Abstract:Purpose: Osteoarthritis (OA) is a common degenerative joint disorder, which causes pain, swelling and reduced motion in joints. Patients with Kashin-Beck disease (KBD), an endemic osteochondropathy, also exhibit pain, swelling and reduced motion in joints, similar to OA. The two disease are considered having similar pathogenesis. Although some key genes and diagnostic markers have been identified in the past decades, these biomarkers still cannot fully reveal the complicated pathogenesis of OA and KBD. To better understand the common key genes and pathways shared in both OA and KBD, we conducted a comprehensive analysis on biological function and interactions of OA and KBD-related genes from the published literature. Methods: We reviewed the literature deposited in PubMed and identified the genes which were reported a statistically significant association with OA or KBD. A protein-protein interactions (PPI) network analysis was then conducted using STRING database v9.05 (https://string-db.org/) to identify the core genes shared with OA and KBD. Moreover, a gene ontology (GO) enrichment analysis and gene-pathway network were conducted towards the common genes related to these two diseases using Cytoscape 3.4.0. Results: We identified 2548 genes with reporting a statistically significant association with OA, 922 genes with KBD and 127 genes related to both OA and KBD. 13 clusters were found in the PPI networks. The 3 most important clusters in the network were related to collagen (Extracellular Matrix), complement, inflammation and immunity, suggesting that inflammation and extracellular matrix damage played important roles in the occurrence of both OA and KBD (Fig. 1). Gene-pathway network analysis also provided similar results to that of PPI analysis, indicating that the common genes shared with OA and KBD were mainly involved in focal adhesion signaling pathway (COL1A1, COL2A1, COL6A6, COL9A1, etc.), complement and coagulation cascades signaling pathways (CD55, CD59, C3, F5, etc.), and AGE-RAGE signaling pathway (IL1B, CXCL8, VEGFA, etc.) (Fig. 2). Conclusions: Several biological pathways (focal adhesion signaling pathway, complement and coagulation cascades signaling pathways, AGE-RAGE signaling pathway) were related to both OA and KBD, which suggests that inflammation and extracellular matrix damage may play important roles in the occurrence of both OA and KBD.
