Abstract:BACKGROUND:Bone morphogenetic proteins(BMPs),multifunctional secreted signaling molecules,are members of transforming growth factor-β(TGF-β) superfamily.BMP could regulate embryonic morphology,organ development and germ cell differentiation by signal cascades.They could also induce stem cell differentiation and apoptosis.The signal transduction pathway of BMPs is various in different stem cells.Although the reason remains uncertain,some studies have suggested that it may be related with the receptor or the integration of various signals.OBJECTIVE:To review the role and transduction mechanism of BMP signal pathway in several stem cells.RETRIEVAL STRATEGY:Using the keyword of "bone morphogenetic protein,the differentiation and proliferation of stem cell",we searched Pubmed database for related English articles dated between April and July 2007.A total of 45 articles were collected,after the first trail only the articles ①related to bone morphogenetic protein signal transduction pathway and ②published recently or in authoritative journal in the same research field were selected.Repeated research and Meta analysis articles were excluded.LITERATURE EVALUATION:Most articles were analysis of BMP signal transduction.Of the 31 selected articles,2 were review articles,and the others were clinical or basic empirical studies.DATA SYNTHESIS:①BMP signals belong to kinase transduction system,which can translate signals by Smads or p38 MAPK pathway and be regulated by many additional intracellular and extracellular parameters.②BMP signal strength,timing and downstream pathway choices can strongly influence the proliferation and differentiation of early embryonic stem cells,which likely relate to Wnts on multiple levels of the cascades.③BMP signals could induce neural stem cells to differentiate towards neuron and neuroglial cells,and inhibit the differentiation of pancreatic progenitors.BMP4 regulates the number of hair cells by inhibition differentiation and stopping progenitor proliferation,and promotes the proliferation and apoptosis of embryonic cells within the eye.Nodal and BMP signals could regulate the development,differentiation and proliferation of mesodermal progenitor cells.CONCLUSION:BMP signals,which are regulated by many additional intracellular and extracellular parameters,could influence the differentiation and proliferation of stem cells and transcriptions of target gene.But BMP is only the regulatory factor but not the marker of the proliferation and differentiation of stem cells.Currently,the molecular and cytology mechanisms,timing and spacing of BMP signal are not yet identified.
