Abstract:The composite hydrogels were produced using the solution casting method due to the non-toxic and biocompatible nature of chitosan (CS)/polyvinyl alcohol (PVA). The best composition was chosen and crosslinked with tetraethyl orthosilicate (TEOS), after which different amounts of graphene oxide (GO) were added to develop composite hydrogels. Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), atomic force microscopy (AFM) and contact angle was used to analyze the hydrogels. The samples were also evaluated for swelling abilities in various mediums. The drug release profile was studied in phosphate-buffered saline (PBS) at a pH of 7.4. To predict the mechanism of drug release, the data were fitted into kinetic models. Finally, antibacterial activity and cell viability data were obtained. FTIR studies revealed the successful synthesis of CS/PVA hydrogels and GO/CS/PVA in hydrogel composite. SEM showed no phase separation of the polymers, whereas AFM showed a decrease in surface roughness with an increase in GO content. 100 μL of crosslinker was the critical concentration at which the sample displayed excellent swelling and preserved its structure. Both the crosslinked and composite hydrogel showed good swelling. The most acceptable mechanism of drug release is diffusion-controlled, and it obeys Fick’s law of diffusion for drug released. The best fitting of the zero-order, Hixson-Crowell and Higuchi models supported our assumption. The GO/CS/PVA hydrogel composite showed better antibacterial and cell viability behaviors. They can be better biomaterials in biomedical applications.

Abstract:Graphene oxide/conducting polymer (GO/CP) composite hydrogels were prepared by in situ chemical polymerization of corresponding aromatic monomers in aqueous dispersions of GO sheets. GO/polypyrrole (PPy), GO/poly(3,4-ethylenedioxythiophene) (PEDOT) and GO/polyaniline (PANi) hydrogels were obtained by this technique, and the mechanism of gelation was discussed. Among them, GO/PPy composite hydrogels were tested to have low critical hydrogel concentrations (<1%, by weight), high storage moduli (>10 kPa) and electrical conductivity, and strong electrochemical activity. A gas sensor based on a typical GO/PPy hydrogel showed high sensitivity towards ammonia gas.

Abstract:In this study, Gum Arabic Aldehyde (GAA) and polyvinyl alcohol (PVA) were used to develop pH-sensitive and self-healable hydrogels. The PVA and GAA based hydrogels have the advantages of non-toxicity, water-solubility, eco-friendly, biodegradability, and pH sensitivity. However, poor stiffness, low mechanical strength, and hydrophilicity limit their applications. Graphene oxide (GO), gelatin, and boric acid were used to improve the limitations of GAA/PVA hydrogels through cross-linking, Schiff-base reaction between aldehyde groups of GAA and amino groups of gelatin, and hydrogen bonding between PVA chains and boric acid. The prepared hydrogels demonstrated high mechanical properties, good self-healing properties, and pH sensitivity, which facilitate their application as favorable biomaterials for drug delivery. The swelling behavior, degradation, mechanical properties, and rheological behaviors were studied for hydrogels. The hydrogels demonstrated well mechanical properties and well pH sensitivity during controlled release investigation of rivastigmine (RIV) drug under intestinal (~ 83% at pH 7.4) media. Incorporating GO in the hydrogel network, enhanced the mechanical properties, self-healing properties, drug entrapment efficiency, and ensure the controlled release of the entrapped drug. The self-healing properties, swelling behavior, degradation, tensile and compressive tests, rheological behaviors, and in vitro drug release behavior were studied to monitor the effect of GO on the hydrogel properties. Our results illustrated that the self-healing nanocomposite hydrogel was expected to be a platform for drug delivery in pH 7.4.Graphical abstract

Abstract:A methodology is described for the preparation of pH-and temperature-responsive semi-interpenetrating hydrogels with surface-functionalized graphene oxide (GO) as the crosslinker, N-isopropylacrylamide (NIPAM) as the monomer and sodium alginate (SA) as an additive. GO sheets were first prepared via an oxidation reaction in aqueous solution and then modified with 3-(trimethoxysilyl) propylmethacrylate (TMSPMA) via a silanization reaction. Free-radical polymerization of NIPAM and SA was then carried out with the presence of TMSPMA-modified GO sheets at 20 degrees C in a water bath, leading to the formation of pH-and temperature-responsive PNIPAM/SA/GO hydrogels crosslinked with TMSPMA-modified GO sheets. The effects of TMSPMA-modified GO sheets content on various physical properties were investigated. Results show that PNIPAM/SA/GO hydrogels undergo a large volumetric change in response to temperature. It also exhibits significantly larger water uptake compared to conventional PNIPAM/ SA hydrogels. Moreover, the PNIPAM/SA/GO hydrogels have a much better mechanical properties than the conventional PNIPAM/SA hydrogels.

Abstract:Polyvinyl alcohol (PVA) hydrogels have been proposed for use as promising biomaterials in biomedical and tissue engineering but their poor mechanical and water-retention properties have hindered their development. Graphene oxide (GO), an excellent nanofiller, was added to PVA to make GO/PVA composite hydrogels by a freeze/thaw method. The mechanical properties of the GO/PVA hydrogels were significantly improved. Compared to pure PVA hydrogels, a 132% increase in tensile strength and a 36% improvement of compressive strength were achieved with the addition of 0.8 wt% of GO, which suggests an excellent load transfer between the GO and the PVA matrix. The incorporation of certain amount of GO into composite hydrogels does not affect the toxicity of PVA to osteoblast cells.

Abstract:Introduction: Stimuli-responsive hydrogels, which indicate a significant response to the environmental change (e.g., pH, temperature, light, …), have potential applications for tissue engineering, drug delivery systems, cell therapy, artificial muscles, biosensors, etc. Among the temperature-responsive materials, poly (N-isopropylacrylamide) (PNIPAAm) based hydrogels have been widely developed and their properties can be easily tailored by manipulating the properties of the hydrogel and the composite material. Graphene oxide (GO), as a multifunctional and biocompatible nanosheet, can efficiently improve the mechanical strength and response rate of PNIPAAm-based hydrogels. Here, hydrogel composites (HCs) of PNIPAAm with GO was developed using the modified starch as a biodegradable cross-linker. Methods: Micro/nanohydrogel composites were synthesized by free radical polymerization of NIPAAm in the suspension of different feed ratio of GO using maleate-modified starch (St-MA) as cross-linker and Tetrakis (hydroxymethyl) phosphonium chloride (THPC) as a strong oxygen scavenger. The HCs were characterized by FT-IR, DSC, TGA, SEM, and DLS. Also, the phase transition, swelling/deswelling behavior, hemocompatibility and biocompatibility of the synthesized HCs were investigated. Results: The thermal stability, phase transition temperature and internal network crosslinking of HCs increases with increasing of the GO feed ratio. Also, the swelling/deswelling, hemolysis, and MTT assays studies confirmed that the HCs are a fast response, hemocompatible and biocompatible materials. Conclusion: The employed facile approach for the synthesis of HCs yields an intelligent material with great potential for biomedical applications.

Abstract:Graphene is a widely used material as a therapeutic agent (drug) carrier due to its low toxicity, high surface area, specific flake-shape, 2-dimensional structure and geometry. In order to evaluate the efficiency of drug delivery improved by using graphene oxide as a drug carrier, drug release experiment using Cytochrome-C (Cyto- C), a model drug, was carried out. First of all, different ratios of Cyto-C were combined with 100 mg GO to investigate the best drug-loading efficiency so that the best loading ratio was confirmed with Cyto-C:GO of 1:2. Alginate hydrogel (Alg hydrogel) and Alginate-Graphene oxide hydrogel (Alg-GO hydrogel) were fabricated by cross-linking in 0.1 M CaCl2 solution. Characterization of the hydrogels was confirmed by Fourier transform infrared (FT-IR) and scanning electron microscope (SEM) analysis. The drug efficiency by the ratio of graphene oxide to Cyto-C was compared by ultraviolet visible spectrophotometer (UV-VIS). The difference in drug release behavior in hydrogels was confrimed by drug release test. The Alg-GO hydrogel exhibited slow and steady release compared to that of alginate hydrogel as control. In this study, we demonstrated that applying graphene oxide as a drug carrier affects in control of drug release behavior.

Abstract:For conductive hydrogels applied in biosensors, wearable devices and so forth, multifunctionality is an inevitable trend of development to meet various practical requirements and enhance human experience. Herein, inspired by nanocomposite, double-network (DN) and mussel chemistry, a new Graphene oxide@Dopamine/Alginate/Poly (acrylic acid-co-acrylamide) [GO@DA/Alginate/P(AAc-co-AAm)] hydrogel was fabricated through one-pot in-situ radical copolymerization. GO@DA nanofillers, prepared via GO confined DA polymerization, imparted the hydrogel with remarkable adhesiveness. Alginate/P(AAc-co-AAm) DN matrix, physically and chemically crosslinked by Fe3+ and N,N'-Methylenebisacrylamide, made hydrogels ultrastretchable, self-healing and biocompatible. With contents of DA and alginate accurately regulated, the tensile strength, elongation, adhesion strength and conductivity of the optimal hydrogel could reach 320.2 kPa, 1198 %, 36.9 kPa and 3.24 +/- 0.12 S/m, respectively. What's more notable was that the synergistic integration of repeatable adhesiveness, strain sensitivity, use stability, self-healing ability and biocompatibility provided such hydrogels with tremendous possibility of practical application for strain sensors.
Synthesis, characterizations, and release mechanisms of carboxymethyl chitosan-graphene oxide-gelatin composite hydrogel for controlled delivery of drug

Abstract:Hydrogels have received immense interest for various biomedical applications owing to their hydrated environment and outstanding properties. However, challenges still exist regarding utilizing hydrogels in the application of drug delivery and release, because of the lack of controllability, actuation and quick-response properties. Here, the functionalized carboxymethyl chitosan-graphene oxide-gelatin composite hydrogel with excellent swelling property, high biocompatibility and drug delivery efficiency was prepared by ionic crosslinking. Our study found that the carboxymethyl chitosan-graphene oxide-gelatin composite hydrogel exhibited porous structures and the pores distribute randomly but arrange regularly in the cross section. In addition, the thermal stability and excellent swelling ratio of the carboxymethyl chitosan-graphene oxide-gelatin composite hydrogel were improved. As a consequence, the CMCS-GO-Gelatin-3 exhibited the maximum cumulative release rate and the cumulative release amount of this composite hydrogel was 74.6 under the condition of buffer solution pH = 6.5 PBS and 89.1 under the condition of buffer solution pH = 7.4 PBS. Therefore, this composite hydrogel showed improved drug delivery efficiency and achieved drug prolonged administration to 24 h. This work highlights that carboxymethyl chitosan-graphene oxide-gelatin composite hydrogel will be a promising candidate for the applications in biomedical engineering.
Construction of a Mesoporous Polydopamine@GO/Cellulose Nanofibril Composite Hydrogel with an Encapsulation Structure for Controllable Drug Release and Toxicity Shielding

Abstract:The development of intelligent and multifunctional hydrogels having photothermal properties, good mechanical properties, sustained drug release abilities with low burst release, antibacterial properties, and biocompatibility is highly desirable in the biomaterial field. Herein, mesoporous polydopamine (MPDA) nanoparticles wrapped with graphene oxide (GO) were physically cross-linked in cellulose nanofibril (CNF) hydrogel to obtain a novel /CNF composite hydrogel for controllable drug release. MPDA nanoparticles exhibited a high drug loading ratio (up to 35 wt %) for tetracycline hydrochloride (TH). GO was used to encapsulate MPDA nanoparticles for extending the drug release time and reinforcing the physical strength of the obtained hydrogel. The mechanical strength of the as-fabricated /CNF composite hydrogel was five times greater compared to that of the pure CNF hydrogel. Drug release experiments demonstrated that burst release behavior was significantly reduced by adding The drug release time of the /CNF composite hydrogel was 3 times and 7.2 times longer than that of the polydopamine/CNF hydrogel and pure CNF hydrogel, respectively. The sustained and controlled drug release behaviors of the composite hydrogel were highly dependent on the proportion of MPDA and GO. Moreover, the rate of drug release could be accelerated by near-infrared (NIR) light irradiation and pH value change. The drug release kinetics of the as-prepared composite hydrogel was well described by the Korsmeyer–Peppas model, and the drug release mechanism of TH from the composite hydrogel was anomalous transport. Importantly, this carefully designed /CNF composite hydrogel showed good biocompatibility through an <i>in vitro</i> cytotoxicity test. In particular, the toxicity of GO was well shielded by the CNF hydrogel. New Polyvinyl Alcohol/Succinoglycan-Based Hydrogels for pH-Responsive Drug Delivery

Abstract:We fabricated new hydrogels using polyvinyl alcohol (PVA) and succinoglycan (SG) directly isolated and obtained from Sinorhizobium meliloti Rm 1021 via the freeze–thaw method. Both the composition of the hydrogels and the freeze–thaw cycles were optimized to maximize the swelling ratio for the preparation of the PVA/SG hydrogels. During the optimization process, the morphology and conformational change in the hydrogel were analyzed by scanning electron microscopy, rheological measurements, and compressive tests. An optimized hydrogel with a maximum swelling ratio of 17.28 g/g was obtained when the composition of PVA to SG was 50:50 (PVA/SG 50/50) and the total number of freeze–thaw cycles was five. The PVA/SG 50/50 hydrogel had the largest pore with 51.24% porosity and the highest cross-over point (28.17%) between the storage modulus (G′) and the loss modulus (G′′). The PVA/SG 50/50 hydrogel showed improved thermal stability owing to its interaction with thermally stable SG chains. The improvement in the thermal stability was confirmed by thermogravimetric analysis and differential scanning calorimetry. In addition, the PVA/SG 50/50 hydrogel showed differential drug release according to the corresponding pH under acidic conditions of pH 1.2 and slightly basic conditions of pH 7.4. Furthermore, the cell viability test on the HEK-293 cell line for that hydrogel demonstrated that the PVA/SG 50/50 hydrogel was non-toxic and biocompatible. Therefore, this hydrogel could be a potential scaffold capable of pH-responsive drug delivery for chronic wound dressing applications.

Abstract:Multiple pH-sensitive composites have been prepared through non-covalently functionalizing chemically converted graphene (CCG) with chitosan. Chtiosan exhibits as polybases and CCG shows characteristics of polyacids. Owing to the synergistic effects of chitosan and CCG, chitosan decorated graphene (CS-G) presents a multiple pH-responsive behavior that it can be dispersed well whether in acidic or in basic solution but aggregated in near-neutral solution. After CS-G was modified through a controlled deposition and cross-linking process of chitosan, the resultant cross-linked chitosan decorated graphene (CLCS-G) can be converted to a different pH-sensitive material that disperses only in acidic solution. Both CS-G and CLCS-G present a reversible switching between dispersed and aggregated states with pH as a stimulus. The unique pH response mechanisms of CS-G and CLCS-G have been further investigated by zeta potential analysis. Based on the unique pH-responsive property of CS-G, a stable and repeatable pH-driven switch was developed for monitoring pH change.

Abstract:A novel pH sensitive antibiotic carrier hydrogel was synthesized having sustained drug releasing capabilities. The in-situ polymerization of Graphene oxide (GO), Polyvinyl pyrrolidone (PVP), and Carboxymethyl cellulose (CMC) of varying stoichiometry having Ca 2+ as a linker yielded ternary nanocomposite hydrogels GO/PVP/CMC-II and GO/PVP/CMC-III. The physicochemical, structural, morphological and swelling properties of the binary nanocomposite GO/PVP and ternary nanocomposite hydrogels, GO/PVP/CMC were investigated using FTIR, XRD, SEM, TEM, TGA and DTA. FTIR and XRD confirm successive formation of ternary hydrogels. The hydrogels swelling studies at various pH levels viz. 4.5, 7 and 9.2, show that the swelling ability can be proportionally correlated with CMC concentration. To ensure the efficacy of this innovative hydrogel drug release application, an in vitro Ampicillin loading and release efficiency experiment at pH 7.2 (close to blood pH 7.3–7.5) was performed. Ternary hydrogels GO/PVP/CMC-II and GO/PVP/CMC-III exhibited sustained drug loading and release kinetics (72 h).

A pH-responsive hydrogel system based on cellulose and dopamine with controlled hydrophobic drug delivery ability and long-term bacteriostatic property

Abstract:An interpenetrating double network hydrogel based on cellulose backbone and poly-dopamine dendritic structure was successfully prepared with the advantages of controlled hydrophobic drug loading, pH-responsive ability, and long-term bacteriostatic property. 13C-NMR was used to process the structural characterization and explain the mechanism of hydrogel system’s construction. Hydrophobic drug of ciprofloxacin with carbonyl group can evenly carry in the hydrogel system, and the highest loaded ratio of ciprofloxacin was reach to 80.56%. Besides, the swelling property and the controlled drug loading ability of our prepared hydrogel were carefully studied, which showed a close relation with the hydrogel design. Moreover, the in vitro drug release experiments demonstrated that the hydrogel had an obvious pH-responsive release property of ciprofloxacin, and the result of bacteriostatic experiment also showed a long-term bacteriostatic effect on Escherichia coli, which showed potential application on biological and medical fields, including wound treatment.Graphical abstract.

Abstract:Cellulose-based biocompatible, tunable and injectable hydrogels embedded with pH-responsive diblock copolymer micelles were constructed to achieve localized drug delivery with prolonged, stimuli-driven and slow-release function. First, we prepared two types of modified carboxymethyl cellulose (CMC) including hydrazide-modified carboxymethyl cellulose (CMC-NH2) and oxidized carboxymethyl cellulose (CMC-CHO) with varying degrees of oxidation. Then, pH-responsive poly (ethylene oxide)-block-poly (2-(diisopropylamino) ethyl methacrylate) (PEO-b-PDPA) copolymers as micelle cores to carry hydrophobic substances were also synthesized through atom transfer radical polymerization (ATRP). An injectable hydrogel composite system was finally obtained by mixing CMC-NH2 and CMC-CHO polymer suspensions containing PEO-b-PDPA copolymer micelles through a Schiff base reaction. This newly-synthesized, tunable, cellulose-based double barrier system exhibits a pH-triggered, prolonged, and slow-release profile based on the release test using both Nile Red dye and doxorubicin. The hydrogel system also exhibited comparable storage moduli and tunable degradation properties.

Abstract:Super-porous hydrogels are considered a potential drug delivery network for the sedation of gastric mechanisms with retention windows in the abdomen and upper part of the gastrointestinal tract (GIT). In this study, a novel pH-responsive super-porous hybrid hydrogels (SPHHs) was synthesized from pectin, poly 2-hydroxyethyl methacrylate (2HEMA), and N, N methylene-bis-acrylamide (BIS) via the gas-blowing technique, and then loaded with a selected drug (amoxicillin trihydrate, AT) at pH 5 via an aqueous loading method. The drug-loaded SPHHs-AT carrier demonstrated outstanding (in vitro) gastroretentive drug delivery capability. The study attributed excellent swelling and delayed drug release to acidic conditions at pH 1.2. Moreover, in vitro controlled-release drug delivery systems at different pH values, namely, 1.2 (97.99%) and 7.4 (88%), were studied. These exceptional features of SPHHs—improved elasticity, pH responsivity, and high swelling performance—should be investigated for broader drug delivery applications in the future.

Abstract:Polymeric materials have been essential biomaterials to develop hydrogels as wound dressings for sustained drug delivery and chronic wound healing. The microenvironment for wound healing is created by biocompatibility, bioactivity, and physicochemical behavior. Moreover, a bacterial infection often causes the healing process. The bacterial cellulose (BC) was functionalized using graphene oxide (GO) by hydrothermal method to have bacterial cellulose-functionalized-Graphene oxide (BC-f-GO). A simple blending method was used to crosslink BC-f-GO with polyvinyl alcohol (PVA) by tetraethyl orthosilicate (TEOS) as a crosslinker. The structural, morphological, wetting, and mechanical tests were conducted using Fourier-transform infrared spectroscopy (FTIR), Scanning electron microscope (SEM), water contact angle, and a Universal testing machine (UTM). The release of Silver-sulphadiazine and drug release kinetics were studied at various pH levels and using different mathematical models (zero-order, first-order, Higuchi, Hixson, Korsmeyer–Peppas, and Baker–Lonsdale). The antibacterial properties were conducted against Gram-positive and Gram-negative severe infection-causing pathogens. These composite hydrogels presented potential anticancer activities against the U87 cell line by an increased GO amount. The result findings show that these composite hydrogels have physical-mechanical and inherent antimicrobial properties and controlled drug release, making them an ideal approach for skin wound healing. As a result, these hydrogels were discovered to be an ideal biomaterial for skin wound healing.
