Abstract:Chemically functionalized single-walled carbon nanotubes (SWNTs) have shown promise in tumor targeted accumulation in mice and exhibit biocompatibility, excretion and little toxicity. Here, we demonstrate in-vivo SWNT drug delivery for tumor suppression in mice. We conjugate paclitaxel (PTX), a widely used cancer chemotherapy drug to branched polyethylene-glycol (PEG) chains on SWNTs via a cleavable ester bond to obtain a water soluble SWNT-paclitaxel conjugate (SWNT-PTX). SWNT-PTX affords higher efficacy in suppressing tumor growth than clinical Taxol in a murine 4T1 breast-cancer model, owing to prolonged blood circulation and 10-fold higher tumor PTX uptake by SWNT delivery likely through enhanced permeability and retention (EPR). Drug molecules carried into the reticuloendothelial system are released from SWNTs and excreted via biliary pathway without causing obvious toxic effects to normal organs. Thus, nanotube drug delivery is promising for high treatment efficacy and minimum side effects for future cancer therapy with low drug doses.

Abstract:Nanotechnology is revolutionising different areas from manufacturing to therapeutics in the health field. Carbon nanotubes (CNTs), a promising drug candidate in nanomedicine, have attracted attention due to their excellent and unique mechanical, electronic, and physicochemical properties. This emerging nanomaterial has attracted a wide range of scientific interest in the last decade. Carbon nanotubes have many potential applications in cancer therapy, such as imaging, drug delivery, and combination therapy. Carbon nanotubes can be used as carriers for drug delivery systems by carrying anticancer drugs and enabling targeted release to improve therapeutic efficacy and reduce adverse effects on healthy tissues. In addition, carbon nanotubes can be combined with other therapeutic approaches, such as photothermal and photodynamic therapies, to work synergistically to destroy cancer cells. Carbon nanotubes have great potential as promising nanomaterials in the field of nanomedicine, offering new opportunities and properties for future cancer treatments. In this paper, the main focus is on the application of carbon nanotubes in cancer diagnostics, targeted therapies, and toxicity evaluation of carbon nanotubes at the biological level to ensure the safety and real-life and clinical applications of carbon nanotubes.

Abstract:Cancer, once considered as an incurable disease, today, becomes not that difficult to be treated due to the newly emerging nanotechnology and the rapid development of “smart” drug delivery systems. New cancer therapeutics ought to overcome the defects of conventional drug delivery systems, such as nonspecific bio-distribution or targeting, short circulation time, etc. Tumor targeted drug delivery systems basically includes two strategies: passive and active targeting. Passive targeting is often referred to the enhanced permeability and retention effect (EPR effect) due to the enhanced vascular penetration of tumor capillaries and limited lymphatic drainage of lymphatic capillaries. Active targeting systems are based on local stimuli characteristic of the target pathological zone (such as, increased temperature or lowered pH values, redox potential changes, characteristic of inflamed, the receptor mediated internalization by target cells, etc.). Intriguingly, the combination of the two strategies above is promising and exciting. Herein, we give a brief review of tumor targeted drug delivery systems based on tumor cell itself and its external milieu. Abstract:Cancer remains a significant risk to human health. Nanomedicine is a new multidisciplinary field that is garnering a lot of interest and investigation. Nanomedicine shows great potential for cancer diagnosis and treatment. Specifically engineered nanoparticles can be employed as contrast agents in cancer diagnostics to enable high sensitivity and high-resolution tumor detection by imaging examinations. Novel approaches for tumor labeling and detection are also made possible by the use of nanoprobes and nanobiosensors. The achievement of targeted medication delivery in cancer therapy can be accomplished through the rational design and manufacture of nanodrug carriers. Nanoparticles have the capability to effectively transport medications or gene fragments to tumor tissues via passive or active targeting processes, thus enhancing treatment outcomes while minimizing harm to healthy tissues. Simultaneously, nanoparticles can be employed in the context of radiation sensitization and photothermal therapy to enhance the therapeutic efficacy of malignant tumors. This review presents a literature overview and summary of how nanotechnology is used in the diagnosis and treatment of malignant tumors. According to oncological diseases originating from different systems of the body and combining the pathophysiological features of cancers at different sites, we review the most recent developments in nanotechnology applications. Finally, we briefly discuss the prospects and challenges of nanotechnology in cancer.

Abstract:Multiwalled carbon nanotubes (MWCNTs) are at the forefront of nanotechnology-based advancements in cancer therapy, particularly in the field of targeted drug delivery. The nanotubes are characterized by their concentric graphene layers, which give them outstanding structural strength. They can deliver substantial doses of therapeutic agents, potentially reducing treatment frequency and improving patient compliance. MWCNTs' diminutive size and modifiable surface enable them to have a high drug loading capacity and penetrate biological barriers. As a result of the extensive research on these nanomaterials, they have been studied extensively as synthetic and chemically functionalized molecules, which can be combined with various ligands (such as folic acid, antibodies, peptides, mannose, galactose, polymers) and linkers, and to deliver anticancer drugs, including but not limited to paclitaxel, docetaxel, cisplatin, doxorubicin, tamoxifen, methotrexate, quercetin and others, to cancer cells. This functionalization facilitates selective targeting of cancer cells, as these ligands bind to specific receptors overexpressed in tumor cells. By sparing non-cancerous cells and delivering the therapeutic payload precisely to cancer cells, this therapeutic payload delivery ability reduces chemotherapy systemic toxicity. There is great potential for MWCNTs to be used as targeted delivery systems for drugs. In this review, we discuss techniques for functionalizing and conjugating MWCNTs to drugs using natural and biomacromolecular linkers, which can bind to the cancer cells' receptors/biomolecules. Using MWCNTs to administer cancer drugs is a transformative approach to cancer treatment that combines nanotechnology and pharmacotherapy. It is an exciting and rich field of research to explore and optimize MWCNTs for drug delivery purposes, which could result in significant benefits for cancer patients.

Abstract:Cancer is considered the world's deadliest disease, and its underlying pathology is intricate. Though cancer patients have had access to chemotherapeutics since aeons, the traditional treatments sometimes cause discomfort, have unwanted side effects, and are not site-specific. In this regard, several nanomaterials have lately been developed and explored specifically for use in cancer treatment ascribable to their unique optically active, magnetic, and electrical properties owing to nano-sized (1 to 100 nm) particles. Targeting, improved bioavailability, and low toxicity are vital reasons for nanomedicines becoming increasingly popular. Nanoparticles actively and passively target the cancer cells and kill them. Various ligands, including aptamers, biomolecules, peptides and/or antibodies have been in use for active targeting of cancer, while the cell characteristics, like leaky vasculature, angiogenesis etc., of cancer cells facilitate passive targeting. Focusing on nano oncology research investigations and clinical applications, this review traces the development and current state of targeted cancer therapy using nanomaterials. Also, this article furnishes account of challenges hindering the reach of these materials to clinical applications.

Abstract:During the past years, great progress has been made in the field of nanomaterials given their great potential in biomedical applications. Carbon nanotubes (CNTs), due to their unique physicochemical properties, have become a popular tool in cancer diagnosis and therapy. They are considered one of the most promising nanomaterials with the capability of both detecting the cancerous cells and delivering drugs or small therapeutic molecules to these cells. Over the last several years, CNTs have been explored in almost every single cancer treatment modality, including drug delivery, lymphatic targeted chemotherapy, thermal therapy, photodynamic therapy, and gene therapy. In this review, we will show how they have been introduced into the diagnosis and treatment of cancer. Novel SWNT-based tumor-targeted drug delivery systems (DDS) will be highlighted. Furthermore, the in vitro and in vivo toxicity of CNTs reported in recent years will be summarized.

Abstract:The incidence and mortality of cancer are gradually increasing. The highly invasive and metastasis of tumor cells increase the difficulty of diagnosis and treatment, so people pay more and more attention to the diagnosis and treatment of cancer. Conventional treatment methods, including surgery, radiotherapy and chemotherapy, are difficult to eliminate tumor cells completely. And the emergence of nanotechnology has boosted the efficiency of tumor diagnosis and therapy. Herein, the research progress of nanotechnology used for tumor diagnosis and treatment is reviewed, and the emerging detection technology and the application of nanodrugs in clinic are summarized and prospected. The first part refers to the application of different nanomaterials for imaging in vivo and detection in vitro , which includes magnetic resonance imaging, fluorescence imaging, photoacoustic imaging and biomarker detection. The distinctive physical and chemical advantages of nanomaterials can improve the detection sensitivity and accuracy to achieve tumor detection in early stage. The second part is about the nanodrug used in clinic for tumor treatment. Nanomaterials have been widely used as drug carriers, including the albumin paclitaxel, liposome drugs, mRNA-LNP, protein nanocages, micelles, membrane nanocomplexes, microspheres et al., which could improve the drug accumulate in tumor tissue through enhanced permeability and retention effect to kill tumor cells with high efficiency. But there are still some challenges to revolutionize traditional tumor diagnosis and anti-drug resistance based on nanotechnology.

Abstract:The pathological processes of cancer are complex. Current methods used for chemotherapy have various limitations, such as cytotoxicity, multi-drug resistance, stem-like cells growth, and lack of specificity. Several types of nanomaterials are used for cancer treatment. Nanomaterials 1–100 nm in size have special optical, magnetic, and electrical characteristics. Nanomaterials have been fabricated for cancer treatments to overcome cytotoxicity and low specificity, and improve drug capacity and bioavailability. Despite the increasing number of related studies, few nanodrugs have been approved for clinical use. To improve translation of these materials, studies of targeted drug delivery using nanocarriers are needed. Cytotoxicity, enhanced permeability and retention effects, and the protective role of the protein corona remain to be addressed. This mini-review summarizes new nanomaterials manufactured in studies and in clinical use, analyses current barriers preventing their translation to clinical use, and describes the effective application of nanomaterials in cancer treatment.

Abstract:<p>Lung cancer is one of the most rapidly growing malignancies in terms of morbidity and mortality. Although traditional treatments have been used for more than 50 years, it is still far from solving the problems of postoperative risks and systemic toxicity. Emerging targeting and immunotherapy are developing continuously and are gaining recognition; eventually, they face the inevitable challenge of drug resistance. Nanotechnology has several important effects on lung cancer treatment, owing to its unique properties. Several nanoparticle-based treatments have successfully become cancer treatments. Good biocompatibility with higher specific surface area can carry substantial amounts of lung cancer treatment medications while avoiding medication toxicity; editable and modified characteristics give rise to multifunctional nanomedicines; excellent photoelectric effects make lung cancer a multimodal treatment. This article summarizes the breakthroughs achieved by nanotechnology, targeted therapy, and immunotherapy, reflecting the importance and necessity of nanotechnology in the treatment of lung cancer.</p>

Abstract:Tumor immunotherapy exerts its anti-tumor effects by stimulating and enhancing immune responses of the body. It has become another important modality of anti-tumor therapy with significant clinical efficacy and advantages compared to chemotherapy, radiotherapy and targeted therapy. Although various kinds of tumor immunotherapeutic drugs have emerged, the challenges faced in the delivery of these drugs, such as poor tumor permeability and low tumor cell uptake rate, had prevented their widespread application. Recently, nanomaterials had emerged as a means for treatment of different diseases due to their targeting properties, biocompatibility and functionalities. Moreover, nanomaterials possess various characteristics that overcome the defects of traditional tumor immunotherapy, such as large drug loading capacity, precise tumor targeting and easy modification, thus leading to their wide application in tumor immunotherapy. There are two main classes of novel nanoparticles mentioned in this review: organic (polymeric nanomaterials, liposomes and lipid nanoparticles) and inorganic (non-metallic nanomaterials and metallic nanomaterials). Besides, the fabrication method for nanoparticles, Nanoemulsions, was also introduced. In summary, this review article mainly discussed the research progress of tumor immunotherapy based on nanomaterials in the past few years and offers a theoretical basis for exploring novel tumor immunotherapy strategies in the future.

Cancer-Cell Targeting and Photoacoustic Therapy Using Carbon Nanotubes As "bomb" Agents

Abstract:A unique approach using the large photoacoustic effect of single-walled carbon nanotubes (SWNTs) for targeting and selective destruction of cancer cells is demonstrated. SWNTs exhibit a large photoacoustic effect in suspension tinder the irradiation of a 1064-nm Q-switched millisecond pulsed laser and trigger a firecracker-like explosion at the nanoscale. By using such an explosion, a photoacoustic agent is developed by functionalizing the SWNTs with folate acid (FA) that can selectively bind to cancer cells overexpressing folate receptor on the surface of the cell membrane and kill them through SWNT explosion inside the cells tinder the excitation of millisecond pulsed laser. The uptake pathway of folate-conjugated SWNTs into cancer cells is investigated via fluorescence imaging and it is found that the FA-SWNTs can enter into cancer cells selectively with a high targeting capability of 17-28. Under the treatment of 1064-nm millisecond pulsed laser, 85% of cancer cells with SWNT uptake die within 20 s, while 90% of the normal cells remain alive due to the lack of SWNTs inside cells. Temperature changes during laser treatment are monitored and no temperature increases of more than +/- 3 degrees C are observed. With this approach, the laser power used for cancer killing is reduced 150-1500 times and the therapy efficiency is improved. The death mechanism of cancer cells caused by the photoacoustic explosion of SWNTs is also studied and discussed in detail. These discoveries provide a new way to use the photoacoustic properties of SWNTs for therapeutic applications.
Targeted therapeutic nanotubes influence the viscoelasticity of cancer cells to overcome drug resistance

Abstract:Resistance to chemotherapy is the primary cause of treatment failure in over 90% of cancer patients in the clinic. Research in nanotechnology-based therapeutic alternatives has helped provide innovative and promising strategies to overcome multidrug resistance (MDR). By targeting CD44-overexpressing MDR cancer cells, we have developed in a single-step a self-assembled, self-targetable, therapeutic semiconducting single-walled carbon nanotube (sSWCNT) drug delivery system that can deliver chemotherapeutic agents to both drug-sensitive OVCAR8 and resistant OVCAR8/ADR cancer cells. The novel nanoformula with a cholanic acid-derivatized hyaluronic acid (CAHA) biopolymer wrapped around a sSWCNT and loaded with doxorubicin (DOX), CAHA-sSWCNT-DOX, is much more effective in killing drug-resistant cancer cells compared to the free DOX and phospholipid PEG (PL-PEG)-modified sSWCNT formula, PEG-sSWCNT-DOX. The CAHA-sSWCNT-DOX affects the viscoelastic property more than free DOX and PL-PEG-sSWCNT-DOX, which in turn allows more drug molecules to be internalized. Intravenous injection of CAHA-sSWCNT-DOX (12 mg/kg DOX equivalent) followed by 808 nm laser irradiation (1 W/cm(2), 90 s) led to complete tumor eradication in a subcutaneous OVCAR8/ADR drug-resistant xenograft model, while free DOX alone failed to delay tumor growth. Our newly developed CAHA-sSWCNT-DOX nanoformula, which delivers therapeutics and acts as a sensitizer to influence drug uptake and induce apoptosis with minimal resistance factor, provides a novel effective means of counteracting the phenomenon of multidrug resistance.

Abstract:The biomedical applications of carbon nanotubes (CNTs) have attracted deep interest in recent years. Antitumor immunotherapy has the potential to improve the prognosis of cancer treatment but the efficacy of current immunotherapy generally needs further improvement. Multi-walled CNTs conjugated to tumor lysate protein are investigated as to whether they would enhance the efficacy of an immunotherapy employing a tumor-cell vaccine in a mouse model bearing the H22 liver cancer. The tumor cure rate is found to be markedly improved by CNTs conjugated to tumor lysate protein. The cellular antitumor immune reaction is also enhanced. Moreover, the observed antitumor immune response is relatively specific against the tumor intended for treatment. These findings suggest that CNTs may have a prospective role in the development of new antitumor immunotherapies.

Abstract:Key PointsNanoparticles are emerging as a new class of therapeutics for cancer because they can perform in ways that other therapeutic modalities cannot.Newer nanoparticle therapeutics are showing enhanced efficacy with lower side effects than traditional small-molecule chemotherapeutics in early clinic studies, and are doing so without creating additional new side effects due to the nanoparticle.Although there are many types of nanoparticles, few will have the proper attributes to reach clinical use because of the issues involved in translating research grade nanoparticles to clinic grade nanoparticles.Newer nanoparticle therapeutics have a greater degree of multifunctionality and involve not only delivery to the tumour but intracellular delivery so that multidrug resistance can be bypassed and therapeutic agents such as siRNA that require intracellular delivery can be utilized.Nanoparticles that contain cancer cell-surface targeting ligands are now in the clinic.

Abstract:The development of new medical cancer treatment technologies is of great significance in reducing cancer mortality. Traditional clinical cancer therapy has a short drug action time, difficulty in accurately targeting tumour tissues and high levels of toxicity in normal tissues. With the development of nanotechnology, nanomaterials have been used as drug carriers to specifically target cancer cells and release drugs into the tumour environment. This technique has become an important research hotspot in cancer treatment. There are several advantages of using nanomaterials for cancer treatment that improve the efficacy of drug delivery, including increased drug concentrations in the targeted tumour area, reduced toxicity in normal tissues and controlled drug release. In this work, we describe the latest research development on the use of nanomaterials for drug delivery in cancer treatment and explore related mechanistic pathways. In addition, the methods used to control drug release into the targeted area using nanocarriers are reviewed in detail. Overall, we present current achievements using nanomaterials and nanotechnologies in cancer treatment, followed by current challenges and future prospects.

Current chemotherapy faces problems such as lack of specificity, cytotoxicity, induction of multi-drug resistance and stem-like cells growth. Nanomaterials are materials in the nanorange 1–100 nm which possess unique optical, magnetic, and electrical properties. Nanomaterials used in cancer therapy can be classified into several main categories. Targeting cancer cells, tumor microenvironment, and immune system, these nanomaterials have been modified for a wide range of cancer therapies to overcome toxicity and lack of specificity, enhance drug capacity as well as bioavailability. Although the number of studies has been increasing, the number of approved nano-drugs has not increased much over the years. To better improve clinical translation, further research is needed for targeted drug delivery by nano-carriers to reduce toxicity, enhance permeability and retention effects, and minimize the shielding effect of protein corona. This review summarizes novel nanomaterials fabricated in research and clinical use, discusses current limitations and obstacles that hinder the translation from research to clinical use, and provides suggestions for more efficient adoption of nanomaterials in cancer therapy.

Abstract:Key PointsNanotechnology concerns the study of devices that are themselves or have essential components in the 1–1,000 nm dimensional range (that is, from a few atoms to subcellular size).Two main subfields of nanotechnology are nanovectors — for the administration of targeted therapeutic and imaging moieties — and the precise patterning of surfaces.Nanotechnology is no stranger to oncology: liposomes are early examples of cancer nanotherapeutics, and nanoscale-targeted magnetic resonance imaging contrast agents illustrate the application of nanotechnology to diagnostics.Photolithography is a light-directed surface-patterning method, which is the technological foundation of microarrays and the surface-enhanced laser desorption/ionization time-of-flight approach to proteomics. Nanoscale resolution is now possible with photolithography, and will give rise to instruments that can pack a much greater density of information than current biochips.The ability of nanotechnology to yield advances in early detection, diagnostics, prognostics and the selection of therapeutic strategies is predicated based on its ability to 'multiplex' — that is, to detect a broad multiplicity of molecular signals and biomarkers in real time. Prime examples of multiplexing detection nanotechnologies are arrays of nanocantilevers, nanowires and nanotubes.Multifunctionality is the fundamental advantage of nanovectors for the cancer-specific delivery of therapeutic and imaging agents. Primary functionalities include the avoidance of biobarriers and biomarker-based targeting, and the reporting of therapeutic efficacy.Thousands of nanovectors are currently under study. By systematically combining them with preferred therapeutic and biological targeting moieties it might be possible to obtain a very large number of novel, personalized therapeutic agents.Novel mathematical models are needed, in order to secure the full import of nanotechnology into oncology.

Abstract:Anti-cancer drugs with various mechanisms emerge in succession, but most of free therapeutics are accompanied by some disadvantages, such as poor safety and effectiveness, inability to target tumor cells, and short circulation time. Nanotechnology provides new solutions for overcoming the intrinsic limits of free therapeutics and navigating biological barriers. Considerable technological and clinical success have been achieved in cancer nanomedicines, but the main obstacle to the clinical translation of nanomedicine is an incomplete understanding of the requirements of clinical trials in new and established research scientists and the latest research of nanomedicine in clinicians. In this review, by searching the authoritative database, we introduce the progress and challenges of nanomedicines that have entered the clinical market or are ubiquitous in clinical trials for cancer therapy, including lipid-based nanoparticles, polymeric nanoparticles and albumin-based nanoparticles. Furthermore, we introduce several biomolecule-based nanomedicines that are promising for clinical application in the future, including biomimetic membranes, virus-like particles and DNA-based nanomachine, and highlight their advanced nanoparticle designs, advantages and challenges.
