Abstract:Self-biting disease occurred in most farmed furanimal in the world caused wound to take effect onmink pelt quality.There is little information concerning the mechanism and rapid detection methods of this disease had not been reported,so to distinguish self-biting individuals from healthy ones was one of the majormeasures to control the disease.Our research group used Marker Assisted Selection(MAS)and gene-assisted selection(GAS)to find the molecular marker and candidate genes of self-biting disease since 2007.So far,we had found sixspecial molecular markers and sevenSNPson three candidate genes to detect the disease.In this paper,literatures concerning recent researches on etiological factor of selfbiting disease from both domestic and oversea were reviewed to provide theoretical basis for further study on molecular breeding and disease prevention of mink.
Examination on Clinical Effect of "MieCanYing"

Abstract:Myiasis is one of the serious disease in sericulture, and application of "MieCanYing" is the major preventive method in cocoon production. "MieCanYing" was sampled from the market in the present experiment, its chemical active ingredients were determined and clinical efficacy was tested. The results showed that effective component content of "MieCanYing" from the the market was significantly lower tha standard. Recommended efficacy index is up to 50.83. Half of the recommended dosage will result in significant decrease in disease control; however doubled recommended dose can not significantly increase in clinical effect but will cause decrease of economic characters of silkworm. Although effective control of disease can be achieved by reducing the dilution fator if "MieCanYing" with insufficient active ingredients,was used, obvious side effects could be observed. Development of new disease control technique to replace the "MieCanYing" is an urgent issue due to the devastating effect of organic phosphorus pesticide on the environment and the security needs of sericulture production.

Abstract:DNA samples obtained from healthy and self-biting Jinzhou Black minks were amplified by PCR with 100 random primers. A DNA fragment corresponding to self-biting cases was identified and marked as S356. The DNA fragment was purified and cloned into PMD18-T vector,subsequently,transformed into E. Coli JM109 and sequenced. We applied the SCAR label technology to detect the molecular marker S356 among the healthy and self-biting minks. Based on the sequence of S356,two pairs of SCAR primers,S3568001 and S3568002,were designed and used in PCR detection. The result showed that the target DNA fragment was amplified from 29 of 30 self-biting minks ( 96. 67% positive correspondence rate) with the SCAR labeling primers S3568001 and S3568002. However,there was no target fragment detected from healthy minks. The DNA fragment was denominated as DS356800,which could be used as a genetic marker to differentiate potential self-biting individuals from healthy minks,and could form a guideline for future anti-disease genetic breeding to eliminate the self-biting disease from mink populations.

Abstract:【Objective】 This study aimed to investigate nervous system pathological changes in mink with self-injurious behavior and to research the relationship between nervous system pathological changes and this abnormal behavior in order to provide reference for pathological mechanism research of self-injurious behavior.【Method】 The cerebrum,cerebellum,medulla oblongata and medulla marrow from each of 20 healthy minks and 20 self-injurious minks were collected in current study.All samples were fixed in 4% paraformaldehyde,followed by dewatering,hyalining,paraffin embedding,slicing and hematoxylin eosin staining.All sections were observed at 200-400 times light microscope.【Result】 Compared with clinical healthy minks,histopathological changes were found in nervous system of minks with self-injurious behavior.Swelling,lysing and microsatellite phenomenon of nerve cells were observed in the cerebrum.Many nuclei in nerve cells became pyknotic(shrunken and dark).Vacuolizations were observed in cerebrum of some minks with self-injurious behavior.Intracellular membrane of neuroglia cell was deeply stained.Swollen and lysed Purkinje cells were investigated in cerebellum,and some of the myelin sheaths were lysed in this region.Neurocyte swelling,membrane ablation of macroglia and serious microsatellite phenomenon were observed in medulla oblongata of SIB mink.Nerve cells pyknotic and membrane lysed were also found in spinal marrow.【Conclusion】 Self-injurious behavior of minks has correlation with pathological changes of membrane of nerve cell and nerve myelin sheath.

Abstract:Self-biting behavior (SBB) is a serious behavioral disorder in farmed minks, but little is known about the biological basis of this disorder. The present study examined for the first time the hepatic and cerebral oxidative stress biomarker levels in SBB minks and compared them with those in normal ones to study the association of oxidative stress and SBB in minks. Calcium-activated adenosine triphosphatase (Ca2+ATPase) activities were also determined to investigate whether the Ca2+ pump in brain is affected. Twenty male SBB minks and 20 male normal minks were chosen in December. The brain and liver of each mink were harvested immediately after slaughter to test oxidative stress biomarkers. All parameters were determined by using a spectrophotometer. Our findings were as follows: SBB minks produced more malondialdehyde with 51% increase in brain (P < 0.01) and 22% increase in liver (P < 0.01) compared with normal minks. Similarly, the hepatic and cerebral protein carbonyls in SBB minks were 18.5% (P < 0.01) and 30% higher (P < 0.001), respectively. Glutathione peroxidase activities in brain and catalase activities in both tissues of SBB minks were markedly lower than those of normal group (P < 0.01). However, SBB minks had higher total superoxide dismutase (SOD), Cu/Zn-SOD, and Mn-SOD activities in both tissues (P < 0.05 or P < 0.01). Significant depletion of glutathione and vitamin E levels were observed in SBB minks (P < 0.01 or P <0 .05). Calcium-activated adenosine triphosphatase activities in the brain of SBB minks were inhibited (P < 0.01). Our results supported the oxidative stress hypothesis in minks with SBB.

Abstract:Pseudomonas aeruginosa is one of the most host-susceptible pathogenic bacteria to cause acute and chronic infections in humans and animals. Notably, its infection can especially cause fatal pathogenic infectious pneumonia in minks. Many previous mink studies have investigated the pathology, pathogenesis, serology, antimicrobial resistance, virulence gene, vaccination, and related diseases of P. aeruginosa. However, the relationship of P. aeruginosa infections with mink immunity and resistance is relatively less studied that needs more elaborations. Therefore, we here provide a comprehensive review about mink immunity and resistance to P. aeruginosa infections and the pathogenesis of mink hemorrhagic pneumonia in four major aspects. They include characterization, infection, immunity, and resistance of P. aeruginosa, and their implication and perspective, which aim to contribute the useful and valuable information to further related research and clinical treatment of P. aeruginosa and to avoid the potential fatal hemorrhagic pneumonia spreading.

Abstract:In order to determine drug resistance,pathogenicity and genetic of the pathogenic bacteria in mink of Heilongjiang province,three strains were isolated from dead minks and identified.Based on colony morphology,microscopic characteristics,biochemical test and 16S rRNA sequencing.The antibiotic susceptibility to 24 antibiotics were detected by using standard K-B paper method,and virulence assay,phylogenetic analysis were carried out at the same time.The result showed that the isolates were highly tolerance to antibacterials,could cause 100％ death of mice in experimental group and were closely related to strains from activated sludge and volcanic deposits.It showed that Serratia marcescens infection of mink existed in Heilongjiang province.Strains resistance phenomenon was serious and had a strong pathogenicity,and might be originated from the environment.It was the first time that the pathogenic Serratia marcescens isolated from the mink was reported.The results provided information for the detection,identification,diagnosis,treatment and prevention of related diseases.

Abstract:Abstract Minks are raised in many countries and have transmitted severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) to humans. However, the biologic properties of SARS-CoV-2 in minks are largely unknown. Here, we investigated and found that SARS-CoV-2 replicates efficiently in both the upper and lower respiratory tracts, and transmits efficiently in minks via respiratory droplets; pulmonary lesions caused by SARS-CoV-2 in minks are similar to those seen in humans with COVID-19. We further found that a spike protein-based subunit vaccine largely prevented SARS-CoV-2 replication and lung damage caused by SARS-CoV-2 infection in minks. Our study indicates that minks are a useful animal model for evaluating the efficacy of drugs or vaccines against COVID-19 and that vaccination is a potential strategy to prevent minks from transmitting SARS-CoV-2.

Abstract:Akkermansia muciniphila(A.muciniphila)has drawn much attention as an important gut microbe strain in recent years.A.muciniphila can influence the occurrence and development of diseases of the endocrine,nervous,digestive,musculoskeletal,and respiratory systems and other diseases.It can also improve immunotherapy for some cancers.A.muciniphila is expected to become a new probiotic in addition to Lactobacillus and Bifidobacterium.An increase in A.muciniphila abundance through direct or indirect A.muciniphila supplementation may inhibit or even reverse disease progression.However,some contrary findings are found in type 2 diabetes mellitus and neurodegenerative diseases,where increased A.muciniphila abundance may aggravate the diseases.To enable a more comprehensive understanding of the role of A.muciniphila in diseases,we summarize the relevant information on A.muciniphila in different systemic diseases and introduce regulators of A.muciniphila abundance to promote the clinical transformation of A.muciniphila research.

Abstract:From July–November 2020, mink (Neogale vison) on 12 Utah farms experienced an increase in mortality rates due to confirmed SARS-CoV-2 infection. We conducted epidemiologic investigations on six farms to identify the source of virus introduction, track cross-species transmission, and assess viral evolution. Interviews were conducted and specimens were collected from persons living or working on participating farms and from multiple animal species. Swabs and sera were tested by SARS-CoV-2 real-time reverse transcription polymerase chain reaction (rRT-PCR) and serological assays, respectively. Whole genome sequencing was attempted for specimens with cycle threshold values <30. Evidence of SARS-CoV-2 infection was detected by rRT-PCR or serology in ≥1 person, farmed mink, dog, and/or feral cat on each farm. Sequence analysis showed high similarity between mink and human sequences on corresponding farms. On farms sampled at multiple time points, mink tested rRT-PCR positive up to 16 weeks post-onset of increased mortality. Workers likely introduced SARS-CoV-2 to mink, and mink transmitted SARS-CoV-2 to other animal species; mink-to-human transmission was not identified. Our findings provide critical evidence to support interventions to prevent and manage SARS-CoV-2 in people and animals on mink farms and emphasizes the importance of a One Health approach to address emerging zoonoses.

Abstract:Foot-and-mouth disease(FMD) is one of the most highly contagious diseases which affects domestic cloven-hoofed animals.And the disease has debilitating effects,including weight loss,decrease in milk production,and loss of draught power,resulting in a loss in productivity for a considerable time.Current methods of disease control include inhibition of animal movement,slaughter of infected and in-contact susceptible animals,and vaccine inoculation.While current vaccine can induce complete protection by 7~10 days,in emergency outbreak situations it is imperative to block or reduce virus shedding as rapidly as possible to contain the outbreak,so some antiviral approach were needed to provide protection for sensitive animals and normal animals during the early of after vaccine inoculation.In this article,progress on antiviral approach of foot-and-mouth disease was reviewed,including IFN,antisense technology and RNA interference.

Abstract:Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is transmitted on mink farms between minks and humans in many countries. However, the systemic pathological features of SARS-CoV-2-infected minks are mostly unknown. Here, we demonstrated that minks were largely permissive to SARS-CoV-2, characterized by severe and diffuse alveolar damage, and lasted at least 14 days post inoculation (dpi). We first reported that infected minks displayed multiple organ-system lesions accompanied by an increased inflammatory response and widespread viral distribution in the cardiovascular, hepatobiliary, urinary, endocrine, digestive, and immune systems. The viral protein partially co-localized with activated Mac-2 + macrophages throughout the body. Moreover, we first found that the alterations in lipids and metabolites were correlated with the histological lesions in infected minks, especially at 6 dpi, and were similar to that of patients with severe and fatal COVID-19. Particularly, altered metabolic pathways, abnormal digestion, and absorption of vitamins, lipids, cholesterol, steroids, amino acids, and proteins, consistent with hepatic dysfunction, highlight metabolic and immune dysregulation. Enriched kynurenine in infected minks contributed to significant activation of the kynurenine pathway and was related to macrophage activation. Melatonin, which has significant anti-inflammatory and immunomodulating effects, was significantly downregulated at 6 dpi and displayed potential as a targeted medicine. Our data first illustrate systematic analyses of infected minks to recapitulate those observations in severe and fetal COVID-19 patients, delineating a useful animal model to mimic SARS-CoV-2-induced systematic and severe pathophysiological features and provide a reliable tool for the development of effective and targeted treatment strategies, vaccine research, and potential biomarkers.

Abstract:Many studies have been conducted to explore outbreaks of SARS-CoV-2 in farmed mink and their intra-/inter-species spread and spillover to provide data to the scientific community, protecting human and animal health. Studies report anthropozoonotic introduction, which was initially documented in April 2020 in the Netherlands, and subsequent inter-/intra-species spread of SARS-CoV-2 in farmed mink, likely due to SARS-CoV-2 host tropism capable of establishing efficient interactions with host ACE2 and the mink hosts' ability to enhance swift viral transmission due to their density, housing status, and occupational contacts. Despite the rigorous prevention and control measures adopted, transmission of the virus within and between animal species was efficient, resulting in the development of mink-associated strains able to jump back and forth among the mink hosts and other animal/human contacts. Current knowledge recognizes the mink as a highly susceptible animal host harboring the virus with or without clinical manifestations, furthering infection transmission as a hidden animal reservoir. A One Health approach is, thus, recommended in SARS-CoV-2 surveillance and monitoring on mink farms and of their susceptible contact animals to identify and better understand these potential animal hosts.

Abstract:Bovine subclinical mastitis (SM) causes great loss in China and global diary industry due to its high frequency,wide distribution and complicated etiology.With the development of molecular biology,computer technology,advanced molecular methods have been applied to the diagnose of SM,including nested PCR,metagenome,and immunological detection technology method which brought the fast,precise,convenient diagnose assays into reality.Moreover,in terms of SE treatment,traditional medicine and new drugs were becoming a tendency,which gradually replaced traditional antibiotics.In this review,the incidence regularity,etiology,diagnose method and treatment of bovine subclinical mastitis were discussed at home and abroad.

Abstract:Aleutian mink disease (AMD) is one of the most serious diseases in minks worldwide, it brings tremendous financial losses in mink farming. AMD virus (AMDV) has unusually high genetic diversity, its genomic structure remains unclear. In 2014, sudden death of breeding minks was occurred in northeast China. After clinical signs evaluation and virus isolation, AMDV was identified in all sudden death minks, we investigated the complete genomic sequence of AMDV-LM isolated from the sudden death case. The full-genome sequence of AMDV-LM was 7 nucleotides (nts) or 8 nts longer than isolates AMDV-BJ and AMDV-G. AMDV-LM contained two unique nucleotide changes in VP2 (G79T, T710C), which led to two amino acid changes G27W and L237S. For NS1, some unique point mutations, such as A374C, A428C, A463C, and T476A were found and resulted in four unique amino acid mutations at N24V, H125P, V143P, K155Q, and V159N, respectively. The predicted secondary structure of the 5 & PRIME; terminal of AMDV-LM formed a large bubble formation near the 5 & PRIME; end, which affected the stability of the U-shaped hairpin. Phylogenetic analysis demonstrated that AMDV-LM was closely related to Chinese isolates and confirmed that AMDV strains circulating in China had different origins of ancestors. This study was first to investigate the association of sudden death of adult breeding minks with AMDV infection. Our findings provide useful suggestions for evaluation of the pathogenic potential of AMDV, additional details on AMDV genome characterization were also presented. Future work should focus on the importance of AMDV-LM strain in mink infection.

Abstract:Melatonin is a potent immunomodulator with anti-inflammatory and antioxidant effects that significantly reduces inflammation. In practice, the subcutaneously implanted melatonin in minks is generally used to stimulate molting in autumn in advance, thereby reducing feed cost to obtain considerable economic benefits. The opportunistic Pseudomonas aeruginosa (P. aeruginosa) infection causes hemorrhagic pneumonia, massive mortality and economic losses in minks, while melatonin treatments to minks could improve host resistance; therefore, the objective of our study is to investigate the relationships of the susceptible infections caused by P. aeruginosa with the improved immunity by melatonin in minks. In this study, 1 × 109 colony forming units of P. aeruginosa were used to infect 12 minks in the case group (IGM group) after 10 mg melatonin dosage per body weight (kg) treatment under the implantation in mink ears. The control group (IG group) contains 12P. aeruginosa infected minks without melatonin treatments. In the meantime, another 24 minks were also included in our study with no P. aeruginosa infections, where 12 of them were treated with melatonin for comparisons. The results showed that melatonin treatments increased mink immunity and resistance to P. aeruginosa infections, alleviated the pathogenesis and pathological lung damages, and enhanced the lung scavenging abilities. The lung indexes of minks after melatonin treatments significantly decreased comparing IGM group with IG group (P < 0.01). Similarly, the bacterial loads of minks in the IGM group were significantly lower than those in the IG group at different infected time points (P < 0.05). In addition, the concentration levels of tumor necrosis factor α (TNF-α) significantly increased after melatonin treatments (P < 0.05). In conclusion, our study explored the relationships between P. aeruginosa infection and host resistance in minks, and provided valuable understandings of the pathogenesis of mink hemorrhagic pneumonia and the potential applications of melatonin in the future practical treatments.
