IL-25 and Its Receptor (IL-25R) in Allergic Inflammation: Its Role in VEGF-mediated Angiogenesis

Abstract:RATIONALE: IL-25 is thought to participate in allergic inflammation by propagating Th2-type responses. We hypothesised that it has distinct additional pro-inflammatory roles including the induction of angiogenesis. METHODS: The time course of accumulation of IL-25+ and IL-25R+ cells, and inparticular IL-25R+, CD31+ vascular endothelial cells was established in a human cutaneous model of the allergen-induced LPR in atopic subjects (n = 10) by immunohistochemistry. RT-PCR was employed to detect expression of IL-25R and VEGF in cultured human endothelial cells (HUVEC). The effect of IL-25 on angiogenesis in vitro was evaluated by assessing the development of microvessel structures in co-cultures of HUVEC and fibroblast cells. RESULTS: Following cutaneous allergen challenge in vivo, cells expressing IL-25 and IL-25R immunoreactivity peaked at 24 hr, while total CD31+ endothelial cells and those expressing IL-25R peaked later at 48 hr. IL-25 expression correlated with IL-25R expression on CD31+ cells at both time points (p < 0.05). In vitro, HUVEC constitutively expressed IL-25R and expression was further upregulated by IL-25 and TNF-α, both of which also increased VEGF expression. In the in vitro angiogenesis assay, IL-25 concentration-dependently increased the numbers, length and area of microvessel structures. Anti-VEGF pretreatment attenuated 80% of IL-25-induced angiogenesis. The PI3 K specific inhibitor LY294002 and the MEK specific inhibitor U0126, but not the p38 MAPK inhibitor SB203580 markedly attenuated IL-25-induced angiogenesis. CONCLUSIONS: IL-25 is elevated in allergic inflammation and contributes to angiogenesis by acting on endothelial cells partially through VEGF induction and through PI3 K and Erk/MAPK pathways.
