Abstract:The title complex [Mn(C4H4O4)(phen)(H2O)(3)] . 2H(2)O has been prepared and its crystal structure determined by X-ray diffraction methods. The Mn(II) atom has octahedron coordination formed by four oxygen and two nitrogen atoms. The coordination mode of the succinate dianion in the complex is remarkably different from those reported previously. One carboxyl group of the succinate dianion coordinates to the Mn(II) atom as a monodentate and the other carboxyl group is free. The crystal structure consists of the complex and crystalline water molecules. All the oxygen atoms in the structure take part in an H-bonding network and assemble to form a hydrophilic layer, while phenanthroline ligands between neighboring hydrophilic layers to form a hydrophobic layer. Tight stacking interactions of phenanthroline rings are observed in the hydrophobic layer as evidenced by the short distance (3.312 Angstrom) between neighbouring parallel ring planes.
Experimental and Theoretical Study of the Terahertz Absorption Spectra of Crystalline Saccharides

Abstract:Terahertz time-domain spectroscopy has been used to measure the full dielectric function of polycrystaltine D-glucose and D-mannose. A number of well-resolved absorption peaks were observed in the frequency range 0.2-3.0 THz, which are interpreted as originating from inter-and intra-molecular vibrational modes. In parallel with the experimental study, density functional theory was applied to obtain structure and vibrational frequencies of the saccharides. The calculated results are in reasonable agreement with the experimental data. The comparison between experimental and theoretical study shows that far-infrared absorption features are highly sensitive to the structure and spatial arrangement of molecules.
Analysis of Sucrose Acetates in A Crude 6-O-acetyl Sucrose Product by On-Line Hydrolysis-High-Performance Liquid Chromatography with Pulsed Amperometric Detection

Abstract:A standard-free and sensitive method was developed for analysis of sucrose acetates in a crude 6-O-acetyl sucrose (S-6-a) product by on-line hydrolysis-high-performance liquid chromatography with pulsed amperometric detection (PAD). Sucrose, three regio-isomers of acetyl sucrose and five regio-isomers of diacetyl sucrose were separated on a C18 column using 3% (v/v) acetonitrile in water as eluent within 25min. After purification with LC followed by semi-preparative HPLC, their chemical structures were identified by 1D, 2D NMR and LC-MS. Moreover, quantification of those regio-isomers was achieved by on-line alkaline hydrolysis to liberate sucrose using a post-column delivery system, and then detected by PAD for indirect estimation of the sucrose acetate content. Under optimal conditions, the linear ranges were from 0.03 to 150μmolL(-1) for sucrose corresponding to sucrose acetates with coefficient of determination as 0.9997 and detection limit as 0.01μmolL(-1) (S/N=3). Good repeatability was obtained (RSD<3%, n=6). Furthermore, this method has been successfully applied to the analysis of sucrose and sucrose acetates in a crude S-6-a product during synthesis, purification and structure elucidation studies. The recoveries were from 94.89% to 102.31% for sucrose and sucrose acetates.

Abstract:The physicochemical properties and biological activities of polysaccharides depend on their structures. Monosaccharide composition analysis is indispensable for the structural characterization of polysaccharides and is helpful in the quality control of polysaccharide preparation. Here, using a model mixture and tamarind seed polysaccharide as examples, we demonstrated that a quantitative 2D NMR method, gsHSQC i (three gradient-selective Heteronuclear Single Quantum Coherence spectra acquired with incremented repetition times, i = 1, 2, 3) can directly quantify a variety of monosaccharides in solution with adequate precision and accuracy, requiring no derivatization, postprocessing steps and column separation. Both anomeric and non-anomeric signals of monosaccharides can be utilized for content determination. More accurate quantification of fructose in a mixture containing nine monosaccharides is obtained, which is difficult to achieve by quantitative 1D 1 HNMR and the common PMP-HPLC method (high-performance liquid chromatography through pre-column derivatization with 1-phenyl-3-methyl-5-pyrazolone) due to the peak overlapping and the poor derivatization efficiency, respectively. The results also revealed that Na[Fe(EDTA)] can serve as a proper relaxation-enhancing agent for saccharide samples to save experimental time. We expect that this approach can be applied as an alternative to analyzing the monosaccharide composition and be helpful in interpreting the structure of polysaccharides.

Abstract:<p>Two-dimensional NMR spectroscopies are one of the most frequently used techniques for the structural determination of carbohydrates. However, the data analysis is challenging because of the signal overlap in the <sup>1</sup>H homonuclear correlation spectra. We attempted to explore a general strategy for the structural determination of carbohydrates by combined multi-dimensional spectroscopies. The strategy was applied to a human milk oligosaccharide lacto-N-difucohexaose I, that has been previously studied by conventional two-dimensional NMR spectroscopy. Assignment of the intra-residue resonances of the hexasaccharide using the three-dimensional spectrum was straightforward. Consequently, data analysis of the multi-dimensional spectra was significantly simplified, leading to a quicker determination of the intra- and inter-residue connections in the hexasaccharide. Application of the NMR strategy to chondroitin sulfate from bovine cartilage revealed two repeating disaccharide regions of the A and C units of chondroitin sulfate, indicating the high potential of this technique for the structural determination of complex polysaccharides.</p>

Abstract:The two sugar molecules sucrose and trehalose are both considered as stabilizing molecules for the purpose of preserving biological materials during, for example, lyophilization or cryo-preservation. Although these molecules share a similar molecular structure, there are several important differences in their properties when they interact with water, such as differences in solubility, viscosity, and glass transition temperature. In general, trehalose has been shown to be more efficient than other sugar molecules in preserving different biological molecules against stress, and thus by investigating how these two disaccharides differ in their water interaction, it is possible to further understand what makes trehalose special in its stabilizing properties. For this purpose, the structure of aqueous solutions of these disaccharides was studied by using neutron and X-ray diffraction in combination with empirical potential structure refinement (EPSR) modeling. The results show that there are surprisingly few differences in the overall structure of the solutions, although there are indications for that trehalose perturbs the water structure slightly more than sucrose.
Revealing elusive conformations of sucrose from hydrogen bond J-coupling in H2O: A combined NMR and quantum mechanics study

Abstract:Hydrogen bonding is a crucial feature of biomolecules, but its characterization in glycans dissolved in aqueous solutions is challenging due to rapid hydrogen exchange between hydroxyl groups and H2O. In principle, the scalar (J) coupling constant can reveal the relative orientation of the atoms in the molecule. In contrast to J-coupling through H-bonds reported in proteins and nucleic acids, research on J-coupling through H-bonds in glycans dissolved in water is lacking. Here, we use sucrose as a model system for H-bonding studies; its structure, which consists of glucose (Glc) and fructose (Frc), is well-studied, and it is readily available. We apply the in-phase, antiphase-HSQC-TOCSY and quantify previously unreported through H-bond J-values for Frc-OH1-Glc-OH2 in H2O. While earlier reports of Brown and Levy indicate this H-bond as having only a single direction, our reported findings indicate the potential presence of two involving these same atoms, namely, G2OH ➔ F1O and F1OH ➔ G2O (where F and G stand for Frc and Glc, respectively). The calculated density functional theory J-values for the G2OH ➔ F1O agree with the experimental values. Additionally, we detected four other possible H-bonds in sucrose, which require different phi, psi (ϕ, ψ) torsion angles. The ϕ, ψ values are consistent with previous predictions of du Penhoat et al. and Venable et al. Our results will provide new insights into the molecular structure of sucrose and its interactions with proteins.
Terahertz Spectroscopic Investigation of Trehalose and Cellobiose

Abstract:In this paper, the absorption coefficient spectra of trehalose and cellobiose are measured in the frequency range from 0.4 to 1.8 THz by using THz time-domain spectroscopy. The two disaccharide are common disaccharide in agricultural products containing sugar. The experimental results show that trehalose and cellobiose have some characteristic absorption peaks in the range. The structure and vibrational frequencies of trehalose and cellobiose molecule in the THz range are simulated by the B3LYP algorithm of density functional theory, and the THz characteristic absorption peaks of trehalose and cellobiose molecule are identified by using Gaussian View software. The analysis show that the experimental results tally with the theoretical ones well.The observed absorption peaks in the THz range are mainly caused by the intramolecular and intermolecular vibrations jointly. The work of the paper presents promising methods for the identification of disaccharide and the quality assurance of agricultural products.

Abstract:Sucrose is the carbon skeletons and energy vector for plants, which is important for plants growth. Among thousands of disaccharides in Nature, why chose sucrose for plants? In this paper, we analyzed the intrinsic structural characteristics of four sucrose isomers with different glycosidic linkage by mass spectrometry (MS) technique. Our results show that sucrose has the most labile glycosidic bond compared with other three isomers, which is helpful for releasing glucose and fructose unit. Besides, sucrose has the most stable integral structure, which is hard to dehydrate and degrade into fragments through losing one or three even four-carbon units, just as its three isomers. In other words, sucrose is more easily holds an integral structure during the transport process, whenever it is necessary, and sucrose can be cleaved into glucose and fructose easily. Besides, we also investigate the internal relationship of sucrose with K(+) by tandem mass spectrometry and viscosity measurement. The related results have shown that the K(+) can stabilize sucrose to a greater extent than the Na(+). Furthermore, under the same conditions, K(+) ions reduce the viscosity of sucrose-water system much more than Na(+). These results suggest that K(+) is a better co-transporter for sucrose. Of course, the transport of sucrose in plants is a very complicated process, which is involved in many proteins. This paper directly accounts for the basic structure feature of sucrose, and the results discovered could provide the novel insight for the answer why Nature chose sucrose for plants.

Abstract:Carboxymethyl xylan (CMX) was synthesized and characterized. A method based on quantitative 13C NMR was developed to obtain detailed structural information. The total degree of substitution (DS), partial DS at 2- and 3-O-positions, and the quantitation of each unit in CMX could be acquired simultaneously by this method without decomposing the polymer backbone, thus avoiding errors caused by polymer hydrolysis with traditional methods using HPLC and 1H NMR spectrometry. Moreover, different from methods of direct detection of carboxyl groups, such as acidometric and potentiometric titrations, this method was not affected by probable impurity introduced during carboxymethylation.

Abstract:Oxidized sucrose (OS) is a bio-based cross-linking agent with excellent biological safety and environmental non-toxicity. However, the precise structure of OS has not been elucidated owing to its structural complexity and low purity. Accordingly, in this study, complete chemical shift assignments were performed by applying various nuclear magnetic resonance techniques, which permitted the structural and quantitative characterization of the two main OS products, each of which contained four aldehyde groups. In addition, we investigated the use of OS as a cross-linking agent in the preparation of a hydrogel from carboxymethyl chitosan (CMC), one of the most popular polysaccharides for use in biomedical applications. The primary amine groups of CMC were immediately cross-linked with the aldehyde groups of OS to form hydrogels without the requirement for a catalyst. It was found that the degree of cross-linking could be easily controlled by the feed amount of OS during CMC hydrogel preparation and the final cross-linking degree affected the thermal, swelling, and rheological properties of the obtained hydrogel. The results presented in this study are therefore expected to be applicable in the preparation of fully carbohydrate-based hydrogels for medical and pharmaceutical applications.

Abstract:A new ent ‐kaurane diterpenoid glycoside has been shown to be the main sweetening constituent of the traditional medicines Aster tongolensis and A. ageratoides native to China. This study involved the total 1 H and 13 C spectral assignment of the compound and its degraded product by means of two‐dimensional NMR techniques (COSY, NOESY, HETCOR and TOCSY) and resulted in unambiguous spectral assignments, particularly in the heavily convoluted regions of the spectra. Concerning the absolute stereochemistry, the sweetener was determined to be (4 R ,5 S ,8 S ,9 S ,10,13 R )‐16,17‐dihydroxykauran‐19‐oic acid β‐D‐glucopyranosyl ester through a combination of circular dichroism and 2D NMR techniques.

Abstract:Sucrose isomerase NX-5 from Erwinia rhapontici efficiently catalyzes the isomerization of sucrose to isomaltulose (main product) and trehalulose (by-product). To investigate the molecular mechanism controlling sucrose isomer formation, we determined the crystal structures of native NX-5 and its mutant complexes E295Q/sucrose and D241A/glucose at 1.70 angstrom, 1.70 angstrom and 2.00 angstrom, respectively. The overall structure and active site architecture of NX-5 resemble those of other reported sucrose isomerases. Strikingly, the substrate binding mode of NX-5 is also similar to that of trehalulose synthase from Pseudomonas mesoacidophila MX-45 (MutB). Detailed structural analysis revealed the catalytic RXDRX motif and the adjacent 10-residue loop of NX-5 and isomaltulose synthase PalI from Klebsiella sp. LX3 adopt a distinct orientation from those of trehalulose synthases. Mutations of the loop region of NX-5 resulted in significant changes of the product ratio between isomaltulose and trehalulose. The molecular dynamics simulation data supported the product specificity of NX-5 towards isomaltulose and the role of the loop330-339 in NX-5 catalysis. This work should prove useful for the engineering of sucrose isomerase for industrial carbohydrate biotransformations.
Identification of characteristic compounds of sweet orange oil and their sweetening effects on the sucrose solution with sweetness meter, sensory analysis, electronic tongue, and molecular dynamics simulation

Abstract:The characteristic aroma compounds of five-fold sweet orange oil were analyzed using gas chromatography-mass spectrometry combined with the odor aroma value (OAV) method. The results indicated that limonene, linalool, dodecanol, (E,E)-2,4-decadienal, (E)-citral, linalool, (E)-2-decenal, and geraniol are important contributors. The sweetening effects of key compounds on sucrose solutions were experimentally investigated. The results showed that the sweetness effects of five compounds (limonene, citronellal, geraniol, β-sinensal and β-caryophyllene) were better than those of (E)-citral, linalool and octanal. Molecular dynamics implied that the hydrogen bonding residues of the T1R2/T1R3-sucrose system were converted from LYS65, GLU302, ASP278, and SER144 to ASP278, SER144, ASP142, and ASP213 after the addition of limonene. Meanwhile, the hydrophobic interaction forces of the system are significantly enhanced. The total energy of the T1R2/T1R3-sucrose system decreased from -32.08 kcal/mol to -63.57 kcal/mol. The synergistic sweetening mechanism of characteristic aroma compounds of sweet orange oil on sucrose was revealed.
Progress in synthesis of food sweetener sucralose

Abstract:With the improvement of life quality,there has been an increasing demand on high quality and high safety food sweeteners. Sucralose is exactly such an ideal artificial sweetener,which has the features of non-nutritive,low-calorie,high sweetness,high safety,and good flavor. Therefore,it is extremely necessary to study the synthetic methods of sucralose. This paper mainly summarizes the recent progress in the synthetic methods of sucralose,and comprehensive evaluation is given. It is concluded that,while the omni-group protection method contains too many synthetic steps and the mono-group protection method of the acetic anhydride method and trimethyl orthoacetate method are still in development, the organotin method has fewer synthetic steps and the organotin can be recycled, therefore it is considered to be the most applicable in industrial production so far. On the other hand, chemical-enzyme synthetic method,especially the sucrose method,is believed to be the most promising one in the future due to its environment friendly and high selectivity aspect. In addition, there are some other synthetic methods,such as enzymatic hydrolysis of sucralose-6-ester,etc.,worthy of further research. Finally,the research trends are also discussed and several development suggestions are proposed.

Chemical Structure and Nmr of A New Saponin from Dipsacus Japonicus (Dipsacaceae)

Abstract:A new triterpenoid saponin containing five sugars was isolated from the ethanolic root extract of Dipsacus japonicus Miq., and its structure was established as 3-O-[beta-D-glucopyranosyl (1-->4)] [alpha-L-rhamnopyransoyl (1 --> 3)]-beta-D-glucopyranosyl (1-->3)-alpha-L-rhamnopyranosyl (1-->2)-alpha-L-arabinopyranosyl-oleanoic acid. The sites of glycosylation and the sequence of sugars in the glycoside can be determined unambiguously and total assignment of severely overlapping proton resonance of sugar residues were achieved by a combined use of the 1D-SEMDY and NOE difference spectroscopy in rotating frame techniques, without having recourse to chemical degration or modification. The results showed that these new NMR techniques are very effective and convenient for the structure determination and spectral assignment of this class of compounds.

Abstract:Hexaacetyl D-mannose hydrazine is one type of important intermediates in saccharide chemistry. In this paper, its single crystal was obtained and furthermore, X-ray diffraction and quantum chemistry calculation were performed. It belongs to orthorhombic system, space group P2(1)2(1)2(1), with a = 16.267(3), b = 19.263(3), c = 7.1948(12). angstrom, M-r. = 446.41, D-c = 1.315 g/cm(3), V = 2254.5(6) angstrom(3) and Z = 4. Meanwhile, the experimental results also provide information for designing a kind of molecular switch based on the mannose nitrogenous derivatives.

Abstract:One new monacolin analog, monacolin V (1), together with two new monacolin‐like natural products, 6‐hydroxyl monacolin P (2) and 3‐keto monacolin S (3), were isolated from the ethyl acetate portion of red yeast rice ethanol extract. Their structures were identified by HRESIMS and NMR experiments, and the complete assignments of 1H and 13C NMR data for three compounds were obtained by the aid of HSQC, HMBC, 1H‐1H COSY, and NOESY data. This is the first time that the NMR data of compounds 2 and 3 have been fully assigned.
