Abstract:Utilizing iron chloride as a Lewis acid catalyst, we developed a straightforward and mild oxidative cross-coupling reaction between quinoxalinones and indoles, yielding a series of versatile 3-(indol-3-yl)quinoxalin-2-one derivatives. This approach allows for the incorporation of a wide array of functional groups into the final products, demonstrating its synthetic versatility. Notably, the method was successfully scaled up to gram-scale reactions while maintaining high yields. Our mechanistic investigation indicates that iron chloride serves as a catalyst to facilitate the formation of key intermediates which subsequently undergo oxidation to afford the desired products. The merits of this protocol include its cost effectiveness, operational simplicity, and the ease of product isolation via filtration.

Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:The development of aerobic oxidative C–H/N–H cross-coupling catalyzed by the first-row transition metals is attractive yet challenging. Here, the first iron-catalyzed intramolecular cross-dehydrogenative coupling for the synthesis of carbazoles and indoles with using air as oxidant is reported. The general and efficient protocol allows for C–N bond formation under green conditions with excellent functional group compatibility, delivering a wide range of N–H carbazoles and indoles in good yields. Impressively, all carbazoles are prepared from 2-cyclohexenyl aminoarenes with losing 6Hs via a C–H amination and aromatization process. Furthermore, it also works for C–O bond formation.

Abstract:Considering the importance of developing powerful catalysts and the pharmacophore characteristics of indole derivatives, we described a switchable approach for the iron-catalyzed oxidative C(sp3)–H functionalization of indolin-2-ones. Selective transformations displayed excellent activity and chemoselectivity using FeCl2 as the catalyst, air as the oxidant, and alcohol as the solvent. By manipulating the reaction conditions, particularly the choice of solvent, catalyst loading, and reaction sequence, a series of valuable indole derivatives, including isatins and symmetrical and nonsymmetrical isoindigos, were selectively synthesized in good to excellent yields. Furthermore, the gram-scale synthesis of compounds with biological anticancer activity under simple conditions highlights their great potential in practical applications.

Abstract:Indoles are critically important heterocyclic building blocks with broad applications in pharmaceuticals, fragrances, agrochemicals, pigments, and materials. Recent advancements have highlighted bis‐indole derivatives and oligomerized indoles as promising scaffolds for anticancer compounds and materials in organic semiconductors. Despite the challenges in forming C–C bonds in indoles, we developed a novel method combining electrochemical and cobalt complex dual catalysis to achieve regioselective C3‐C3/C2‐C3 coupling. Using 1‐methylindole as a model substrate, we optimized reaction conditions to achieve a 90% yield of the tetramerized product. The method proved effective for various substituted indoles, with satisfactory yields for 6‐position substituents and poorer results for 5‐position due to steric hindrance, as confirmed by DFT calculations. The proposed mechanism involves initial C3‐C3 coupling facilitated by cobalt catalysis, followed by C2‐C3 coupling under electrochemical conditions, providing an efficient and environmentally benign strategy for synthesizing complex indole derivatives.

Abstract:2,3'‐Bisindoles with C–C linkages have attracted interest in medicinal chemistry, yet their synthesis is intricate with many steps. Notably, direct C–H/C–H cross‐coupling of non‐directed heteroaromatics remains challenging, often requiring precious metals and oxidants to enhance coupling efficiency. Herein, we present a copper‐catalyzed C–H/C–H cross‐coupling method for N‐substituted indoles without directing groups, facilitated by molecular oxygen under gentle conditions. It showed reasonable functional group compatibility and provided one‐pot access to a variety of 2,3'‐bisindoles derivatives in moderate to good yields.

Abstract:A mild and general alpha-arylation of alpha-amino carbonyls with indoles catalyzed by Fe(ClO4)(3) has been developed. C-H activation is smoothly fulfilled by using TBHP as the oxidant with good yields. Two hydrogen dissociations make this transformation more environmentally benign because of high atom efficiency.

Abstract:Developing an efficient catalytic system using molecular oxygen as the oxidant for rhodium-catalyzed cross-dehydrogenative coupling remains highly desirable. Herein, rhodium-catalyzed oxidative annulation of 2- or 7-phenyl-1H-indoles with alkenes or alkynes to assemble valuable 6H-isoindolo[2,1-a]indoles, pyrrolo[3,2,1-de]phenanthridines, or indolo[2,1-a]isoquinolines using the atmospheric pressure of air as the sole oxidant enabled by quaternary ammonium salt has been accomplished. Mechanistic studies provided evidence for the fast intramolecular aza-Michael reaction and aerobic reoxidation of Rh(I)/Rh(III), facilitated by the addition of quaternary ammonium salt.

Abstract:In continuation of our research interest in the green oxidation of indoles, we further explore the direct oxidation of 2-methylindoles to 2-formyl indoles promoted by NCS and associated with H2O as the oxygen source. This methodology was demonstrated to be a robust protocol consisting of chlorination, SN2', and oxidation processes, and presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of high added-value versatile building blocks susceptible to further functionalization.

Abstract:Oxindole units were common structure features of natural products and drugs. A novel method for synthesizing 2‐oxindoles from 1,3‐dicarbonyl compounds based on ferrous salt and iodide catalyst was described. Using Fe(OAc) 2 /NaI/Na 2 S 2 O 8 , various 2‐alkyl‐3‐phenylamino‐3‐oxopropanoates bearing fluoro, chloro, bromo, alkyl, alkoxy, trifluoromethyl, cyano etc . on the benzene ring were converted smoothly to 2‐oxindoles.

Abstract:Oxidative rearrangement of 3-substituted indoles is a fundamental organic transformation for the synthesis of 2-oxindole scaffolds. However, this time-consuming and complex task produces a single type of product via two-electron oxidation using stoichiometric chemical oxidants. Herein, we report a unified and efficient copper-catalyzed selective single-electron transfer strategy for three oxidation reactions of 3-substituted indoles using air (O2) as the terminal oxidant, providing a streamlined and practical synthetic approach to access 3-hydroxyl-, 3-alkoxyl-, and 3-hygrogenous-2-oxindoles via a 3-radical-2-hydroxyl indoline intermediate. This copper catalysis protocol demonstrates high chemoselectivity, good functional group tolerance, and broad application potential in the modular synthesis of biologically 3-functionalized oxindole derivatives.

Abstract:We report the development of a rapid approach for directly converting indoles into 2-oxindoles promoted by HOCl formed in situ from the combination of PIFA and n-Bu4NCl·H2O. The procedure is widely functional group tolerant and provides 2-oxindoles in up to 94% yield within 5 min. The potential applications of the developed methodology are demonstrated by the gram-scale preparation of 3-methyl-2-oxindole (11a), the one-pot two-step syntheses of spiro-oxindoles 26a and 26b, and the formal synthesis of (-)-folicanthine (2).

Abstract:Mild conditions have been developed to achieve Pd-catalyzed homocoupling of indoles with excellent regioselectivity in the presence of Cu(OAc)(2) x H(2)O in DMSO at room temperature in high efficiency. This method provides a simple route to 2,3'-biindolyls from the electron-rich to moderately electron-poor indoles. The reaction tolerates the bromide substituent on indoles. In addition, a one-pot approach to C3-position acetoxylated biindolyls is realized via palladium catalysis by the use of AgOAc under oxygen atmosphere as oxidants.

Abstract:An efficient organoiodine‐catalysed intramolecular direct C–N bond oxidative coupling reaction is presented. Structurally diverse 3‐(mono)substituted oxindoles were rapidly obtained in a complex oxidative system in up to 87% yield. Various N‐alkoxy‐2‐phenylpropanamides were well‐tolerated. This study showed that the electronic effects on the aromatic ring of reactants have a crucial effect on the selectivity (C−N/C−O) of cyclisation products. In addition, gram‐scale synthesis and late‐stage modification of 3‐(mono)substituted oxindole derivatives revealed the practical application of this transformation.

Abstract:A highly effective and straightforward method to construct a wide range of functionalized 2,3-disubstituted indoles has been developed. The method involves the tandem reaction of rhodium(II)-catalyzed denitrogenative annulation of triazole-based benzyl anilines and oxygen-mediated oxidative aromatization. The developed method can also be used to synthesize 2,3-disubstituted benzofurans by replacing the benzyl anilines with benzyl phenols.

Abstract:A new electrochemical proton-coupled electron transfer method for the intermolecular C SP 2 –H amination of heteroarenes without oxidants, metal catalysts and external electrolytes has been developed. Various new N-containing heteroarenes were prepared in medium to high yields, and the indole-containing product could be converted into practical 2-oxindole by simple basic hydrolysis. Mechanistic investigation indicated that ester sulfonyl-substituted N -radicals could be formed by the combination of 2,6-lutidine and electrochemical oxidation, which is the key to achieve the desired chemoselectivity.
