Cardiac Ang Ⅱ and NADPH Oxdiase-derived ROS Are Involved in Pressure Overload-induced Cardiac Hypertrophy

Abstract:Objective To investigate the role of NADPH oxidase-derived ROS in angiotensin Ⅱ modulating cardiac hypertrophy. Methods The model of rat ventricular hypertrophy was established through Aorta-construction(AC). Rats were treated with angiotensin converting enzyme inhibitor(ACEI)-captopril or NADPH oxidase inhibitor-apocynin for 8 weeks. We tested the content of angiotensin Ⅱ in cardiac tissues using an radioimmunoassay kit ;determined the O2·-level of left ventricular myocardium with a laser scanning confocal microscopy ; detected the expression of NADPH oxidase using immunohistochemistry method. Results (1) The content of AngⅡ,expression of NADPH oxidase and the level of O2·-in rat cardiac tissues after AC were significantly elevated;(2) Captopril depressed AngⅡ content,O2·-level and the expression of NADPH oxidase significantly in cardiac tissues,and inhibited the degree of ventricular hypertrophy; (3)Apocynin depressed O2·-level and inhibited ventricular hypertrophy in some degree,while it did not affect the AngⅡ content and the expression of NADPH oxidase. Conclusions The elevated cardiac AngⅡ induced by persistent pressure overload may promote the up-regulation of NADPH oxidase,which in turn enhance the production of O2·-. This may be one of the regulation pathways in pressure overload-induced cardiac hypertrophy.
