Abstract:Objective To investigate the expression of astrocyte-related gene during the differentiation of primary cultured ventral mesencephalic precursors(VMPs) from inbred rat fetuses in vitro.Methods Primary cultured VMPs cells were isolated from inbred rat fetuses.The cells were harvested and cultured for 7 days,and then induced for differentiation for 7 days.Total RNA of the cells was extracted on day 7,day 11 and day 14,then gene expression profiles were determined by Affymetrix Rat Expression Array 230 2.0 chips.Results The expressions of astrocyte related genes,such as GFAP gene,S100b gene,gap junction protein alpha1(Gja1) gene,Clu gene were all up-regulated during differentiation of VMPs,while the expressions of GDNF family members were not significantly changed.Conclusion During differentiation of VMPs into astrocytes,the expressions of GFAP,S100b,Gja1 and Clu were all up-regulated significantly,further investigation remains needed.
