Immunoregulatory Roles of T Cells Vaccine in Collagen Induced Arthritis

Abstract:The study aims to explore the immunoregulatory roles of T cells vaccine(TCV) on collagen-induced arthritis.DBA/1 mice were treated with TCV,and then used to establish collagen induced arthritis(CIA).After disease onset,the incidence,severity,and joint histopathology were evaluated.We detected the percentage of Th subsets such as Treg,Th1 and Th17.ELISA assay was used to detect the levels of related cytokines in the culture supernatants.Meanwhile,the mRNA expression levels of transcription factors of those T cells were determined by real-time quantitative PCR.Mice in the TCV-treated group showed alleviated disease symptoms,and less infiltration of inflammatory cells in the joint sections.The proliferation of CII-specific cells and the levels of anti-CII antibodies was inhibited in the TCV treatment group.TCV treatment also increased the percentage of Treg cells,but decreased the percentage of Th1 and Th17.The results of this study indicated that TCV treatment can reduce the incidence and severity of CIA by up-regulating Treg and down-regulating Th1 and Th17.
