Abstract:Objective Preparing of a rat model for study of mechanismand process of insulin re-sistance by different diet composition and caloric restriction. Methods 132 rats were divided into 3groups :control , obesity and caloric restriction. Euglycemic-hyperinsulinemic clamp technique andOGTT were taken every two months . At the same ti me ,plasma and tissue samples and body weightwere taken fromeach group .Insulinlevels and fat tissue mass were determined . Results The orderof insulin sensitivity and glucose tolerance ability were caloric restriction control obesity group .The order of plasma insulin level was obesity control caloric restriction groups .The body weightand visceral fat mass wereincreased significantlyin obesity group . Conclusions The protocols of di-ets andfeeding patterns adoptedinthis study are able toinduce a rat model with different insulin sensi-tivity .The accumulation of body fat mass is related to the metabolismabnormalities in vivo .
