Original Studies on Dormancy and Germination Properties of Cistanche deserticola Seeds

Abstract:In order to know parasitic property of Cistanche deserticola,we studied dormancy and germination properties of Cistanche deserticola seeds.Embryo was observed through whole transparent law;seed vitalities were detected through TTC way;seed germination was induced through physical ways (such as water soaking,changed temperature,stratification)and chemical ways(such as hormone,strigol analogue——GR24,drawing liquid of Chinese herbal medicine、roots of Nitraria tangutorum and roots of Haloxylon ammodend).The results showed: seeds of Cistanche deserticola was smaller,the 1000-seed weight was only 0.096g,embryo of matured seeds developed incompletely,embryo lied in spherical embryo stage all the time before and after germination.Percentages of viable seeds were 25.3%.Germination rate of seeds was the highest when seeds were soaked in water for 30 days,cultured in GA_3 (0.5μg/ml)and 25℃ constant temperature condition,seed germination rate was up to 16.7%.Some of Chinese herbal medicine、drawing liquid of Nitraria tangutorum may stimulate seeds germination,but GR24、drawing liquid of Haloxylon ammodend roots had no action in seed germination.The results testified seeds of Cistanche deserticola had dormancy phenomenon,seeds germination induction need a certain stimulating signal from host roots except long period seedripening course.Dormancy and germination properties of Cistanche deserticola seeds were different from non-parasitic pants and other parasitic plant.
