Effect of siRNA Targeted to Fibrinogen α Chain Gene on Proliferation and Apoptosis of Human Hepatocellular Carcinoma Cells

Abstract:Background: Fibrinogen(FG) is a glucoprotein synthesized and secreted by hepatocytes.Besides blood clotting and fibrinolysis,FG also plays important role in cell proliferation,angiogenesis,tumorigenesis,progression and metastasis.Plasma level of FG is higher in patients with hepatocellular carcinoma(HCC) than in healthy adults.Aims: To study the effect of RNA interference targeted to FG α chain(FGA) gene on proliferation and apoptosis of human HCC cells.Methods: Recombinant plasmid carrying FGA siRNA(pU6-FGA siRNA) was constructed and stably transfected into human HCC cell line HepG2.HepG2 cells transfected with blank plasmid vector pU6 were served as controls.Efficiency of RNA interference was identified by RT-PCR and Western blotting at mRNA and protein levels.Cell proliferation and apoptosis were determined by CCK-8 assay and flow cytometry,respectively.Results: Expression levels of FGA mRNA and protein were significantly lower in HepG2 cells transfected with pU6-FGA siRNA than in HepG2 cells transfected with pU6.Cell proliferation was suppressed and apoptosis was enhanced in HepG2 cells transfected with pU6-FGA siRNA(P<0.001).Conclusions: RNA interference can effectively down-regulate FGA expression,and subsequently inhibit proliferation and promote apoptosis in human HCC cells.FGA might be a potential target for treatment of HCC.
