The Relationship Of Skeletal Muscle Akt/As160 Signaling Activity And Exercise Ameliorating Insulin Resistance

Abstract:Insulin resistance (IR) is a common pathophysiological feature of type 2 diabetes mellitus (T2DM). It is well known that regular exercise can attenuates IR, whereas the underlying mechanism is still not fully understood. Repetitive evidences also have revealed that dysfunction in glucose uptake is major cause of IR. AS160 plays an important role in mediating GLUT4 trafficking and glucose uptake. PURPOSE: To investigate the effect of 6 week aerobic exercise on Akt/AS160 signaling activity and glucose transport in skeletal muscle of normal control and IR mice. METHODS: IR animal model were created by feeding C57BL/6 mice with high-fat diet for 10 weeks. We then compared the effect of 6-week treadmill training (60 min/day) at 75% VO2max on mice in normal diet (NE) and high-fat diet (HE) groups with their sedentary control groups (NC and HC). IR was evaluated by Oral Glucose Tolerance Test (OGTT) and fasting insulin concentration. Skeletal muscle Akt2, AS160, GLUT4 gene expression were detected by RT-PCR Akt, pAkt-Ser473, AS160, pAS160-Thr642 and GLUT4 protein expression were detected by Western blot. RESULTS: Fasting serum insulin level was elevated by 77.19% (P<0.01) in mice of HC group whereas it was decreased by 25.33% (P<0.05) in HE group comparing with their control groups. An impaired glucose tolerance in mice of HC group was reversed by 6 week exercise (HE vs HC). 6 weeks exercise resulted in both Akt2 and GLUT4 mRNA expression changing. Akt2 expression in NE and HE groups increased 8.58% (P<0.05) and 12.71% (P<0.05) respectively comparing with their control groups. With regular exercise, GLUT4 gene expression increased 26.25% (P<0.01) and 24.20% (P<0.01) in NE and HE group, and Akt and GLUT4 proteins expression significantly increased by 3.86% (P<0.05) and 19.91% (P<0.05) in HE group comparing with their sedentary. In addition, GLUT4 protein expression also significantly increased in NE group after aerobic exercise; however, Akt protein was not affected in NE group. pAkt-Ser473 protein expression was increased by 14.65% (P<0.05) and 7.29% (P<0.05) in NE and HE groups compared to their sedentary control. And high fat diet down regulated pAS160-Thr642 protein expression by 30.38% (P<0.01) when compare HC and NC groups. pAS160-Thr642 protein expression were up-regulated by 16.20% (P<0.01) (NE vs NC) and by 15.56% (P<0.01) (HE vs HC) after 6-week exercise. However, total AS160 gene and protein expression were not affected by exercise (NE and HE) comparing with their sedentary control. CONCLUSION: 6-week aerobic exercise ameliorated negative condition of IR by activating Akt/AS160 signaling pathway, increasing glucose metabolism by promoting GLUT4 translocation.
