Abstract:A hydrophilic bis(1,2,3-triazolyl)fluorene derivative was synthesized as a multi-photon-absorbing, zinc-ion-sensing fluorescent probe. The fluorescence response was approximately five-fold greater in presence of Zn(2+), resulting in a large binding constant (1 × 10(9)) for a 1:2 ligand to zinc complex. A four-fold increase in the two-photon absorption cross section was achieved upon binding Zn(2+). In vitro two-photon fluorescence microscopy imaging revealed a significant fluorescence increase upon introduction of Zn(2+) into HeLa cells and reversible Zn(2+) binding, demonstrating the potential of this probe for zinc ion sensing.

Abstract:A novel fluorescent sensor (AQTF1) based on the N-(quinolin-8-yl) tetrahydrofuran-2-carboxamide was designed and synthesized. This new sensor demonstrated high selectivity for the Zn2+ without the interference from Cd2+. The detection limit of this probe was calculated to be 10.8nM for Zn2+. The in situ prepared AQTF1-Zn2+ complex was used for detection of H2PO4− and displayed good selectivity from the common anions. Furthermore, the AQTF1 displayed good ratiometric response for the relay recognition for Zn2+ and H2PO4−.

Abstract:We have shown previously that 8-(5′-N,N-dimethylamino-1′-naphthalene)-sulfonamidoquinoline (DANQUIN) demonstrated a remarkable selectivity and sensitivity for the Zn(II) ion. In this work, the crystal structures of DANQUIN, Cu(DANQUIN)2 and Cu(DANPY)2 (DANPY, N-2-picolyl-(5′-N,N-dimethylamino-1′-naphthalene)-sulfonamide) are reported and compared with the simulated structure of Zn(DANQUIN)2, which is important for the understanding of the factors that govern the fluorescence of DANQUIN. Free DANQUIN mainly displays the fluorescence of the dansyl group at 547nm while the Zn(II)-DANQUIN complex mainly shows the enhanced fluorescence of aminoquinoline at 469nm, while the emission of the dansyl group shifted to 517nm with an almost constant intensity. This result demonstrates the advantage of this hybrid fluorescent chemosensor for Zn(II), and also makes it a potential candidate for ratiometric Zn(II) detection.
A novel one-stepped synthesized Schiff-base fluorescence probe for specific recognition of zinc ions with highly sensitive and its application in living cells

Abstract:Fluorescent turn-on receptors are extensively employed for the detection of Zn ions contamination in the environment due to its simplicity, convenience and portability. However, developing highly sensitive and cell-imageable fluorescent turn-on probe for the recognition of Zn ions in living organisms remains a significant challenge. Herein, we have successfully synthesized a novel Schiff base probe (H2L) with a significant fluorescence turn-on response (Zn ions) by one-step synthetic method. In this work, H2L exhibited high sensitivity to Zn2+ ions upon interaction with various common metal ions in HEPES buffer solution. Its detection limit is 1.87 × 10-7 M, which is lower than the requirement of Environmental Protection Agency (EPA) and World Health Organization (WHO) guidelines. The fluorescence titration and Job's plot analysis suggested a 1:1 binding ratio between the probe and Zn ion, and the single-crystal structures obtained further confirmed this inference. In addition, the fluorescent sensor demonstrated recyclability, maintaining its fluorescence intensity for up to 6 cycles without significant decrease, which holds promise for future investigations on reversible fluorescent chemosensors. Notably, fluorescence imaging experiments demonstrated that H2L could be successfully used for the detection of Zn2+ in live cells.

Abstract:A novel turn-on fluorescence probe L has been designed that exhibits high selectivity and sensitivity with a detection limit of 9.53 × 10−8 mol/L for the quantification of Zn2+. 1H-NMR spectroscopy and single crystal X-ray diffraction analysis revealed the unsymmetrical nature of the structure of the Schiff base probe L. An emission titration experiment in the presence of different molar fractions of Zn2+ was used to perform a Job’s plot analysis. The results showed that the stoichiometric ratio of the complex formed by L and Zn2+ was 1:1. Moreover, the molecular structure of the mononuclear Cu complex reveals one ligand L coordinates with one Cu atom in the asymmetric unit. On adding CuCl2 to the ZnCl2/L system, a Cu-Zn complex was formed and a strong quenching behavior was observed, which inferred that the Cu2+ displaced Zn2+ to coordinate with the imine nitrogen atoms and hydroxyl oxygen atoms of probe L.

Abstract:A novel fluorescent chemosensor with dansyl-appended tetrapeptide (identified as L), was designed and synthesized using solid phase synthesis (SPPS) technology. At room temperature, L displayed high water solubility and highly selective fluorescence "turn-on" response towards Zn2+ detection in 100% aqueous solutions. Conversely, L exhibited highly selective detection of Cu2+ through fluorescence quenching by a mechanism intrinsic to the paramagnetic species. The binding stoichiometry with L and Zn2+/Cu2+ ions were determined from the Job's plot to be 2:1, and further verified with fluorescence titration, ESI-HRMS and DFT-Calculations studies. The lowest measured detection limits for L were 13.2 nM for Zn2+ and 4.1 nM for Cu2+, respectively, with excellent sensitivity. In addition to that, L was successfully analyzed quantitatively for Zn2+ and Cu2+ ions using logic gate construction. Furthermore, L exhibited excellent cell permeation and low biotoxicity during bioimaging studies, L was able to selectively identify Zn2+ and Cu2+ ions in living LNcap cells, suggesting that the chemosensor L will be a promising biomolecular chemosensor for multifunctional detection of Zn2+ and Cu2+ through differential responses in environmental and biological systems.

Abstract:In the field of optical sensors, small molecules responsive to metal cations are of current interest. Probes displaying aggregation-induced emission (AIE) can solve the problems due to the aggregation-caused quenching (ACQ) molecules, scarcely emissive as aggregates in aqueous media and in tissues. The addition of a metal cation to an AIE ligand dissolved in solution can cause a “turn-on” of the fluorescence emission. Half-cruciform-shaped molecules can be a winning strategy to build specific AIE probes. Herein, we report the synthesis and characterization of a novel L-shaped fluorophore containing a benzofuran core condensed with 3-hydroxy-2-naphthaldehyde crossed with a nitrobenzene moiety. The novel AIE probe produces a fast colorimetric and fluorescence response toward zinc (II) in both in neutral and basic conditions. Acting as a tridentate ligand, it produces a complex with enhanced and red-shifted emission in the DR/NIR spectral range. The AIE nature of both compounds was examined on the basis of X-ray crystallography and DFT analysis.

Abstract:Zinc ions (Zn2⁺) is actively involved in diverse biological processes. Therefore, the precise detection of Zn2⁺ ion is an important object of increasing investigation. Although numerous fluorescent zinc ion detection probes have been developed, simple, biocompatible, and sensitive probes are still urgently needed. Herein, we reported two novel fluorescent probes, ZnTP1 and ZnTP2, by incorporating a quinoline fluorophore into a membrane-permeable zinc chelator tris(2-pyridylmethyl)amine. ZnTP1 exhibited a significant fluorescence enhancement in the presence of zinc ions through chelation-enhanced fluorescence (CHEF) processes, whereas probe ZnTP2 did not show any significant change in fluorescence due to the insertion of the carbonyl group. Further investigations revealed that ZnTP1 can effectively penetrate cell membranes and detect Zn2+ with high sensitivity in diverse biological samples, including living cells, plant tissues, and animal model zebrafish. This work suggests that ZnTP1 as a simple and efficient chemical probe has great potential for zinc ions detection in various biological contexts, thus providing a new tool for probing zinc ions in biosystems.

Abstract:Zinc (Zn2+) ions play a very important role in the environment and organisms, and it is of great significance to realize the high sensitivity and selectivity detection of Zn2+ ions. Herein, a new peptide-based fluorescent probe named DCH was rationally designed and synthesized by solid phase peptide synthesis with Fmoc chemistry. DCH exhibited excellent fluorescent "turn-on" responding to Zn2+ with highly selective and exhilaratingly sensitive in aqueous media, with detection limits (LOD) as low as 30.49 nM. ESI-MS, Job's plot curves and fluorescence titration results indicated that the stoichiometric ratio between DCH and Zn2+ was 2:1. DCH could be regenerated by adding EDTA solutions to the complex formed during interaction, and indicated that DCH was promising reversibility probe. Furthermore, DCH has excellent practical application potential through two actual water sample experiments. Importantly, DCH was successfully used in monitor cellular Zn2+ and EDTA in living HK2 cells and SV-HUC-1 cells based on good biocompatibility and exciting low toxicity.

Abstract:An efficient quinoline-based fluorescent chemosensor (QLNPY) was successfully developed for the detection of zinc ions (Zn2+). This novel chemosensor displayed higher sensitivity and selectivity toward Zn2+ over other competitive metal ions accompanying with obvious fluorescence enhancement. The QLNPY-Zn2+ complex can be further used as a new fluorescent "turn-off" sensor for pyrophosphate (PPi) and sulfur ion (S2-) via a Zn2+ displacement approach. The limits of detection were calculated to be 3.8 x 10(-8) M for Zn2+, 3.7 x 10(-7) M for PPi and 4.9 x 10(-7) M for S2-. The binding mechanism of QLNPY and Zn2+ was investigated through NMR, HR-MS analysis and further studied by crystallographic analysis. Additionally, further application of QLNPY for sequential bioimaging of Zn2+ and PPi was studied in HepG2 cells, suggesting that the quinoline-based chemosensor possesses great potential applications for the detection of intracellular Zn2+ and PPi in vivo. Abstract:A simple quinoline-based derivative (AQDPA) was developed as a highly efficient fluorescent sensor for sensing Zn2+ and sequential recognition of pyrophosphate and sulfur ions. This chemosensor was synthesized from diisopropylamine and 2-chloro-N-(quinolin-8-yl)acetamide. The non-fluorescence sensor displayed significant and specific fluorescence enhancement after binding to Zn2+ with high selectivity. The fluorescence titration analysis shows a detection limit for Zn2+ at 4.1 x 10(-8) M. The sensing mechanisms of AQDPA binding to Zn2+ were supported by H-1 NMR, HR-MS analysis, and density functional theory (DFT) study. Moreover, the fluorescence imaging in HeLa cells suggested that this simple sensor had great potential in the application of biological imaging.

Abstract:A novel ratiometric fluorescent sensor for Zn2+, 2-(2-(2-hydroxy benzoyl)hydrazinyl)-N-(quinolin-8-yl)acetamide (F-1) with quinoline as a fluorophore and salicylhydrazide as a receptor, was designed and synthesized. F-1 shows high selectivity and sensitivity for Zn2+ over Cd2+ in a methanol-tris-HCl (0.05 M) buffer solution (1 : 9, v/v, pH = 7.0). Upon the addition of Zn2+, an overall 5-fold increase in the fluorescence emission ratio (I-500/I-420) was observed. An intermolecular charge transfer (ICT) mechanism is suggested for this ratiometric sensor.

Abstract:A novel fluorescent probe containing an imine structure was synthesized through a condensation reaction based on the skeleton of antipyrine. Due to the synergistic effect of photoinduced electron transfer (PET), excited-state intramolecular proton transfer (ESIPT), and E/Z isomerization, the probe itself has weak fluorescence. When zinc ions are added to the ethanol solution of the probe, the formed complex inhibits PET, ESIPT, and E/Z isomerization while activating chelation-enhanced fluorescence (CHEF), resulting in fluorescent "turn-on" at 462 nm. Under optimal detection conditions, the probe can rapidly respond to zinc ions within 3 min, with a linear range of 60–220 μM and a lower limit of detection (LOD) of 0.63 μM. It can also specifically identify zinc ions in the presence of 13 common metal ions.

Abstract:A novel oxo-spirocyclic structure bridged ditopic Schiff base fluorescent probe L has been successfully synthesized and characterized. Interestingly, probe L exhibits a specific turn-on response to zinc(II) ion in fluorescence with a high selectivity and sensitivity. Further study show that probe L could be applied in imaging of live cells for sensing zinc(II) ion. By contrast, those compounds (L1, L2, and L3) with very similar structure to L lack this properties. The fluorescence intensity of L increases with Zn2+, which is less affected by other metal ions. The binding property of L with Zn2+ was studied by a combination of technologies, such as Job's plot, 1H NMR, ESI-MS, and DFT calculations. Moreover, we have successfully applied the probe L to detect Zn2+ in cancer cell. We are anticipating that this new oxo-spirocyclic structure bridged ditopic Schiff base may provide a new idea for the design of fluorescent probe.

Abstract:A readily available Schiff base quinoline derivative, quinoline-2-carboxaldehyde (pyridine-2-carbonyl)-hydrazine (QCPCH), has been synthesized via a one-step condensation reaction. It is used as a turn-on fluorescent probe for detecting Zn2+ with high selectivity, sensitivity, and rapid response time of less than 3 s. In addition, the probe QCPCH shows a low detection limit of 72 nM and a large linear detection range (0–180 μM). The mechanism of probe QCPCH for sensing Zn2+ is proposed to be based on the combined effects of chelation-induced enhanced fluorescence (CHEF) and inhibiting photoinduced electron transfer (PET); this is further demonstrated by the Job's plot analysis, Benesi-Hildebrand study, FT-IR spectrum, and 1H NMR titration. Furthermore, the QCPCH is successfully applied to determine Zn2+ in real water samples, and is made into test strips for fast, convenient, quantitative and qualitative detection of Zn2+.
