Threshold of rats with partial sciatic nerve ligation as well as SCN9a gene and glial cell expression

Abstract:Objective To observe the pain threshold in rats with partial sciatic nerve ligation (SNL) as well as changes of SCN9a gene expression in rat dorsal root ganglion (DRG) and activation of spinal cord glial cells.Methods Twenty-six male SD rats,weighing 210-250 g,were randomly divided into two groups:sham operation group (Sham group,n =8) and SNL model group (SNL group,n =18).Mechanical paw withdrawal threshold (MWT) value of left hind limb was tested 1 day before,and 3,6,9,12,and 14 days after sugery.Two rats in Sham group were killed each at 0,8,10,14 day respectively,and 6 rats in SNL group were killed each at 8,10,and 14 days.The DRG and lumbar enlargement spinal cord tissue were taken out,and the expression of DRG Nav1.7,spinal cord glial fibrillary acidic protein [glial fibrillary acidic protein (GFAP),astrocyte activation flag] and OX42 (microglia activation flag) was detected by Western blotting.Results As compared with Sham group (11.31 ± 0.17),the MWT of left hind limb in SNL group (3.87 ±0.12) was decreased significantly at 3-14 day after sugery with the difference being statistically significant (P ＜ 0.01).As compared with Sham group (0.38 ± 0.03),the protein levels of Nav1.7 in DRG were significantly increased in SNL group at 8th day (0.70 ± 0.04),10th day (0.81 ±0.06) and 14th day (0.85 ±0.05) with the difference being statistically significant (P ＜0.01).As compared with Sham group (0.40 ±0.05),the GFAP expression in spinal cord was significantly increased in SNL group at 10th day (0.75 ± 0.06) and 14th day (0.85 ± 0.10) with the difference being statistically significant (P ＜ 0.01).As compared with Sham group (0.35 ± 0.05),the OX42 expression in spinal cord was increased in SNL at 8th day (0.45 ± 0.02),10th day (0.44 ± 0.03),and 14th day (0.43 ± 0.03) with the difference being statistically significant (P ＜ 0.01).Conclusion The Nav1.7 and spinal cord glial cells involved in the occurrence and development of SNL in rats.Microglia involved in the occurrence of SNL,and astrocytes involved in the development of SNL.
