Thiamine Deficiency Induced β-amyloid Deposition and Tau Hyperphosphorylation

Abstract:Aim:To explore the effects of thiamine deficiency(TD)on β-amyloid protein(Aβ)deposition and tau hyperphosphorylation.Methods:TD animal model was produced by administrating thiamine deficiency diet combined with daily intraperitoneal injections of pyrithiamine.The control mice were fed with thiaminesupplemented diet and injecting saline intraperitoneally.All animals were sacrificed and the brain tissues were obtained at the 13th day after treatment.The pathological lesions of vulnerable regions in brain were observed by HE staining.Aβdeposition,tau hyperphosphorylation andβ-secretase expression were observed by immunohistochemical staining.Results:TD led to pathological lesions characterized by symmetric petechial hemorrhage mainly in submedial thalamus,and induced A βdeposition in cortex,hippocampus and thalamus.Thalamus as the vulnerable regions of TD exhibited more Aβdeposition.TD also exacerbated tau hyperphosphorylation and upregulatedβ-secretase expression.Control mice neither appear any damage of brain region nor Aβdeposition. Meanwhile,phosphorylated tau andβ-secretase positive cells were significantly lower than that of the TD mice(P<0.05).Conclusion:TD could induce Aβdeposition and promote tau hyperphosphorylation.The upregulateion of β-secretase expression may be involved in Aβdeposition induced by TD.
