Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:RCM + AD = T2: In the presence of the C16-methylene group, regioselective ring-closing metathesis (RCM) formed the (12E)-endocyclic double bond, which underwent Os-catalyzed asymmetric dihydroxylation (AD) to give the desired 12,13-diol intermediate required for the total synthesis of amphidinolide T2 in 16 linear steps in 8.0% overall yield.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:A five-step, practical, and concise total synthesis of Mubritinib is described. The synthesis utilized Friedel-Crafts acylation, Click reaction, reduction and demethylation for the construction of the triazole ring system as key steps. Other important features of this synthesis is Bredereck oxazole synthesis. The main advantages of this process are the improved yield and decreased number of reaction steps, which paves the way for the industrial-scale synthesis of Mubritinib.

Abstract:The efficient total syntheses of Nigrasin I and Kuwanon C, two biologically interesting natural flavonoids with two regioisomeric isoprenyl side chains, were realized for the first time starting from commercially available 1-(2,4,6-trihydroxyphenyl)ethanone via a linear reaction sequence of 11 steps with the overall yields of 22% and 21%, respectively, in which the selection of protective groups and the Claisen rearrangement of the allylated 16 featured the synthetic strategy. The use of acetic anhydride as the solvent and the employment of microwave irradiation are disclosed to be critically important in the efficient and selective Claisen rearrangement.

Abstract:The total synthesis of (-)-goniomitine has been accomplished in 11 steps starting from commercially available diethyl L-malate. The synthesis features a chiral pool approach to prepare the chiral C-9 unit containing a quaternary carbon center, an Ir-catalyzed C-H borylation to synthesize the 2-indoleboronic acid pinacol ester, and a Suzuki reaction to couple together the two key intermediates. Notably, the high degree of convergence of this strategy makes it particularly amenable to the total synthesis of other aspidosperma family natural products.

Abstract:A cascade oxidation/Pictet-Spengler condensation/annulation process has been developed for the one-pot total synthesis of nitramarine, nitraridine, and their analogues. The procedure proceeded with easily available quinolines and tryptophan derivatives. A simple and metal-free approach, wide substrate scope, and functional group tolerance make it applicable for the synthesis of diverse bioactive nitramarine, nitraridine, and their derivatives. Furthermore, the bioactivity evaluation has identified two promising leading compounds 5d and 5e with potent antitumor proliferative activity against breast cancer cells.

An efficient total synthesis of (+)-minfiensine was realized using this strategy. The present method enables access to the common core structure of a series of monoterpene indole alkaloids, such as vincorine, echitamine, and aspidosphylline A.

Abstract:AbstractWith over 5000 members isolated to date, sesquiterpene lactones represent a prolific source of medicinal agents with several derivatives in human clinical trials. The guaianolides, a major subset of this group, have been intensely investigated from both medicinal and chemical‐synthesis perspectives for decades. To date, the myriad stereochemical permutations presented by this enormous family have precluded the synthesis of many unique members. Herein we report the total synthesis of the trans‐fused 8,12‐guaianolide (+)‐mikanokryptin in 10 steps from (+)‐carvone. Notably, this synthesis is the first gram‐scale total synthesis of a guaianolide natural product.

Following a Claisen rearrangement approach to construct the 6,9-bicycle, a transannular [2+2] photocycloaddition and the ensuing ring-opening reaction were implemented to forge the characteristic 5-6-8 propellane-like skeleton. Subsequent late-stage alkylations and reduction completed the synthesis.

Abstract:The first and protective group-free total synthesis of (±)-yezo'otogirin C has been achieved from 3-methyl-4-prenylcyclohex-2-enone in eight steps with 23% overall yield. The tricyclic core of (±)-yezo'otogirin C was established via a bioinspired oxidative cascade cyclization strategy using Mn(II)/Mn(III) and O2, followed by reduction of the peroxy-bridged intermediate using thiourea in refluxing methanol.

Abstract:A facile, one-pot three component catalytic method is developed for the synthesis of N-substituted tetrazole using RuO2/MMT nanocomposite. It is characterized with low and wide angle XRD which suggests that RuO2 nanoparticles are evenly dispersed on the surface of MMT while FESEM images indicate a spherical morphology having size in the range of 40-50 nm. The catalytic efficiency is evaluated for three component one-pot synthesis of the tetrazole using various amine, sodium azide and triethyl-ortho-formate under solvent free condition. This strategy has resulted in good to excellent yields (84 – 97%) of N-substituted tetrazoles within moderate reaction time. Moreover, the catalyst possesses excellent reusability up to five cycles with only 5% decrease in the yield of tetrazole after 5th cycle. The beneficial catalytic activity of the bifunctional nanocomposite is attributed to the uniformly dispersed RuO2 nanoparticles on the surface of MMT where RuO2 site is responsible for coordination of isocyanide intermediate while strong acidic character of MMT induces condensation and cyclization steps in a synergic manner. Thus, it can be argued that RuO2/MMT nanocomposite possesses potential applications for Multi Component Reactions (MCR) in terms of efficient and sustainable manner.

Abstract:The concise total synthesis of aplykurodinone-1 with an unusual cis-fused hydrindane moiety has been accomplished without the need for any protecting group chemistry using a unique SmI2 mediated reductive cascade cyclization reaction and a direct cuprate mediated 1,4-addition. This work represents the first example of the use of a SmI2-mediated intramolecular cascade cyclization reaction between "halide, alkene and aldehyde" groups.

Abstract:Dimeric indolosesquiterpenoids drew significant interest in the synthetic community due to their higher potential of biological activity than their monomeric congeners. Biosynthetically it has been shown that xiamycin A (2a) could dimerised under the oxidative condition to form the more potent all possible N-N, C-N, and C-C dimers. The pursuit of effective methods for creating these linkages has driven the advancement of synthetic chemistry, leading to new strategies and tools that enhance the ability to produce these biologically significant molecules. Till date the synthesis and biological evaluation of dimer of the oridamycin A remains unexplored. Herein we have described a synthetic design towards the concise total synthesis of dioridamycin (1a) and dixiamycin (1b) following a strategic use of hypervalent iodine(III) to forge intramolecular cross amination leading to the indolosesquiterpenoid pentacyclic core under mild and atom economical condition. Our study reveals the utility of PhI(OAc)2 for introduction of intramolecular C-N bond for indolosesquiterpenoids is functional group effective, provides a wider variety of products in good to excellent yields and also have conventionally been viewed as challenging.
