Abstract:Heterocyclic ketene aminals bearing a secondary amino moiety acted as hetero-ene components to react with a number of enones under very mild conditions. The reaction of five- and six-membered heterocyclic ketene aminals 5 and 8 with methyl vinyl ketone proceeded effectively through the aza-ene addition, imine-enamine tautomerization and intramolecular cyclization to give good yields of 8-aroyl-5-hydroxy-5-methyl-1,2,3,5,6,7-hexahydroimidazo[1,2-a]pyridines 6 and 9-aroyl-6-hydroxy-6-methyl-1,2,3,4,7,8-hexahydroxy-6H-pyrido[1,2-a]pyrimidines 9, respectively. When methyl vinyl ketone was present in excess, preliminarily formed product 6 underwent a second aza-ene reaction followed by intramolecular cyclization and water-participating debenzoylation, to yield the cisoid-4,9-dihydroxy-4,9-dimethyl-1,2,5,6,6a,7,8,10 alpha-octahydro-4H,9H-imidazo[1,2,3-ij][1,8]naphthyridin-10 alpha-ylium benzoate 7 as the sole product. In refluxing acetonitrile, heterocyclic ketene aminals 5 underwent aza-ene addition and cyclocondensation reaction with a number of substituted alpha,beta-unsaturated ketones to afford 1,2,3,7-tetrahydroimidazo[1,2-a]pyridine derivatives 23 while tricyclic imidazo[1,2-a]quinoline derivatives were easily obtained when 2-benzylidene-cyclohexanone and -cyclohexane-1,3-dione were used as enophiles. The effect of the structures of heterocyclic ketene aminals, particularly of the intramolecular hydrogen bond, on the aza-ene reactivity is also discussed.

Abstract:Reactions of heterocyclic ketene aminals with activated carbonyl compounds are strongly influenced by the structure of the heterocycle moiety. Six-membered heterocyclic ketene aminals with or without an N-methyl group such as 3 and 4 underwent efficient addition and consecutive cyclocondensation reactions with diethyl oxomalonate 9 to produce gamma-lactam-fused pyrimidine derivatives 10 and 11 in moderate to excellent yield. For the five-membered enediamines, however, those compounds without any N-substituent such as 6 underwent the same reaction slowly and no reaction at all was observed with N-methyl and N,N'-dimethyl-substituted analogs 7 and 8. Heterocyclic ketene aminals 3 also reacted with glyoxylic acid esters 17 to afford 8-aroyl-7-hydroxy-6-oxo-1,2,3,4,6,7-hexa pyrrolo[1,2-a]pyrimidines 18 in good yield. No asymmetric induction-was found, however, when (1R,2S,5R)-(-)-menthyl glyoxylate 17b was employed as a chiral substrate. An aza-ene reaction mechanism involving heterocyclic ketene aminals as the aza-ene component (H-N-C=C) has been proposed. The effects of intramolecular hydrogen bonding and heterocyclic ring size on the reactivity are also discussed.

Abstract:Cu-II-catalysed diazo transfer and Cu-I-catalysed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) "click chemistry" were used to synthesis C7N aminocyclitol-derivatised 1,2,3-triazoles. In the course of this work, the -N=N- moiety was transferred onto C7N aminocyclitols such as validamine, valienamine and valiolamine by employing imidazole-1-sulfonyl azide as the diazo transfer reagent with catalysis by Cu-II, Zn-II and Ni-II, in moderate to good yields. The obtained azidocyclitols were coupled with various terminal alkynes under modified Meldal's conditions with good to excellent yields. The stereo- and regiochemistry of the products were confirmed by 2D-NMR (NOESY and HMBC). One-pot syntheses of the corresponding 1,2,3-triazoles, as safer and more efficient procedures, were also investigated and gave moderate to good yields.

Abstract:Heterocyclic ketene aminals having at least one secondary amino group underwent the aza–ene reaction with ethyl propiolate under various conditions to yield the corresponding adducts, which were readily transformed into the δ-lactam fused heterocyclic products with or without the aid of sodium ethoxide in refluxing ethanol, while the ester- and cyano-substituted tertiary enediamines acted as the strong Michael donor to add to ethyl propiolate in a polar or a protic solvent. The unusual substituent effect on the reactivity and mechanism was discussed.

Abstract:The 1-aza-2-azoniaallene salts, generated from alpha-chlotoazo compounds by treatment with halophilic Lewis acids, undergo intramolecular C-H amination reactions to form pyrazolines in good to excellent yields. This intra-molecular amination occurs readily at :both benzylic and tertiary aliphatic positions and proceeds at an enantio enriched chiral center with retention Of stereochemistry. Competition experiments show that insertion occurs more readily At an electron-rich benzylic position than it does at an electron-deficient one The C-H amination reaction occurs only with certain tethers connecting the heteroallene cation and the pendant aryl groups. With a longer tether or when the reaction is intermolecular, electrophilic aromatic substitution occurs instead of C-H amination. The mechanism and origins of stereospecificity and chemoselectivity were explored with density functional theory (B3LYP and MO6-2X). The 1-aza-2-azoniaallene cation undergoes C-H amination through a hydride transfer transition state to form the N-H bond, and the subsequent C-N bond formation occurs spontaneously to generate the heterocyclic product. This concerted two-stage mechanism was shown by IRC and quasi-classical molecular dynamics trajectory studies.

Abstract:A regio- and enantioselective formal [4 + 3] annulation reaction between enals and in situ formed azoalkenes has been achieved. A diverse set of 1,2-diazepine derivatives were synthesized in good yields with excellent enantioselectivities (often 99% ee). Alternatively, modifying the standard NHC catalyst switched the reactivity toward a formal [4 + 1] annulation to afford functionalized pyrazoles. The electronic and steric properties of the N-heterocyclic carbene organocatalyst play a vital role in controlling the reaction pathway (homoenolate vs acyl-anion reactivity of enal), allowing selective access to diverse 1,2-diazepine and pyrazole derivatives from identical substrates.

Abstract:A direct dearomative [4 + 2] annulation of electron-poor N-heteroarenes with azoalkenes generated in situ from α-halogeno hydrazones was developed under mild conditions. Accordingly, a series of fused polycyclic tetrahydro-1,2,4-triazines with potential biological activity were obtained in up to 96% yield. Various α-halogeno hydrazones and N-heteroarenes, such as pyridines, quinolines, isoquinolines, phenanthridine, and benzothiazole, were tolerated by this reaction. The general applicability of this method was shown by upscale synthesis and product derivatization.

Abstract:A synthetic approach to the previously unknown thieno[2,3-f][1,2,4]triazolo[1,5-a]azepine derivatives was efficiently accomplished by cycloaddition of the bicyclic 1-aza-2-azoniaallene salts 6, derived from 6,7-dihydrobenzo[b]thiophen-4(5H)-one hydrazones 4, to the triple bond of nitriles, followed by ring enlargement. The X-ray crystal diffraction analysis of the picrate 10a provided affirmative proof for the structural assignment made.

Abstract:A number of enamine-functionalized 1,2,3-triazole derivatives have been prepared via the Cu-catalyzed three-component reaction of terminal alkyne, azide, and 2H-azirine. The reaction proceeds through insertion of vinyl nitrene into the C-Cu bond of the triazolyl-Cu species, providing an efficient and step- and atom-economic approach to the enamine-bearing polysubstituted 1,2,3-triazoles. The resulting triazoles were easily transformed to trisubstituted pyrazoles in the presence of a Rh catalyst.

Abstract:Chiral azaarene compounds are extremely important due to their prevalence in pharmaceutical ingredients. Herein, an array of chiral molecules bearing azaaryl groups is synthesized in moderate-to-excellent yields with moderate-to-excellent Z/E ratios, high dr, and excellent enantioselectivity by a copper(I)-catalyzed asymmetric conjugate addition of 1,4-dienes to (E)-β-substituted alkenyl azaarenes. The reaction is carried out under mild proton-transfer conditions, which enjoys very high atom economy. Moreover, the reaction features a broad substrate scope on (E)-α,β-unsaturated azaarenes as various azaarenes are well tolerated, such as benzothiazole, thiazole, N-methyl-benzimidazole, benzoxazole, quinoline, isoquinoline, pyrimidine, pyrazine, and triazine. Interestingly, the reaction with (Z)-α,β-unsaturated azaarenes affords the same products in excellent results but with a reversed absolute configuration. DFT calculations indicate that the C-C bond-forming nucleophilic addition is a Z-/E- and enantio-selectivities-determining step and provides a rationale for the origin of selectivities. At last, the synthetic utilities of the product are showcased by several transformations, including olefin metathesis, [4 + 2] cyclization, [2 + 1] cyclization, and cleavage of the benzothiazole ring.

Abstract:The regiospecific allylation of benzoyl-substituted heterocyclic ketene aminals 2 is reported. Compound 2 reacted with allyl bromide in refluxing acetonitrile to give, exclusively, C-allylated products 5 in moderate to goad yields, while only N-allylated compounds 7 were obtained in dimethylformamide with the use of sodium hydride. Under neutral conditions, reaction between 2 and allyl bromide proceeded via a six-membered transition state involving the secondary enamine segment (HNC=C) and carbon-bromine bond (C-Br). Without the secondary amino group, C-allylation was inhibited, which was illustrated by the fact that 1,3-dimethyl-2-(aroylmethylene)imidazolidines did not react with allyl bromide. No 3-aza-Cope rearrangement of N-allylated heterocyclic ketene aminals 7 was observed at temperatures up to 140 degrees C. Only with the use of zinc chloride did the rearrangement take place at 140 degrees C to give allyl-shifted products 5. N-Benzylated heterocyclic ketene aminal analogues did not undergo the rearrangement under the same conditions, indicating that the 3-aza-Cope rearrangement proceeds through a charge-accelerated concerted mechanism.

Abstract:We report a Cu(II)-catalyzed C-H amidation/cyclization of azomethine imines with dioxazolones as acyl nitrene transfer reagents under additive- and ligand-free conditions. An array of 1,2,4-triazolo[1,5-a]pyridine derivatives were afforded in moderate to good yields with excellent functional group tolerance. In addition, scale-up reaction and photoluminescence properties were discussed.

Abstract:1,2,3-Triazines and 1,2,3,5-tetrazines react rapidly, efficiently, and selectively with amidines to form pyrimidines/1,3,5-triazines, exhibiting an orthogonal reactivity with 1,2,4,5-tetrazine-based conjugation chemistry. Whereas the mechanism of the reaction of the isomeric 1,2,4-triazines and 1,2,4,5-tetrazines with alkenes is well understood, the mechanism of the 1,2,3-triazine/1,2,3,5-tetrazine-amidine reaction as well as its intrinsic reactivity remains underexplored. By using 15N-labeling, kinetic investigations, and kinetic isotope effect studies, complemented by extensive computational investigations, we show that this reaction proceeds through an addition/N2 elimination/cyclization pathway, rather than the generally expected concerted or stepwise Diels-Alder/retro Diels-Alder sequence. The rate-limiting step in this transformation is the initial nucleophilic attack of an amidine on azine C4, with a subsequent energetically favored N2 elimination step compared with a disfavored stepwise formation of a Diels-Alder cycloadduct. The proposed reaction mechanism is in agreement with experimental and computational results, which explains the observed reactivity of 1,2,3-triazines and 1,2,3,5-tetrazines with amidines.
