detection of metal ions such as Fe(3+)ions at the cellular level is of vital importance for monitoring life activities and disease diagnosis. Coumarin-based molecules are uniquely selective fluorescence probes for detecting Fe(3+)ions; however, due to their lower water solubility, applying them in biological detection is a bottleneck problem. Herein, a centrally hydrophobic/externally hydrophilic strategy is proposed and a novel coumarin-based fluorescence probe was successfully synthesized by using LaOBrviaa nucleophilic substitution reaction. The hydrophobic part of each coumarin molecule faces inward, and the hydrophilic part (iron-ion-capturing group) faces outward, allowing each coumarin molecule to be fully stretched. Compared with the coumarin-basedN-[2-hydroxy-1,1-bis(hydroxymethyl)ethyl]-7-hydroxycoumarin-3-carboxamide (DAT) solution, the water solubility of the LaOBr/DAT composite solution was greatly improved, with enhanced fluorescent properties and detection performance. In a water solvent, the detection limit of the composite considerably improved from 300 nM to 440 pM, and the composite demonstrated better stability and selectivity for detecting Fe3+. The composite probe was applied in Fe(3+)detection at the cellular level and in invisible anti-counterfeiting. This material design is of universal significance for improving the performance of other organic molecules.

Abstract:A highly selective and sensitive coumarin-based chemodosimeter 1 for Cu2+ in water is reported in this work. 1 was designed and facilely synthesized by a one-step reaction with coumarin as a fluorophore and 2-picolinic acid as the binding moiety, which showed very week fluorescence in buffer solution, and its fluorescence was considerably enhanced by the addition of Cu2+ at room temperature in 5min. Mechanism study suggested that Cu2+ promoted the hydrolysis of 1 via the catalytic sensing cycle, generating a highly fluorescent product 7-hydroxycoumarin with fluorescence signal greatly amplified. The probe exhibited remarkably selective fluorescence enhancement to Cu2+ over other metal ions at 454nm, with a detection limit of 35nM Cu2+. Under optimal condition, 1 was successfully used for the determination of Cu2+ in fetal equine serum and two water samples.

Abstract:A new chemosensor based on coumarin FB has been designed and synthesized for the detection of Fe3+ and PPi. FB displayed a high affinity to Fe3+ in the presence of other competing cations. The resulting FB-Fe3+ complex displayed highly sensitivity to PPi via Fe3+ displacement approach. The Fe3+ and PPi recognition processes is rapid and reversible, the detection limits of FB to Fe3+ and FB-Fe3+ complex to PPi were estimated to be 8.73 x 10(-8) M and 1.25 x 10(-8) M, respectively. The good biocompatibility of FB enables the investigation of fluorescent response for Fe3+ and PPi in living cells by confocal microscope. Abstract:A simple, fast responsive fluorescent chemical sensor for 3-aldehyde-4-hydroxyphenylpropylpyran-2-keto-4-hydroxybenzoyl (JT) was designed and synthesized, which was characterized by 1 H NMR and MS. The fluorescent chemical sensor JT showed outstanding selectivity and high sensitivity for identifying Al 3+ and the limit of detection (LOD) was 9.76 × 10 −8 M in EtOH/H 2 O (9:1, v/v) system. Furthermore, the in situ generated [JT + Al 3+ ] system was used for the relay detection of Fe 3+ without separation and purification, and the experimental results showed a new fluorescence quenching signal under the same conditions and the LOD to Fe 3+ was 5.21 × 10 −7 M. In addition, density functional theory (DFT) calculations were carried out to obtain optimized structures and orbital energy for the chemical sensor JT, [JT + Al 3+ ] and [JT + Fe 3+ ] complexes. Moreover, the test strips experiments were given for rapid detection of Al 3+ and continuous monitoring of Fe 3+ . Ultimately, it is worth mentioning that we have successfully achieved the recognition of Al 3+ and relay identification Fe 3+ by the sensor using zebrafish as a biological carrier, which indicate the sensor own excellent practical application prospect.

Abstract:A new fluorescent probe RH-Fe based on rhodamine B lactam was developed as a colorimetric and fluorescent chemosensor for Fe3+. The optical feature of RH-Fe for Fe3+ was investigated using UV-vis absorption and fluorescence emission spectroscopy. Upon the addition of Fe3+, the RH-Fe displayed a distinct color change from colorless to pink and a significant fluorescence enhancement, which can be directly detected by the naked eye. The stoichiometry of RH-Fe to Fe3+ complex was found to be 1 : 1 and the detection limit was determined to be as low as 0.27 mM. The results suggest that the RH-Fe probe may provide a convenient method for the visual detection of Fe3+ with high sensitivity.

Abstract:Three Rhodamine B derivatives were synthesized and characterized by ESI-MS, NMR, HR-MS and IR. The probes exhibit high selectivity and sensitivity towards Fe3+ over other metal ions in CH3CN-water. Upon the addition of Fe3+, the spirocyclic ring of the probe was opened and a significant enhancement of visible color and fluorescence within the range of 540-700 nm was observed. The colorimetric and fluorescent response to Fe3+ can be conveniently detected even by the naked eye, which provides a facile method for the visual detection of Fe3+. Job's plot, fluorescence titration and MS indicated the formation of 1:2 complexes between the probes and Fe3+. Abstract:A new fluorescent derivative (1) containing coumarin exhibits Fe(III)-selective strong yellow-green fluorescence in ethanol. This compound could be used as an "off-on" chemosensor and allow the detection of Fe3+ by monitoring changes in absorption and fluorescence spectra. Upon addition of Fe3+, an overall emission change of 125-fold was observed. High selectivity and sensitivity were observed over other metal ions, mainly due to the spirolactam ring-opening power of Fe3+. The detection limit was as low as 5.6 ppb. Photo-induced electron transfer, coupled with intramolecular charge transfer are proposed to account for the observed spectral response.

Abstract:Two new rhodamine-based fluorescent probes were synthesized and characterized by NMR, high resolution mass spectrometer (HR-MS) and IR. The probes displayed a high selectivity for Fe3+ among environmentally and biologically relevant metal ions in aqueous solution (CH3OH-H2O = 3 : 2, v/v). The significant changes in the fluorescence color could be used for naked-eye detection. Job's plot, IR and H-1 NMR indicated the formation of 1: 1 complexes between sensor 1 and Fe3+. The reversibility establishes the potential of both probes as chemosensors for Fe3+ detection. The probe showed highly selectivity in aqueous solution and could be used over the pH range between 5 and 9. Abstract:A Fe3+ chemosensor L1 was successfully synthesized with a quinoline moiety bound to rhodamine 6G hydrazide. The sensor L1 shows high selectivity and sensitivity to Fe3+ in aqueous solution in the presence of other trace metal ions in organisms, abundant cellular cations and prevalent toxic metal ions in the environment. In addition, biological imaging and micro computed tomography (MCT) technology studies have demonstrated that L1 could act as a turn-on fluorescent chemosensor for Fe3+ in living cells.

Abstract:Based on the fluorescence quenching mechanism of Fe3+ to moxifloxacin (MOX), a fluorescence probe has been found and exhibits high sensitivity, unique selectivity and good stability toward Fe3+. Moreover, the MOX probe can rapidly response to the change of Fe3+ concentration in pure aqueous solution. The MOX probe was utilized for quantitative determination of Fe3+ in real water sample, showing a good linear relationship (R=0.9980) between (F0–F) and [Fe3+]1/2 at the Fe3+ concentration from 0.05–15μM. The detection limit was found to be 0.032μM, which is much lower than that of most literature reported. Furthermore, by merging the MOX probe and a Y-tape fiber-optic spectrometer, a fluorescent fiber-optic chemosensor has been developed to quantitatively monitor Fe3+ in HepG2 living cells in vivo. The monitoring results are consistence with that obtained from the confocal imaging technique. The established method possesses the advantages of fast responsive, high sensitivity, unique selectivity, very stability and good repeatability for quantitative monitoring Fe3+ in real water and HepG2 living cells. In addition, the complex mechanism of MOX toward Fe3+ has also been explored by Fourier transform infrared spectroscopy (FT-IR) and mass spectra (MS) in the present work.

Abstract:A novel benzothiazole-functionalized fluorescein derivative was designed and synthesized. Sensor 1 displays highly selective and sensitive OFF-ON fluorescence response and naked-eye color change to Fe3+ in aqueous solution at pH 7.2. The mechanisms have been supported by Job's plot evaluation, fluorescence titrations, MS, and H-1 NMR spectroscopic studies. The detection limits of sensor 1 to Fe3+ were estimated to be 7.4 nM. 1 exhibits low cytotoxicity and cell-membrane permeability, which makes it capable of Fe3+ bioimaging in living Hep G2 cells. Abstract:Fe3+ plays a crucial role in many vital cell functions and its detection has attracted considerable attention. In this work, a water-soluble rhodamine-based chemosensor RL has been designed and synthesized as an "off-on" chemosensor for Fe3+ detection. Upon the addition of Fe}, RL displayed obvious color change, quick fluorescence enhancement, high sensitivity and anti-interference capacity over other metal ions (Li+, Na+, K+, Mg2+ Ca2+, Ba2+, Al3+, Sn2+, Pb2+, Bi3+, Cr3+, Mn2+, Co2+, Ni2+, Pd2+, Cu2+, Ag+, Zn2+, Cd2+, Hg2+). The detection limit for Fe3+ was calculated to be 0.28 mu M with the binding constant K-a to be 4.67 x 10(8) M-2. Abstract:Trimeric phenolic coumarin (TPC) was designed through the internal hydrophobic-external hydrophilic strategy, achieving greatly enhanced water solubility and sensing performance with detection limit of 9.6 pM, which is much lower than that of coumarin molecule probes and other reported iron ion probes. Sun-excited naked eye Fe3+ detection and Fe3+/Fe2+ detection in the cellular environment were obtained.

Abstract:A rhodamine derivative bearing a thiazole receptor (probe 1) was developed and characterized. Probe 1 rendered a prominently chromogenic and turn-on fluorescent signal toward Fe3+ ion in aqueous media based on coordination reaction. Probe 1 also showed high sensitivity toward Fe3+ ion with the detection limit as low as 92nM. By means of the Job's plot, 2:1 binding mode of 1-Fe3+ complex was determined and a possible sensing mechanism was further proposed based on the changes in the Fourier Transform Infrared (FT-IR) and mass spectra. The turn-on fluorescence and sensing mechanism for 1-Fe3+ complex were well demonstrated by theoretical calculations. Importantly, probe 1 has been tested highly suitable for mapping Fe3+ ion in human colon cancer cells SW480, providing a wonderful candidate for tracking Fe3+ in biological processes.
