Arsenic Trioxide Inhibits the Growth of Transplanted Tumor of Human Gallbladder Carcinoma and Promotes Its Apoptosis in Nude Mice

Abstract:Objective:To observe the inhibitory effect of arsenic trioxide(As2O3)on the growth of transplanted tumor of human gallbladder carcinoma and to probe into its mechanism in nude mice.Methods: Twenty BALB/c nude mice,with the resembling volume of subcutaneous transplanted tumor,were randomly assigned into eithor nature saline(NS) group or As2O3 groups.0.2ml NS or 10 mg·kg-1 As2O3 was injected into the tail vain of every mouse in NS group or As2O3 group,once a day,for seven days.All mice were put to death fourteen days after the perfusion was performed completely,then the weight and dimension were assayed as well as the expression of Cyclin D1 were determined by immunohistochemical staining and Western Blot.Results: The weight and volume of transplanted tumor was 0.992±0.401 g and 1.50±0.57 mm3,respectivey in NS group,which reduced to 0.526±0.269g and 0.83±0.39 mm3 respectivey in As2O3 group.The expression of Cyclin D1 also decreased significantly in As2O3 group.Conclusions: As2O3 can significantly inhibit the growth of transplanted tumor of human gallbladder carcinoma and its mechanism may be associated with down-regulation of expression of Cyclin D1.
