Abstract:To evaluate the edible safety of anti-radiation preparations of tea polyphenol,the experiment included acute toxicity test,Ames test,micronucleus test,sperm shape abnormality test and thirty days feeding test were performed.Results showed that the oral maximum tolerance dose(MTD) was higher than 20.0 g/kg·bw both in female and male mice and rats.The results on toxicity of Ames test,micronucleus test,sperm shape abnormality test and mutagencity were negative.No toxicological symptom was appeared in tested animals from thirty days feeding test.It is concluded that the anti-radiation preparations of tea-polyphenol was classified as non-toxicology and genotoxicity,high edible safety.

Abstract:The edible safety of mulberry-leaf tea bag was investigated according to the Standard of Health Food Test and Asses.Acute toxicity test in mice showed that LD50 was higher than 30 g/kg Bw,all three genetic tests showed negative results,30 days feeding test showed that the outer appearance,behaviors,weights,organ coefficients,the indexes of hematology and biochemistry in rats have no significant difference comparing with the control group,besides,significant pathological changes of inherent organs were not discovered.All results suggested that mulberry-leaf tea bag has no acute toxicity,no genotoxicity,and high in edible safety.

Abstract:Accumulation of potentially toxic elements in soil and tea leaves is a particular concern for tea consumers worldwide. However, the contents of potentially toxic elements and their potential health and ecological risks in Chinese tea gardens have rarely been investigated on the national scale. In this study, we collected 225 paired soil and tea plant samples from 45 tea gardens in 15 provinces of China to survey the current risk of potentially toxic element accumulation in Chinese tea gardens. The results suggest that the average contents of most trace metals in rhizosphere soils meet the risk control standard for agricultural land in China. However, the mean contents of As, Cr, Cd, Zn, Cu, and Ni in rhizosphere soils were 1.94, 2.14, 1.23, 1.15, 1.18, and 1.19 times their corresponding background soil values in China. Cd had the highest geo-accumulation index, followed by As, Zn, Cr, Ni, Cu, Pb, and Mn in rhizosphere soils. Nearly 2.22% and 4.44% of soils were moderately to heavily contaminated with As and Cd, respectively. The risk index ranged from 18.0 to 292, with an average value of 90.0, indicating low to moderate ecological risk in Chinese tea gardens. This is the first national-scale reconnaissance of trace metals in tea across China. Our findings provide a useful reference for ensuring the quality and safety of tea production and mitigating the risk of toxic element accumulation in tea.
Tea Consumption And The Risk Of Endometrial Cancer: An Updated Meta-Analysis

Abstract:Although the relationship between tea consumption and the risk of endometrial cancer has been previously analyzed in certain studies, the resulting information is still conflicting, and a previous meta-analysis yielded inconsistent results. Therefore, here, we aimed to perform an updated meta-analysis of studies on this subject in order to elucidate this relationship. We searched the literature on the PubMed, Web of Science, Scopus, and Chinese National Knowledge Infrastructure databases for studies that were published prior to September 25, 2019, and all the relevant references were examined. Ultimately, we included eight studies, and seven of them were on black tea. We used the overall relative risk values (RR) and 95% confidence intervals (CI) to evaluate the risk. The synthetic RR of the eight eligible studies demonstrated that tea consumption was not relevant to the incidence rate of endometrial cancer (RR 1.06, 95% CI 0.96, 1.18). No publication bias was found. We detected significant heterogeneity among the studies (Q = 15.84,p = 0.027,I-2= 55.8%). In conclusion, the results of this meta-analysis indicate that tea consumption is not relevant to the incidence of endometrial cancer. Further research and cohort studies should be conducted to validate our result.

Abstract:The flower of tea (Camellia sinensis L.) plant has been paid an increasing attention in the last twenty years, since it was found that tea flowers contained representative constituents similar to those of tea leaves, such as catechins, caffeine and amino acids. Tea flower is theoretically valuable although it has been considered as an industrial waste over a long period of time. This review summarizes the research findings conducted until now on physiological genetics, chemical composition, health benefits and toxicology of tea flowers, aiming to foresee their future applications. A lot of genes are involved in flower development and the synthesis and transmission of various chemicals in tea flowers. The chemical composition of tea flower consists mainly of catechins, polysaccharides, proteins, amino acids and saponins and thus tea flower possesses various health benefits such as antioxidant, anti-inflammatory, immunostimulating, antitumor, hypoglycemic, anti-obesity and anti-allergic activities. Moreover, tea flower contains a protease that can elevate the free amino acids content in the tea infusion by almost two folds. More importantly, the enzymatic activity of the protease is much higher than that of the commercially available proteases. Additionally, aqueous extracts of tea flower are demonstrated to safe to animals. Thus, the potential uses of tea flowers in food and medical fields are warranted.

Abstract:The purpose of this study was to evaluate the mutagenicity and safety of water extract of the fruit hull of Camellia oleifera Abel (WECO), which was prepared using hot-reflux method. The oral maximum tolerated dose (MTD) of WECO was above 20 g/kg body weight both in rats and in mice, which can be regarded as virtually nontoxic. No mutagenicity was found in Ames test, mouse bone marrow cell micronucleus test and mouse sperm abnormality test. In the subacute study, the SD rats were administered orally at 0.5, 1, or 2 g/kg/BW for 30 d. There were no treatment-related toxic effects from WECO. No significant differences were found in parameters of body weight, hematology value, clinical chemistry value, and organ/body weight ratio. The level of no observed adverse effect level (NOAEL) for WECO was 2 g/kg/BW for subacute toxicity study.

Abstract:Fermented puerh tea, having undergone a long period of secondary oxidization and fermentation, has become more and more popular in recent years. In the present paper, a safety evaluation of aqueous extracts from fermented puerh tea (EFPT) was performed, including an oral acute toxicity study in rats and mice, mutation tests, a mouse micronucleus test, mouse sperm abnormality test and a 30 day feeding study in rats. Meanwhile, the antihyperlipidemia effect of EFPT was investigated as well. It was found that the oral maximum tolerated dose of EFPT was more than 10.0 g per kg body weight both in rats and mice. And it had no mutagenicity as judged by negative experimental results of the mutation test. No abnormal symptoms, clinical signs or deaths have been found in rats in each group throughout the experiments. In addition, EFPT in this study showed certain effects on hyperlipidemia.

Study on the Antioxidant Activity of Tea Flowers (camellia Sinensis)

Abstract:Major chemical compounds in different extracts from tea flowers (Camellia sinensis) were analyzed. Distilled water or 70% ethanol extracts were then fractionated with chloroform, ethyl acetate and n-butanol, respectively. Each extract fraction was tested its scavenging activities on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl free radicals. The results showed that ethyl acetate fraction of ethanol-extract of tea flower (EEA) exhibited the highest quenching activity to hydroxyl radicals (SC50 11.6 mug/ml), followed by ethanol-extract (EE) of tea flower (SC50 19.7 microg/ml). Same tea flower extract showed big different scavenging activities on different free radicals. EEA quenched 80% of hydroxyl radicals generated by Fenton's reaction, however, only 40% of DPPH radical was scavenged in the Fe (II)-H2O2 -luminol system. The contents of flavones, polyphenols and catechins in EE and EEA fractions were higher than those in other fractions. We suggest that the stronger scavenging abilities to free radicals might be due to polyphenols, EGCG, ECG and flavones. However, the water extracts of tea flower and their fractions showed lower antioxidant activity for their inhibitory effect on hydroxyl radicals and DPPH radicals.

Abstract:Ethnopharmacological relevance: Pu-erh green tea, originally produced in the Yunnan province of P.R. China for about 1700 years, is believed to be beneficial to health in Asian countries. The potential toxicity of Pu-erh green tea when administered at high doses via concentrated extract, however, has not been completely investigated.The aim of the study: The present study was aimed to evaluate the potential toxicity of Pu-erh green tea extract (PGTE) of sub-chronic administration to Sprague Dawley (SD) rats.Materials and methods: Growing SD rats were administrated orally by gavage with PGTE at doses of 0, 1250, 2500, and 5000 mg/kg/day for 91 consecutive days. Clinical observations, including survival, hematology, serum biochemistry, urinalysis and histopathological examination were measured to monitor treatment-related adverse effects in rats.Results: The results showed that oral administration of high dose of PGTE led to body weight gain suppression, liver and calcium deposition dysfunctions.Conclusions: In conclusion, the no-observed-adverse-effect level for Pu-erh green tea extract derived from the results of the present study was 2500 mg/kg/day for both genders. Abstract:Water-soluble polysaccharide from tea plant flower (TFP) was screened for its biological and biophysical effects. TFP-1, one of the major constituents, yielded after removing the proteins and purification by Sephadex G-100 (Sigma-Aldrich Pty Ltd., Castle Hill, Australia). It was subjected to chemical identification and the result indicated its average molecular weight (132 kDa) and its formation which consisted of xylose, glucose, arabinose and galactose with the molar ratio of 1.0: 2.9: 4.6: 21.8. Presence of TFP-1 displayed reactive oxygen radicals scavenging activity and it showed dose-dependent antioxidant activity by comparing with three commonly used antioxidants. The research showed that administration of TFP-1 for 28d continuously was found not only to be protective on liver lipid peroxidation induced by bromobenzene in mice through increasing the activity of superoxidase dismutase, total antioxidant capacity, but also markedly to attenuate the enhancement of malondialdehyde content in dose-dependent manner. Altogether, these results suggest that TFP-1 could be considered as a potential antioxidant.

But its potential toxicity when administered at a high dose as concentrated extract has not been completely investigated.Aim of the study: The present study was aimed at evaluating potential reproductive and developmental toxicities of Pu-erh black tea extract (BTE) in Sprague Dawley rats.Materials and methods: Growing rats were given BTE by gavage at levels of 0, 200, 700 and 2500 mg/kg/day as the F-0 generation in reproductive toxicity study. Additionally, BTE was administered to mate female rats from gestation day 0.5 through 19.5 at the doses of 0, 200, 700 and 2500 mg/kg/day to evaluate the developmental toxicity.Results: In the reproductive toxicity study, only 2500 mg/kg/day BTE reduced the body weight gain and altered the relative organ weights including testes, prostata and ovary both for F-0 parents and F-1 offspring compared to the controls. High dose of BTE (2500 mg/kg/day) administration caused developmental disturbances in embryo-to-foetus period including resorbed embryos, decreased embryo size and skeletal anomalies.Conclusion: In conclusion, the no-observed-adverse-effect level of BTE is 700 mg/kg/day both for reproductive toxicity and developmental toxicities. Study on Teratogenicity of Tea Flower in Rat

Abstract:To study the teratogenicity of tea flower in rats, SD pregnant rats were divided randomly into four groups, each containing at least 12 rats. Tea flower extract was administered to rats at dose levels of 0, 0.50, 1.00 and 2.00 g/kg by gavage during days 7-16 of gestation. Body weight of the rats was recorded during the pregnancy and females were euthanized by cervical dislocation on the 20th day of gestation. The uterine horns were exteriorized through a midline abdominal incision, opened and examined for number of implantation sites, living and dead fetuses, resorption sites. The weight, length and external malformations of each living fetus were also measured and examined. Half of each litter was stained by alizarin red for skeletal examination, and the other half dyed by Bouins staining solution to measure visceral anomalies. The body length of fetuses from low dosage group (0.50 g/kg) was higher than that of the control (P<0.01). No abnormalities were observed in other three dosages groups, compared with the negative control group. It suggested that tea flower had no embryonic and teratogenic toxicity to SD pregnant rats.

Abstract:Pu-erh teas are believed to be beneficial beverages for health since they possess several pharmacological properties such as antioxidation, hypocholesterolemia, and antiobesity properties, but their potential toxicities when administered at a high dose as concentrated extracts have not been completely investigated. In this study, the chemical components in Pu-erh green tea and Pu-erh black tea were analyzed and compared, and the safety of tea extracts was evaluated in Wistar rats. The polysaccharide, tea pigment, and flavonoid levels were substantially increased in the Pu-erh black tea, while the polyphenol and free amino acid levels were higher in unfermented green tea. Low toxicities of Pu-erh green tea extract (GTE) were observed at doses of 2500 and 5000 mg/kg/day with a 28-day subacute study. Serum biochemical data including alanine aminotransferase increased to 5000 mg/kg/day GTE males, and creatinine (Cr) increased in all 5000 mg/kg/day GTE groups and 2500 mg/kg/day GTE males. Slight bile duct hyperplasia in the liver was also observed. The target organs of GTE were considered to be the liver and kidney. Comparatively, no adverse effects were observed in Pu-erh black tea extract (BTE)-treated rats. In conclusion, a dose of 1250 mg/kg/day for GTE and 5000 mg/kg/day for BTE following oral administration could be considered safe under the conditions of this study.

Abstract:Camellia sinensis L., its fresh leaves and buds are used to make tea, is an important industrial crop with a long history. However, less attention has been paid to tea flowers. Indeed, tea flower extract (TFE) is a rich source of functional molecules, but its nutritional value remains unclear. This study, from the perspective of "whole food", aimed to investigate the composition of TFE and further explore its possible health-promoting effects on cyclophosphamide-induced mice. It was found that TFE was mainly composed of carbohydrates (34.02 +/- 1.42%), phenolic compounds (11.57 +/- 0.14%), crude proteins (27.72 +/- 3.07%) and saponins (2.81 +/- 0.00%). Supplementation of TFE at 200 mg/kg.BW/d regulated intestinal homeostasis by improving the intestinal barrier, alleviating dysbacteriosis (reverse 44 of 68 disordered genera), stimulated immunoreactions with significant enhancement of serum TNF-alpha, IFN-gamma, IL-1 beta, IL-2 and IL-6. Furthermore, TFE could improve the liver function through decreasing the hepatic malondialdehyde and aminotransferase levels and increasing the levels of catalase, myeloperoxidase, superoxide dismutase and reduced glutathione. Notably, the ameliorating effects of TFE on cyclophosphamide-induced immunosuppression and the hepatic injury were associated with its modulation of gut microbiota. The results provide the evidence for the application of tea flower as potential functional food.
Acute and Subacute Safety Evaluation of Black Tea Extract (herbt Tea Essences) in Mice

Abstract:Theabrownin (TB) is a heterogeneous biomacromolecule, extracted from tea, with many functional groups. Importantly, TB possesses diverse health benefits, such as antitumor activity and blood lipid-lowering effects. Presently, the content of TB in tea extract is relatively low. Here, we obtained a deep-processed black tea extract with a high content of TB (close to 80%), which was named Herbt Tea Essences (HTE). Currently, this study was designed to evaluate the biosafety of high-content TB products on mice. We implemented acute and subacute toxic experiments to assess its safety on organs, the serum biochemical and molecular levels. In the acute exposure study, we found that the median lethal dose (LD50) value of HTE was 21.68 g/kg (21.06–24.70 g/kg, greater than 5 g/kg), suggesting that HTE had a low acute toxicity. In the 28-day subacute exposure study, our results showed that no abnormal effects were observed in the 40 and 400 mg/kg/day HTE-treated groups. However, we observed slight nephrotoxicity in the 4000 mg/kg/day HTE-treated group. The HTE-induced nephrotoxic effect might involve the inflammatory response activation mediated by the nuclear transcription factor kappa-B (NF-κB) signaling pathway. This study would provide valuable data for the TB safety assessment and promote this natural biomacromolecule application in daily drinking.
Structural characterization and antioxidant activities of 2 water-soluble polysaccharide fractions purified from tea (Camellia sinensis) flower.

Abstract:The water-soluble crude polysaccharide tea flower polysaccharide (TFP), obtained from tea (Camellia sinensis) flower by boiling-water extraction and ethanol precipitation, was fractionated by Sephadex G-100 column chromatography, giving 2 polysaccharide fractions termed TFP-1 and TFP-2. The structural features of TFP-1 and TFP-2 were investigated by high-performance liquid chromatography (HPLC), gel-permeation chromatography (GPC), rheometer, infrared (IR) spectra, nuclear magnetic resonance (NMR) spectroscopy, atomic force microscope (AFM), and scanning electron microscope (SEM). Results indicated that TFP-1 was composed of glucose: xylose: rhamnose: galactose = 1.0:1.2:0.81:0.98 with a molecular weight of 167.5 KDa, while TFP-2 comprised glucose: xylose: rhamnose: arabinose = 1.0:0.76:2.3:2.3 with a molecular weight of 10.1 KDa. The 1H NMR revealed that TFP-1 contained alpha-L-Rhap, alpha-D-Galp, alpha-D-GalpNAc, alpha-D-Xylp, alpha-D-Glcp, and beta-D-Glcp residues, while TFP-2 was illustrated to have alpha-L-Rhap, alpha-L-Arap, alpha-D-Xylp, alpha-D-Glcp, and alpha-D-GlcpNAc residues. Antioxidant activities of these fractions were investigated using various in vitro assay systems compared with ascorbic acid. In conclusion, TFP-2 exhibited the higher antioxidant activities and could be explored as a novel potential antioxidant. Practical Application At present, commonly low-grade tea is preferred to extract the tea polysaccharide, to take full advantage of tea flower resource to extract polysaccharides can greatly reduce the cost of tea products. Thus, the search for plant-derived biomaterials from this study could generate natural value-added products from underutilized tea plant waste and used as a medicinal agent against chronic health problems, such as cancers, aging, and atherosclerosis caused by reactive free radicals that produced from oxidation.
Toxicological Research and Safety Evaluation for an Antioxidant Preparation Derived from Puerh Tea

Several tests were performed, including acute toxicity test, Ames test, micronucleus test, aperm ahape abnormality test and thirty days feeding test. It was found that oral maximum tolerated dose( MTD) was higher than 20.0 g/kg·bw both in male or female mice and rats. Ames test,micronuleus test and sperm shape abnormality test showed positive results. No significant toxicity was detected in thirty days feeding test. Results indicated that antioxidant preparation in puerh tea had no acute toxicity, no genotoxicity, and high edible safety.
Anti-Inflammatory Effects of a Polyphenols-Rich Extract from Tea (camellia Sinensis) Flowers in Acute and Chronic Mice Models

Abstract:While beneficial health properties of tea leaves have been extensively studied, less attention is paid to the flowers of tea. In this study, the anti-inflammatory effects of hot water extract of tea (Camellia sinensis) flowers were investigated. Pharmacological studies found that administration of tea flowers extract (TFE) could effectively inhibit croton oil-induced ear edema and carrageenin-induced paw edema. Furthermore, administration of TFE also protected against Propionibacterium acnes (P. ances) plus lipopolysaccharide-(LPS-) induced liver inflammation by reversing the histologic damage and plasma alanine aminotransferase (ALT) increase. Moreover, the levels of nitric oxide (NO), tumor necrosis factor-(TNF)-α and interleukin-(IL-) 1β mRNA in mouse liver were markedly suppressed after treatment with TFE in mice with immunological liver inflammation. These results indicated that tea flowers had potent anti-inflammatory effects on acute and immunological inflammation in vivo, and may be used as a functional natural food.
Isolation and identification of compounds from the ethanolic extract of flowers of the tea (Camellia sinensis) plant and their contribution to the antioxidant capacity

Abstract:While beneficial health properties of tea leaves have been extensively studied, less attention has been given to that of flowers of the tea (Camellia sinensis) plant. In this work, the ethanolic extract and its ethyl acetate-soluble fraction (EEA) from the tea flowers were found to possess the potent antioxidant activity using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging assay. The compounds present in EEA had comparatively strong DPPH scavenging activity and strongly contributed to the antioxidant activity of the tea flowers. From EEA, besides eight catechins, five flavonol glycosides were isolated and their structures were elucidated on the basis of mass spectrometry and nuclear magnetic resonance spectroscopy as myricetin 3-O-β-d-galactopyranoside, quercetin 3-O-β-d-galactopyranoside, kaempferol 3-O-β-d-galactopyranoside, kaempferol 3-O-β-d-glucopyranoside, and kaempferol 3-O-[α-l-rhamnopyranosyl-(1-6)-β-d-glucopyranoside]. In addition, epigallocatechin gallate and epicatechin gallate were found as the major active components responsible for the antioxidant activity of tea flowers through the use of a combination of preparative liquid chromatography separation and DPPH assay.
