Abstract:A method for analysis of phosphatidylcholine(PC) in rapeseed phospholipid by highly effective liquid phase(HPLC) was developed.In order to avoid the absorption of mobile phase at UV 200～214 nm,which interferes the detection of PC,evaporation light examination(ELSD) was used for quantitative analysis of PC.Results show the procedure for the developed method are as follows: Alltima silica 5 u,250 mm×4.0 mm column,eluent is composed of 10%(v/v) methanol and 90%(v/v) acetonitrile,degree of elution flow rate of 0.5 mL/min;ELSD detector drift temperature of 70 ℃,carrier gas flow rate 1.5 L/min and gain of 1.This detection procedure of PC has less interference and higher detection sensitivity,and is timesaving,completed in about 15 min.At PC range of 0.206 8～1.034 mg/mL,linear correlation coefficient is 1.00,coefficient of variation for repeat is 1.16%,good linear relationship and repeatability.This method shows good practicality.
Quantification of Soybean Phospholipids in Soybean Degummed Oil Residue by Hplc with Evaporative Light Scattering Detection

Abstract:yA high-performance liquid chromatographic method coupled with evaporative light-scattering detector was used for the accurate quantification of soybean phospholipids. This method is based on normal-phase chromatography with silica gel as stationary phase and a ternary gradient with n-hexane, isopropanol, and water as mobile phase. Major soybean phospholipid classes were separated by optimizing the solvent systems. The ternary solvent of n-hexane: isopropanol: water (53:42:5, by volume) was suitable for the separation of neutral lipids (NL), glycolipids (GL), and phosphatidylethanolamine (PE). By using hexane: isopropanol: water (17:66:17, by volume), phosphatidylcholine (PC), sphingomyelin (SM), and lysophosphatidylcholine (LPC) were separated. PE and phosphatidylinositol (PI), phosphatidic acid (PA), and PC were base line resolved by carefully adjusting the flow rate and switch time of intermediate gradients. The analysis was completed in 32min, and repeated injections of the samples were possible. The method has good repeatability and accuracy from the view point of high recovery, and low coefficients of variance in retention time and peak area for each phospholipid. Comparison was made between three detectors, i.e., ELSD, UV, and RI. The results show that ELSD is the best in phospholipids analysis.

Abstract:Lysophosphatidic acid (LPA) is a class of lipids that play multiple biological functions. Several reports show that they are potential biomarkers for diagnosing ovarian cancer. Therefore, it is necessary to accurately quantify their levels in biological samples. Here we report a high throughput LC/ESI/MS/MS (liquid chromatography electrospray tandem mass spectrometry) method employing a reversed phase C18 column to quantify LPA. In this method, a [13C16] labeled 16:0 LPA is used as the internal standard and the lipids are extracted out from biological samples using Bligh–Dyer method under highly acidic condition. The total run time is 8min. The detection limits of the assay reach fmol level and the CV% of the assay are within 10%. Using this method, we quantify the levels of six LPA species (16:0, 18:2, 18:1, 18:0, 20:4, and 22:6 LPA) in plasma samples. We find that some unknown compounds present in plasma can interfere with the quantification of LPA if they are not well separated from LPA. These unknown compounds are more hydrophobic than LPA and can be removed by thin-layer chromatography (TLC). We also find that the levels of LPA species in human plasma generally follow the order: 18:2 LPA>16:0 LPA, 20:4 LPA>18:1 LPA, 22:6 LPA, and 18:0 LPA.

Abstract:Objective To establish a method for determining the content of phosphatidylserine and phosphatidyl cholines by HPLC-ELSD.Methods Column:LiChrospher 100 Diol(100 mm×4.6 mm,5 μm);mobile phase:N-hexane-isopropanol-glacial acetic acid-triethylamine(825:170:5:0.8,A phase)and isopropanol-water-acetic acid-triethylamine(845:150:5:0.8,B phase)using gradient elution.The flow rate was 1.0 mL· min-1and detected by ELSD.Results The linear range of phosphatidylserine was 0.1-0.8 mg·mL -1with correlation coefficient 0.999 8.The linear range of phosphatidyl cholines was 0.1-0.8 mg·mL -1with correlation coefficient 0.999 6.The average recovery of phosphatidylserine was 96.40%(RSD=2.31%),and the average recov-ery of phosphatidyl cholines was 96.58%(RSD=1.55%).Conclusion The method has high specificity and accuracy,and can be used as a method for controlling the content of phosphatidylserine and phosphatidyl cholines.
Determination of phospholipids in peanut by direct-injection HPLC-ELSD

Abstract:A method was established to determine phosphatidylcholine (PC),phosphatidyl ethanolamine (PE),phosphatidylinositol (PI) and phosphatidate (PA) in peanut by direct-injection HPLC-ELSD.A modified Folch method was used to extract the total fat and then the extract dissolved in the mobile phase was taken directly into HPLC.The HPLC method was performed on a column of L-Si (250 mm ×4.6 mm,5 μm) by gradient elution using hexane-isopropanol (volume ratio 2∶3)and hexane-isopropanol-25 mmol/L ammonium acetate(volume ratio 3.6∶ 5.4∶1)as the mobile phases and detected by evaporative light-scattering detector.The correlation coefficient of standard curve for mass concentration and peak area of each phospholipid in linearity range was above 0.995,as well as the average recovery rates of the four phospholipids in peanut were in the ranges of 93.59％-108.57％ with the relative standard deviations (RSDs) less than 10％.The method had advantages of good reproducibility,high stability and simple pretreatment and it should be used for separation and determination of phospholipids.

Abstract:OBJECTIVE:To establish a RP-HPLC method for the determination of total luteolin-glucosides content in Elsholtzia blanda Benth extracts (EBBE). METHODS The luteolin content was determined on an ODS C18 column, the mobile phase consisted of 0.01 mol· L-1 phosphorate buffer (pH 2.0) -tetrahydrofuran-isopropynol (70:30:5) as mobile phase with detection wavelength 360 nm. RESULTS The recoveries of the method were 95.56% - 105.58% , and the assay was liner at concentration from 2 to 400 μg·mL-1( r = 0.999 9) .The detection limits was 50 ng·mL-1(S/N≥3) , the limits of quantification was( 2.40±0.13)μg·mL-1,the content of luteolin in EBBE was (3.12±0.15)% . CONCLUSION This method is simple, accurate and can be used to optimize extraction procedure and determine the content of luteolin-glucosides in Elsholtzia blanda Benth extracts.
Selective separation of phosphatidylserine and phosphatidylcholine with ionic liquids as extractants

Abstract:Homologues separation is a key process in the production of high-purity drugs and chemicals. High-purity phosphatidylserine (PS) is widely used as drugs and functional additives; however PS often appears in a mixture with structurally similar homologues and the selective separation of phospholipid homologues remains challenging. Here, a novel extraction method was developed to separate PS and phosphatidylcholine (PC) with ionic liquids (ILs) as extractants. The effects of structure and concentration of ILs and temperature on extraction performance were investigated. The results showed that the structure of the IL's anion had a significant impact on the separation selectivity, and an obvious synergistic extraction effect between IL and polar diluent was observed. In 1-ethyl-3-methylimidazolium bromide([EMIm]Br)-methanol-hexane biphasic systems, the selectivity of PC to PS (SPC/PS) reached up to 29.48, when the mole fraction of IL was only 5％. Additionally,ab initio calculations were used to explore the extraction mechanism, and the results indicated that the multiple hydrogen-bonding and electrostatic interactions were formed between phospholipids and ILs, which is probably beneficial for the selective separation of phospholipid homologues.

Abstract:Comparison of HPLC coupled with each of the detectors ultraviolet detector(UV),differential refractive index detector(RI) and evaporative light scattering detector(ELSD) was carried out for the determination of monoglycerides.Serious interference from impurities,low signal intensity and weak sensitivity were observed when using UV detection.However,RI detection showed good linearity over a high concentration range despite not being suitable for the detection of low-concentration samples.Similarly,ELSD detection exhibited excellent linearity over the concentration range of 0.040 to 5.000 mg/mL,with a correlation coefficient of 0.9998;the recovery of ELSD detection was 92.94%,with a relative standard deviation of 2.24% and a coefficient of variation of 2.41%.HPLC-ELSD was sensitive,accurate and repeatable and applicable for the determination of monoglycerides in emulsion.

Abstract:A method for the determination of egg-yolk phosphatidylcholine by normal-phase high performance liquid chromatography (NP-HPLC) with evaporative light scattering detection (ELSD) was developed. Yolk phosphatidylcholine was successfully separated on a Nova-Pak Silica 60A column (3.9 mm i.d. x 150 mm, 4 microm), using hexane-isopropanol-water containing 3% acetic acid (35: 65: 8, v/v/v) as mobile phase at a flow rate of 1.0 mL/min. The drift tube temperature and air carrier gas pressure of the ELSD were set at 50 degrees C and 350 kPa, respectively. The linear detection range was 0.16 - 1.61 g/L (r2 = 0.997 9) and the limit of detection (LOD) was 0.64 microg with excellent relative standard deviation (RSD) of 3.2% (n = 5). The recovery was in the range of 98.2% - 128.2%. The results indicated that this method is accurate, rapid, simple and reproducible, and thus suitable for the analysis of egg-yolk phosphatidylcholine in real samples.

Abstract:A hyphenated method of nonaqueous capillary electrophoresis coupled to electrospray ionization mass spectrometry (NACE-ESI-MS) is described for the simultaneous analysis of phospholipids. The best results were obtained with a mixed solution of methanol/ACN (40:60 v/v) containing 20 mM ammonium acetate and 0.5% acetic acid, under the applied voltage of 30 kV and capillary temperature of 25 degrees C. ESI-MS measurements were performed in the negative mode with methanol/ACN (40:60 v/v) containing 50 mM ammonium acetate as sheath liquid at a flow rate of 2 mu L/min. Different phospholipid classes have been successfully separated within 16 min, and the molecular species of every single class have been identified by using MS2 or MS3, which generates characteristic fragments through CID. The developed method has been applied to analyze the phospholipids extracted from rat peritoneal surface and the molecular species of phospholipid classes are presented.

Abstract:A high-performance liquid chromatographic method with evaporative light-scattering detector was used to quantify dipalmitoylphosphatidylcholine of pulmonary surfactants in bronchoalveolar lavage fluid of rat.Chromatographic analysis was carried out on YWG-SiO2,methanol-water(11∶2,V/V) as the mobile phase A,and pure chloroform as the mobile phase B(A∶B=1∶39,V/V).The samples were dissolved by chloroform-methanol-water.Correlation coefficients for dipalmitoylphosphatidylcholine were 0.9971.The method precision ranged from 10.9% to 13.4%,the recoveries ranged from 98% to 115%.The method described in the present study is sensitive and reproducible for the determination of dipalmitoylphosphatidylcholine of pulmonary surfactants in bronchoalveolar lavage fluid of rat.

Abstract:In this study, we developed an AC-induced interfacial electro-epoxidation reaction for localization of double-bonds in nega-tively and positively charged forms of lipids simultaneously. An AC voltage plays multiple roles in this method, including initiation of the electrochemical epoxidation of double-bonds in both charge states of lipids, and protonation and deprotona-tion of lipid for detection in both ion modes. The advantages of simultaneous detection of negatively changed and positively charged unsaturated lipids, voltage-controlled electrochemical derivatization, and the low sample consumption allow its wide applications in a broad range of lipid-related research.

Abstract:Phospholipids are the main constituents of biological membranes and their biological function has been increasingly recognized. Therefore, there is an unmet need to develop methods capable of quantifying a wide range of phospholipids with high sensitivities and high throughput. We employed an ultrahigh-performance liquid chromatography system coupled to a triple-quadrupole mass spectrometer (UHPLC-MS) and developed a method that can quantitatively analyze 10 major classes of phospholipids in biological samples in 11 min. These are phosphatidic acid, phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylinositol, phosphatidylserine, sphingomyelin, lysophosphatidic acid, lysophosphatidylcholine and lysophosphatidylethanolamine. The limit of detection (LOD) and limit of quantitation (LOQ) are 0.04-33 pmol mL-1 and 0.1-110 pmol mL-1, respectively. The method takes three steps: first and second steps identified phospholipid structures in a mixture containing aliquots of all the samples using the combinations of multiple reaction monitoring (MRM), product ion scan and retention time in the positive and negative ion modes. These steps enabled the identification of phospholipids present in the samples and provided information on efficient sample analysis in the final step of sample quantitative analysis. We have developed fast and sensitive label-free quantitation with normalization of the acyl chain length to achieve more accurate quantification. The method developed was applied to analyze 6 different biological samples (plasma, cells and tissues) for applicability validation, where a total of 308 phospholipid species across 10 phospholipid classes were identified and 295 phospholipid species were quantified. The method is highly efficient, sensitive, and is universally applicable.

Abstract:Dimyristoylphosphatidylglycerol (DMPG), dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE) were characterized by electron impact (EIMS), field desorption (FDMS) and liquid secondary ion (LSIMS) mass spectrometry. The results showed that the fragmentation processes of the three phospholipids are very similar. In the EI mass spectra, no molecule-ion peaks were found, the peaks of the fatty acid chains being dominant. The results of FDMS and LSIMS showed that both techniques worked particularly well with all three kinds of phospholipids. The FD mass spectra of all three phospholipid gave strong MH+ peaks, and no fragmental peaks were found except that of DMPC. With LSIMS, the fragmentations of all the phospholipids were extensive and clear, and gave dominant peaks of MH+. The cleavages around phosphate groups were the most abundant. An interesting phenomenon was found in the LSIMS measurements that all three kinds of phospholipids formed intense peaks of molecule-ion clusters, [2M + H](+), [3M + H](+) and [4M + H](+), etc. It was found that the formation of the molecule-ion clusters was related to several factors: concentration, ionic strength, pH, etc., among which concentration is the most important. The degree of cluster formation increased with increase in concentration of phospholipid. The results also showed that the nature of the matrix is critical to a successful LSIMS analysis of phospholipids, and (S)-glycerol was confirmed as a very satisfactory matrix for these phospholipids. Separation and Determination of Phospholipids in Plant Seeds by Nonaqueous Capillary Electrophoresis

Abstract:A method has been developed for the separation and determination of phospholipids by nonaqueous capillary electrophoresis in a separation medium of acetonitrile-2-proponol (3:2, v/v), 0.3% acetic acid and 60 mM ammonium acetate. To optimize the separation conditions, the composition of separation medium including alcohols, acetic acid, n-hexane and ammonium acetate was studied. The solvation interaction and ion-dipole interaction were also investigated. The contents of phospholipids in soybean, sunflower, peanut, apricot kernel, filbert and walnut were determined by the recommended method. The results obtained by the nonaqueous capillary electrophoreses were in good agreement with those determined by micellar electrokinetic chromatography.

Abstract:The protein sample extracted from rat liver tumor tissues and its enzymic hydrolysates were detected by high performance liquid chromatography-laser induced fluorescence detector(HPLC-LIF) (lambda(ex) = 473nm, lambda(em) = 525 nm) after labeled with 4-fluoro-7-nitro-2, 1, 3-benzoxadiazole (NBD-F) in dark at room temperature for 1 h with borate buffer (50 mmol/L, pH = 8.5) as reaction medium, and the results were compared with those obtained from HPLC-UV (at 214nm) without derivatization. The analysis of enzymic hydrolysates shows that LIP detector can greatly improve the detection by its high sensitivity: the peak intensities of most components are 10 times higher than UV detection and 42 peaks more were observed. For the analysis of protein sample, most of the peaks detected by UV are located in strong polarity area, which indicates that these components with strong polarity have high UV absorption. The signals obtained from LIF are located in weak polarity or non-polarity area. One reason is that derivatization can change the polarity of the components, and the other is that components with weak or non-polarity might be labeled more easily. This result also suggests selectivity difference between these two methods over the real protein sample. In addition, selective label of complex sample by changing reaction conditions (such as pH and reaction media) enables to achieve high sensitive detection of low abundance proteins and peptides with better selectivity.
