Abstract:Glutamine is a major energy source for rapidly dividing cells, such as hematopoietic stem cells and cancer cells. Reliance on glutamine is therefore regarded as a metabolic hallmark of proliferating cells. Moreover, reprogramming glutamine metabolism by various factors, including tissue type, microenvironment, prooncogenes, and tumor suppressor genes, can facilitate stem cell fate decisions, tumor recurrence, and drug resistance. However, the significance of glutamine metabolism in cardiomyocytes, an end-differentiated cell type, is not fully understood. Existing evidence suggests important roles of glutamine metabolism in the development of cardiovascular diseases. In this review, we have focused on glutaminolysis and its regulatory network in proliferating cells. We have summarized current findings about the role of glutamine utilization in cardiomyocytes and have discussed possibilities of targeting glutamine metabolism for the treatment of cardiovascular diseases. Effects of Glutamine on the Growth Performance and Intestinal Mucous Morphology in Early Weaned Piglets

Abstract:Seventy-two piglets weaned at 21d were allocated to two treatments,each of which had 3 replicates,12 piglets per replicate.The control treatment was fed basal diet,while the experimental group was fed basal diet supplemented with 1% glutamine(Gln).The experiment lasted for 21days.The results showed that 1% Gln in diets could significantly improve growth performance and relative intestinal weight at 7d post weaning.Piglets fed with glutamine had significantly higher intestinal villus height and lower crypt depth at 7d post weaning,but at 14d post weaning,the effects of Gln on growth performance and intestinal mucous development were not significant.

Abstract:The intestinal barrier integrity is essential for the absorption of nutrients and health in humans and animals. Dysfunction of the mucosal barrier is associated with increased gut permeability and development of multiple gastrointestinal diseases. Recent studies highlighted a critical role for glutamine, which had been traditionally considered as a nutritionally non-essential amino acid, in activating the mammalian target of rapamycin cell signaling in enterocytes. In addition, glutamine has been reported to enhance intestinal and whole-body growth, to promote enterocyte proliferation and survival, and to regulate intestinal barrier function in injury, infection, weaning stress, and other catabolic conditions. Mechanistically, these effects were mediated by maintaining the intracellular redox status and regulating expression of genes associated with various signaling pathways. Furthermore, glutamine stimulates growth of the small intestinal mucosa in young animals and also enhances ion transport by the gut in neonates and adults. Growing evidence supports the notion that glutamine is a nutritionally essential amino acid for neonates and a conditionally essential amino acid for adults. Thus, as a functional amino acid with multiple key physiological roles, glutamine holds great promise in protecting the gut from atrophy and injury under various stress conditions in mammals and other animals.

Abstract:This experiment was conducted to study the effects of glutamine (Gln) and chitosan (CS) on small intestine disaccharidase activity, intestine villus height and faecal microflora of weanling piglets. One hundred and ninety-two 21-day-old weanling piglets with an average body weight of (5. 27 ± 0. 13) kg were randomly allocated into 4 treatments with 3 replicates in each treatment and 16 piglets in each replicate. The experiment which lasted for 14 days included two phases: 21～28 d and 29 ～ 35 d. The treatments were basal diet (CT), basal diet + 1. 00% glutamine (GlnT), basal diet + 0. 04% chitosan (CST) and basal diet + 1. 00% glutamine + 0. 04% chitosan (GCT), respectively. Results were as followed: 1) compared with CT, duodenum lactase and maltase activities, jejunum lactase and sucrase activities, ileum lactase and sucrase activities were significantly increased in GCT (P0. 05); 2) the intestine villus height from CST, GlnT to GCT was increased, but the differences between treatments were not significant (P0, 05); 3) the quantities of E. coli and Staphylococci in GCT decreased but those of Lactobacillus increased whereas the differences were not significant (P0. 05). These results indicated that the addition of 1.00% glutamine and 0. 04% chitosan significantly increased small intestine disaccharidase activities (P0. 05) and intestine villus height and improved faecal microflora, and the effects were better than single addition.
Effects of glutamine supplementation on growth performance and micro-morphology of small intestine of broilers

Abstract:Three hundred 1-day old broilers(Avian)were randomly divided into four treatments,each including three replicates of twenty-five birds.Control group was fed with basal diet,trial groups were fed with basal diet supplemented with 0.5 %,1.0 % and 1.5 % glutamine for 42 days.The effects of glutamine supplementation on growth performance and micro-morphology of small intestine of broilers were studied.The results indicated that dietary glutamine supplementation significantly increased the ADG and feed efficiency on broilers of earlier stage(P < 0.05).Compared with control group,supplementation of glutamine increased the villus height,decreased the depth of recess and crypt;duodenum and ileum were more sensitive to glutamine than jejunum(P < 0.05).It concluded that glutamine could improve micro-morphology of small intestine and growth performance of broilers.

Abstract:Glutamine has an essential role with a beneficial function in improving the nutrition status of young mammals. The influence of L-Glutamine (GM) on the Coefficient of Total Tract Apparent Digestibility (CTTAD) and Apparent Ileal Digestibility (AID), the jejunal enzyme activity associated with nutrient absorption and the energy production in weaned piglets has not been sufficiently studied. The aim of the present study is to provide a profile of the effects of Gln on CTTAD, AID, the activities of jejunal enzymes in connection with nutrient digestion and absorption and energy production. The piglets were weaned at 21 days of age. There were two groups in Experiment 1 and 2 representing supplementation with 0 or 1% Gln to the basal diet. In Experiment 1, the CTTAD of the dietary components and energy was assessed at 3, 5, 10, 15 and 30 days after weaning. In Experiment 2, productive performance, AID, jejunal enzyme activities and expression of Peroxisome Proliferator-Activated Receptor gamma (PPAR gamma) were measured at 10 and 30 days post-weaning. Results showed that dietary Gln supplementation significantly improved the CTTAD of DM, OM, GE and AA and the CTTAD increased significantly with the extension of days after weaning. For the entire experiment, the average daily gain increased by 12.40% (p = 0.049) in the Gin group. Dietary Gln supplementation increased the AID of GE, Leu, Lys, Cys and Pro by 12.50 (p = 0.047), 7.03% (p = 0.041), 5.95% (p = 0.036), 9.30 (p = 0.025), 11.17% (p = 0.009), respectively at 10 days post-weaning; Pro by 6.11% (p = 0.044) at 30 days post-weaning. Jejunal brush border membrane-bound alkaline phosphatase activity increased in the Gln-supplemented pigs by 30.36% (p = 0.048) and 6.21% (p = 0.30) at 10 and 30 days post-weaning, respectively. Compared with the control pigs, the mRNA level of PPAR gamma decreased by 10.85% (p = 0.14) and 41.88% (p = 0.023) after the administration of 1% Gln for 10 and 30 days, respectively. The activity of glutamine synthetase decreased by 48.89% (p = 0.044) at 10 days post-weaning and pyruvate kinase by 13.13% (p = 0.036) at 30 days post-weaning in the Gln-supplemented pigs. In conclusion, 1% Gln supplementation to the post-weaned piglet diet enhanced the CTTAD and AID of diet, improved intestinal absorption and modified jejunal enzyme activities related to Gin metabolism and energy production.

Abstract:Sixty crossbred piglets(Duroc×Landrace×Yorkshire)weaned at 21 days of age were divided into 2 treatments based on weight and gender.Piglets in treatments were fed basal diet and basal diet supplemented with 0.5 % glutamine respectively.The effect of glutamine on morphology of intestine mucosal of early weaned piglets was studied.The VH and LPD traits of intestine mucosal at the age of 21,28,35 d were measured.The results showed that both VH and LPD of weaned pigs increased with supplementation of glutamine.

Abstract:The gut is composed of a single layer of intestinal epithelial cells and plays important roles in the digestion and absorption of nutrients, immune and barrier functions and amino acid metabolism. Weaning stress impairs piglet intestinal epithelium structural and functional integrities, which results in reduced feed intake, growth rates and increased morbidity and mortality. Several measures are needed to maintain swine gut development and growth performance after weaning stress. A large body of evidence indicates that, in weaning piglets, glutamine, a functional amino acid, may improve growth performance and intestinal morphology, reduce oxidative damage, stimulate enterocyte proliferation, modulate cell survival and death and enhance intestinal paracellular permeability. This review focuses on the effects of glutamine on intestinal health in piglets. The aim is to provide evidentiary support for using glutamine as a feed additive to alleviate weaning stress.

Abstract:Following extensive bowel resection, the intestinal tract undergoes a variety of adaptive responses to enhance bowel function. The purpose of this study was to determine the effect of glutamine-supplemented parenteral nutrition on mucosal cellularity and gut function. In addition, enterocyte gene expression of two relevant systems was also characterized and related to the structural and functional changes that occurred. Male Wistar rats underwent a 60% small bowel resection and jugular vein catheterization and were randomized into two groups. The control group (n = 10) received a standard intravenous nutritional solution and the study group (n = 10) received a similar solution but enriched with alanylglutamine dipeptide. After 7 days blood was taken for amino acid analysis, and bowel was harvested to determine mucosal morphology and expression of mucosal cell glutaminase and IGF-I mRNA. Mesentery lymphnodes were cultured to determine the presence of bacteria and thus access bacteria translocation. Serum glutamine concentration and mucosal architecture were maintained in the study group compared to the controls. Seventy percent of lymphnodes were cultured positive in control vs. only 20% in the study group (P < 0.05). Jejunal mucosal glutaminase and ileum mucosal IGF-I mRNA increased twofold and threefold respectively compared to control animals. Parenteral nutrition supplemented with alanyl-glutamine dipeptide supports mucosal cellularity and regional immune function in rodents following intestinal resection, These alterations are associated with enhanced enterocyte expression of glutaminase and IGF-I. These changes may facilitate the structural and functional alterations which were observed in the glutamine treated animals.

Abstract:Glutamine, an amino acid, is the principal energy substrate for small intestinal cells. It also acts as a nitrogen carrier through its amide nitrogen. Arterial glutamine is supported by net synthesis in skeletal muscle. Glutamine is rapidly metabolized by the intestine, whether supplied from the lumen or from the arterial circulation. Intestinal uptake of glutamine increases after trauma and operative stress. The consumption of glutamine by the gut may in large part be dependent on mucosal glutaminase activity and on enterocyte glutamine transport. Glutaminc has been shown to improve gut morphology and outcome in animal models of encerocolitis. It may play a similar role in aiding repair of human intestinal injury in persons with sufficient glutamine in their diet compared to those who arc glutamine deficient. Glutamine may have a positive effect on the immune function of the intestinal mucosal-associated lymphoid tissue. Glutamine is not currently available in nutritional preparations for routine clinical use, yet it has recently been shown to benefit maintenance of nitrogen balance in humans. Due to the instability and low solubility of glutamine, dipeptides have been studied. L-alanyl-L-glutamine seems to be the most promising glutamine precursor for parenteral use in humans, as it is safe and rapidly hydrolyzed in vivo to release free glutamine. The exact role of glutamine as a therapeutic agent to promote intetitinal well-being has yet to be determined. However, preliminary evidence suggests that glutaminc will be helpful in a variety of clinical scenarios.

Abstract:Sixty 21-day weaning piglets were selected to study the effects of 1% glutamine supplementation in diets on the immunity.The results showed:on 10th day postweaning,IgG and IgM concentration of serum in control decreased 21.44%(P0.05) and 28.23%(P0.05) than preweaning,IgA in 1.0% Gln group increased 28.13%(P0.05) than preweaning,IgA,IgG,IgM concentration of serum in Gln group was 6.49%(P0.05),34.67%(P0.05) and 32.58%(P0.05) higher respectively than those in control.Supplementation of Glutamine enhanced IgA,IgG and IgM concentration in serum by 15.22%(P0.05),2.28%(P0.05),30.66%(P0.05) as compared to the control on 20th day post weaning.Glutamine supplementation significantly increased the T lymphocyte proliferation rate by 75.06%(P0.05) in vitro.The DNA content of mesenteric lymph nodes in Glu group was 9.18%(P0.05) higher than that in control.

Abstract:Glutamine is the most abundant free amino acid in the body,which is regarded as a conditionally essential amino acid under the conditions of stress and hypermetabolism.It has been proved that glutamine can enhance the proliferation of immune cells and the repair of impaired mucosa and can also improve the nitrogen balance and skeletal muscle protein synthesis.Therefore,it is now more and more widely used in the bone marrow transplantation.The roles of glutamine in the prevention and treatment of bone marrow transplantation-related mucositis and veno-occlusive disease are reviewed in this article.

Abstract:BACKGROUND:Dysfunction of tight junction integrity is associated with decreased nutrient absorption and numerous gastrointestinal diseases in humans and piglets. Although l-glutamine has been reported to enhance intestinal-mucosal mass and barrier function under stressful conditions, in vivo data to support a functional role for l-glutamine on intestinal tight junction protein (TJP) expression in weanling mammals are limited.OBJECTIVE:This study tested the hypothesis that glutamine regulates expression of TJPs and stress-related corticotropin-releasing factor (CRF) signaling in the jejunum of weanling piglets.METHODS:Piglets were reared by sows or weaned at 21 d of age to a corn and soybean meal-based diet that was or was not supplemented with 1% l-glutamine for 7 d. Growth performance, intestinal permeability, TJP abundance, and CRF expression were examined.RESULTS:Weaning caused increases (P < 0.05) in intestinal permeability by 40% and in CRF concentrations by 4.7 times in association with villus atrophy (P < 0.05). Western blot analysis showed reductions (P < 0.05) in jejunal expression of occludin, claudin-1, zonula occludens (ZO) 2, and ZO-3, but no changes in the abundance of claudin-3, claudin-4, or ZO-1 in weanling piglets compared with age-matched suckling controls. Glutamine supplementation improved (P < 0.05) intestinal permeability and villus height, while reducing (P < 0.05) jejunal mRNA and protein levels for CRF and attenuating (P < 0.05) weanling-induced decreases in occludin, claudin-1, ZO-2, and ZO-3 protein abundances.CONCLUSION:Collectively, our results support an important role for l-glutamine in regulating expression of TJPs and CRF in the jejunum of weanling piglets.

Effects of Glutamine on Growth Performance of Weanling Piglets

Abstract:An experiment was conducted to investigate the effects of glutamine on growth performance of weanling piglets. Sixty piglets weaned at 21 days of age were randomly assigned to two groups (10 piglets per pen, 3 pens per group). The control group received a maize-soybean meal-based diet. The treatment group received a maize-soybean meal-based diet supplemented with 1.0% free -glutamine. Piglets were fed the diets for 20 days. Results showed that piglets fed the glutamine diet had lower diarrhoea ratio and shorter diarrhoea duration than those fed the control diet during 20 days after weaning. During the first ten days after weaning, pigs supplemented with glutamine had a 12.05% lower feed:gain ratio than those fed the control diet ( P 0.05) compared to the control. During the second ten days after weaning, serum urea nitrogen was reduced by 4.27% and serum concentrations of total protein increased by 18.70% in pigs supplemented with glutamine compared to the control (P > 0.05). There were no differences in albumin, T 3, T 4 and growth hormone.

Glutamine-derived peptides: Current progress and future directions

Abstract:Glutamine, the most abundant amino acid in the body, plays a critical role in preserving immune function, nitrogen balance, intestinal integrity, and resistance to infection. However, its limited solubility and instability present challenges for its use a functional nutrient. Consequently, there is a preference for utilizing glutamine-derived peptides as an alternative to achieve enhanced functionality. This article aims to review the applications of glutamine monomers in clinical, sports, and enteral nutrition. It compares the functional effectiveness of monomers and glutamine-derived peptides and provides a comprehensive assessment of glutamine-derived peptides in terms of their classification, preparation, mechanism of absorption, and biological activity. Furthermore, this study explores the potential integration of artificial intelligence (AI)-based peptidomics and synthetic biology in the de novo design and large-scale production of these peptides. The findings reveal that glutamine-derived peptides possess significant structure-related bioactivities, with the smaller molecular weight fraction serving as the primary active ingredient. These peptides possess the ability to promote intestinal homeostasis, exert hypotensive and hypoglycemic effects, and display antioxidant properties. However, our understanding of the structure-function relationships of glutamine-derived peptides remains largely exploratory at current stage. The combination of AI based peptidomics and synthetic biology presents an opportunity to explore the untapped resources of glutamine-derived peptides as functional food ingredients. Additionally, the utilization and bioavailability of these peptides can be enhanced through the use of delivery systems in vivo. This review serves as a valuable reference for future investigations of and developments in the discovery, functional validation, and biomanufacturing of glutamine-derived peptides in food science.
Glutamine protects intestinal immunity through microbial metabolites rather than microbiota

Abstract:Glutamine has anti-inflammatory properties as well as the ability to maintain the integrity of the intestinal barrier. In our previous study, we found that 1.0% glutamine promoted SIgA (secretory immunoglobulin A) synthesis in the gut via both T cell-dependent and non-dependent processes, as well as via the intestinal microbiota. The purpose of this study was to investigate whether the intestinal microbiota or microbial metabolites regulate SIgA synthesis. In the mouse model, supplementation with 1.0% glutamine had no significant effect on the intestinal microbiota, but KEGG function prediction showed the difference on microbiota metabolites. Therefore, in this study, untargeted metabolomics techniques were used to detect and analyze the metabolic changes of glutamine in intestinal luminal contents. Metabolomics showed that in the positive ion (POS) mode, a total of 1446 metabolic differentials (VIP ≥ 1, P < 0.05, FC ≥ 2 or FC ≤ 0.5) were annotated in samples treated with glutamine-supplemented group compared to control group, of which 922 were up-regulated and 524 down-regulated. In the negative ion (NEG) mode, 370 differential metabolites (VIP ≥ 1, P < 0.05, FC ≥ 2 or FC ≤ 0.5) were screened, of which 220 were up-regulated and 150 down-regulated. These differential metabolites mainly include bile secretion synthesis, ABC transporters, diterpenoids and other secondary metabolites. KEGG analysis showed that propionic acid metabolism, TCA cycle, endoplasmic reticulum protein processing, nitrogen metabolism and other metabolic pathways were active. The above metabolic pathways and differential metabolites have positive effects on intestinal development and intestinal immunity, and combined with our previous studies, we conclude that glutamine supplementation can may maintain intestinal homeostasis and improving intestinal immunity through intestinal microbial metabolites.

Abstract:Mammalian target of rapamycin (mTOR) plays an important role in regulating protein synthesis. This paper discusses the effects of glutamine and electrical stimulation on protein content and the mTOR pathway in C2C12, providing theories on how glutamine improves sports performance and recovery. After 5 days of differentiation, C2C12 myotubes were incubated in 4 mM or 10 mM doses of glutamine for 48 hours, and then stimulated to contract for 90 mins at 15v, 3Hz, and 30ms. After electrical stimulation, samples of Es and Gln+Es were harvested at 0, 6, 18, and 24 hours. The BCA Protein Assay Kit was used to measure total protein content (μg/ cm2). The expression of mTOR and S6K1 were measured by RT-PCR. The expression of p-S6 and myosin heavy chain (MHC) were measured by western blot. The results showed that total protein content, mTOR, p-S6, MHC expression of Gln were greater than that in Con (P<0.05). Compared with the protein content found in Con, that of Es18 and Es24 increased significantly (P<0.01). When the expression of mTOR and p-S6 were compared, it was apparent that Gln+Es and Es had similar trends, which was a decline in protein followed by a subsequent rise. The difference was that Es reached the peak in 18 hours, while Gln+Es did so in 6 h. The MHC expression in both Es and Gln+Es changed stable with a similar trend, decline followed by rise. Electrical stimulation and glutamine promote expression of MHC through the mTOR pathway, increase protein synthesis, and promote the rise of protein content and muscle recovery.
Effects of Dietary l-Glutamine Supplementation on the Intestinal Function and Muscle Growth of Piglets

Abstract:The aim of this study was to investigate the effects of dietary l-glutamine (Gln) supplementation on the morphology and function of the intestine and the growth of muscle in piglets. In this study, sixteen 21-day-old piglets were randomly divided into two groups: the Control group (fed a basal diet) and the Gln group (fed a basal diet supplemented with 0.81% Gln). Blood, gut, and muscle samples were collected from all piglets on Day 20 of the trial. Compared with the Control group, the supplementation of Gln increased (p < 0.05) the villus height, villus width, villus surface area, and villus height/crypt depth ratio of the small intestine. Furthermore, the supplementation of Gln increased (p < 0.05) total protein, total protein/DNA, and RNA/DNA in both the jejunum and ileum. It also increased (p < 0.05) the concentrations of carnosine and citrulline in the jejunal mucosa, as well as citrulline and cysteine concentrations in the ileum. Conversely, Gln supplementation decreased (p < 0.05) Gln concentrations in both the jejunum and ileum, along with β-aminoisobutyric acid and 1-Methylhistidine concentrations, specifically in the ileum. Subsequent research revealed that Gln supplementation increased (p < 0.05) the mRNA levels for glutathione-S-transferase omega 2 and interferon-β in the duodenum. In addition, Gln supplementation led to an increase (p < 0.05) in the number of Lactobacillus genus in the colon, but a decrease (p < 0.05) in the level of HSP70 in the jejunum and the activity of diamine oxidase in plasma. Also, Gln supplementation reduced (p < 0.05) the mRNA levels of glutathione-S-transferase omega 2 and interferon stimulated genes, such as MX1, OAS1, IFIT1, IFIT2, IFIT3, and IFIT5 in both the jejunum and ileum, and the numbers of Clostridium coccoides, Enterococcus genus, and Enterobacterium family in the colon. Moreover, Gln supplementation enhanced (p < 0.05) the concentrations of total protein, RNA/DNA, and total protein/DNA ratio in the longissimus dorsi muscle, the concentrations of citrulline, ornithine, arginine, and hydroxyproline, and the mRNA level of peptide transporter 1, while reducing the contents of hydrogen peroxide and malondialdehyde and the mRNA level of glutathione-S-transferase omega 2 in the longissimus dorsi muscle. In conclusion, dietary Gln supplementation can improve the intestinal function of piglets and promote the growth of the longissimus dorsi muscle.
