Abstract:Objective To clone and sequence the extracellular domain of H-2K~d gene, and construct an associated expression vector. Methods The 891bp fragment of the extracellular domain of H-2K~d gene cDNA was amplified from the total RNA of BALB/c mice spleen by using RT-PCR. The cDNA was inserted into PBS-T vector directly and the recombinant plasmid was identified by restriction endonucleases digestion and DNA sequencing. Results Restriction analysis and DNA sequencing confirmed that the extracellular domain of H-2K~d gene had been inserted into vector PBS-T. Conclusion The extracellular domain of H-2K~d gene was successfully cloned.
