Abstract:A residue depletion study was performed to investigate the tissue kinetics of quinocetone (1) and its major metabolites. Quinocetone and its major metabolites were simultaneously quantitated with a high-performance liquid chromatography-ultraviolet (HPLC-UV) method. A total of 25 pigs, 30 broilers, and 50 carp were fed 100 mg/kg quinocetone for 90, 42, and 60 days, respectively. Liver, kidney, muscle, and fat (skin) tissues were collected at five different withdrawal times for analysis. Results revealed that quinocetone, 1-desoxyquinocetone (2), carbonyl-reduced 4-desoxyquinocetone (4), 3-methylquinoxaline-2-carboxylic acid (5), and carbonyl-reduced dideoxyquinocetone (6) could be depleted quickly in tissues; by contrast, dideoxyquinocetone, 3, persisted for a long time in the liver. Therefore, the liver is possibly the target tissue of quinocetone, and 3 is the residual marker; the recommended withdrawal times (WDTs) are 0 days in pigs and carp and 3 days in broilers. These results provided clear monitoring tools and technical standards to evaluate the food safety of quinocetone.
Acute and sub-chronic oral toxicological evaluations of quinocetone in Wistar rats.

Abstract:To provide a detailed toxicity with wide spectrum of doses for quinocetone, a new antimicrobial growth promoting agent, acute and sub-chronic toxicological studies were conducted. For acute study, quinocetone was administered singly by oral gavage to Wistar rats and Kunming mice. Calculated LD50 was 8687.31mg/kg b.w./day in rats and 15848.93mg/kg b.w./day in mice. In sub-chronic study, quinocetone was fed to Wistar rats at dietary levels of 0, 50, 300 and 1800mg/kg or olaquindox (300mg/kg), approximately equivalent to quinocetone 5, 30, 180 or olaquindox 30mg/kg b.w./day. There was significant decrease in body weight in both genders, total protein and creatinine in females and alkaline phosphatase in males fed with 1800mg/kg diet, while alkaline aminotransferase values decreased in all treated groups. Significant increase in relative weights of liver and kidneys in both genders and testis in male rats were noted at 1800mg/kg diet. Histopathological observations revealed that 1800mg/kg quinocetone diet and 300mg/kg olaquindox diet could induce proliferation of bile canaliculi in the portal area. In conclusion, quinocetone can induce hepatic histological changes as well as leaking of different serum enzymes. The no-observed-adverse-effect level of quinocetone was considered to be 300mg/kg diet.

Abstract:Quinocetone, a new quinoxaline 1,4-dioxide derivative, has been approved as an animal growth promoter in China since 2003. To investigate the genotoxicity of quinocetone in vitro, its effects on the extent of DNA injury in human hepatoma (HepG2) cells accompanied by chromosomal damage and genomic DNA alterations were tested. The cell viability test indicated that quinocetone inhibited cell proliferation as a function of dose and time. In the comet assay, significant DNA fragment migration was observed in a dose-dependent manner. A dose-dependent increase of the micronucleated (MN) cell frequency was shown in cytokinesis-block micronucleus (CBMN) test. The gain/loss of randomly amplified polymorphic DNA (RAPD) bands and the change of band intensity in RAPD profiles were obtained after HepG2 cells were exposed to quinocetone at concentrations of 1.25, 2.5 and 5μg/mL. The results demonstrated that quinocetone exerted genotoxic effects on HepG2 cells. Thus, the use of quinocetone as a growth promoter in animal feed should be seriously considered.

Abstract:Excretion, disposition, and metabolism of [3H]-quinocetone in rats, pigs, broilers, and carp following oral administration were investigated. After a single p.o. dose, total radioactivity was rapidly excreted, with ⩾94% in all species within 14days. Fecal excretion of radioactivity was 68% and 65% of the administered dose in rats and pigs, respectively, with the remainder excreted in the urine. Six hours after the last of seven daily oral administrations of 3H-labeled QCT, radioactivity was found to be distributed throughout all tissues, with the majority of radioactivity cleared within 7days, and elimination was the slowest from the liver and kidney. QCT was extensively metabolized in all of the species, and the primary changes included N–O group reduction, carbonyl group reduction, double bond reduction, and hydroxylation. The major tissue metabolites of QCT were Q2, Q4, Q5, Q8, and Q9 in rats; Q1, Q2, Q3, Q4, and Q5 in pigs; Q1, Q2, Q3, Q4, and Q7 in broilers; and Q1, Q2 in carp. This confirmed the potential link between QCT metabolism through N–O group reduction and its organ toxicity. The results of the present study provide important data that could help understand the relationship between the toxicities and metabolic disposition of QCT.

Identification of the Major Metabolites of Quinocetone in Swine Urine Using Ultra-Performance Liquid Chromatography/electrospray Ionization Quadrupole Time-of-flight Tandem Mass Spectrometry.

Abstract:Quinocetone (QCT), 3-methyl-2-cinnamoylquinoxaline-1,4-dioxide, is a quinoxaline-N,N-dioxide used in veterinary medicine as a feed additive. QCT is broadly used in China to promote animal growth, but few studies have been performed to reveal the metabolism of QCT in animals until now. In the present study, the metabolites of QCT in swine urine were investigated using ultra-performance liquid chromatography/electrospray ionization quadrupole time-of-flight mass spectrometry (UPLC/ESI-QTOF-MS). Multiple scans of metabolites in MS and MS/MS modes and accurate mass measurements were performed simultaneously through data-dependent acquisition. Most measured mass errors were less than +/-5 mDa for both protonated molecules and product ions using external mass calibration. The structures of metabolites and their product ions were easily and reliably characterized based on the accurate MS(2) spectra and known structure of QCT. As expected, extensive metabolism was observed in swine urine. Thirty-one metabolites were identified in swine urine, most of which were reported for the first time. The results reveal that the N-O group reduction at position 1 and the hydroxylation reaction occurring at the methyl group, the side chain or on the benzene ring are the main metabolic pathways of quinocetone in swine urine. There was abundant production of 1-desoxyquinocetone and hydroxylation metabolites of 1-desoxyquinocetone. The proposed metabolic pathway of quinocetone in vivo can be expected to play a key role in food safety evaluations.

Abstract:Quinocetone (3-methyl-2-quinoxalinbenzenevinylketo-1,4-dioxide; QCT) is a new promising antimicrobial growth promoter for quinoxalines. The identification of the major metabolites of QCT has resulted in a number of studies regarding its metabolic pathway. However, little is known about the systematic synthesis, characterization, and simultaneous determination of its metabolites. To obtain system data for the four main metabolites of QCT, a convenient synthesis of these compounds was performed. All synthesized compounds were characterized by infrared spectroscopy, nuclear magnetic resonance, and high-resolution mass spectroscopy. The theoretical NO bond dissociation enthalpies (BDEs) and octanol–water partition coefficient (Kow) were estimated. A cytotoxicity assay for these compounds in hepatocytes isolated from rats was proposed, and the cytotoxicity results were evaluated based on the calculated NO BDEs.

Abstract:To investigate the reproductive toxicity and teratogenic potential of quinocetone, a growth promoting agent, Wistar rats were fed different diets containing 0, 50, 300 and 1800mg/kg quinocetone or 300mg/kg olaquindox. Groups of 15 males and 30 females (F0) were fed through a 10-week prebreed period as well as during mating, gestation, parturition and lactation. At weaning, 12 males and 24 females of F1 generation weanlings per group were selected randomly as parents for F2 generation. Selected F1 weanlings were exposed to the same diet and treatment as their parents. At the highest quinocetone group, body weights in F0 and F1 rats, fetal body weight on day 21 after birth and number of viable fetuses in F0 and F1 generation significantly decreased. In teratogenicity study, groups of 12 males and 24 females were fed with the same diets through a 12-week prebreed period and matting periods. Pregnant rats were subjected to cesarean section on GD 20 for teratogenic examination. At the highest quinocetone group, body weights and feed efficiency, fetal body lengths, tail lengths, litter weights and number of viable fetuses significantly decreased. The NOAEL for reproduction/development of quinocetone for rats was estimated to be 300mg/kg diet.
Quinocetone-induced Nrf2/HO-1 pathway suppression aggravates hepatocyte damage of Sprague–Dawley rats

Abstract:Quinocetone (3-methyl-2-quinoxalin benzenevinylketo-1,4-dioxide, QCT) is a widely used veterinary drug in PR China that promotes feed efficiency and growth of various animals. However, its potential toxicity has been concerned recently. In the present study, we investigated QCT-induced hepatocyte changes and its related mechanism, especially the expression of Nrf2/HO-1 pathway. Oxidative stress induced by QCT in hepatocyte led to DNA damage, inflammation and apoptosis. Nevertheless, hepatocyte has a self-repair system to protect itself from oxidative stress. In the 50mg/kg/day QCT group, the morphology and function of liver were approximately maintained on normal level, which indicated that the damaged cell might have a self-repair mechanism. Notably, nuclear factor-erythroid 2-related factor 2/heme oxygenase-1 (Nrf2/HO-1) pathway plays a critical role in protecting cells against reactive oxygen species (ROS) generation. However, higher doses of QCT (800mg/kg/day and 2400mg/kg/day) inhibited the expression of Nrf2/HO-1 pathway, which resulted in excessive ROS generation and irreversible oxidative DNA damage, inflammation and apoptosis. In conclusion, although QCT-induced oxidative stress activates the expression of Nrf2/HO-1 pathway initially, persistent QCT exposure will inhibit this expression and aggravate hepatocyte damage. Simultaneously, inflammation and apoptosis continues to progress, liver dysfunction and tissue damage will be occurred eventually.
Comparative Study of the Toxicity Mechanisms of Quinolone Antibiotics on Soybean Seedlings: Insights from Molecular Docking and Transcriptomic Analysis.

Abstract:The ecological effects of quinolone antibiotics (QNs) on non-target organisms have received widespread attention. The toxicological mechanisms of three common QNs, that is, enrofloxacin, levofloxacin, and ciprofloxacin, on soybean seedlings were investigated in this study. Enrofloxacin and levofloxacin caused significant growth inhibition, ultra structural alterations, photosynthetic suppression, and stimulation of the antioxidant system, with levofloxacin exhibiting the strongest toxic effects. Ciprofloxacin (<1 mg & BULL;L-1) did not have a significant effect on the soybean seedlings. As the concentrations of enrofloxacin and levofloxacin increased, antioxidant enzyme activities, malondialdehyde content, and hydrogen peroxide levels also increased. Meanwhile, the chlorophyll content and chlorophyll fluorescence parameters decreased, indicating that the plants underwent oxidative stress and photosynthesis was suppressed. The cellular ultrastructure was also disrupted, which was manifested by swollen chloroplasts, increased starch granules, disintegration of plastoglobules, and mitochondrial degradation. The molecular docking results suggested that the QNs have an affinity for soybean target protein receptors (4TOP, 2IUJ, and 1FHF), with levofloxacin having the highest binding energy (-4.97, -3.08, -3.8, respectively). Transcriptomic analysis has shown that genes were upregulated under the enrofloxacin and levofloxacin treatments were mainly involved in ribosome metabolism and processes to synthesize oxidative stress-related proteins. Downregulated genes in the levofloxacin treatment were primarily enriched in photosynthesis-related pathways, indicating that levofloxacin significantly inhibited gene expression for photosynthesis. Genes expression level by quantitative real-time PCR analysis was consistent with the transcriptomic results. This study confirmed the toxic effect of QNs on soybean seedlings, and provided new insights into the environmental risks of antibiotics.

Abstract:Physiologically based pharmacokinetic (PBPK) models are scientific methods used to predict veterinary drug residues that may occur in food-producing animals, and which have powerful extrapolation ability. Quinocetone (QCT) and mequindox (MEQ) are widely used in China for the prevention of bacterial infections and promoting animal growth, but their abuse causes a potential threat to human health. In this study, a flow-limited PBPK model was developed to simulate simultaneously residue depletion of QCT and its marker residue dideoxyquinocetone (DQCT) in pigs. The model included compartments for blood, liver, kidney, muscle and fat and an extra compartment representing the other tissues. Physiological parameters were obtained from the literature. Plasma protein binding rates, renal clearances and tissue/plasma partition coefficients were determined by in vitro and in vivo experiments. The model was calibrated and validated with several pharmacokinetic and residue-depletion datasets from the literature. Sensitivity analysis and Monte Carlo simulations were incorporated into the PBPK model to estimate individual variation of residual concentrations. The PBPK model for MEQ, the congener compound of QCT, was built through cross-compound extrapolation based on the model for QCT. The QCT model accurately predicted the concentrations of QCT and DQCT in various tissues at most time points, especially the later time points. Correlation coefficients between predicted and measured values for all tissues were greater than 0.9. Monte Carlo simulations showed excellent consistency between estimated concentration distributions and measured data points. The extrapolation model also showed good predictive power. The present models contribute to improve the residue monitoring systems of QCT and MEQ, and provide evidence of the usefulness of PBPK model extrapolation for the same kinds of compounds.

Abstract:Quinoxaline-1,4-dioxides derivatives have been widely used as animal growth promoter. This study was conducted to investigate the cytotoxicity and genotoxicity of quinoxaline-1,4-dioxides derivatives, namely carbadox, olaquindox and quinocetone, in Vero cells. The cell viability results from MTT assay demonstrated the severe inhibitory effects by these chemicals in both dose and time dependent manner. Among these chemicals quinocetone exhibited the highest cytotoxicity followed by olaquindox and carbadox. DNA damage analyses using alkalic comet assay revealed pronounced increase of DNA fragmentation in all three compound treated cells. In contrast, DNA damage was significantly decreased after incubation with S9 mix. These findings suggest that the intermediate metabolites of these compounds exerted lower genotoxicity than their parent drugs. We further described chromosomal damage induced by these drugs employing cytokinesis-block micronucleus assay (MN assays). The micronucleus frequency was significantly higher in these drugs treated cells than that of controls and the nuclear division index was also markedly reduced with increasing drug concentration applied. Similar to the observation in comet assay, incorporation of S9 mix in the MN assays was able to markedly alleviate the chromosome damage. In conclusion, our results strengthened previous reports on the cytotoxicity and genotoxicity of carbadox, olaquindox and quinocetone.

Abstract:<p>Quinoline is a critical raw material for the dye, metallurgy, pharmaceutical, rubber, and agrochemical industries, and its use poses a serious threat to human health and the ecological environment. Quinoline has carcinogenic, teratogenic and mutagenic effects on the human body through food accumulation. However, due to the steric hindrance of its bicyclic fused structure and its long photooxidation half-life, quinoline is too difficult to decompose naturally. To date, numerous technologies have been used to degrade quinoline, whereas only a few have been reviewed. Therefore, this paper is focused on offering a comprehensive overview of the state of quinoline degradation in an effort to improve its degradation efficiency and fully utilize the carbon and nitrogen within quinoline without causing any damage to the environment. Accordingly, the strains, research progress and mechanisms of various methods for degrading quinoline are explored and elucidated in detail, especially quinoline biodegradation and the combination of these technologies for efficient removal. The state-of-the-art processes and new findings of our team on the biofortification of quinoline degradation are also presented. Finally, research bottlenecks and gaps for future research were identified along with the prospects and resource utilization of quinoline. These discussions facilitate the realization of the zero discharge of quinoline.</p>

Abstract:Quinocetone (QCT), a member of the quinoxaline 1,4-di-N-oxides (QdNOs) family, can cause genotoxicity and hepatotoxicity, however, the precise molecular mechanisms of QCT are unclear. This present study investigated the protective effect of quercetin on QCT-induced cytotoxicity and the underlying molecular mechanisms in human L02 and HepG2 cells. The results showed that quercetin treatment (at 7.5-30 mu M) significantly improved QCT-induced cytotoxicity and oxidative damage in human L02 and HepG2 cells. Meanwhile, quercetin treatment at 30 mu M significantly inhibited QCT-induced loss of mitochondrial membrane potential, an increase in the expression of the CytC protein and the Bax/Bcl-2 ratio, and an increase in caspases-9 and -3 activity, and finally improved cell apoptosis. Quercetin pretreatment promoted the expression of the phosphorylation of p38, Nrf2, and HO-1 proteins. Pharmacological inhibition of p38 significantly inhibited quercetin-mediated activation of the Nrf2/HO-1 pathway. Consistently, pharmacological inhibitions of the Nrf2 or p38 pathways both promoted QCT-induced cytotoxicity and partly abolished the protective effects of quercetin. In conclusion, for the first time, our results reveal that quercetin could improve QCT-induced cytotoxicity and apoptosis by activating the p38/Nrf2/HO-1 pathway and inhibiting the ROS/mitochondrial apoptotic pathway. Our study highlights that quercetin may be a promising candidate for preventing QdNOs-induced cytotoxicity in humans or animals.

Abstract:Used heavily in food animal production, quinolones occur widely in food products of animal origin. Development of highly sensitive and selective analytical techniques for the detection of quinolone residues, often at trace levels, in food samples is necessary to ensure food safety and understand their public health risk. With complex matrices, food samples typically require a series of pre-treatment steps, such as powdering, homogenization, deproteinization, and filtration. This review summarizes the recent advances in extraction, concentration, and detection techniques for quinolones in food product samples, and briefly compares the advantages and limitations of major techniques. Recent development of quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method and immunoassay- and biosensor-based detection methods for the determination of quinolones residues in food products is discussed in details. A perspective on the trends and needs of future research is also presented.

Abstract:Quinocetone (QCT), a new antimicrobial growth promotant of quinoxalines, can effectively improve the growth and feed efficiency of food animals with more safety than is provided by olaquindox and carbadox. To clarify its metabolism and residue levels in animals, a liquid chromatographic method with UV-Vis detection was developed for the determination of QCT and its main metabolites, desoxyquinocetone (DQCT) and 3-methylquinoxaline-2-carboxylic acid (MQCA), in muscle, liver, kidney, and fat of swine and chicken. For sample pretreatment, QCT and DQCT were extracted with ethyl acetate and purified with iso-octane; after alkaline hydrolysis of the tissue, MQCA was extracted with ethyl acetate and citric acid buffer (pH 6.0), and the extract was purified over a cation-exchange column (AG MP-50 resin). Detection was at 312 and 320 nm for QCT and DQCT, respectively, and at 320 nm for MQCA. The observed limit of detection for the 3 compounds was 0.025 microg/g in various tissues. The methods were linear over the concentrations range of 0.01-0.64 microg/mL with mean recoveries of approximately 71-86% and relative standard deviations of about 4-12% at the levels of 0.05, 0.10, and 0.20 microg/g. The method is highly selective and can be applied to the determination of QCT and its main metabolites in animal tissues, which would accelerate the pharmacokinetic and residue study of QCT in food animals.

Abstract:We examined the tissue distribution and elimination of quinocetone (QCT) and its major metabolites 1-desoxyquinocetone (1-DQCT), di-desoxyquinocetone (BDQCT), and 3-methyl-quinoxaline-2-carboxylic (MQCA) in ducks. The analytes were simultaneously quantitated using a UPLC-MS/MS method after oral administration of QCT at 100 mg·kg day for 7 days. We found that QCT and its major metabolites were widely distributed in duck tissues. The concentrations indicated that the primary compound in the liver, kidney, and heart was MQCA and the primary compound in the stomach, intestine, spleen, and lung was QCT. We also identified that MQCA was the most appropriate compound for QCT residue monitoring. The liver and kidney are the primary QCT target organs in ducks, and this study provides clear monitoring tools and important data to evaluate its safety.
