Abstract:Background: Angiopoietin-like protein 1 (ANGPTL1) has been reported to suppress migration and invasion in lung and breast cancer, acting as a novel tumor suppressor candidate. Nevertheless, its effects on colorectal cancer (CRC) remain poorly defined. In this study, we aim to demonstrate the biological function of ANGPTL1 in CRC cells.Methods: We explored ANGPTL1 mRNA expression in human CRC tissues and its association with prognosis. CRC cell lines overexpressing ANGPTL1 or with ANGPTL1 knocked down were constructed and analyzed for changes in proliferation, colony formation, migration and invasion. ANGPTL1-regulated microRNAs were analyzed, and microRNA inhibitor and mimics were used to explore the role of microRNA in ANGPTL1-associated biological function.Results: ANGPTL1 mRNA expression was down-regulated in CRC tissues, and high ANGPTL1 expression predicted better survival in CRC patients. ANGPTL1 overexpression resulted in suppressed migration and invasion in vitro, and it prolonged overall survival in mouse models. By contrast, its down-regulation enhanced migration and invasion of CRC cells. MicroRNA-138 expression was positively correlated with ANGPTL1 mRNA level in CRC tissues and up-regulated by ANGPTL1 in CRC cells. In addition, the microRNA-138 inhibitor or mimics could reverse or promote the ANGPTL1-mediated inhibition of the migratory capacity of CRC cells, respectively.Conclusions: This study is the first to demonstrate the biological function of ANGPTL1 in CRC cells. ANGPTL1 expression was down-regulated in CRC tissues and inversely correlated with poor survival. ANGPTL1 repressed migration and invasion of CRC cells, and microRNA-138 was involved in this process.

Abstract:Angiopoietin-like proteins are a family of proteins that are closely related to lipid, glucose and energy metabolism, as well as angiogenesis. To date, eight Angptls have been discovered, namely Angptl1 to Angptl8 that play key roles in metabolic regulation and marker assisted selection. In this review, we summarized current progress on the structure, signaling pathways, upstream regulatory genes and metabolic network of Angptl1-8. Finally, in combination with our work, the status and problems of animal breeding as well as the future prospects for Angptls were discussed.

Abstract:Angiopoietin-like proteins (ANGPTLs), a class of secreted glycoproteins with a similar structure to angiopoietin, play a pivotal role in solid tumors invasion, metastasis, angiogenesis and apoptosis. As the prominent feature of the tumor microenvironment, hypoxia affects tumor progression and therapeutic effect through mediating multiple biological processes. Some ANGPTLs can act as downstream target genes of hypoxia-inducible factor 1 to regulate tumor physiological functions. Therefore, elucidating the effect and mechanism of ANGPTLs in tumor is of essential significance for improving the tumor cure rate and prognosis. In this article, a brief outline of the biological characteristics of the ANGPTLs family and their possible roles in tumorigenesis, development and hypoxic microenviroment is presented.

Abstract:Angiogenin, as a member of the ribonuclease superfamily, is an angiogenic protein. The angiogenic ability of angiogenin plays an important role in many physical and pathological processes. Angiogenin can induce endothelial cell migration, proliferation, tubulation, as well as inhibition of cellular apoptosis. Angiogenin can be used to modulate the angiogenetic process of tissue engineered constructions via local delivery. Furthermore, angiogenin can also be regarded as a biomarker for diagnostic evaluation of malignancy, or as a target for cancer therapy. This paper presents a comprehensive overview of the angiogenic mechanisms of angiogenin, as well as its potential application in the process of wound healing and treatment of ischemic diseases and malignancy.

Abstract:Long-term controversy regarding the role of angiopoietin-like protein 8 (ANGPTL8) in beta-cell proliferation and diabetes progression made it a research spotlight. Recently, the controversy was resolved. Although ANGPTL8 could not control beta-cell expansion and islet function, ANGPTL8 was still considered as a novel but atypical member in the ANGPTL family because of its unique structure and crucial effects on lipid metabolism. Besides, ANGPTL8 also participated in some other disorders such as non-alcoholic fatty liver disease and renal dysfunction. Understanding the features of ANGPTL8 may offer new diagnostic and therapeutic approaches to metabolic-related diseases. Therefore, we reviewed most recent findings about ANGPTL8 and aimed to provide an integrated picture of ANGPTL8.

Abstract:. The angiopoietins are an important family of growth factors specific for vascular endothelium. Most of them bind to the TIE2 receptor and are related to regulation of angiogenesis. During large-scale DNA sequencing of the human fetal brain cDNA library, we cloned a novel human angiopoietin-like cDNA and termed it human angiopoietin-like 5 ( ANGPTL5 ). Like other members of the angiopoietin family, ANGPTL5 -deduced protein also has an N-terminal cleavable signal peptide, a predicted coiled-coil domain, and a fibrinogen-like domain. The search against the human genome database indicated that ANGPTL5 maps to 11q22. Expression analysis of ANGPTL5 shows that it is mainly expressed in adult human heart.

Abstract:OBJECTIVE: To review the advance of the relationship between ANGPT-TIE signal system and the development of tumor.METHODS:Totally 287 English articles and 122 Chinese articles from 2000-01-2011-10 were retrieved with the Key words angiopoietin;tie;inflammation;metastasis by Pubmed,Science Direct,Springer Data and CNKI.Twenty-four articles were included in the reference with the selection criteria: 1)the biological characters and mechanism of action of Angiopoietin and Tie;2) The relationship between Angiopoietin,Tie and angiogenesis,inflammation,tumor metastasis and prognosis.RESULTS: ANGPT-TIE signal system is one of the most important regulators for vascularization.It is critically involved in maintaining both vascular homeostasis and vessel maturation as well as angiogenesis and inflammation.CONCLUSIONS:ANGPT-TIE signal system can play a critical role in tumor-induced angiogenesis,and the suppression of ANGPT-TIE signal system caused by using specific blocking agents could suppress the growth of tumor.The signal system is a attractive target for anti-angiogenic therapy.

Abstract:Lipin family including at least three members Lipin 1, Lipin 2, and Lipin 3 is a critical regulatory enzyme identified recently, which plays dual roles in lipid metabolisms. Lipin family has physiological effects not only on regulating lipid metabolism, but also on maintaining normal peripheral nervous functions, liver lipoprotein secretion, cell morphous, reproductive functions, and energy homeostasis. Since mutations in Lipin gene express may be associated with AIDS, insulin resistance, obesity, diabetes mellitus, and the other diseases of metabolic syndrome, Lipin may be a new useful target in treatment of above-mentioned clinical-related diseases. In this article, we focused on discovery, construction features, expression, regulatory mechanism, and biological functions of Lipin, as well as its correlation research with clinical-related diseases.

Abstract:As a member of the vertebrate-specific secreted ribonucleases, angiogenin (ANG) was first isolated and identified solely by its ability to induce new blood vessel formation, and now, it has been recognized to play important roles in various physiological and pathological processes through regulating cell proliferation, survival, migration, invasion, and/or differentiation. ANG exhibits very weak ribonucleolytic activity that is critical for its biological functions, and exerts its functions through activating different signaling transduction pathways in different target cells. A series of recent studies have indicated that ANG contributes to cellular nucleic acid metabolism. Here, we comprehensively review the results of studies regarding the structure, mechanism, and function of ANG over the past three decades. Moreover, current problems and future research directions of ANG are discussed. The understanding of the function and mechanism of ANG in a wide context will help to better delineate its roles in diseases, especially in cancer and neurodegenerative diseases.

Abstract:Background: Angiopoietin-like 4 (ANGPTL4) belongs to the family of angiopoietin- like proteins. The involvement of ANGPTL4 in various aspects of lipid metabolism and inflammation has become an important area of research. Methods: A thorough search on PubMed related to ANGPTL4, lipid metabolism, and inflammation was performed. Results: Over the past two decades, the recognition of ANGPTL4 as a potent regulator of lipid metabolism has substantially increased. As part of the senescence-associated secretory phenotype, ANGPTL4 also serves as an inflammatory mediator. Considering the advancements in ANGPTL4 research, we have highlighted that ANGPTL4 acts as a key node linking lipid metabolism and inflammation. ANGPTL4 impacts inflammation by regulating lipid metabolism. It affects critical enzymes (lipoprotein lipase, hepatic lipase, endothelial lipase, and acetyl-CoA carboxylase), regulatory factors (AMPK, cAMP, SLC7A11, GPX4, and mTOR), and receptors (LepR, CD36, and PPARγ) of lipid oxidation, synthesis, and peroxidation, thereby affecting immune cells and inflammatory pathways. Conclusion: Understanding the potential association and the therapeutic value of ANGPTL4 for regulating lipid metabolism and inflammation could contribute to drug discovery and therapeutic development.

Abstract:Angiopoietin family is a recently discovered type of cellular factors that specifically bind to the TIE-2 receptors located exclusively in endothelial cell membrane. The protein structures of this family members are similar. They can be structurally divided into three domains: an N-terminal region lacking homology to any known structures, an alpha-helical rich coiled-coil segment, and a fibrinogen-like domain. The distribution and biological activity of these factors are different in organism. Angiopoietin-1 as a agonist, mostly locates in close proximity with vascular endothelial cells, keeps the stability of blood vessels, enhances the affinity of vascular endothelial cells with surrounding cells and matrix, decreases the leakage of vessel. Ang-2 is a naturally occurring antagonist of Ang-1, exists in the angiogenic remodeling region and is related to the decrement of the stability of vessel. Ang-3 is widely distributed in multiple mouse tissues, while Ang-4 is expressed only in lung. Although Ang-3 and Ang-4 are structurally diverged from each other, they appear to represent the mouse and human counterparts of the same gene locus. Biological functions of Ang-3 and Ang-4 have not been elucidated yet. Angiopoietin family has potentially clinical applications for incurring illnesses which lead to vessel wound and vascular abnormal development.

Abstract:Abstract In the present commentary, we discuss new observations stating that angiopoietin-like protein 1 (ANGPTL1) attenuates cancer metastasis and stemness through Forkhead box O-3a (Foxo3a)–SRY-related HMG-box-2 (Sox2) axis in colorectal cancer (Clin. Sci. (2022) 136, 657–673, https://doi.org/10.1042/CS20220043). ANGPTL1 has been reported to play a critical role in cancer progression and metastasis. However, the underlying mechanisms remain controversial. Here, we integrate the possible mechanisms for ANGPTL1 inhibiting colorectal cancer liver metastasis and discuss the regulation of ANGPTL1 on the Foxo3a–Sox2 pathway. Although ANGPTL1 showed multifunctional potential, there is still a long way to go for ANGPTL1 to be an effective treatment strategy in the clinic.

Abstract:Angiopoietin-like protein 3 and-4 (ANGPTL-3 and-4) are generally related to lipid metabolism as well as angiogenesis in animals, however very less was known about their mRNA expression characterizations in tissue development. In this study, the mRNA expressions of ANGPTL-3 and ANGPTL-4 (ANGPTL-3 and-4) and tissue distribution in Peking duck (Anas platyrhynchos) of 1 to 8 weeks of age were analyzed using quantitative real-time PCR methods. It was observed that ANGPTL-3 and-4 mRNAs were broadly expressed in duck liver and adipose tissues and were most abundant in infra abdominal adipose tissue (AD). ANGPTL-3 and-4 had different expression patterns in tissues. These data suggested that both duck ANGPTL-3 and-4 could be related to the development of tissues, and ANGPTL-4 may contribute to the development of adipose tissue through promoting adipocyte differentiation.

Abstract:Since its discovery in 2000 for its role in lipid metabolism, ANGPTL4 has gained attention for its involvement in glucose metabolism, angiogenesis, wound healing, and tumor growth. This review highlights ANGPTL4's key functions in metabolic regulation and tumor progression, offering insights into its potential as a therapeutic target. Since ANGPTL4 was discovered to be involved in lipid metabolism in 2000 for the first time, Angptl4 has attracted the attention of researchers. With the further research, it was found that angptl4 was also involved in many biological activities (glucose metabolism, angiogenesis, wound healing, tumor growth, etc.) in vivo. In this review, we provide an overview of the fundamental role of ANGPTL4 in metabolic regulation and its impact on tumor growth. These insights may provide a way for exploring ANGPTL4 as a potential therapeutic target for future disease treatments.

Abstract:Angiogenin was originally isolated from cloned human carcinoma cells, which can effectively promote the proliferation, differentialion and migration of endothelial cells and can mediate cell adhesion and tubular morphogenesis. Angiogenin is a member of the pancreatic ribonuclease superfamily, but its ribonucleolytic activity is relatively weak. There are two modes of action when exogenous angiogenin binds to the endothelial cells. First, it can activate the classical signal transduction, and it can also be transported into the nucleus by its nuclear localization sequence. Angiogenin palys an important role under many physiological and pathological conditions, such as embryogenesis, endometrium alteration, wound healing, development of diabetes and cancer.

Abstract:Abstract Angiogenin (ANG) is first isolated and identified by its ability to stimulate the growth of blood vessels. As a member of the vertebrate-specific secreted ribonuclease (RNase) family, ANG is encoded by a single exon and is usually located in the middle of its family gene cluster. Particularly, ANG gene locus has a unique gene arrangement, featured by shared promoters and 5’-untranslated regions (5’-UTR) which direct two distinct exons enconding ANG and ribounclease 4 (RNASE4, the 4th member of RNase family) respectively. However, the gene structure and regulation of these genetic regions are largely unknown. Here, we have characterized the promoters, defined the transcription start site, and identified a unique mechanism of transcription regulation. We demonstrated that two Pol III elements within the promoter regulate ANG and RNASE4 expression in a position- and orientation-dependent manner. Moreover, an intragenic chromatin loop formed between the two CCCTC-binding factor (CTCF)-binding sites located in two introns flanking ANG coding exon, which preferentially enhances ANG transcription. These results suggest a multilayer transcriptional regulation of ANG and RNASE4 gene locus. The data also add more direct evidence to the notion that Pol III elements are able to directly influence Pol II gene transcription. Furthermore, our data indicate that a CTCF-dependent chromatin loop is able to differentially regulate transcription of genes that share the same promoters. On the other hand, the secreted ANG undergoes a receptor-mediated endocytosis from the cell surface to the nucleus and accumulates in the nucleoli. The nucleoli ANG can promote 47S pre-rRNA transcription by binding the ABE (Angiogenin Binding Element) and UCE (Upstream Core Element) region on the promoter of ribosomal DNA (rDNA), where ANG increases the number of actively transcribing rDNA and promotes the assembly of initiation complex by epigenetic activation through promoter methylation and histone modification. We have also shown that the surplus ANG in nucleus also related to mRNA transcription. ANG binds the first exon region of estrogen receptor-related receptor gamma (ERRγ) and inhibits the ERRγ expression. To screen and identify the mRNAs regulated by ANG at a genome-wide level, we carried out chromatin immunoprecipitation-chip assay (ChIP-on-chip) and found a total of 699 genes that may be regulated by ANG. These genes were significantly enriched to tumorigenesis, Wnt and TGF-beta pathways by the KEGG (Kyoto Encyclopedia of Genes and Genomes) analysis. Based on the findings that ANG is able to remodel the histone modification through direct binding to the histone protein, we propose that ANG might act as a chromatin remodeling activator to regulate RNA transcription. [Cloning of a novel gene, ANGPTL4 and the functional study in angiogenesis].

Abstract:OBJECTIVE:To clone a novel HCC-related gene and study its functions. METHODS:Using a combination of cell growth based functional screening of human placental cDNA library and RACE, a novel gene, angiopoietin-like 4 (ANGPTL4) was isolated. The gene was mapped by RH-PCR; also it was expressed with E. coli. Effects on angiogenesis was studied by MTT assay, in vitro wound migration assay, in vitro invasion assay, in vitro tube formation assay and in vivo mouse matrigel plug assay. RESULTS:ANGPTL4 was cloned and mapped in 19p13.3. ANGPTL4 had no significant effect on proliferation of PAEC (P = 0.0504), while it can significantly stimulate migration and invasion of PAEC with the dose of 2 microgram/ml (P < 0.05) and promote in vitro tube formation of PAEC and angiogenesis of mouse in vivo. CONCLUSIONS:ANGPTL4 can stimulate angiogenesis.

Abstract:BACKGROUND:ETS-1,the first subgroup in the ETS family transcription factor,plays a key role in angiogenesis.Recent studies demonstrate that ETS-1 also has the important regulative functions in apoptosis of vascular endothelial cells.When the blood vessel begins to form,ETS-1 decides the apoptosis of partial endothelial cells.OBJECTIVE:To summarize the function and the research progress of vascular growth factor ETS-1 in angiogenesis and apoptosis.RETRIEVAL STRATEGY:A computer-based search in Pubmed,Sciencedirect and Ovid database was underwent to identify the articles from January 1990 to September 2007,with the Key word "ETS-1,angiogenesis,apoptosis,osteoblast,osteoclast" in English.Simultaneously,Chinese Journal Full-text Database and Wanfang database were retrieved for the related articles from January 2000 to June 2007,with the key word "ETS-1,angiogenesis,apoptosis,endothelial cell" in Chinese.Altogether 128 articles were collected and carried on the first examination.Inclusive criteria:①The articles closely correlated with ETS-1 in angiogenesis and apoptosis.②The articles published recently or in the authoritative magazine were chose for the identical domain.Exclusive criteria:Duplicated researches.LITERATURE EVALUATION:The articles were mainly the experimental study of EST-1 in angiogenesis and apoptosis.Among 31 selected articles,2 were reviews while other 29 for clinical or basic experimental studies.DATA SYNTHESIS:①When orthodontic tooth moves,alveolar bone rebuilding is the dynamical equilibrium process between bone absorption and new bone formation.This process mechanism is complex and adjusted by many kinds of factors.②Among numerous regulatory factors,ETS-1 is the dominant factor and has important effect on angiogenesis.③The research indicates,four kinds of typical vascular growth factors(acid fibroblast growth factor,basic fibroblast growth factor,vascular endothelial growth factor and epidermal growth factor) can induce the ETS-1 expression,and ETS-1 promotes vessel production through inducing the expressions of the related factors with angiogenesis,such as matrix metalloproteinase and integrin β3.④Many progresses have been achieved in the regulatory effect of ETS-1 in angiogenesis,but it still has many problems,such as how the ETS-1 precisely regulate angiogenesis,how the ETS-1 precisely adjust apoptosis in gene expression and so on.CONCLUSION:As a kind of important regulatory factors,ETS-1 has important regulatory effects on angiogenesis and apoptosis.
