Abstract:A simple method has been developed to detect the activity of alkaline phosphatase (ALP) by the changing of fluorescence intensities of the quantum dots (QDs). In this system, the fluorescence intensities of the QDs were quenched by p-nitrophenol (pNP) which was produced in the process of ALP catalytic reaction. A series of linear calibration curves of the activity of ALP were obtained in different pH buffer solutions. The wide linear range was 3–1000UL−1 and the detection limit was 3UL−1 (S/N=3). Furthermore, the experimental conditions of biosensor were optimized, and anti-interference ability was presented. The activity of ALP was also detected in serum and the recovery of ALP in serum samples was more than 95%. The excellent performance of this biosensor indicates that it can be used in practice detection of ALP.
Facile and Sensitive Fluorescence Sensing of Alkaline Phosphatase Activity with Photoluminescent Carbon Dots Based on Inner Filter Effect

Abstract:A simple and sensitive fluorescent assay for detecting alkaline phosphatase (ALP) based on the inner filter effect (IFE) has been proven, which is conceptually different from the previously reported ALP fluorescent assays. In this sensing platform, N-doped carbon dots (CDs) with a high quantum yield of 49% were prepared by one-pot synthesis and were directly used as a fluorophore in IFE. p-Nitrophenylphosphate (PNPP) was employed to act as an ALP substrate, and its enzyme catalytic product (p-nitrophenol (PNP)) was capable of functioning as a powerful absorber in IFE to influence the excitation of fluorophore (CDs). When in the presence of ALP, PNPP was transformed into PNP and induced the absorption band transition from 310 to 405 nm, which resulted in the complementary overlap between the absorption of PNP and the excitation of CDs. Because of the competitive absorption, the excitation of CDs was significantly weakened, resulting in the quenching of CDs. The present IFE-based sensing strategy showed a good linear relationship from 0.01 to 25 U/L (R(2) = 0.996) and provided an exciting detection limit of 0.001 U/L (signal-to-noise ratio of 3). The proposed sensing approach was successfully applied to ALP sensing in serum samples, ALP inhibitor investigation and phosphatase cell imaging. The presented IFE-based CDs fluorescence sensing strategy gives new insight on the development of the facile and sensitive optical probe for enzyme activity assay because the surface modification or the linking between the receptor and the fluorophore is no longer required.

Abstract:Herein, green-fluorescent N-doped carbon quantum dots (N-CQDs) with quantum yield of 30.6% were prepared via a simple bottom-up electrochemical (EC) method using the mixtures of pyrocatechol and ethylenediamine as both precursors and electrolytes. The ON-OFF fluorescence (FL) principle that the FL intensity of the N-CQDs was quenched in the presence of Fe3+ was further extended to the ON-OFF-ON and ON-OFF-ON-OFF ones for pyrophosphate anions (PPi) and alkaline phosphatase (ALP) detection, respectively, based on the specific competitive interaction of N-CQDs with Fe3+ and PPi and the hydrolysis of PPi in the presence ALP. Under the optimized conditions, limit of detection (LOD) of 1.2 μM and 0.5 μM were achieved for assays of Fe3+ and PPi, respectively. Meanwhile, measurable evaluation of ALP activity was reached in a wide range from 5 to 360 U/L with a LOD of 1.1 U/L. The as-proposed sensors showed many advantages including label free, rapid, simple, low-cost, high sensitive, which has great promising application potential in vivo and in vitro clinical diagnosis and will promote the application of EC derived carbon nanomaterials.
Synergistic effect-mediated fluorescence switching of nitrogen-doped carbon dots for visual detection of alkaline phosphatase

Abstract:The natural enzyme alkaline phosphatase (ALP), which is associated with a large number of diseases, plays an essential role in clinicopathological examinations. Therefore, it is of great significance to quantify the ALP activity levels. In this work, a switchable fluorescence assay of detecting alkaline phosphatase (ALP) activity was proposed using Fe3+ to adjust the fluorescence of nitrogen-doped carbon dots (CDs). Wherein, CDs chelated with Fe3+, leading to a remarkable and dynamic fluorescence quenching. However, ascorbic acid (AA) and phosphate group (Pi), resulting from the enzymatic reaction between ascorbic acid-2-phosphate (AAP) and ALP, enabled the great fluorescence recovery, which was possibly related with the synergistic effect including the chemical redox of AA towards Fe3+ and the formation of a stable FePO4 complex. Based on this synergistic effect-mediated switching mechanism, the indirect detection of ALP activity was developed in the range of 15–445 U/L with the limit of detection at 2.576 U/L (3σ/k) and the limit of quantitation at 8.587 U/L (10σ/k), which could be observed by the naked-eyes. Then, the proposed method was further applied to the detection of ALP activity in human serum and urine samples with satisfactory results, which was in good agreement with the result obtained by ALP commercial kit. This proposed synergistic effect from AA and Pi provides a new insight for ALP activity detection.

Abstract:A fluorescence turn-on assay for alkaline phosphatase (ALP) activity is developed through the controlled release of polyethyleneimine-capped copper nanoclusters (PEI-capped CuNCs) from the MnO2 nanosheets. In an aqueous solution, the positively charged PEI-capped CuNCs could be adsorbed onto the surface of the negatively charged MnO2 nanosheets. Such adsorption through favorable electrostatic interactions could efficiently quench the nanocluster fluorescence emission via resonance energy transfer from the PEI-capped CuNCs to the MnO2 nanosheets. 2-Phospho-l-ascorbic acid (AAP) could be hydrolyzed to l-ascorbic acid (AA) in the presence of ALP. AA could reduce MnO2 into Mn2+ and trigger the disintegration of the MnO2 nanosheets. As a result, the CuNCs were released and the quenched fluorescence was recovered efficiently. The detection strategy is simple, inexpensive, sensitive, selective, with low toxicity, and has better biocompatibility. The newly fabricated biosensor for ALP activity will potentially make it a robust candidate for numerous biological and biomedical applications.
Co, N-doped Carbon Dot Nanozymes with Acid Ph-Independence and Substrate Selectivity for Biosensing and Bioimaging

Abstract:Nanozymes are more stable, economical, and easier to produce than natural enzymes; however, their low activity in non-acidic environments and lack of substrate selectivity severely limit the applications of nanozymes. In this study, we developed Co- and N-doped carbon dot nanozymes (CoNCDs) that specifically exhibited peroxidase-like activity toward o-phenylenediamine in a non-acidic environment. Based on the peroxidase-like activity and fluorescence property of CoNCDs in a neutral environment, a one-pot multi-mode sensing platform (colorimetric and ratiometric fluorescence imaging) was developed to detect cholesterol and xanthine in solution and human serum samples. This platform was simple to operate, and the CoNCDs demonstrated a reduced detection time and an improved analytical performance compared to most of the reported nanozymes. Moreover, the improved peroxidase activity at neutral pH, good biocompatibility, and photostability of CoNCDs facilitated their use for monitoring endogenous H2O2 levels in vivo. This study not only provides a novel approach for developing an acid pH-independent and substrate selectivity nanozyme but also demonstrates the application of ratiometric fluorescence imaging using the developed CoNCDs both in vitro and in vivo.

Abstract:Alkaline phosphatase (ALP) is an important biomarker whose abnormal level in activity is associated with hepatobiliary, skeletal, and renal diseases as well as cancer. Herein, we synthesized ZnSe@ZnS quantum dots (ZnSe@ZnS QDs) and Mn-doped ZnS quantum dots (Mn:ZnS QDs) as fluorophores to establish the ratiometric fluorescent assay for ALP activity detection in biological samples. p -Nitrophenyl phosphate (PNPP) was used as a substrate for ALP, and the overlaps between absorption spectra of PNPP and excitation spectra of QDs resulted in sharp fluorescence quenching. Under the catalysis of ALP, PNPP was hydrolyzed into p -nitrophenol (PNP), which caused a red shift of absorption band of PNPP and fluorescence recovery of Mn:ZnS QDs (585 nm). However, the overlaps between absorption spectra of PNP and emission spectra of ZnSe@ZnS QDs led a further quenching of ZnSe@ZnS QDs (405 nm). Therefore, the ratiometric fluorescent signals ( F 585 / F 405 ) were associated with activity of ALP based on bidirectional responses of QDs to the concentration of PNPP. Under the optimum conditions, the method exhibited a good linear relationship from 4 to 96 U per L ( R 2 = 0.9969) with the detection limit of 0.57 U per L. Moreover, the method was successfully applied for detecting the ALP activity in a complex biological matrix (human serum and HepG2 cells) with impressive specificity. In particular, the complicated chemical modifications of QDs and pretreatments of biological samples were not required in the whole detection procedures. Therefore, it not only provided a sensitive, specific and simple approach to clinical ALP activity detection, but it also provided support for early diagnosis of diseases.

The probe was constructed by combining Ce 3+ -crosslinked copper nanoclusters (Ce 3+ -CuNCs) which exhibit the aggregation-induced emission (AIE) feature with carbon dots (CDs). The introduction of phosphate (Pi) induced the generation of CePO 4 precipitation, resulting in significant decrease of fluorescence emission of CuNCs at 634 nm. At the same time, the fluorescence of CDs at 455 nm was obviously enhanced, thus generating ratiometric fluorescence response. Based on the fact that the hydrolysis of pyrophosphate (PPi) by ALP can produce Pi, the CD/Ce 3+ -CuNCs ratiometric probe was successfully used to determine ALP. A good linear relationship between the ratiometric value of F 455 / F 634 and ALP concentrations ranging from 0.2 to 80 U·L − 1 was obtained, with a low detection limit of 0.1 U·L − 1 . The ratiometric responses of the probe resulted in the visible fluorescence color change from orange red to blue with the increase of ALP concentration. The smartphone-based RGB recognition of the fluorescent sample images was used for ALP quantitative determination.

Abstract:Herein, a simple and sensitive ratiometric fluorescence sensing platform to detect alkaline phosphatase (ALP) activity is developed on the basis of yellow fluorescent nitrogen-doped carbon quantum dots (YNCDs). The hydrolysis of ascorbic acid 2-phosphate (AAP) into ascorbic acid (AA) is catalyzed by ALP. Then, AA will react with o-phenylenediamine (OPD) to form 3-(1,2-dihydroxyethyl)furo[3,4b]-quinoxaline (QXD) which is a blue fluorescent quinoxaline derivative with emission at 435 nm in the presence of Cu 2+ . YNCDs have yellow fluorescence emission at 555 nm, and can maintain stable in QXD reaction system. Therefore, by utilizing the fluorescence of YNCDs at 555 nm as reference signal and the fluorescence of QXD at 435 nm as report signal, we can detect the ALP activity by monitoring the fluorescence ratio ( F 435 / F 555 ). The linear range is 0.5–5 U/L, and the limit of detection is 0.14 U/L. An application of this method for the analysis of ALP in human serum has given satisfactory results.

Abstract:In this work, a novel and sensitive fluorescence sensing system for alkaline phosphatase (ALP) was constructed using a bifunctional copper metal-organic framework (Cu@MOF) nanozyme, which had excellent oxidase-mimetic activity and fluorescence properties. Owing to the presence of 2-amino-1,4-benzenedicarboxylic acid (1,4-BDC-NH2) ligand, Cu@MOF displays excellent fluorescence performance at 444 nm. Additionally, Cu2+ endows the oxidase-like activity of Cu@MOF, which could trigger p-phenylenediamine (PPD) to be oxidized to a brown product (PPDox) and quench the photoluminescence of Cu@MOF through the inner filtration effect (IFE). As the preferential affinity of ATP for Cu2+, the catalytic activity of Cu@MOF was significantly reduced once ATP was added, thus PPD could not be oxidized and fluorescence was recovered. In the presence of ALP, ATP was hydrolyzed to adenosine and Pi, which allowed Cu@MOF to regain its catalytic activity and continued to catalyze the generation of PPDox. The fluorescence of Cu@MOF was therefore weakened once again. The ALP activity was directly proportional to the degree of decrease in fluorescence intensity. Thus, this novel fluorescence sensing strategy had a linear range of 0.5-60 U/L and the limit of detection was 0.14 U/L. The established sensing method could also be used to for ALP inhibitors screening, and achieved satisfactory results in determining the level of ALP activity in human serum.
Fluorescence assay for alkaline phosphatase activity based on energy transfer from terbium to europium in lanthanide coordination polymer nanoparticles.

Abstract:In this study, bimetallic lanthanide coordination polymer nanoparticles (Tb-GMP-Eu CPNs) were developed for sensing alkaline phosphatase (ALP) by utilizing guanine monophosphate (GMP) as the bridge ligand to self-assemble with Tb3+ and Eu3+ in Tris-HCl buffer solution. In this polymer framework, an efficient energy transfer from Tb3+ to Eu3+ occurred because the emission energy of Tb3+ matched well with the absorption energy of Eu3+. As a result, the synthesized Tb-GMP-Eu CPNs exhibited the characteristic red emission of Eu3+ when excited at 290 nm. When ALP was introduced into the Tb-GMP-Eu CPNs system, it would destruct the combination between Tb and GMP by a dephosphorylation process, resulting in interruption of energy transfer from Tb3+ to Eu3+ and therefore the fluorescence quenching of Eu3+. Based on these facts, a fluorescence assay based on bimetallic lanthanide coordination polymer nanoparticles for ALP detection was developed. Such a Tb-GMP-Eu CPNs sensor exhibited sensitive response to ALP with a detection limit down to 0.004 U L-1. Moreover, the successful application of ALP inhibitor evaluation and determination of ALP in serum samples have been demonstrated. Therefore, the proposed method has great potential application for ALP detection in biological and clinical diagnosis.

Fluorescence Turn-on Detection of Alkaline Phosphatase Activity and Al3+ Using Vitamin B6 Cofactor Conjugated GSH Capped Mn-doped ZnS Quantum Dots

Abstract:The glutathione (GSH) functionalized Mn-doped ZnS quantum dots (GSH_Mn_ZnS QDs) was conjugated with pyridoxal 5'-phosphate (PLP). The -CHO group of vitamin B6 cofactor PLP interacted with the -NH2 group of GSH functionalized Mn_ZnS QDs. The conjugation of PLP quenched the fluorescence emission of GSH_Mn_ZnS QDs at 601 nm. Addition of alkaline phosphatase (ALP) catalytically dephosphorylated the PLP into pyridoxal that restored the fluorescence emission of GSH_Mn_ZnS QDs. With a sensitivity of 0.035 U/L, the PLP conjugated GSH_Mn_ZnS QDs was applied to quantify ALP activity in human serum and plasma. Further, the developed nanoprobe PLP conjugated GSH_Mn_ZnS QDs was also applied to detect Al3+. The complexation-induced fluorescence enhancement was observed at 492 nm upon the interaction of Al3+ with the PLP conjugated GSH_Mn_ZnS QDs. Without any interference from other tested metal ions, this nanoprobe can be employed to detect Al3+ down to 2.30 µM.

Abstract:Sulfur quantum dots (SQDs) are a kind of pure elemental quantum dots, which are considered as potential green nanomaterials because they do not contain heavy metal elements and are friendly to biology and environment. In this paper, SQDs with size around 2 nm were synthesized by a microwave-assisted method using sulfur powder as precursor. The SQDs had the highest emission under the excitation of 380 nm and emit blue fluorescence at 470 nm. In addition, the SQDs had good water solubility and stability. Based on the synthesized SQDs, a fluorescence assay for detection of alkaline phosphatase (ALP) was reported. The fluorescence of the SQDs was initially quenched by Cr (VI). In the presence of ALP, ALP-catalyzed hydrolysis of 2-phospho-L-ascorbic acid to generate ascorbic acid. The generated ascorbic acid can reduce Cr (VI) to Cr (III), thus the fluorescence intensity of SQDs was restored. The assay has good sensitivity and selectivity and was applied to the detection of ALP in serum samples. The interesting properties of SQDs can find a wide range of applications in different sensing and imaging areas.

Label-free fluorescence turn-on detection of alkaline phosphatase activity using the calcein-Ce3+ complex

Abstract:This study introduces an innovative approach for the real-time and efficient detection of alkaline phosphatase (ALP) activity, using a calcein fluorescence probe and leveraging the static quenching properties of calcein fluorescence by Ce3+ metal ions. In this method, calcein serves as the signal element, with its fluorescence effectively preserved through energy transfer or charge transfer when coordinated with Ce3+. Conversely, ALP catalyzes the phosphopeptide substrate to generate a substantial amount of Pi, preventing calcein fluorescence quenching due to the higher affinity between Pi and Ce3+ compared with that between calcein and Ce3+. The fluorescence intensity ratio (F-F0/F0) exhibited excellent linearity, facilitating sensitive ALP detection. The proposed ALP detection method covers a range from 0 to 1.4 mU/mL (R2 = 0.9942), with the limit of detection at 0.069 mU/mL (S/N = 3). Additionally, this method was successfully applied for detecting ALP in serum samples and studying its inhibitors. This research introduces a novel clinical diagnosis approach for ALP sensing while broadening the potential applications of calcein.

Abstract:A method is described for the determination of the activity of alkaline phosphatase (ALP). It is based on the reversible modulation of the fluorescence of WS2 quantum dots (QDs). The fluorescence of the QDs is quenched by Cr(VI) but restored by free ascorbic acid (AA). The detection scheme relies on the fact that ALP hydrolyzes the substrate ascorbic acid 2-phosphate to produce AA, and that enzymatically generated AA can restore the fluorescence of the QDs. The signal (best measured at excitation/emission peak wavelengths of 365/440 nm) increases linearly in the 0.5 to 10 U·L−1 ALP activity range, with a detection limit of 0.2 U·L−1. Abstract:Carbon dots (CDs) emitting red fluorescence (610 nm) were synthesized by solvent thermal treatment of p-phenylenediamine in toluene. Upon 440 nm excitation, quercetin (QCT) alone endowed slight effects on the red fluorescence of CDs. Once Zn2+ was further introduced, the QCT-Zn2+ complex was quickly formed. This complex absorbs excitation light and emits bright green fluorescence at 480 nm. The red fluorescence of CDs was greatly quenched owing to the inner-filter effect. The ratio of fluorescence intensity at 480 nm and 610 nm (I480/I610) gradually increases with increasing concentration (c) of Zn2+. Al3+ exhibits the same phenomen like Zn2+. Fluoride ions form a more stable complex with Al3+ than QCT-Al3+ complex but have a negligible effect on the QCT-Zn2+ complex. The possible interference of Al3+ on Zn2+ can thus be avoided by adding certain amount of F-. The CD-QCT-F- system was constructed as a ratio-metric fluorescent nanoprobe toward Zn2+ with determination range of 0.14-30 μM and limit of detection (LOD) of 0.14 μM. Due to the stronger affinity of adenosine triphosphate (ATP) to Zn2+ than QCT, the I480/I610 value of CD-QCT-F--Zn2+ system gradually decreases with increasing cATP. The ratiometric fluorescent nanoprobe toward ATP was established with detection ranges of 0.55-10 and 10-35 μM and a LOD of 0.55 μM. The above two probes enable the quantitative determination of Zn2+ and ATP in tap and lake water samples with satisfactory recoveries. Abstract:Alkaline phosphatase (ALP) is a commonly used marker in clinical practice, and this enzyme is a key indicator for diagnosing various diseases. In this study, we describe the development of a reliable and novel fluorescent assay for ALP detection based on chitosan carbon dots (C-CDs, peak emission, 412 nm) and calcein (peak emission, 512 nm). In the presence of Eu3+ (which binds calcein), the fluorescence intensity of calcein is quenched. Utilizing the ALP-triggered generation of phosphate ions (PO43−) from the substrate p-nitrophenyl phosphate (pNPP), the Eu3+ ions bind PO43− (which shows a higher affinity toward Eu3+ than calcein), and the fluorescence of calcein is recovered. As a consequence, C-CDs fluorescence is decreased by inner filter effect (IFE). Exploiting these changes in the fluorescence intensity ratio of C-CDs and calcein, we developed a high sensitivity, accurate, and easily synthesized ratiometric fluorescence probe. Our novel fluorescent bioassay demonstrates good linear relationship in the 0.09–0.8 mU mL−1 range, with a low detection limit of 0.013 mU mL−1. The excellent applicability of this novel assay in HepG2 cells and human serum samples demonstrates that our novel method has excellent biomedical research and disease diagnosis prospects.

Abstract:Water-soluble luminescent CdTe–ZnS quantum dots (QDs) modified by mercaptopropyl-acid was used to determinate H+ in aqueous solutions. The fluorescence (FL) of the water-soluble QDs could be irreversibly quenched by H+ and the fluorescence intensity of the water-soluble QDs decreased linearly as the pH decreased in the range of 8.0–5.0. Based on this phenomenon, a simple, rapid and specific method to quick determination of enzyme reaction kinetics was proposed. The modified QDs was successfully used as pH probe in monitoring the hydrolysis of glycidyl butyrate catalyzed by porcine pancreatic lipase (PPL). The proposed method demonstrated improved sensitivity, stability and a wider monitoring range for H+ determinations as compared to the already described analytical methods based on PNP.
A ratiometric fluorescent probe for simultaneous detection of L-ascorbic acid and alkaline phosphatase activity based on red carbon dots/polydopamine nanocomposite

Abstract:Herein, efficient red carbon dots (R-CDs) were synthesized by one-step hydrothermal treatment of N -(4-amino phenyl) acetamide and (2,3-difluoro phenyl) boronic acid. The optimal emission peak of R-CDs was at 602 nm (under 520 nm excitation) and the absolute fluorescence quantum yield of R-CDs was 12.9%. Polydopamine, which was formed by the self-polymerization and cyclization of dopamine in alkaline condition, emitted characteristic fluorescence with peak position of 517 nm (under 420 nm excitation) and affected the fluorescence intensity of R-CDs through inner filter effect. L-Ascorbic acid (AA), which was the hydrolysis product of L-ascorbic acid-2-phosphate trisodium salt under the catalytic reaction of alkaline phosphatase (ALP), effectively prevented the polymerization of dopamine. Combined with the ALP-mediated AA production and the AA-mediated polydopamine generation, the ratiometric fluorescence signal of polydopamine with R-CDs was correlated closely with the concentration of both AA and ALP. Under optimal conditions, the detection limits of AA and ALP were 0.28 μM during linear range of 0.5–30 μM and 0.044 U/L with linear range of 0.05–8 U/L, respectively. This ratiometric fluorescence detection platform can efficiently shield the background interference of sophisticated samples by introducing a self-calibration as reference signal in a multi-excitation mode, which can detect AA and ALP in human serum samples with satisfactory results. Such R-CDs/polydopamine nanocomposite provides a steadfast quantitative information and makes R-CDs be excellent candidate for biosensors via combining target recognition strategy.
