Abstract:Nanoarrays (NAs) with stable signal output have become the most promising photoelectrochemical (PEC) biosensing substrate. However, a general issue is that interfacial charge-carrier recombination in a single-component semiconductor cannot be easily prevented, resulting in a low photocurrent density. Herein, a biosensor utilizing a Ag-embedded TiO2-Ag2S nanojunction (TiO2-Ag- Ag2S) as a signal converter was developed for the detection of CD44 protein -a transmembrane glycoprotein highly expressed in breast cancer cells. The ternary Z-scheme heterojunction was prepared by a distinctive scheme in which the Ag layer is introduced onto the surface of rutile TiO2 NAs by magnetron sputtering, whereas the Ag2S is rooted in the local sulfuration of Ag. With a sufficient density of oriented nanorods, TiO2-Ag-Ag2S exhibits a smooth photocurrent output and minimal variation among different batches; it is undoubtedly a satisfactory PEC sensing carrier, which enables highly specific identification of target CD44 on the surface of MDA-MB-231 cells due to DNA strand displacement reactions (SDRs) and host-guest recognition between hyaluronic acid (HA) and CD44. The biosensor shows a sensitive PEC response to CD44 over a wide range of 37 to 5.0 x 10(5) cells/mL. We can conclude that this approach will provide an alternative solution to breast cancer diagnosis.
Versatile Immunosensor Using A Quantum Dot Coated Silica Nanosphere As A Label for Signal Amplification

Abstract:A versatile immunosensor using a CdTe quantum dots (QDs) coated silica nanosphere (Si/QD) as a label was proposed for ultrasensitive detection of a biomarker. In this approach, silica nanospheres with good monodispersity and uniform structure were employed as the carrier for immobilization of QDs and antibodies. Rabit IgG served as a model protein to demonstrate the performance of the immunosensor. Goat antirabbit IgG antibody was covalently bound to CdTe QDs on the surface of silica nanospheres. CdTe QDs coated with a silica nanosphere label (Si/QD/Ab2) were attached onto the gold electrode surface through a subsequent "sandwich" immunoreaction. This reaction was confirmed by scanning electron microscopic (SEM) and fluorescence microscopic images. Due to signal amplification from the high loading of CdTe QDs, 6.6- and 5.9-fold enhancements in electrochemiluminescent (ECL) and square-wave voltammetric (SWV) signals for IgG detection were achieved compared to the unamplified method. The detection limits for IgG were 1.3 and 0.6 pg mL(-1) for ECL and SWV measurements, respectively. The resulting versatile immunosensor possesses high sensitivity, satisfactory reproducibility and regeneration, and good precision. This simple and specific strategy has vast potential to be used in other biological assays.

Abstract:This paper presents the synthesis of aqueous CdTe QDs embedded silica nanoparticles by reverse microemulsion method and their applications as fluorescence probes in bioassay and cell imaging. With the aim of embedding more CdTe QDs in silica spheres, we use poly(dimethyldiallyl ammonium chloride) to balance the electrostatic repulsion between CdTe QDs and silica intermediates. By modifying the surface of CdTe/SiO(2) composite nanoparticles with amino and methylphosphonate groups, biologically functionalized and monodisperse CdTe/SiO(2) composite nanoparticles can be obtained. In this work, CdTe/SiO(2) composite nanoparticles are conjugated with biotin-labeled mouse IgG via covalent binding. The biotin-labeled mouse IgG on the CdTe/SiO(2) composite nanoparticles surface can recognize FITC-labeled avidin and avidin on the surface of polystyrene microspheres by protein-protein binding. Finally, the CdTe/SiO(2) composite nanoparticles with secondary antibody are used to label the MG63 osteosarcoma cell with primary antibody successfully, which demonstrates that the application of CdTe/SiO(2) composite nanoparticles as fluorescent probes in bioassay and fluorescence imaging is feasible.

Abstract:Core-shell structured quantum dot (QD)-silica fluorescent nanoparticles have attracted a great deal of attention due to the excellent optical properties of QDs and the stability of silica. In this study, core-shell structured CdTe/CdS@SiO2@CdTe@SiO2 fluorescent nanospheres were synthesized based on the Stober method using multistep silica encapsulation. The second silica layer on the CdTe QDs maintained the optical stability of nanospheres and decreased adverse influences on the probe during subsequent processing. Red-emissive CdTe/CdS QDs (630nm) were used as a built-in reference signal and green-emissive CdTe QDs (550nm) were used as a responding probe. The fluorescence of CdTe QDs was greatly quenched by added S2-, owing to a S2--induced change in the CdTe QDs surface state in the shell. Upon addition of Cd2+ to the S2--quenched CdTe/CdS@SiO2@CdTe@SiO2 system, the responding signal at 550nm was dramatically restored, whereas the emission at 630nm remained almost unchanged; this response could be used as a ratiometric off-on' fluorescent probe for the detection of Cd2+. The sensing mechanism was suggested to be: the newly formed CdS-like cluster with a higher band gap facilitated exciton/hole recombination and effectively enhanced the fluorescence of the CdTe QDs. The proposed probe shows a highly sensitive and selective response to Cd2+ and has potential application in the detection of Cd2+ in environmental or biological samples.

Abstract:A versatile immunosensor using CdTe quantum dots as electrochemical and fluorescent labels has been developed for sensitive protein detection. This sandwich-type sensor is fabricated on an indium tin oxide chip covered with a well-ordered gold nanoparticle monolayer. Gel imaging systems were successfully introduced to develop a novel high-efficient optical immunoassay, which could perform simultaneous detection for the samples with a series of different concentrations of a target analyte. The linear range of this assay was between 0.1 and 500 ng/ mL, and the assay sensitivity could be further increased to 0.005 ng/mL with the linear range from 0.005 to 100 ng/mL by stripping voltammetric analysis. The immunosensor showed good precision, high sensitivity, acceptable stability, and reproducibility and could be used for the detection of real sample with consistent results in comparison with those obtained by the ELISA method.

Abstract:Fluorescent labeling via quantum dot (QD)-based nanospheres is intended to improve the detection performance for the lateral flow immunoassay (LFIA). In this thematic issue, current synthetic strategies for fluorescent nanospheres are rigorous, which might even cause the breaking of the photophysical properties of QDs. Here, a facile method was tailored to bright ultrastable QDs/silica nanospheres (UQSNs) via self-controlled sequential growth, used for sensitive LFIA. Specifically, in dendritic mesoporous silica nanoparticles (DMSNs) modified with a thiol compound beforehand, the in situ growth of highly fluorescent CdTe QDs is achieved; accompanying QD growth, under alkaline and high-temperature conditions, the -Si-O- bonds on the surface of DMSN will break and rapidly remodel to form a dense SiO2 outer layer (protecting the QDs from leakage and improving their stability). UQSN exhibits uniform size and high colloidal/optical stability, even under strong acid/base conditions; meanwhile, the size, fluorescence peak, and ligand of UQSN can also be controlled well. Importantly, it was found that the UQSN-115 has an optimal particle size, compared with the other three DMSNs with various diameters (80, 220, and 340 nm), for LFIA detection of the prostate-specific antigen (PSA, a valuable biomarker for prostate cancer diagnosis), reaching a detection limit of 0.138 ng/mL, besides the routine C-reaction protein (CRP) detection. This self-controlled sequential growth strategy provides new insights into producing promising fluorescent materials and developing the next sensitive bioassay.

Abstract:We propose a novel sandwich-structured electrochemiluminescence immunosensing platform using CdTe quantum dots (QDs)-embedded mesoporous silica nanospheres as enhanced signal probes for labeling the secondary antibodies and magnetite-silica-polystyrene (Fe3O4@SiO2@PS) nanocomposites as magnetic separable carriers for primary antibodies. The mSiO2/CdTe was successfully fabricated by loading CdTe QDs into mesoporous silica (mSiO2) and characterized by ultraviolet-visible spectra, photoluminescence spectra, transmission electron microscopy, fourier transform infrared spectrum and zeta potential. The electrochemiluminescence intensity of mSiO2/CdTe is much higher than that of CdTe QDs because a single mSiO2 can encapsulate hundreds of QDs. Therefore, mSiO2/CdTe has been chosen as ECL labels for conjugation with secondary antibodies. Meanwhile, Fe3O4@SiO2@PS was used as a carrier of broad primary antibodies owing to convenient magnetic separation capability, large specific surface area, and the special force between the PS and antibodies. Carcinoembryonic antigen as a model tumor mark has been detected to prove the feasibility of our immunosensing platform. The immunosensor for carcinoembryonic antigen was successfully confirmed by cyclic voltammograms and electrochemical impedance spectroscopy. Under the optimal conditions, the immunosensor presented a linear range of 0.001 to 80ng/mL for carcinoembryonic antigen detection with a low detection limit of 0.3pgmL−1 (S/N=3). Furthermore, the immunosensor also displayed outstanding selectivity, good stability, and acceptable reproducibility, indicating its potential applications in clinical diagnostics.

Abstract:The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for the synthesis of green- to near-infrared-emitting CdSeTe/CdS/ZnS QDs with time- and component-tunable photoluminescence was proposed. And, on the basis of the adjustable optical properties of both CdSeTe/CdS/ZnS QDs and GNRs, excellent overlap between donor emission and acceptor absorption can be obtained to ensure effective ECL-ET quenching, thus improving the sensing sensitivity. This method represents a novel approach for versatile detection of biomolecules at low concentrations.

Abstract:In this paper, an immunosensor using CdTe@SiO2 core-shell nanoparticles as labels was constructed for highly sensitive detection of prostate-specific antigen (PSA). In this approach, CdTe@SiO2 core-shell nanoparticles were synthesized using the sol-gel method. The additional Cd ions and sulfur source in SiO2 shell can greatly enhance the fluorescence intensity of CdTe nanocrystals. The reason is the formation of CdS-like cluster in SiO2 shell, which reduced the quantum size effect. The obtained CdTe@SiO2 nanoparticles also exhibited excellent biocompatibility, which was ideal for applying in biomarker detection. Furthermore, PSA capture antibodies functionalized magnetic Fe3O4 nanoparticles (Fe3O4-Ab1) were utilized in the proposed immunosensor to capture and enrich the PSA. The captured PSA was then immuno-recognized by CdTe@SiO2 labeled with PSA detection antibodies (CdTe@SiO2-Ab2) by forming the sandwich complex Fe3O4-Ab1/PSA/Ab2-CdTe@SiO2. The construction of this immunosensor was confirmed by fluorescence spectroscopy. The proposed immunosensor showed a good linear relationship between the fluorescent intensity and the target PSA concentration ranging from 0.01 to 5 ng/mL, and a detection limit as low as 0.003 ng/mL was achieved. The sensor also exhibited good specificity to PSA. This highly sensitive and specific immunosensor has great potential to be used in other biological detection.

Abstract:CdTe@SiO2 fluorescent particles were synthesized via hydrolysis and condensation of tetraethyl orthosilicate (TEOS) in water-in-oil (W/O) emulsion. Uniform luminophore-doped silica nanoparticles with 100nm in diameter were obtained using microemulsion method and characterized by SEM. Antibody proteins were successfully conjugated to the fluorescent particles by the reaction of avidin and biotin, which were confirmed by fluorescence spectra. CdTe@SiO2 fluorescent particles were potentially useful for the applications in biolabeling and imaging.

Abstract:A signal amplification strategy for sensitive detection of tumor necrosis factor-alpha (TNF-α) using quantum dots (QDs)-polymer-functionalized silica nanosphere as the label was proposed. In this approach, silica nanospheres with good monodispersity and uniform structure were employed as carriers for surface-initiated atom transfer radical polymerization of glycidyl methacrylate, which is readily available functional monomer that possessing easily transformable epoxy groups for subsequent CdTe QDs binding through ring-open reaction. Then, human anti rabbit TNF-α antibody (anti-TNF-α, Ab2, served as a model protein) was bonded to CdTe QDs-modified silica nanospheres coated with polymer to obtain QDs-polymer-functionalized silica nanosphere labels (Si/PGMA/QD/Ab2). The Si/PGMA/QD/Ab2 labels were attached onto a gold electrode surface through a subsequent "sandwich" immunoreaction. This reaction was confirmed by scanning electron microscopy (SEM) and fluorescence microscopic images. Enhanced sensitivity could be achieved by an increase of CdTe QD loading per immunoassay event, because of a large number of surface functional epoxy groups offered by the PGMA. As a result, the electrochemiluminescence (ECL) and square-wave voltammetry (SWV) measurements showed 10.0- and 5.5-fold increases in detection signals, respectively, in comparison with the unamplified method. The detection limits of 7.0 pg mL(-1) and 3.0 pg mL(-1) for TNF-α antibodies by ECL and SWV measurements, respectively, were achieved. The proposed strategy successfully demonstrated a simple, reproducible, specific, and potent method that can be expanded to detect other proteins and DNA.

Abstract:CdS nanocrystals (NCs) are decorated homogeneously on the TiO2 nanotube arrays (TiNT) by chemical bath deposition method. The resultant CdS/TiNT composite shows largely amplified electrochemiluminescence (ECL) signals due to the vertically aligned nanotubular structure. The ultrasensitive antigen detection is achieved on the composites via an efficient ECL quenching process introduced by sandwiched immunoreaction, and shows a detection limit of 1.0fgmL−1.

Abstract:Herein, we synthesized Pt dendrimer-encapsulated nanoparticles (Pt DENs) using amine-terminated sixth-generation polyamidoamine dendrimers. The enhanced and stable anodic electrochemiluminescence (ECL) of 3-mercaptopropionic acid-capped CdTe quantum dots (QDs) in a tripropylamine solution was achieved owing to Pt DENs. The reason may be that Pt DENs exhibit high catalytic electrochemical oxidation in the presence of tripropylamine and excellent conductive property. Inspired by this, Pt DENs were conjugated with Fe3O4@SiO2 nanoparticles and served as nano-carriers. The capture antibodies were immobilized on the Fe3O4@SiO2-Pt DEN nanocomposites, which possess many attractive advantages such as the ease of bioconjugation, large specific surface area, and convenience of magnetic separation. Fluorescence microscopy images and UV-vis spectra were used to verify the immobilization of capture antibodies on the nanocomposites. The CdTe QDs were applied as signal labels for conjugation of nanocomposites with detection antibodies, which were characterized by agarose gel electrophoresis. Electrochemical impedance spectroscopy and cyclic voltammetry demonstrated the successful preparation of an ECL immunosensor. Under the optimal conditions, the proposed immunosensor provided a wide linear range from 0.005 ng mL-1 to 150 ng mL-1 with a detection limit of 0.2 pg mL-1 (S/N = 3) for the detection of carcinoembryonic antigen. Moreover, the immunosensor showed good performance for the detection of carcinoembryonic antigen in serum samples as well as great potential in clinical bioassay.
Preparation of Bioconjugates of CdTe Nanocrystals for Cancer Marker Detection

Abstract:Highly fluorescent CdTe quantum dots (Q-dots) stabilized by 3-mercaptopropionic acid (MPA) were prepared by an aqueous solution approach and used as fluorescent labels in detecting a cancer marker, carcinoembryonic antigen (CEA), expressed on human colon carcinoma cell line LS 180. Nonspecific adsorptions of CdTe Q-dots on carcinoma cells were observed and effectively eliminated by replacing MPA with a thiolated PEG ( poly( ethylene glycol), Mn = 750) synthesized according to literature. It was unexpectedly found out that the PEG- coated CdTe Q-dots exhibited very strong and specific affinity to anti-CEA monoclonal antibody rch 24 ( rch 24 mAb). The resultant CdTe -(rch 24 mAb) conjugates were successfully used in detections of CEA expressed on the surface of cell line LS 180. Further experiments demonstrated that the fluorescent CdTe Q-dots exhibited much better photostability and a brighter fluorescence than FITC, which consequently led to a higher efficiency in the cancer marker detection.

Abstract:In this paper, based on selecting the appropriate type of quantum dots (QDs), a novel method is developed to enhance the quantum yield (QY) of silica-coated QD nanoparticles (SQDNPs). The effect of varying types of QDs on the QY after silica encapsulation is systematically studied. The results show that QDs with appropriate structure and composition of shells can much better retain the initial QY after silanization. The seven-layered shell/core QDs with QY of 47.8% nearly completely retain the original QY and is the best type among six types of QDs for silica modification. In the aspect of shell composition, the CdS plays an important role for QY retention since the lattice mismatch between CdSe and CdS is lower than that of CdSe and ZnS. After the appropriate type of QDs is chosen for silica coating, the highly fluorescent SQDNPs are chemically modified with amine, thiol and carboxyl groups, and then labeled by antibodies for particle-based immunofluorescence assay. The results indicate that the SQDNPs-antibody bioconjugates are alternative fluorescent probes useful for biodetection.

Abstract:The detection of Epstein-Barr virus-derived latent membrane protein 1 (LMP-1) is essential for understanding its contribution to the development of malignancy in epithelial cells and the early screening of nasopharyngeal carcinoma tumors. It is important to explore novel means for enhancing detection sensitivity of LMP-1.The current strategy for enhancing sensitivity is based on signal amplification of LMP-1/cadmium telluride (CdTe) quantum dots (QDs) functionalized silica nanosphere labels (Si/QD/Ab2). Si/QD/Ab2 was fabricated by covalently binding LMP-1 antibody (denoted Ab2) to CdTe QDs, which have been previously coated onto the surface of silica nanoparticles with EDC Chemistry. The as-prepared Si/QD/Ab2 label can be brought to a modified gold slice by a "sandwiched" immunoreaction, which was confirmed by SEM images and detected by square wave voltammetry (SWV).The amount of captured Si/QD/Ab2 by sandwiched immunoreaction was related to the concentration of LMP-1 in the incubation solution. The calibration range for LMP-1 detection was found to be 0.001 to 10 ng/ml with a correlation coefficient of 0.9897 and the lowest detectable concentration of 0.001 ng/ml. Compared with traditional sandwich immunoassay, the detection sensitivity of presented approach was enhanced largely due to the large surface area of silica nanoparticle carriers, which increased in CdTe QDs loading per sandwiched immunoreaction.The ease of functionalization, good monodispersed sizes and uniformity of the silica nanoparticles allows the QDs coated silica nanospheres to be highly suited for immunological labeling of trace protein analysis. The proposed method is simple, selective, reproducible, and can be extended to study protein-protein, peptide-protein, and DNA-protein interaction.

Using alpha-fetoprotein as a model and monitoring the variation of ECL intensity before and after immunoreaction, a label-free ECL biosensor was developed.

Abstract:To improve the detection sensitivity of porous silicon microcavity biosensors, CdSe/ZnS quantum dots are used to label complementary DNA molecules for the refractive index amplification and angular spectrum method for detection. In this method, the TE mode laser is used as the detection light and the light source is changed into a parallel beam by collimating and expanding the beam, which illuminates the PSM surface and receives the reflected light from the PSM surface through the detector. The angle corresponding to the weakest reflected light intensity before and after the biological reaction between probe DNA and complementary DNA of different concentrations labeled by quantum dots was measured by the detector, and the relationship between the angle change before and after the biological reaction and the complementary DNA concentration labeled by quantum dots was obtained. The experimental results show that the angle change increases linearly with increasing complementary DNA concentration. The detection limit of the experiment, as determined by fitting, is approximately 36 pM. The detection limit of this method is approximately 1/300 of that without quantum dot labeling. Our method has a low cost because it does not require the use of a reflectance spectrometer, and it also demonstrates high sensitivity.
