Abstract:Carcinogenic Sudan I has been added illegally into spices for an apparent freshness. 1 H solution and solid-state (SS) nuclear magnetic resonance (NMR) spectroscopies were applied and compared for determination of Sudan I in paprika powders (PPs). For solution NMR, PPs spiked with Sudan I were extracted with acetonitrile, centrifuged, rotor-evaporated, and re-dissolved in DMSO-d6 for spectral collection. For SSNMR, Sudan I contaminated PPs were mixed with DMSO-d6 solution and used for spectral collection. Linear regression models constructed for quantitative analyses resulted in the average accuracies for unknown samples as 98% and 105%, respectively. Limits of detection for the solution NMR and SSNMR spectrometers were 6.7 and 128.6 mg kg −1 , while the limits of quantification were 22.5 and 313.7 mg kg −1 . The overall analysis time required by both methods was similar (35 and 32 min). Both NMR techniques are feasible for rapid and accurate determination of Sudan I adulteration in PPs.

Abstract:One hundred random samples of chili products and other related foods either local or imported (spices, mixtures of spices, tomato sauces, dressing, crisps, oils ,pickles and sausages) were collected from retail Egyptian market and analyzed for the presence of Sudan dyes by HPLC procedure . Sudan dyes have been found in a large number of food products samples containing contaminated chilli powder. The prevalence of illegal dyes were detected in a very high percentage of food products out of one hundred samples tested, more than forty five samples were found to contain detectable levels of illegal dyes. Sudan I (Sudan Red) was found as a highest ratio in red hot chili powder (220 mg/kg), also Sudan IV (Scarlet Red) was achieved the highest ratio (1120 mg/kg) in the another chilli powder sample and 326 mg /kg for Sudan II in paprika . Sudan III was detected as the highest ratio in Indian Curry paste sample(165 mg/kg) .

Rapid analysis of four Sudan dyes using direct analysis in real time-mass spectrometry

Abstract:A simple direct analysis in a real time-mass spectrometry (DART-MS) method was developed for the rapid determination of four Sudan dyes (I-IV) in chili powder. Simple liquid extraction by hexane without further clean-up was used for sample preparation. DART parameters were systematically optimized to achieve the best detection performance. A DIP-it sampler was used for automatic sampling. The matrix effect was measured by comparing the limit of detection (LOD) in matrix solution with that in pure organic solution. Eventually, the identification of the Sudan dyes was confirmed by MS/MS results and the LODs for four analytes in matrix solution were similar to 0.5 mu g mL(-1). The method showed good linearity with correlation coefficients (R-2) greater than 0.99 for concentrations ranging from 1 to 20 mu g mL(-1). The whole analytical process could be completed within 15 minutes with good recoveries (88-116%) and satisfactory repeatability (<26%, n = 3).

Abstract:Analytical techniques as strong, precise, and expensive are necessary for monitoring food and water safety for contaminants, microorganisms, and allergies that might be harmful if used. Sudan dyes are commonly utilized as an ingredient in food dye substances and a variety of industrial items. These colors are classified as three carcinogens and are linked to liver and bladder cancers. They are not authorized for human consumption by the International Agency for Research on Cancer (IARC) and are not permitted to be used by the Food Standards Agency or the European Union. This article describes electrochemical dye analysis beside the numerous electrochemical sensors utilized to identify these dyes as a food colorant and water. As a result, the qualities, chemistry, and toxicity of dyes as food colorants and industrial goods in Sudan have been investigated in this study. Sudan dyes have been thoroughly studied, and many electrochemical sensors have been developed to define and monitor these dyes in food colorants. As a result, current electrochemical sensors have been found to be neither mass-production nor cost-effective. Mostly, the synthesis of high-performance materials needs high knowledge, and the production of electrode surfaces is remained difficult due to labor-intensive and time-consuming activities.

[Rapid Determination of Sudan Dyes Adulterated in Natural Paprika Red by FTIR].

Abstract:How to rapidly determine synthesized pigments or dyes adulterated in natural pigments is a difficult problem for food analysts. Natural paprika red is widely used in foods because of its coloration and pharmaceutical effect, but it is sometimes adulterated with Sudan dyes for its poor stability under light or heat treatment. In the present paper, the macro-fingerprint characteristic of infrared spectroscopy was utilized to identify Sudan dyes adulterated in paprika red. The strong peaks at 1 621, 1 500 and 751 cm(-1) in FTIR spectra and at the fingerprint region of 753, 684 and 496 cm(-1) in the secondary deriative FTIR spectra were remarkable characteristics for Sudan dyes adulterated in paprika red, of which the limit of determination was about 1%.

Abstract:The addition of Sudan dyes with carcinogenic effects to food threatens human health. Herein, a ratiometric fluorescence strip consisting of core-shell upconversion particles (NaYF4:Yb,Tm@NaYF4:Yb,Er), metal-organic frameworks and dual-template molecularly imprinted polymers was developed to selectively and sensitively detect four Sudan dyes based on inner filter effect (detection time only takes 8 min). The high adsorption capacity of metal-organic frameworks and the greater overlap between the emission of NaYF4:Yb,Tm@NaYF4:Yb,Er and the absorbance of four Sudan dyes enable the signal responses to be more sensitive. The limits of detection in chilli powder samples are as low as 29.87 ng/g, 37.55 ng/g, 47.89 ng/g and 51.02 ng/g, with satisfactory recovery (93.32-103.4%) and minor relative standard deviations (≤4.3%). This method broadens the idea for low-cost and portable detection of multiple illegal additives in complex substrates with high selectivity and sensitivity based on one kind of fluorescent strip.

Abstract:A novel kind of magnetic porous carbon nano-fibers (Fe3O4@P-CNFs) materials was successfully prepared and used as an adsorbent. Based on the above-mentioned adsorbent, a simple and effective magnetic disperse solid-phase extraction (MSPE) method was developed and first utilized to the enrichment and purification of five Sudan dyes (including Sudan I, Sudan II, Sudan III, Sudan IV, and Sudan Red 7B) in foodstuffs for the first time. High-performance liquid chromatography was used to determine the content of the Sudan dyes. The parameters affecting the extraction performance were studied and optimized, including the amount of the adsorbent and inorganic salt, type and the volume of the eluent, pH of the sample solution and extraction time. Under the optimized experimental conditions, the results show that the proposed method has a good linear relationship (r ≥ 0.9993). The limits of detection range from 0.88 μg L−1 to 1.27 μg L−1. The recoveries range from 86.6% to 99.7% with the relative standard deviations ranging from 0.6% to 7.9% in the methodology validation. The above-mentioned results indicate that the proposed method is a sensitive and reliable procedure with good reproducibility for the detection of Sudan dyes residues in foodstuffs.

Abstract:A simple high-performance liquid chromatography method was developed for quantitative determination of para red, Sudan I, Sudan II, Sudan III, Sudan IV, canthaxanthin, and astaxanthin in feedstuff. The sample was extracted using acetonitrile and cleaned up on a C(18) SPE column. The residues were analyzed using ultra-performance liquid chromatography coupled to a diode array detector at 500 nm. The mobile phase was acetonitrile-formic acid-water with a gradient elution condition. The external standard curves were calibrated. The mean recoveries of the seven colorants were 62.7-91.0% with relative standard deviation 2.6-10.4% (intra-day) and 4.0-13.2% (inter-day). The detection limits were in the range of 0.006-0.02 mg/kg.

Abstract:Sudan dyes are strictly prohibited from being added to edible products as carcinogens and tetracycline hydrochloride (TC) remaining in animal-derived food may cause harm to the human body. Therefore, it is necessary to establish a high-sensitivity, simple and convenient method for the detection of Sudan dyes and TC in foods for safety purposes. In this work, multifunctional blue fluorescent carbon dots (B-CDs) were prepared by a one-step hydrothermal synthesis using glucose as the carbon source. The results show that the fluorescence intensity of B-CDs was significantly affected by the acidity of the solution and can be quenched by Sudan I, IV and TC through selective studies. Interestingly, the fluorescence quenching intensities of B-CDs have a good linear relationship with the concentration of Sudan I and IV at pH = 3–7. The wide range of pH is beneficial to broaden the application of B-CDs in a practical samples analysis. The method has been successfully applied to real food samples of tomato paste, palm oil and honey, and the detection limits are 26.3 nM, 54.2 nM and 31.1 nM for Sudan I, Sudan IV and TC, respectively. This method integrates Sudan dyes and TC into the same multifunctional B-CDs, which shows that the sensor has a great potential in food safety detection.

Preparation of new hydrophobic deep eutectic solvents and their application in dispersive liquid-liquid microextraction of Sudan dyes from food samples

Abstract:In this work, a new generation of hydrophobic deep eutectic solvents (DESs) was prepared using eugenol (as hydrogen bond donor) and benzyltriethylammonium bromide, benzyltributylammonium bromide, benzyltriethylammonium chloride and benzyltributylammonium chloride (as hydrogen bond acceptor) in different molar ratios. These DESs were applied to vortex-assisted dispersive liquid-liquid microextraction of Sudan dyes from food samples, followed by high-performance liquid chromatographic determination. The influencing parameters, including the type of DES, amount of DES, extraction time, solution pH and salt addition, were investigated and optimized. Under the optimized conditions, a linear range of 2-1000 ng mL-1 with determination coefficients of <0.999 was obtained. Limits of detection and limits of quantification were in the range of 0.5 to 1 ng mL-1 and 2 to 3 ng mL-1, respectively. The proposed method was successfully used in the determination of Sudan dyes in chili sauce, chili powder and ketchup, and satisfactory recoveries of between 89.9 and 119.3% were obtained, with relative standard deviations in the range of 0.1-6.8%. The proposed method is simple, green and efficient, and can be applied to determine Sudan dyes in complex matrices.

Abstract:The new method of determining banned additives in the food such as Sudan red Ⅰ and Ⅱ was established with thin layer chromatography（TLC） and high performance liquid chromatography（HPLC） two methods combined.Sudan red and impurities can be separated by silica gel plate.Under the condition of chromatography in setting,the concentration in the 0.1~20.0 μg/mL range,the peak areas（Y） and concentration（X） present good linear relationship,Sudan red Ⅰ,Ⅱ regression equations Y1=-18785＋138962X,Y2=-2623＋131432X.Correlation coefficient R1=0.9997,R2=0.9998.Its application of determing the Sudan red Ⅰ and Ⅱ in chili powder,chili oil,and tomato sauce was studied,and got the detection limit of 0.024 μg/g,0.027 μg/g,respectively.

Abstract:Objective:To establish a quick,accurate,sensitive method for analysis of Sudan I,Sudan II,Sudan III and Sudan IV in foodstuffs by ultra performance liquid chromatography-tandem mass spectrometry.Methods:The ultra performance liquid chromatography-quadrupole mass spectrometry method was used.The mobile phase consisted of acetronitrile-water,at a flow rate of 0.3 ml/min,with analysis time of 5 min.Electrospay ionization(ESI) source was applied and operated in the positive ion mode.The multiple reaction monitoring(MRM) mode using the transition of m/z 249→93,277→120,353→197,381→224 was used to quantify Sudan I,Sudan II,Sudan III and Sudan IV in foodstuffs.Results:The assay was linear from 1.1616~290.4,1.2608~315.2,1.184~296.0,1.1664~291.6 ng/ml for Sudan I,Sudan II,Sudan III and Sudan IV,respectively.Sudan I,Sudan II,Sudan III and Sudan IV were found in three of twenty-five food samples.Conclusion:The method is selective and sensitive and can be used to detect and determine the contents of Sudan I,Sudan II,Sudan III and Sudan IV in foodstuffs.
Ultrasensitive Detection of Sudan I in Food Samples by a Quantitative Immunochromatographic Assay

Abstract:An immunochromatographic assay (ICA) based on surface-enhanced Raman scattering (SERS) for ultrasensitive determination of Sudan I in food samples was reported. Gold-silver core-shell bimetallic nanorods (referred to as Au@Ag NRs) were synthesized, characterized and used as the substrate for preparation of the ICA. Polyclonal antibody against Sudan I was immobilized on the surface of the Au@Ag NRs carrying the Raman reporter 5,5'-dithiobis (2-nitrobenzoic acid). The Raman scattering intensity on the test line was used for quantitation of Sudan I. The assay was completed in 15 min. IC50 and limit of detection (LOD) were 30 pg mL(-1) and 0.2 pg mL(-1), respectively. There was no cross-reactivity (CR) of the assay with Sunset Yellow, Lemon Yellow and Brilliant blue FCF, but only 3.53%-9.74% CR with Sudan II, III and IV. The recoveries of Sudan I from spiked food samples were in the range of 88.9-107.6% with relative standard deviation of 3.7-8.7% (n=3).

Abstract:The health threat from Sudan red dyes has been the subject of much attention in recent years and is crucial to design and establish reliable measurement technologies. In the present study, a new magnetic nanomaterial, polyamidoamine dendrimer-modified magnetic nanoparticles (Gn-MNPs), was synthesized and characterized. The nanomaterials had good adsorption capacity for Sudan dyes from natural waters. G1.5-MNPs possessed excellent adsorption capacity and a linear adsorption relationship over the range from 0.02 to 300 μg L −1 of Sudan dyes with relative coefficients all larger than 0.996. The sensitivity of the proposed method was excellent with detection limits over the range from 1.8 to 5.5 ng L −1 and the precision was less than 3.0%. G1.5-MNPs showed a remarkable application potential for the enrichment of trace environment pollutants in aqueous samples and the developed method based on this nanomaterial could be a robust and reliable alternative tool for routine monitoring of such pollutants.

Abstract:A simple, rapid, inexpensive and robust solid phase extraction (SPE) cleanup method coupled with high performance liquid chromatography–diode array detector (HPLC–DAD) for the determination of four Sudan dyes in chili products was developed. The interference substances were removed completely by normal SPE-Alumina N, resulting in a clean and interference-free baseline. Without the use of expensive LCMS/MS instrument the low levels of Sudan dyes were identified and quantified in chili foods with good precision and recovery. Depending on the Sudan dyes involved, the limit of determination (LOD) and limit of quantification (LOQ) were in the range of 4.1–5.8 and 13.2–19.1μg/kg, respectively. The recovery, repeatability and reproducibility varied from 93.2% to 103%, from 0.3% to 4.4%, and from 0.6% to 4.9%, respectively. This method is suitable for the routine analysis of Sudan dyes for the common factory laboratory due to its sensitivity, simplicity, convenience and low-cost.

Abstract:Novel self-assembled water-soluble nanomicelles that contain fluorescent conjugated polymers (poly(9,9-dioctylfluorene) (PFO) or poly[2,7-(9,9-dihexylfluorene)-alt-4,4'-phenylether] (PF-PE)) have been obtained and used as the highly sensitive/selective platform for Sudan dye detection. The Fluorescent nanomicelles exhibited a highly selective fluorescence quenching by the prohibited food additive Sudan 1, while not for the natural pigments: Capsanthin and Betacarotene, due to the more suitable matching of the LUMOs (lowest unoccupied molecular orbital) of the conjugated polymers with that of Sudan I molecules. The Stern-Volmer constants (K-SV) of PF-PE/F127 and PFO/F127 for Sudan I were 1 040 480 and 665 000 M-1, respectively, which were more than 100 times higher than those of the same conjugated polymers in the orgainc solvents. The significantly enhanced sensitivity was due to the collective effect of the F127 micelles to both chromophore and analyte, through which the fluorophone analyte binding interaction is significantly strengthened and efficient photoinduced charge transfer occurs. The as-proposed materials and approach may be potentially applied in the real-time food safety screening.

Abstract:In this manuscript, a novel strategy to design and prepare magnetic molecularly imprinted polymers (MMIPs) is proposed. Based on the synthesized MMIPs, a fluorescent sensor for low-cost and fast detection of Sudan I was established. Under the optimal conditions, the fluorescence intensity of MMIPs decreased linearly with the concentration of Sudan I in the range of 1–40 μM, and the detection limit was 0.38 μM. The present fluorescent sensor demonstrated satisfactory performance for the quantification of Sudan I in chili powder, covering a concentration range of 49.66–993.12 mg kg −1 . The LOD was determined to be 17.38 mg kg −1 . Furthermore, the average percentage recovery for both intraday and inter-day precision studies ranged from 98% to 104% and 99% to 105%, respectively. The experimental results indicate that the prepared MMIPs exhibited broad potential for low-cost and fast detection in food safety analysis.

Abstract:Ionic liquid/CdO/NPs carbon paste electrode (IL/CdO/NPs/CPE) has been used as a high sensitive sensor for the professional quantitative determination of sudan I in food samples in aqueous solution. In the first step, CdO/NPs synthesis using chemical precipitation method and characterized with different methods such as transmission electron microscopy (TEM) and X-ray diffraction (XRD). The propose sensor shows a better electrochemical response with lower over-potential and high sensitivity for sudan I compared with unmodified carbon paste electrode using linear sweep voltammetry (LSV). The electro-oxidation of sudan I occurred in a pH-dependent e(-) and H+ process, and the electrode reaction followed a diffusion-controlled pathway. Under the optimum conditions in square wave voltammetry (SWV), the voltammetric oxidation peak current of sudan I showed linear dynamic ranges with a detection limit of 0.05 mu M for sudan I. The modified electrode was successfully used for the determination of the analytes in real samples with satisfactory result.
