Abstract:As already known, complex prescription has been the outstanding feature of traditional Chinese medicine(TCM). This fact therefore brings complicated formula with multicomponents from multimaterals, which produces some difficulties to detect the components and to control the quality. Shuanghuanglian preparations(SHLs), including oral liquid, injection powder, capsule, and tablet, belong to a family of refined traditional Chinese patent medicine and a combined herbal remedy comprised of three herbs: Flos Lonicerae, Radix Scutellariae, and Fructus Forsythiae. They are often prescribed to treat acute upper respiratory tract infection caused by virus or bacteria. Apart from the classic methods, ultraviolet spectrophotography(UV), thin layer chromatography(TLC), and high performance liquid chromatography(HPLC), other new brand methods have been developed to determine the components mentioned above, such as microemulsion electrokinetic chromatography with ultraviolet detection, and capillary electrophoresis(CE) with electrochemical detection. All these, however, can only be applied to the simultaneous assay of the multicomponents in the same herb, other than to the complex prescription of TCM. For example, it is stipulated by Chinese Pharmacopoeia that three main active components, baicalin, chlorogenic acid, and phillyrin in SHLs(Chemical structures are shown in Scheme 1) should be detected with HPLC, respectively, by triplicate manipulations, which is a time-consuming and tedious procedure. Recently, ultra performance liquid chromatography(UPLC) as a novel separation device has presented more advantages in the rapid and simultaneous determination of multicomponents. In this study, we developed an UPLC method with time-of-flight -mass spectrometry (TOF-MS) as detector, for the simultaneous detection of the three components in SHLs, and it proved reliable, sensitive, specific, and convenient.
