Abstract:Alginate (Alg) hydrogels have demonstrated great potential in drug delivery, wound dressings, and tissue engineering. However, the practical applications of conventional Alg hydrogels suffer from rapid degradation, insufficient controlled drug release, and poor adhesion to tissues. In this paper, the physicochemical properties of Alg hydrogels were comprehensively modulated through chemical modification and the incorporation of various polymers. The results showed that introducing neutral polymers stabilized Alg hydrogels at room temperature. However, this stabilizing effect did not persist at 37 °C. The cationic chitosan (CS) can bind to Alg through electrostatic interaction. The incorporation of CS substantially improved the stability of Alg hydrogels and remarkably prolonged the release of BSA from hydrogels to 14 days at 37 °C. Meanwhile, introducing dopamine (DA)-modified Alg enhanced the adhesion of hydrogel to porcine skin. The optimized hydrogels were utilized to encapsulate platelet-rich plasma (PRP). The PRP-loaded composite hydrogel (2 wt% Alg, with a Ca2+/carboxyl group molar ratio of 0.2, a DA/carboxyl group molar ratio of 0.2, and a CS content of 30 % of Alg) exhibited an excellent therapeutic effect in the healing of rabbit shoulder cuff injury. These findings provide valuable insights into the modulation of physicochemical properties of hydrogels for biomedical applications.

Abstract:Alginates are natural polysaccharides widely participating in food, pharmaceutical, and environmental applications due to their excellent gelling capacity. Their excellent biocompatibility and biodegradability further extend their application to biomedical fields. The low consistency in molecular weight and composition of algae-based alginates may limit their performance in advanced biomedical applications. It makes microbial alginate production more attractive due to its potential for customizing alginate molecules with stable characteristics. Production costs remain the primary factor limiting the commercialization of microbial alginates. However, carbon-rich wastes from sugar, dairy, and biodiesel industries may serve as potential substitutes for pure sugars for microbial alginate production to reduce substrate costs. Fermentation parameter control and genetic engineering strategies may further improve the production efficiency and customize the molecular composition of microbial alginates. To meet the specific needs of biomedical applications, alginates may need functionalization, such as functional group modifications and crosslinking treatments, to achieve enhanced mechanical properties and biochemical activities. The development of alginate-based composites incorporated with other polysaccharides, gelatin, and bioactive factors can integrate the advantages of each component to meet multiple requirements in wound healing, drug delivery, and tissue engineering applications. This review provided a comprehensive insight into the sustainable production of high-value microbial alginates. It also discussed recent advances in alginate modification strategies and alginate-based composites for representative biomedical applications.

Abstract:Abstract Alginate is a linear, water-soluble acidic seaweed polysaccharide composed of L-guluronic acid and its C5 epimer D-mannuronic acid, a general term for hydrophilic derivatives of alginic acid. It is widely found in various brown algae. Alginate lyase and its enzymatic hydrolysate have a variety of biological activities and become a new research hotspot. This paper reviews the source, classification, enzymatic mechanism and enzymatic properties of alginate lyase, and summarizes the function and application of alginate lyase and its enzymatic hydrolysate brown algal oligosaccharide, in order to provide a base for the development and application of alginate lyase and its products.

Abstract:The degradation of high molecular weight organic matter (HMWOM) is a core process of oceanic carbon cycle, which is determined by the activity of microbial communities harboring hundreds of different species. Illustrating the active microbes and their interactions during HMWOM processing can provide key information for revealing the relationship between community composition and its ecological functions. In this study, the genomic and transcriptional responses of microbial communities to the availability of alginate, an abundant HMWOM in coastal ecosystem, were elucidated. The main degraders transcribing alginate lyase (Aly) genes came from genera Alteromonas , Psychrosphaera and Colwellia . Meanwhile, some strains, mainly from the Rhodobacteraceae family, did not transcribe Aly gene but could utilize monosaccharides to grow. The co-culture experiment showed that the activity of Aly-producing strain could promote the growth of Aly-non-producing strain when alginate was the sole carbon source. Interestingly, this interaction did not reduce the alginate degradation rate, possibly due to the easily degradable nature of alginate. This study can improve our understanding of the relationship between microbial community activity and alginate metabolism function as well as further manipulation of microbial community structure for alginate processing.

Abstract:Alginate, the most abundant polysaccharides of brown algae, consists of various proportions of uronic acid epimers α-L-guluronic acid (G) and β-D-mannuronic acid (M). Alginate oligosaccharides (AOs), the degradation products of alginates, exhibit excellent bioactivities and a great potential for broad applications in pharmaceutical fields. Alginate lyases can degrade alginate to functional AOs with unsaturated bonds or monosaccharides, which can facilitate the biorefinery of brown algae. On account of the increasing applications of AOs and biorefinery of brown algae, there is a scientific need to explore the important aspects of alginate lyase, such as catalytic mechanism, structure, and property. This review covers fundamental aspects and recent developments in basic information, structural characteristics, the structure–substrate specificity or catalytic efficiency relationship, property, molecular modification, and applications. To meet the needs of biorefinery systems of a broad array of biochemical products, alginate lyases with special properties, such as salt-activated, wide pH adaptation range, and cold adaptation are outlined. Withal, various challenges in alginate lyase research are traced out, and future directions, specifically on the molecular biology part of alginate lyases, are delineated to further widen the horizon of these exceptional alginate lyases.

Abstract:Alginate is a brown seaweed-based linear polysaccharide of D – mannuronic acid and L – guluronic acid residues. Hydrolysis products of the polysaccharide, specifically oligosaccharides, have been receiving increasing interest, due to their significant bioactivity and potential utilisation routes. The bioactivity of alginate oligosaccharides is closely linked to structural characteristics, namely: molecular weight, degree of polymerisation, and ratio of the monomers (the M/G ratio). Hence, potential applications (such as utilisation as a biostimulant fertilizer) depend on these parameters. This review focuses on recent advances in producing alginate oligosaccharides using chemical or enzymatic methods. The literature survey includes utilisation of these methods at both laboratory and industrial scale. For the chemical methods, we assessed the standard laboratory scale procedures of alginate oligosaccharide production, the potential of scaling up to an industrial level, and the subsequent challenges. For the enzymatic route, we provide an overview of alginate lyases and the application perspectives of enzymatic hydrolysis of alginate.

Abstract:The cell wall of brown algae contains alginate as a major constituent. This anionic polymer is a composite of β-d-mannuronate (M) and α-l-guluronate (G). Alginate can be degraded into oligosaccharides; both the polymer and its products exhibit antioxidative, antimicrobial, and immunomodulatory activities and, hence, find many commercial applications. Alginate is attacked by various enzymes, collectively termed alginate lyases, that degrade glycosidic bonds through β-elimination. Considering the abundance of brown algae in marine ecosystems, alginate is an important source of nutrients for marine organisms, and therefore, alginate lyases play a significant role in marine carbon recycling. Various marine microorganisms, particularly those that thrive in association with brown algae, have been reported as producers of alginate lyases. Conceivably, the marine-derived alginate lyases demonstrate salt tolerance, and many are activated in the presence of salts and, therefore, find applications in the food industry. Therefore, this review summarizes the structural and biochemical features of marine bacterial alginate lyases along with their applications. This comprehensive information can aid in the expansion of future prospects of alginate lyases.

Abstract:Algin oligosaccharides have been applied in diverse industries and could be innovative synthesized by alginate-degrading bacteria. For enhance the alginate degradation efficiency to produce more algin oligosaccharides, a mutant strain ( Cobetia sp. cqz5-12-M1) was obtained through the complex mutagenesis using UV and the alkylating agent 1-methyl-3-nitro-1-nitrosoguanidine. The enzyme activity of the fermentation supernatant of mutant exhibited a significant 38.09% (53.98 ± 0.69 U/mL) increase, and its optimal growth conditions were determined as: 5 g/L sodium alginate, 5 g/L yeast powder, 30 g/L NaCl, 2 g/L K 2 HPO 4 , 2 g/L KH 2 PO 4 , 1 g/L MgSO 4 •7H 2 O, 0.01 g/L FeSO 4 •7H 2 O, pH 6.5, and 34 °C. Moreover, its optimal degradation conditions were identified as: 5 g/L sodium alginate, 5 g/L yeast powder, 30 g/L NaCl, 2 g/L K 2 HPO 4 , 2 g/L KH 2 PO 4 , 1 g/L MgSO 4 •7H 2 O, 0.01 g/L FeSO 4 •7H 2 O, pH 6.5, 31 °C and 72 h, yielding an enzyme activity of 120.98 ± 1.40 U/mL in the fermentation supernatant. Conclusive experiments on reagent tolerance revealed the growth of the mutant strain was significantly inhibited by 3% hydrogen peroxide, 5% carbolic acid, and 10 mg/mL gatifloxacin. Additionally, the alginate degradation capacity of mutant strain was highly significantly inhibited by 75% ethanol and all tested antibiotics.

Abstract:Natural polysaccharide-based hydrogels have drawn much concern in the biomedical fields. Among them, alginate, a natural polyanionic polysaccharide, has become one of the research hotspots, because of its abundant source, biodegradability, biocompatibility, solubility, modification flexibility, and other characteristics or physiological functions. Recently, through adopting various physical or chemical crosslinking strategies, selecting suitable crosslinking or modification reagents, precisely controlling the reaction conditions, or introducing organic or inorganic functional materials, a variety of alginate-based hydrogels with excellent performance have been continuously developed, considerably expanding the breadth and depth of their applications. Here, various crosslinking strategies in the preparation of alginate-based hydrogels are comprehensively introduced. The representative application progress of alginate-based hydrogels in drug carrier, wound dressing and tissue engineering is also summarized. Meanwhile, the application prospects, challenges and development trends of alginate-based hydrogels are discussed. It is expected to provide guidance and reference for the further development of alginate-based hydrogels.

Comparative study on the in vitro effects of Pseudomonas aeruginosa and seaweed alginates on human gut microbiota

Abstract:Alginates pertain to organic polysaccharides that have been extensively used in food- and medicine-related industries. The present study obtained alginates from an alginate overproducing Pseudomonas aeruginosa PAO1 mutant by screening transposon mutagenesis libraries. The interaction between bacterial and seaweed alginates and gut microbiota were further studied by using an in vitro batch fermentation system. Thin-layer chromatography (TLC) analysis indicated that both bacterial and seaweed alginates can be completely degraded by fecal bacteria isolated from study volunteers, indicating that a minor structural difference between bacterial and seaweed alginates (O-acetylation and lack of G-G blocks) didn't affect the digestion of alginates by human microbiota. Although, the digestion of bacterial and seaweed alginates was attributed to different Bacteroides xylanisolvens strains, they harbored similar alginate lyase genes. Genus Bacteroides with alginate-degrading capability were enriched in growth medium containing bacterial or seaweed alginates after in vitro fermentation. Short-chain fatty acid (SCFA) production in both bacterial and seaweed alginates was also comparable, but was significantly higher than the same medium using starch. In summary, the present study has isolated an alginate-overproducing P. aeruginosa mutant strain. Both seaweed and bacterial alginates were degraded by human gut microbiota, and their regulatory function on gut microbiota was similar.

Genome Analysis of a Potential Novel Vibrio Species Secreting pH- and Thermo-Stable Alginate Lyase and Its Application in Producing Alginate Oligosaccharides

Abstract:Alginate lyase is an attractive biocatalyst that can specifically degrade alginate to produce oligosaccharides, showing great potential for industrial and medicinal applications. Herein, an alginate-degrading strain HB236076 was isolated from Sargassum sp. in Qionghai, Hainan, China. The low 16S rRNA gene sequence identity (<98.4%), ANI value (<71.9%), and dDDH value (<23.9%) clearly indicated that the isolate represented a potential novel species of the genus Vibrio. The genome contained two chromosomes with lengths of 3,007,948 bp and 874,895 bp, respectively, totaling 3,882,843 bp with a G+C content of 46.5%. Among 3482 genes, 3332 protein-coding genes, 116 tRNA, and 34 rRNA sequences were predicted. Analysis of the amino acid sequences showed that the strain encoded 73 carbohydrate-active enzymes (CAZymes), predicting seven PL7 (Alg1-7) and two PL17 family (Alg8, 9) alginate lyases. The extracellular alginate lyase from strain HB236076 showed the maximum activity at 50 °C and pH 7.0, with over 90% activity measured in the range of 30-60 °C and pH 6.0-10.0, exhibiting a wide range of temperature and pH activities. The enzyme also remained at more than 90% of the original activity at a wide pH range (3.0-9.0) and temperature below 50 °C for more than 2 h, demonstrating significant thermal and pH stabilities. Fe2+ had a good promoting effect on the alginate lyase activity at 10 mM, increasing by 3.5 times. Thin layer chromatography (TLC) and electrospray ionization mass spectrometry (ESI-MS) analyses suggested that alginate lyase in fermentation broth could catalyze sodium alginate to produce disaccharides and trisaccharides, which showed antimicrobial activity against Shigella dysenteriae, Aeromonas hydrophila, Staphylococcus aureus, Streptococcus agalactiae, and Escherichia coli. This research provided extended insights into the production mechanism of alginate lyase from Vibrio sp. HB236076, which was beneficial for further application in the preparation of pH-stable and thermo-stable alginate lyase and alginate oligosaccharides.

Abstract:This review article presents past and current alginate-based materials in each application, showing the widest range of alginate's usage and development in the past and in recent years. The first segment emphasizes the unique characteristics of alginates and their origin. The second segment sets alginates according to their application based on their features and limitations. Alginate is a polysaccharide and generally occurs as water-soluble sodium alginate. It constitutes hydrophilic and anionic polysaccharides originally extracted from natural brown algae and bacteria. Due to its promising properties, such as gelling, moisture retention, and film-forming, it can be used in environmental protection, cosmetics, medicine, tissue engineering, and the food industry. The comparison of publications with alginate-based products in the field of environmental protection, medicine, food, and cosmetics in scientific articles showed that the greatest number was assigned to the environmental field (30,767) and medicine (24,279), whereas fewer publications were available in cosmetic (5692) and food industries (24,334). Data are provided from the Google Scholar database (including abstract, title, and keywords), accessed in May 2023. In this review, various materials based on alginate are described, showing detailed information on modified composites and their possible usage. Alginate's application in water remediation and its significant value are highlighted. In this study, existing knowledge is compared, and this paper concludes with its future prospects.

Abstract:Alginate is a natural polysaccharide present in various marine brown seaweeds. Alginate oligosaccharide (AOS) is a degradation product of alginate, which has received increasing attention due to its low molecular weight and promising biological activity. The wide-ranging biological activity of AOS is closely related to the diversity of their structures. AOS with a specific structure and distinct applications can be obtained by different methods of alginate degradation. This review focuses on recent advances in the biological activity of alginate and its derivatives, including their anti-tumor, anti-oxidative, immunoregulatory, anti-inflammatory, neuroprotective, antibacterial, hypolipidemic, antihypertensive, and hypoglycemic properties, as well as the ability to suppress obesity and promote cell proliferation and regulate plant growth. We hope that this review will provide theoretical basis and inspiration for the high-value research developments and utilization of AOS-related products.

Abstract:ABSTRACT Alginates are salts of alginic acid derived mainly from sea algae of the genus brown algae. They are also synthesized by some bacteria. They belong to negatively charged polysaccharides exhibiting some rheological properties. High plasticity and the ability to modify the structure are the reasons for their application in numerous industries. Moreover, when in contact with the living tissue, they do not trigger an immune response, and for this reason they are the most often tested materials for medical applications. The paper discusses the latest applications, including 3D bioprinting, drug delivery systems, and sorptive properties. Recognizing alginates as biomaterials, it emphasizes the necessity for precise processing and modification to industrialize them for specific uses. This review aims to provide a thorough understanding of the advancements in alginate research, underscoring their potential for innovative applications.

Abstract:Antibacterial finishing of textiles has been introduced as a necessary process for various purposes especially creating a fabric with antimicrobial activities. Currently, the textile industry continues to look for textiles antimicrobial finishing process based on sustainable biopolymers from the viewpoints of environmental friendliness, industrialization, and economic concerns. This paper reviews the role of alginate, a sustainable biopolymer, in the development of antimicrobial textiles, including both basic physicochemical properties of alginate such as preparation, chemical structure, molecular weight, solubility, viscosity, and sol-gel transformation property. Then different processing routes (e.g. nanocomposite coating, ionic cross-linking coating, and Layer-by-Layer coating) for the antibacterial finishing of textiles by using alginate are revised in some detail. The achievements in this area have increased our knowledge of alginate application in the field of textile industry and promoted the development of green textile finishing.

Abstract:Alginate oligosaccharides (AOS) made from the degradation of alginate, to some extent, makes up for the poor solubility and bioavailability of alginate as a macromolecular substance and possess several beneficial biological activities that are absent in alginate. These properties include prebiotic, glycolipid regulatory, immunomodulatory, antimicrobial, antioxidant, anti-tumor, promoting plant growth and other activities. Consequently, AOS has significant potential for use in the agricultural, biomedical, and food industries, and has been the focus of research in the field of marine biological resources. This review comprehensively covers methods (physical, chemical, and enzymatic methods) for the production of AOS from alginate. More importantly, this paper reviews recent advances in the biological activity and potentially industrial and therapeutic applications of AOS, providing a reference for future research and applications of AOS.

Abstract:Sodium alginate(AlgNa) was purified via the acetone precipitation,and the product was tested by means of FT-IR and GPC.Then,the contants of residual protein in both the unpurified and the purified samples were measured,and several three-dimension porous calcium alginate scaffolds were fabricated through such three crosslinking systems as CaCl2-AlgNa,CaSO4/GDL-AlgNa and Ca-EDTA/GDL-AlgNa.The results indicate that the porous Ca-EDTA/GDL-AlgNa scaffold with a sodium alginate content of 2.5% ～ 3.0% possesses uniform micropore distribution,large specific surface area and excellent mechanical behavior,which means that it is suitable for the cell culture in tissue engineering and cell engineering.

Abstract:OBJECTIVE To review the current situation of alginate-based biomedical materials, especially focus on the clinical strategies and research progress in the clinical applications and point out several key issues that should be concerned about. METHODS Based on extensive investigation of domestic and foreign alginate-based biomedical materials research and related patent, literature, and medicine producted, the paper presented the comprehensive analysis of its research and development, application status, and then put forward several new research directions which should be focused on. RESULTS Alginate-based biomedical materials have been widely used in clinical field with a number of patients, but mainly in the fields of wound dressings and dental impression. Heart failure treatment, embolization, tissue engineering, and stem cells culture are expected to become new directions of research and products development. CONCLUSION Development of alginate-based new products has good clinical feasibility and necessity, but a lot of applied basic researches should be carried out in the further investigations.
